Nucleotide Insertion Fidelity of Human Hepatitis B Viral Polymerase

  • The hepadnaviruses replicate their nucleic acid through a reverse transcription step. The MBP-fused HBV polymerase was expressed in E. coli and purified by using amylase affinity column chromatography. The purified protein represented DNA-dependent DNA polymerase activity. In this report, the MBP-HBV polymerase was shown to lack 3'$\rightarrow$5' exonuclease activity, like other retroviral RTs. The ratio of the insertion efficiency for the wrong versus right vase pairs indicates the misinsertion frequency (f). The nucleotide insertion fidelity (1/f), observed with the MBP-HBV polymerase and HIV-1 RT, was between 60 and 54,000, and between 50 and 73,000, respectively, showing that they are in close range. A relatively efficient nucleotide incorporation by the MBP-HBV polymerase was observed with a specificity of three groups: (1) A : T, T : A>C : G, G : C (matched pairs), (2) A : C, C : A>G: T, T : G (purine-pyrimidine and pyrimidine-purine mispairs), and (3) C : C, A : A, G : G, T : T>T : C, C : T>A : G, G : A (purine-purine or pyrimidine-pyrimidine mispairs), and their order is (1)>(2)>(3). The data from the nucleotide insertion fidelity by the MBP-HBV polymerase suggest that the HBV polymerase may be as error-prone as HIV-1 RT.

Studies on the Biological Attributes of Scelionid Egg Parasitoid Psix striaticeps (Dodd) for the Control of Stink Bug Canthecona furcellata (Wolff) in Sericulture

  • Scelionid eggParasitoids Play an important role in biological control of some economically important pentatomids. Stink bug (Canthecona furcellatta Wolff,) is an important predator of Tasar silkworm larvae (Antheraea mylitta D.) and causes 30-40 percent loss in tasar silk industry. Psix striaticeps (Scelionid: Hymenoptera) has been found to be an important egg parasitoid. The parasitization potential of f striaticeps is mere than 60 percent and the sex ratio is female oriented. Studies indicate that the maintenance of existing scelionid population by avoiding harmful practices and the augmentation of scelionids either directly releasing them in the field or by indirectly making the field environment more favorable for them is an important aspect to control the pentatomid population in sericulture. Female bias sex ratio is advantageous to increase the parasitoid population in the field. 24-hour-old egg of stink bug and one-day old parasitoid is suitable for producing maximum progeny. 1 : 30 of parasitoid: host ratio is needed to regulate the estimated population in silkworm rearing field.

  • 신경 교세포에서 발생하는 가장 흔한 뇌 악성종양인 교모세포종은 조기 진단 및 치료계획 수립이 중요하다. 주로 조영제를 통해 촬영된 T1CE 영상을 통해 암의 진단이 이뤄지는데, 최근 가돌리늄 기반 조영제 주입의 위험이 보고되고 있다. 의료영상에서 새로운 영상을 합성하는 GAN 모델과 영역분할에 대한 심층신경망 모델에 대한 연구가 활발히 진행되고 있다. 본 연구에서는 교모세포종 환자의 T1CE 영상의 생성과 암의 영역분할을 동시에 학습하는 하나의 모델을 제안한다. 제안된 모델의 성능은 평균 제곱오차, 최대신호대잡음비 등의 유사성 측정을 통해 평가되어 0.002, 55dB의 평균 결과 값을 보여준다.

  • 연약 점성지반 위에 상대적으로 낮은 하중의 구조물이 얹어질 때, 상부에 모래층을 두어 지지력을 증진시킬 수 있다. 이와같은 점토 위 모래층이 놓이는 조건에 대한 지지력 검증은 현장에서 평판재하시험 등을 통해 지지력 검증이 어렵기 때문에 설계단계에서부터 정확한 지지력 예측이 필요하다. 따라서, 본 연구는 점토 위 모래층이 놓이는 지반의 지지력 거동을 파악하기 위해 2차원 실내토조실험과 FEM 해석을 수행하였다. 주요 인자로 깊이비 H/B와 지지력비 $q_c/q_s$를 선정하여, 모래층 높이, 점토의 비배수전단강도 그리고 재하 폭을 변수로 하였다. 그 결과, 실내토조실험은 FEM 해석과 적합성이 상당히 높게 나타났다. 이론식과의 비교에 있어서 보다 최신 연구인 Okamura et al.(1998)의 메커니즘보다 관입전단 메커니즘과 그 유사성이 크게 나타났으며, 하중확산 메커니즘의 적용성을 위하여 등가하중확산각을 제시하였다. 또한, $q_c/q_s$에 대하여 무차원 단위의 한계깊이비 $H_f$의 선형 회귀식을 제안하였다.

