Influence of Alcohol Consumption on Oxidative Stress and Antioxidant Status in Cancer Patients - Case-control Study from Western Nepal

  • Aim: The present study assess the effect of consumption of alcohol on oxidative stress and antioxidant status in patients suffering from different types of cancer. Methods: This hospital based case control study conducted in the Western part of Nepal covered a total of 93 cancer patients with or without alcohol intake and smoking habits, along with 94 age, sex and habit-matched individuals serving as controls. Plasma thiobarbituric acid reacting substances (TBARS), total antioxidant activity (TAA), vitamin C, ${\alpha}$-tocopherol and erythrocyte reduced glutathione (GSH) were estimated and compared. Results: The TBARS level was found to be significantly higher ($p{\leq}0.001$) in all types of cancer patients when compared to controls, being aggravated in alcoholics with a smoking habit. No statistical significance ($p{\geq}0.05$) was observed in the level of vitamin C and ${\alpha}$-tocopherol. GSH and TAA level were significantly decreased ($p{\leq}0.001$) in all the groups except those who consumed both branded as well as homemade alcohol and non-alcoholics without smoking habit. Conclusion: Alcohol, irrespective of its commercial brand, increases oxidative stress in all types of cancer patients. This is even higher when alcohol intake is combined with a smoking habit. Decreased TAA and GSH are major risk factors for cancer development.

Reproductive Performance of Dairy Buffaloes Supplemented with Varying Levels of Vitamin E

  • The effect of vitamin E supplementation on plasma ${\alpha}$-tocopherol level, total antioxidant level and reproductive performance in Murrah buffaloes was studied during periparturient period. Twenty-four advance pregnant buffaloes were randomly divided into four equal groups as $T_1$, $T_2$, $T_3$ and $T_4$ and were supplemented with 0, 1,000, 1,500 and 2,000 IU of ${\alpha}$-tocopheryl acetate (Merck) from 60 days prepartum to 30 days postpartum and 0, 500, 750 and 1,000 IU from 30 to 60 days postpartum, respectively. Blood samples were collected at -60, -45, -30, -15, -7, 0, 7, 15, 30 and 60 days of parturition and were analyzed for plasma ${\alpha}$-tocopherol and total antioxidant activity (TAA). The intake of DM, CP and TDN did not vary among different groups. Plasma ${\alpha}$-tocopherol and TAA around parturition (-7 to 15 day) in $T_3$ and $T_4$ were significantly higher than the control group. There was 17% reduction in retention of fetal membranes (RFM) and metritis in $T_4$ than control. The post partum estrus interval averaged 58.00, 55.33, 51.83 and 43.00 days in $T_1$, $T_2$, $T_3$ and $T_4$ respectively. There was significant reduction in days open in both $T_3$ and $T_4$ in comparison to $T_1$ group (127,130 Vs.146). All the vitamin E supplemented groups showed reduction in days open than their previous lactation performance. Supplementation of vitamin E at $1,500IU\;d^{-1}$ from 60 day prepartum to 30 day post partum to buffaloes exhibited beneficial effect on plasma ${\alpha}$-tocopherol level and TAA around parturition and continuation of its supplementation at $1,000IU\;d^{-1}$ from 30 to 60 days of lactation improved post partum reproductive performance of buffaloes.

Freshness Preserving of Table Grape using Corrugated Paperboard Box Laminated with Functional MA Film (기능성 골판지 상자로 포장한 포도의 신선도 유지효과)

  • Weight loss of table Grape packed with control(Corrugated paperboard box), LDPE, CE(MA film masterbatched by ceramic powder treated cemical reagent) and FC box(laminated by CE film pouch) at 25 C after 10 days were 5.6%, 0.4%, 0.6%, and 0.7%, respectively. Weight loss of control was 7.29times higher than that of FC box. Total ascorbic acid content(TAA) of table grape after 10 days was 3.42 mg% for control, was 5.33 for LDPE and 5.14mg% for CE, 4.98mg% for FC. TAA of LDPE and CE showed to higher than that of control, especially TAA of FC was 33% higher than that of control. Titratable acid of LDPE and CE was high compare to control, and acidity of FC was 6% higher than that of control. References in overall appearance of LDPE, CE and FC were better than that of control. Corrugated paperboard box(FC) laminated with functional MA film showed to be able to used as packaging box of table grape.

