Syzygium claviflorum 추출물의 항산화 활성 및 각질형성세포 분화유도 효과 (Identification of Antioxidant Activities and Stimulation of Human Keratinocytes Differentiation Effects of Syzygium claviflorum Extract)

  • 사이자이지움 클래비플로룸(Syzygium claviflorum (Roxb.) Wall. ex A.M. Cowan & Cowan, S. claviflorum)의 추출물 (잎, 줄기, 열매, 꽃)의 화장품 소재로써 활용되기 위한 생리활성 능력을 검증하였다. 첫번째로, S. claviflorum 추출물은 DPPH와 ABTS assay법을 이용한 항산화 실험에서 다양한 농도로 처리한 결과, 약 80% 이상의 자유 라디칼을 제거하였다. 사람 피부 표피 각질형성세포(human epidermal keratinocytes)를 이용한 세포독성 실험에서는 S. claviflorum 추출물은 낮은 세포독성을 보였다. 또한, S. claviflorum 추출물은 각질형성세포의 분화인자 (keratin (KRT)1, KRT2, KRT9, KRT10)와 피부장벽의 기능 유지에 중요한 involucrin (IVL), loricrin (LOR), filaggrin (FLG)과 claudin1 (CLDN1) 유전자의 발현을 현저히 증가시켰다. 특히, in vitro 아토피 피부염 실험에서 interleukin (IL)-4/IL-13에 의해 억제된 FLG 단백질 발현이 S. claviflorum 추출물에 의해 회복되었다. 따라서, 뛰어난 항산화 효능과 피부장벽 개선 기능을 보유한 S. claviflorum 추출물은 향후 아토피 피부염 치료제 및 화장품 개발에 유용한 소재가 될 것이다.

Studies on Genetic Stability of Micropropagated Plants and, Reintroduction in an Endemic and Endangered Taxon: Syzygium travancoricum Gamble (Myrtacae)

  • Tissue culture techniques arguably are an important approach for ex situ conservation of rare and endangered plant species. However, there is utmost importance on maintaining the genetic integrity of the introduced plants especially in tree species. To examine the genetic integrity of the micropropagated plants, we randomly screened few hardened plants of Syzygium travancoricum, a critically endangered tree taxon, using Randomly Amplified Polymorphic DNA (RAPD) markers. Twenty-three random. primers were tried and twenty-five polymorphic loci were identified. The dendrogram based on the Unweighted Pair-Group Method Arithmetic Average and Nei's similarity index depicted about 97% homology between the mother plants and micropropagated plants. Further, an attempt was made to reintroduce the micropropagated plants in the wild. Over three hundred small trees could be successfully established.

Insecticidal Activity of Extracts Isolated from Syzygium Aromaticum

  • This study separated the crude extract (70% ethanol) of and its three fractions (hexane, chloroform and ethyl acetate extracts) on the basis of polarity indexes, and examined for their insecticidal activities against aphid (Uroleucon lactucicola). For crude extraction, the 70% ethanol extract showed the best extract yield (58.0%) and insecticidal activity (69.0%) among the various concentrations tested (water, 30% ethanol, 50% ethanol, 70% ethanol and 95% ethanol). The major chemical compounds of different fractions (hexane, chloroform and ethyl acetate extracts) were identified as eugenol by head space-GC-MS analysis. The hexane extract showed the highest eugenol content (43.7%) and insecticidal activity (80.0%). The insecticidal activity is accordingly believed to be attributable to the eugenol component. This may provide a useful starting point for the development of bio-pesticides.

Comparative observations on Cephaleuros parasiticus and C. virescens (Trentepohliaceae, Chlorophyta) from India

  • Cephaleuros parasiticus and C. virescens were collected from Kerala and Tamil Nadu, India. Macroscopic and microscopic features were observed and their comparative features were discussed. The lesions of C. parasiticus occur on the upper and lower leaf surfaces although zoosporangia form only on the lower surface. The thalli grow subepidermally and intramatrically, causing necrosis of whole leaf tissue. On the other hand C. virescens thalli develop on the upper surface and zoosporangia form on the upper surface, the thalli grow subcuticularly, and only the host epidermal and palisade cells are necrosed. Syzygium aromaticum and Polyalthia longifolia are new host plants of C. parasiticus and C. virescens, respectively.