반경비 및 각속도의 변화에 따른 Taylor 유동에 관한 연구 (A STUDY ON TAYLOR FLOW ACCORDING TO RADIUS RATION AND ANGULAR VELOCITY)

  • This paper represents the numerical study on Taylor flow according to the radius ratio and the angular velocity for flow between tow cylinder. The numerical model is consisted of two cylinder which inner cylinder is rotating and outer cylinder is fix, and the axial direction is used the cyclic condition because of the length for axial direction is assumed infinite. The diameter of inner cylinder is assumed 86.8 mm, the numerical parameters are angular velocity and radius ratio. The numerical method is compared with the experimental results by Wereley, and the results are very good agreement. The critical Taylor number is calculated by theoretical and numerical analysis, and the results is showed the difference about ${\pm}10\;%$. As $Re/Re_c$ is increased, Taylor vortex is changed to wavy vortex, and then the wave number for azimuthal direction is increased. Azimuthal wave according to the radius ratio is showed high amplitude and low frequence in case of small radius ratio, and is showed low amplitude and high frequence in case of large radius ratio.

Association between Interferon-Inducible Protein 6 (IFI6) Polymorphisms and Hepatitis B Virus Clearance

  • CD8+T cells are key factors mediating hepatitis B virus (HBV) clearance. However, these cells are killed through HBV-induced apoptosis during the antigen-presenting period in HBV-induced chronic liver disease (CLD) patients. Interferon-inducible protein 6 (IFI6) delays type I interferon-induced apoptosis in cells. We hypothesized that single nucleotide polymorphisms (SNPs) in the IFI6 could affect the chronicity of CLD. The present study included a discovery stage, in which 195 CLD patients, including chronic hepatitis B (HEP) and cirrhosis patients and 107 spontaneous recovery (SR) controls, were analyzed. The genotype distributions of rs2808426 (C > T) and rs10902662 (C > T) were significantly different between the SR and HEP groups (odds ratio [OR], 6.60; 95% confidence interval [CI], 1.64 to 26.52, p = 0.008 for both SNPs) and between the SR and CLD groups (OR, 4.38; 95% CI, 1.25 to 15.26; p = 0.021 and OR, 4.12; 95% CI, 1.18 to 14.44; p = 0.027, respectively). The distribution of diplotypes that contained these SNPs was significantly different between the SR and HEP groups (OR, 6.58; 95% CI, 1.63 to 25.59; p = 0.008 and OR, 0.15; 95% CI, 0.04 to 0.61; p = 0.008, respectively) and between the SR and CLD groups (OR, 4.38; 95% CI, 1.25 to 15.26; p = 0.021 and OR, 4.12; 95% CI, 1.18 to 14.44; p = 0.027, respectively). We were unable to replicate the association shown by secondary enrolled samples. A large-scale validation study should be performed to confirm the association between IFI6 and HBV clearance.

$MgB_2$ 초전도 박막의 홀 전도도의 온도 의존성 (Temperature dependence of the Hall conductivity in $MgB_2$ superconducting thin films)

  • We have measured the Hall conductivity (${\sigma}_{xy}$) of c-axis-oriented $MgB_2$ thin films as functions of temperatures and magnetic fields. We found that the Hall conductivity (${\sigma}_{xy}$) is expressed by the sum of two terms, ${\sigma}_{xy}=C_l/H+C_3H$, where the coefficient $C_1\;and\;C_3$ are independent of the magnetic fields and have positive values. The coefficient $C_1$ is strongly dependent on the temperature, while the $C_3$ is weakly dependent on the temperature. We have obtained that the $C_1$ is proportional to $(1- T/T_c)^n$ with n = 4.2, which is consistent with the data observed in $La_{2-x}Sr_xCuO_4$ superconductors with low anisotropy ratio.

육성비육돈에 대한 생균제의 첨가급여가 분 악취 및 파리유충 발생에 미치는 영향 (Effects of Feeding Probiotics on the Odor-Generating Substances and Development of maggots(Fly's larvae) in Pig's Feces)