Involvement of Pyridoxine/Pyridoxamine 5′- Phosphate Oxidase (PDX3) in Ethylene-Induced Auxin Biosynthesis in the Arabidopsis Root

  • As sessile organisms, plants have evolved to adjust their growth and development to environmental changes. It has been well documented that the crosstalk between different plant hormones plays important roles in the coordination of growth and development of the plant. Here, we describe a novel recessive mutant, mildly insensitive to ethylene (mine), which displayed insensitivity to the ethylene precursor, ACC (1-aminocyclopropane-1-carboxylic acid), in the root under the dark-grown conditions. By contrast, mine roots exhibited a normal growth response to exogenous IAA (indole-3-acetic acid). Thus, it appears that the growth responses of mine to ACC and IAA resemble those of weak ethylene insensitive (wei) mutants. To understand the molecular events underlying the crosstalk between ethylene and auxin in the root, we identified the MINE locus and found that the MINE gene encodes the pyridoxine 5′-phosphate (PNP)/pyridoxamine 5′-phosphate (PMP) oxidase, PDX3. Our results revealed that MINE/PDX3 likely plays a role in the conversion of the auxin precursor tryptophan to indole-3-pyruvic acid in the auxin biosynthesis pathway, in which TAA1 (TRYPTOPHAN AMINOTRANSFERASE OF ARABIDOPSIS 1) and its related genes (TRYPTOPHAN AMINOTRANSFERASE RELATED 1 and 2; TAR1 and TAR2) are involved. Considering that TAA1 and TARs belong to a subgroup of PLP (pyridoxal-5′-phosphate)-dependent enzymes, we propose that PLP produced by MINE/PDX3 acts as a cofactor in TAA1/TAR-dependent auxin biosynthesis induced by ethylene, which in turn influences the crosstalk between ethylene and auxin in the Arabidopsis root.

Hepatoprotective Effect of Uncaria rhynchophylla on Thioacetamide-Induced Liver Fibrosis Model

  • Liver fibrosis is a wound-healing response to chronic liver injury, which is caused by the continuous and excess deposition of extracellular matrix (ECM). The aim of this study is to investigate whether Uncaria rhynchophylla water extract (UR) can ameliorate thioacetamide (TAA)-induced liver fibrosis. The liver fibrosis model was induced on C57BL/6 mice by intraperitoneal injection with TAA three times a week for 8 weeks. UR (200 mg/kg) or silymarin (50 mg/kg) was administered orally daily for 8 weeks. Biochemical analyses including AST, ALT, MPO, and Ammonia levels were measured in serum. In the mice liver tissues, western blot and histological staining were analyzed. As a result, UR dramatically reduced the levels in serum AST, ALT, MPO, and Ammonia levels. UR treatment regulated NADPH oxidase factors expression, and antioxidant enzymes except for GPx-1/2 were significantly increased via Nrf2 activation. Furthermore, pro-inflammatory mediators, such as COX-2 and iNOS were markedly suppressed through the inhibition of NF-κB activation. Expressions of ECM-related protein including α-SMA and Collagen I were noticeably decreased. The additional histological evaluation confirmed that hepatocyte damage and collagenous fiber accumulation were attenuated. Taken together, these data suggest that UR possessed hepatoprotective effects in TAA-induced liver fibrosis via the NF-κB inactivation and Nrf2 activation. Therefore, UR may act as a potential therapeutic drug against liver fibrosis.

Protective Effect of Spatholobi Caulis in Thioacetamide induced Acute Liver Injury of Rat (Thioacetamide로 유발한 간손상 모델에서 계혈등(鷄血藤)의 간보호 효과)

  • Objectives : This study was undertaken to investigate the hepatoprotective effect of Spatholobi Caulis water extract (SC) to thioacetamide (TAA)-induced acute liver injury (ALI) in rats. Methods : The rats were injected intraperitoneally with TAA (200 mg/kg body weight) and orally administered SC (100 or 200 mg/kg b.w.) daily for 3 days. Liver biomarkers were assessed by serum glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and ammonia levels. Malondialdehyde (MDA) was measured both serum and liver tissue. In addition, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase, anti-oxidant, and inflammation-related proteins were investigated by western blot analysis. Histological examination further confirmed though hematoxylin and eosin stain. Results : The SC treatment reduced liver function markers like GOT and GPT and also remarkably decreased ammonia level. Moreover, the elevated MDA level in TAA-induced group was significantly reduced by SC treatment. NADPH oxidase expression associated with oxidative stress including NOX2, NOX4, and p47phox markedly inhibited by SC administration. SC treatment exerted anti-oxidant effect through the increase of anti-oxidant enzyme including superoxide dismutase (SOD), Catalase, and heme oxygenase-1 (HO-1). The protein expressions of inflammatory cytokines such as tumor necrosis factor-�� (TNF-��), IL-6, and IL-1�� induced by nuclear factor-kappa B (NF-��B) activation were modulated through blocking the phosphorylation of inhibitor of nuclear factor ��B�� (I��B)��. SC treatment also improved histological alterations. Conclusion : These findings suggested that SC administration may be a potential candidate for the prevention or treatment of ALI.