고지방식이로 유도된 비만 마우스에서 정향(丁香)의 항비만 효과 (The Anti-Obesity Activity of Syzygium aromaticum L. in High-Fat Diet-induced Obese Mice)

  • Objectives : This study aims to analyze the anti-obesity effect of Syzygium aromaticum L. (SA) in obese mice made by a 60% high-fat diet (HFD). Methods : The antioxidant activities of SA were evaluated in vitro. To assess the anti-obesity effect of SA, male C57BL/6 mice were divided into five groups: Normal, Control, GC100 (Garcinia cambogia 100 mg/kg/day), SA100 (SA 100 mg/kg/day), SA200 (SA 200 mg/kg/day). All groups underwent a 6-week regimen of HFD and oral administration, except for the Normal group. Subsequently, we performed blood analysis, western blotting, and histopathological staining. Results : SA demonstrated effectiveness in antioxidant measurements. SA treatment resulted in a significant decrease in body weight gain, along with reductions in liver and epididymal fat weights. Serum triglyceride (TG), total cholesterol (TC), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and leptin levels were reduced with SA treatment. Moreover, in the SA100 group, the reduction of both TG and TC synthesis was caused by inhibiting the sterol regulatory element-binding transcription factor 1 (SREBP-1) and sterol regulatory element-binding transcription factor 2 (SREBP-2) through the Sirtuin 1 (Sirt1)/phospho-AMP-activated protein kinase (p-AMPK) pathway. Furthermore, SA treatment at a dose of 100 mg/kg reduced the accumulation of lipid droplets in the liver and the adipocyte size of the epididymal fat. Conclusion : Our research reveals the anti-obesity effects of SA by demonstrating its ability to inhibit body weight gain and lipid accumulation, suggesting that SA might be promising for obesity treatment.

IgE 매개 RBL-2H3 세포 활성화에 대한 정향 에탄올 추출물의 억제 효과 (Inhibitory Effects of Syzygium aromaticum Ethanol Extracts on IgE Mediated RBL-2H3 cell Activation)

  • 목적 : 본 연구에서는 정향 에탄올 추출물이 RBL-2H3 세포 매개 알레르기 반응에 대해 미치는 영향과 그 작용기전에 대해 연구했다. 방법 : 정향 에탄올 추출물의 RBL-2H3 세포에 대한 독성 여부는 MTT 분석을 통해 평가했다. 정향 에탄올 추출물의 항알러지 작용은 효소결합면역 분석방법(ELISA)을 이용해 ${\beta}$-Hexosaminidase과 Histamine의 분비량을 측정하여 평가하였다. 정향 에탄올 추출물의 작용기전에 대해서는 유사 분열물질-활성화단백질인산화효소(mitogen-activated protein kinase, MAPK)를 western blot 법을 이용하여 측정함으로써 평가하였다. 결과 : 정향의 에탄올 추출물은 RBL-2H3 세포에 대해 독성을 나타내지 않는 농도에서 RBL-2H3 세포의 탈과립과 히스타민 분비를 유의하게 억제하였으며, p38 MAPK의 활성을 차단하였다. 결론 : 본 연구의 결과 정향의 에탄올 추출물은 비만세포에서 유래된 알러지 반응을 억제하는 효과가 있으며, 또한 그 작용기전은 p38 MAPK 인산화와 연계되어 있을 것으로 사료된다.

정향 추출물 도포가 DNCB로 유발된 알레르기성 접촉 피부염에 미치는 영향 (The Effects of Syzygium aromaticum extract Spread on the Allergic Contact Dermatitis induced by DNCB)

  • Objectives : The purpose of this study is to investigate effects of Syzygium aromaticum (SAE) spread on the allergic contact dermatitis caused by 2,4-dinitro-chlorobezene (DNCB). Methods : Forty-two mice were divided into six groups ; normal, negative control (DNCB-treated), positive control (DNCB + 1% pimecrolimus), experimental group I, II and III. control and experimental groups were induced allergic contact dermatitis by DNCB. Experimental group I(DNCB + 0.2% SAE), II(DNCB + 1% SAE) and III(DNCB + 5% SAE) were spread SAE and positive control was spread the 1% pimecrolimus. In this study, effect of SAE on clinical aspects on the skin, histopathological change, the blood level of IgE, cytokines, histamine were investigated. In addition, effect of SAE on spleen $CD4^+/CD8^+$ T cell subset was investigated. Results : 1. In experimental group I, II and III, erythemas and edema were more reduced than negative control. 2. In experimental group I, II and III inflammatory edema and the numbers of infiltrated inflammatory cells were more reduced than negative control. 3. In experimental group I, II and III, clinical skin score was more reduced than negative control. 4. In experimental group II and III, the thickness of skin was statistically significant reduced than negative control. 5. In experimental group II and III, histamine release was statistically significant reduced than negative control in dose-dependantly. 6. In experimental group II and III, cytokines (IL-1${\beta}$, TNF-${\alpha}$, IL-4, IL-6, IL-10) were statistically significant reduced than negative control in dose-dependantly. 7. In experimental group I, II and III, the level of total IgE was statistically significant reduced than negative control in dose-dependantly. 8. In experimental group III, $CD4^+$ and $CD8^+$ T cells were statistically significant decreased similar to the positive control. Conclusions : According to above experiments, Syzygium aromaticum(SAE) was effective on allergic contact dermatitis.