  • This research was conducted to investigate the effects on the amount of nitrogen and phosphorus and the number of maggots in the feces from the experimental pigs fed probiotics additives, and the variance of NH3-concentration(ppm) generated in the experimental pig-pens. The experiment was performed twice in the W-farm(under the condition of sufficient energy and protein in Spring) and B-farm(insufficient energy and protein in Summer). The experiment was designed with 5 treatments (control and 4 treatments according to the kinds of additives used). In the W-farm experiment, 75 three-way crossbred pigs weighing average 38kg were randomized with 3 replications for 102 days. In the B-farm experiment, 105 three-way crossbred pigs weighing average 33kg were randomized with 3 replications for 130 days. The results obtained are as follows: 1. The amount of dry matter, organic matter, nitrogen and phosphorus in the feces was not significantly different among the treatments. But the amount of OM, N and P shows a tendency to be somewhat lowered in the treatment A. On the otherhand, the amount of dry matter was somewhat lowered in the treatment B. The amount of such matters in the W-farm was somewhat lowered than those in B-farm. But there was not significant difference between W-farm and B-farm. 2. On the variance of the number of maggots developed in the pig's feces according to each treatment, the number of maggots in the treatments added probiotics were markedly decreased in comparison with those of the control(T), and it was significantly different(p<0.01). Especially the decrease in the treatment A and B was much remarkable. 3. The addition of probiotics to the pigs' feeds made the NH3-concentration generated in the pig-pens decrease markedly. especially the NH3-concentration in the treatment A had the highest decreasing ratio among all the treatments. The decreasing ratio of NH3-concentration of the treatments added probiotics as compared to control(T) was 59.1∼80.0%(average 70.3%) in the treatment A, 56.8∼80.0%(ave. 68.5%) in the treatment B and 24.4∼46.0%(ave. 35.0%) in the treatment C and D. On the variance of the NH3-concentration according to the experimental times)seasonal conditions), the NH3-concentration generated in the pig-pens was gradually increased from April to August, on the other hand it was decreased in the September. By the result of this study, it was appeared that the probiotics added to the pigs' feeds made the NH3-concentration decrease in the pig-pens to the extent of safety degree(25 ppm and less) permitted to swine especially in the treatment A and B.

래버린스 위어를 이용한 침사지 내 침전효율 개선 (Improvement of Sedimentation Rate in the Settling Basin by Labyrinth Weir)

  • 본 연구에서는 기존의 침사지 내부 수공구조물(격벽)로 사용되고 있는 둔턱을 대체하여 일반형 사다리꼴 래버린스 위어 (generic labyrinth weir)와 변형된 형태인 경사형 마루 래버린스 위어(inclined crest labyrinth weir)를 제시하였으며, 래버린스 위어 적용시 발생하는 수리학적 흐름특성을 이용하여 침사지에서 침전율을 개선하고자 하였다. 본 연구에서는 전수두비 (headwater ratio) $H_t/P$, 와 확폭비(magnification ratio) L/W, 그리고 총 마루길이 당 유입유량 $q_L$을 침전효율의 주요 분석지표로 제시하였으며, 기존의 침사지 형상과 래버린스 위어를 적용한 래버린스 침사지 형상을 제작하여 실험을 수행하였다. 실험결과 래버린스 위어를 적용한 침사지는 확폭비 변화에 따른 전수두비의 변동이 침전효율(%)에 크게 영향을 미치는 것으로 분석되었으며, 래버린스 위어 반복배열수(N)가 2~4개 일 때 침전효율이 개선되는 것으로 분석되었다. 또한 회기분석을 통해 침전효율이 개선되는 유입조건 및 형상은 총 마루길이 당 유입유량이 $q_L{\geq}3.5cm^2/s$인 조건과 확폭비가 $L/W{\leq}3.5$ 인 경우 로 나타났으며, 이러한 침전효율이 개선되는 구간을 도표상으로 표현 및 분석하였다. 본 연구에서 제시한 분석지표를 통해 오탁수 유입량 및 침사지 소요면적 등 설계조건에 따른 적합한 침전효율을 갖는 구조적으로 최적화된 래버린스 침사지의 기초 설계에 활용될 수 있을 것이다.

Influence of Temperature and Relative Humidity on the Rearing Performance and Disease Incidence in CSR Hybrid Silkworms, Bombyx mori L.

  • Fifth instar larvae of the new bivoltine hybrid (CSR2 ${\times}$ CSR5) were reared under different temperature and humidity viz., $20^{\circ}C$ and 85 ${pm}$ 5% R.H (T1), $25^{\circ}C$ and 70 ${pm}$ 5% R.H (T2-Optimum), $30^{\circ}C$ and 80% ${pm}$ 5 R.H (T3) and $35^{\circ}C$and 50 ${pm}$ 5% R.H (T4). The cocoon yield, cocoon characters and disease incidence were studied in normal (non infectious source, i.e control) rearing as well as in 1% infectious source of rearing. The results indicated that V instar larval duration was prolonged and cocoon weight was improved in T1. ERR and shell ratio were significantly improved and disease incidence was minimised in T2. Further significant difference was observed among the treatments with regard to spread of diseases.
