Actinoplanes missouriensis KCTC 1780의 응집 균체에 의한 과당생산 (Production of High Fructose Syrup by Flocculated Actinoplanes missouriensis KCTC 1780)

  • The whole cells of Actinoplanes missouriensis KCTC 1780 which produce glucose isomerase was immobillized by flocculation method for the effective production of high fructose syrup using packed-bed bioreactor system. Among the flocculation methods used In this study, the glucose Isomerase activity of flocculated cells using 5% polyethylenimmine and 0.2% glutaraldehyde was the highest as 46.3 unit, and the flocculant was 10.3g(wet weight) per 100m1 of broth, and the residual activity was 92.5%. In the batch operation of glucose isomerization using the flocculated cells, the optimum pH, temperature and isomerization ratio were 7.0, 75$^{\circ}C$ and 31%, respectively. The optimum concentration of Mg2+ which was activator on the glucose isomerization of flocculated cells was 0.1M, and glucose isomerase activity was increased by about 40% compared to none of Mg2+. In the packed-bed bioreactor system with 1.2 hour of residence time at 7$0^{\circ}C$, the reaction stability maintained until 96 hour without toss of activity, and the equilibrium was kept up to 120 hours of the operation.

콩다식 제조시 당의 종류와 당의 양, 반죽 횟수에 따른 물리적 특성 연구 (Study of Physical Characteristics on the Kind, Amount of Sugar and Number of Kneading by Processing Method of Soybean Dasik)

  • The purpose of this study was to establish the most favorable processing method for soybean Dasik. Factors were kind of sugar (honey, syrup, oligo sugar), amount of sugar (50 g, 60 g, 70 g) and number of kneading (20 times, 30 times, 40 times). Physical characteristics were measured by moisture content, color difference and texture. Moisture content was increased in order of honey, oligo sugar and syrup. Lightness of soybean Dasik was different with amount of sugar. The tendency of L value and b value were dominent by honey. Lightness, redness and yellowness had no difference with the kind of sugar and number of kneading. Hardness was not a significant factor. Springiness was significantly different by the kind and amount of sugar. Cohesiveness, gumminess and chewiness was influenced by the kind and amount of sugar, the interaction of kind and amount of sugar.

감미료의 종류를 달리한 데리야끼소스의 품질 특성 (Quality Characteristics of $Teriyaki$ sauce Added with Different Sweeteners)

  • 본 연구는 소비자들의 건강에 대한 요구를 충족시키기 위해 데리야끼소스에 설탕을 대신 할 저열량 감미료로 프락토올리고당, 자일리톨, 에리스리톨, 아가베시럽, 스테비오시드을 선정하였다. 설탕과 프락토올리고당, 자일리톨, 에리스리톨, 아가베시럽, 스테비오시드을 첨가하여 데리야끼소스를 제조한 후 수분과 점도를 측정한 결과 스테비오시드를 첨가한 것의 수분 함량이 가장 작았고 점도는 가장 높았다. 설탕과 자일리톨을 첨가한 소스의 당도가 같고 염분을 가장 낮았다. pH는 유의적인 차이가 없었다. 자일리톨을 첨가한 소스는 관능검사 결과 단맛, 간장의 향, 맛, 감칠맛에서 다른 감미료보다 선호되어 데리야끼소스에 자일리톨을 첨가하는 것이 가장 좋은 것을 알 수 있었다.

  • Aspergillus saitoi가 생산(生産)하는 섬유소분해효소(纖維素分解酵素)의 생산조건(生産條件)과 밀감내과피(內果皮)의 탈피효과(脫皮效果)를 검토한 결과(結果)는 다음과 같다. 1. Asp. saitoi의 배양최적(培養最適) 온도(溫度)는 $30^{\circ}C$, 시간(時間)은 72시간(時間)에서 섬유소(纖維素) 생성능(生成能)이 가장 좋았다. 2. 본효소(本酵素)를 10배(倍) 농도로 희석하여 (희석효소액 : 밀감 2 : 1) $40^{\circ}C$의 항온조(恒溫槽)에서 $3.5{\sim}4$시간(時間) 작용(作用)시킨 것이 탈피정도(脫皮程度), 풍미(風味), 광택(光澤)이 우수하여 밀감 syrup 통조림 제조조건으로 최적(最適)이었다.

이소말토올리고당을 사용한 시럽에 집청한 약과의 특성 (Effects of dipping syrups prepared with isomaltooligosaccharides on the Yackwa quality)

  • 설탕의 0%, 25%, 50%를 IMO로 대체하여 제조한 시럽에 집청한 약과의 물리적, 관능적 특성을 검토하였다. IMO를 사용한 시럽에 집청한 약과는 대조군 약과에 비해 환원당 함량이 증가했으며, 탈수율은 높았으나 집청액 흡수율은 낮았다. 약과의 관능적 특성은 윤기, 단맛, 고소한 맛, 전체적인 기호도 등이 유의적 차이를 나타냈다(p<.05). IMO를 사용한 시럽에 집청한 약과는 대조군 약과에 비해 윤기가 강하고 단맛이 약했으며, 고소한 맛이 강하고 전체적인 기호도가 높았다. Pearson correlation coefficient를 산출하여 관능적 특성간의 상관관계를 검토한 결과, 촉촉한 정도는 윤기(p<.01)와 정의 상관관계를, 부착성은 윤기 및 촉촉한 정도와 정의 상관관계(p<.01)를 나타내었다. 단맛은 윤기와 부의 상관관계(p<.01)를, 냄새는 윤기, 부착성, 응집성과 정의 상관관계(p<.01)를 나타내었다. 전체적인 기호도는 윤기(p<.01), 촉촉한 정도(p<.05), 부착성(p<.05), 응집성(p<.01), 고소한 맛(p<.01)과 정의상관관계를, 단맛과는 부의 상관관계(p<.05)를 나타내어, 윤기가 강하고 더 촉촉하며 부착성과 응집성이 크고, 단맛은 약하고 고소한 맛이 강한 약과에 대한 기호도가 더 높게 나타났다.