Antifungal Activities of the Essential Oils in Syzygium aromaticum (L.) Merr. Et Perry and Leptospermum petersonii Bailey and their Constituents against Various Dermatophytes

  • This study was carried out in order to investigate the potential of using plant oils derived from Leptospermum petersonii Bailey and Syzygium aromaticum L. Merr. Et Perry as natural antifungal agents. The antifungal effects of essential oils at concentrations of 0.05, 0.1, 0.15, and 0.2 mg/ml on the dermatophytes Microsporum canis (KCTC 6591), Trichophyton mentagrophytes (KCTC 6077), Trichophyton rubrum (KCCM 60443), Epidermophyton floccosum (KCCM 11667), and Microsporum gypseum were evaluated using the agar diffusion method. The major constituents of the active fraction against the dermatophytes were identified by gas chromatography-mass spectrometry and high-performance liquid chromatography analysis. The antifungal activities of S. aromaticum oil (clove oil) against the dermatophytes tested were highest at a concentration of 0.2mg/ml, with an effectiveness of more than 60%. Hyphal growth was completely inhibited in T. mentagrophytes, T. rubrum, and M. gypseum by treatment with clove oil at a concentration of 0.2 mg/ml. Eugenol was the most effective antifungal constituent of clove oil against the dermatophytes T. mentagrophytes and M. canis. Morphological changes in the hyphae of T. mentagrophytes, such as damage to the cell wall and cell membrane and the expansion of the endoplasmic reticulum, after treatment with 0.11 mg/ml eugenol were observed by transmission electron microscopy (TEM). At a concentration of 0.2 mg/ml, L. petersonii oil (LPO) was more than 90% effective against all of the dermatophytes tested, with the exception of T. rubrum. Geranial was determined to be the most active antifungal constituent of L. petersonii oil. Taken together, the results of this study demonstrate that clove and tea tree oils exhibited significant antifungal activities against the dermatophytes tested in this study.

저육(猪肉)과 잘 어울리는 췌장라이페이즈 억제능을 가진 한약재의 비교평가 (Comparative Evaluation of Korean Medicine Well-matched with Pork through an Inhibition of Pancreatic Lipase)

  • Objectives : The aim of this study is to search Korean Medicine with the superior inhibitory effect of pancreatic lipase including a possibility as a well-matched sauce material with pork. Methods : We chose 5 samples which have the potent inhibitory effect of pancreatic lipase enzyme among 9 samples. 5 samples were Glycyrrhizae rhizoma, Cinnamomi cortex, Ganoderma lucidum, Syzygium aromaticum, and Schisandra chinensis. Animals were divided into eight groups (n=7). The experimental groups except for normal group were fed 60% high-fat diet. 5 samples were orally administered at a dose of 200 mg/kg body weight and orlistat were orally administrated at a dose of 60 mg/kg body weight for 7 days. Biochemical anaylses of 5 samples were executed based on lipid parameters such as triglyceride (TG) and total cholesterol (TC). Results : Korean Medicines with $IC_{50}$ of below $1mg/m{\ell}$ were Glycyrrhizae rhizoma, Cinnamomi cortex, Ganoderma lucidum, Syzygium aromaticum, and Schisandra chinensis. Body weight change of all drug-treated groups except Glycyrrhizae rhizoma reduced significantly during the experimental period. Orlistat treatment reduced significantly both TG and TC in serum. The significant emission of TG in fece showed in Cinnamomi cortex treatment, whereas Ganoderma lucidum and Syzygium aromaticum showed a tendency to increase without a significance. Besides, Ganoderma lucidum and Schisandra chinensis emitted significantly TC to fece. Conclusions : In conclusion, Cinnamomi cortex may exert anti-obesity effect by directly inhibiting pancreatic lipase, which would prevent the absorption of lipid from the small intestine. Accordingly, Cinnamomi cortex may suggest a high possibility as a well-matched sauce material with pork.