Protective effect of Saenggangeonbi-tang on liver fibrosis induced by thioacetamide (Thioacetamide로 유도된 간섬유화 모델에서 생간건비탕(生肝健脾湯)의 보호 효과)

  • Objective : In modern society, liver diseases such as liver fibrosis are on the rise as inflammation and wound healing processes of the liver are repeated due to factors such as drinking, smoking, and stress. This study was conducted to evaluate the effect of Saenggangeonbi-tang (SGGBT) on thioacetamide (TAA)-induced liver fibrosis. Methods : The mice were divided into 4 groups for examination (n=6): Normal group (Nor), distilled water-treated liver fibrosis mice (Con), silymarin 50 mg/kg-treated liver fibrosis mice (Sily), SGGBT 200 mg/kg-treated liver fibrosis mice (S200). Liver fibrosis was established in the mice via TAA for 8 weeks (1 week 100 mg/kg, 2,3 weeks 200 mg/kg, 4-8 weeks 400 mg/kg, three times a week, intraperitoneal injection) and they were administered silymarin and SGGBT (every day, oral administration) with the TAA. Results : SGGBT significantly decreased the levels of aspartate aminotransferase, alanine aminotransferanse, ammonia, and myeloperoxidase in serum increased by liver fibrosis. As a result of confirming H&E and MT staining, it was confirmed that SGGBT reduced damage and inflammatory cell infiltration in liver tissue, and alleviated changes in collagen fiber deposition and histological fibrosis. Also, it was confirmed through PAS staining that it reduced glycogen deposition in liver tissue. In addition, SGGBT significantly decreased the NADPH oxidases as well as significantly modulated the expression of MMP-2 and TIMP-2. Conclusions : These results suggest that SGGBT regulates the expression of MMP/TIMP protein through inhibition of oxidative stress and alleviates liver fibrosis by reducing collagen and glycogen deposition in liver tissue.

Research for Selecting The Optimized Vitamin C Analysis Method (최적 비타민 C 분석법 선정에 관한 연구)

  • 2,4-Dinitrophenylhydrazine (DHP), indophenol, and HPLC methods, which are generally used to analyze vitamin C, were evaluated by comparing changes in vitamin C contents in various samples kept at 10 and $20^{\circ}C$. Amount of total ascorbic acid (TAA) in yoghurt A kept at $10^{\circ}C$, as measured by DNP method, decreased from 26.7mg/100mL for early period to 22.8mg/100mL for final period of storage, whereas, on 3rd and 10th storage days, increased contrary to general results. Using indophenol, TAA measured at 28.6 and 9.5mg/100mL, and 30.5 and 14.6mg/100mL using HPLC far early and final periods, respectively. With indophenol and HPLC, TAA amount showed steady tendency to decrease. In conclusion, HPLC is the most suitable method to analyze vitamin C content, and indophenol method can be also used during early storage stage.

Quantitative Comparison of Ginsenosides and Nitrogen Compounds in Korean Ginsengs and Related Origin (고려인삼류와 유사생약중 ginsenoside 및 질소화합물의 함량 비교)

  • To differentiate the quality of Korean ginseng from those of other habitats, the quantitative analysis of free amino acids(FAA) and total amino acids(TAA) in addition to ginsenoside Rb1 and Rg1 was carried out using amino acid analyzer and HPLC, respectively. FAA pattern in Korean ginseng was much different from that of Panax notoginseng. The difference in total content of FAA was also found that Korean ginseng contained 26.3-39.8mg/g while Panax notoginseng contained 6.5mg/g. This FAA content had a tendency to increase with the age of radix. The contents of FAA and TAA in Korean ginseng(6 years old) from Kumsan were shown to be the highest than other ginseng origins tested. The content in the 6 years Panax ginseng from China was about same with that of 4 years Korean ginseng of Kumsan. However, regarding to gisenoside Rbl and Rg1, which have been accepted as the characteristic components of Panax ginseng-Panax notoginseng showed considerably higher content than those of any other ginseng origin.