Identification of Two Novel BCKDHB Mutations in Korean Siblings with Maple Syrup Urine Disease Showing Mild Clinical Presentation

  • Maple syrup urine disease (MSUD) is a disorder that involves the metabolism of branched chain amino acids, arising from a defect in branched-chain ${\alpha}$-keto acid dehydrogenase complex. Mutations have been identified in the BCKDHA, BCKDHB, or DBT genes, which encode different subunits of the BCKDH complex. Although encephalopathy and progressive neurodegeneration are its major manifestations, the severity of the disease may range from the severe classic type to milder intermediate variants. We report two Korean siblings with the milder intermediate MSUD who were diagnosed with MSUD by a combination of newborn screening tests using tandem mass spectrometry and family genetic screening for MSUD. At diagnosis, the patients' plasma levels were elevated for leucine, isoleucine, valine, and alloisoleucine, and branched-chain ${\alpha}$-keto acids and branched-chain ${\alpha}$-hydroxy acids were detected in their urine. BCKDHA, BCKDHB, and DBT analysis was performed, and two novel mutations were identified in BCKDHB. Our patients were thought to have the milder intermediate variant of MSUD, rather than the classic form. Although MSUD is a typical metabolic disease with poor prognosis, better outcomes can be expected if early diagnosis and prompt management are provided, particularly for milder forms of the disease.

Chemical Composition and Feeding Value of Chopped Date Fronds (CDF) as Affected by Urea and Date Syrup Extract Treatment

  • In the first of two experiments, chemical composition (Ash, CP and CF contents), nutritive value (in vitro organic matter disappearance "lVOMD" and energy content "ME") and nitrogen retention of chopped date fronds (CDF) as affected by chemical treatment (CT) and storage duration (SD) were investigated. The experimental design was a completely randomized block, arranged in a $3{\times}4$ factorial, with 3 (CT) and 4 (SD) as the main factors using 2 replications / treatment. Three chemical treatments, were used : Control (With zero or no chemical treatment); treatment with 4% urea solution (w /w) and treatment with 4% urea + Date syrup extract (DS). The 4 (SD) were: Control (immediately after opening - without any storage or zero time storage); one month storage period; two months storage period and three months storage period. Both (CT) and (SD) had significant effects (p < 0.05) on CP and ash contents of the CDF, however there were no significant effects (p > 0.05) due to (CT) and (SD) on CF and nutritive value. High retention value (> 80%) for nitrogen was reported for the treated CDF, immediately after opening the incubated material (zero SD). However, nitrogen retention decreased with increasing (SD) to 67% after one month (SD) but tended to stay at a fairly constant level of 67% until 3 months of storage. In experiment 2, the feeding value of treated CDF was evaluated in comparison to Rhodes grass hay, using growing goats and sheep. A $4{\times}2$ factorial design was used (Four roughage sources were used : Rhodes grass hay, untreated CDF, urea-treated CDF and CDF treated with urea + DS with sheep and goats as two animal species, using three animals / treatment). Untreated CDF had a similar feeding value to Rhodes grass hay. However treatment of CDF with urea alone or with urea + DS depressed animal performance of both goats and sheep, apparently due to depression of feed intake (appetite). Feeding untreated CDF decreased feeding cost by 29% and cost/kg gain by 23%. CDF showed a good potential as a cheap local roughage and emergency feed for ruminants in the Sultanate.

광학용 아크릴 점착제 제조 및 점착특성에 관한 연구 (Preparation of Acrylic Pressure Sensitive Adhesives for Optical Applications and Their Adhesion Performance)

  • 본 연구는 2-ethylhexyl acrylate와 2-hydroxyethyl acrylate 기본구조에 isobornyl acrylate와 tetrahydrofurfuryl acrylate(THFA)의 조성비를 변화시키면서 광중합된 4원 공중합체를 합성한 후 가교제로 1,6-hexanediol diacrylate가 혼합된 시럽을 제조하였다. 합성한 시럽에 UV-광원을 조사시켜 점착제를 제조한 후 점착특성과 광학특성을 고찰하였으며 시럽의 기본물성인 고형분, 점도, 및 분자량 분석도 수행하였다. THFA의 함량이 증가할수록 시럽의 분자량은 감소하였으나 고형분은 반대로 증가하였다. 점착제의 박리강도는 THFA의 함량에 따른 반비례관계를 나타내고 점착제의 표면에너지는 비례관계를 나타내었다. 또한 모든 점착제에서 92% 이상의 광투과도와 1.0%이하의 haze값, 그리고 1.0 이하의 색차계값을 보여주었다.
