EGF, T3, HB-EGF 가 치주인대섬유모세포에 미치는 영향 (The Effect of EGF, T3 and HB-EGF on Human Periodontal Fibroblasts)

  • 치주인대섬유모세포들은 완전탈구 된 치아의 성공적인 재식을 위한 중요한 요소이다. 따라서, 외상으로 인해 완전탈구된 치아의 보존을 위한 보관액을 선택하는 것이 중요하다. 성장인자들과 호르몬들은 치주인대섬유모세포들의 생존을 위한 치료적 제제로 고려되고 있다. Epidermal growth factor(EGF)는 다른 조직에서 재생과 상처 치유 과정의 중요한 역할인자로 대두되고 있다. 따라서, 완전탈구 된 치아를 위한 치료적 적용을 위해 EGF의 세포 증식에 미치는 영향을 평가하였다. 또한 EGF와 tri-iodothyronine(T3)의 혼합액, EGF와 Heparin-binding epidermal growth factor-like growth factor(HB-EGF)의 혼합액이 세포 증식에 미치는 상승 효과를 평가하였다. 치주인대섬유모세포의 세포증식은 EGF 농도가 증가함에 따라 증가하였고, 10 ng/ml 농도에서 최대 세포증식을 보였다. EGF는 상처치유분석에서 상처 치유촉진과 이동성을 보여주었다. EGF에 T3와 HB-EGF를 첨가한 혼합액에서 배양한 세포는 EGF만 처리한 경우보다 세포 증식이 상승되었다. EGF와 T3 혼합액의 상승효과 기전을 유추하기 위해서 RT-PCR로 EGF 수용기의 발현을 확인하였고, T3가 EGF 수용기 발현을 증가시켰음을 확인하였다. 따라서 EGF와, EGF와 T3, EGF와 HB-EGF의 혼합액은 완전탈구된 치아의 치료에 있어 유용한 선택이 될 것이다.

이온도입에 의한 염산프로카인의 경피전달 증대 -고전압전류 및 초음파 병행의 상승효과 비교- (Enhanced Transdermal Delivery of Procaine Hydrochloride by lontophoresis -Comparison of Synergic Effect of High Voltage Current and Ultrasound-)

  • The purpose of this study was to determine the effects of iontophoresis on transdermal delivery of procaine hydrochloride in healthy volunteers, as well as to the synergic effect of high voltage current or ultrasound on the efficacy of transdermal delivery of iontophoresis. Forty healthy volunteers were randomly assigned to four groups topical application group (TA), iontophoresis group (IT), pre-treatment of high voltage current stimulation with iontophoresis (HVS + IT), and pre-treatment of ultrasound application with iontophoresis (US + IT). All subjects received procaine iontophoresis on the forearm using direct current with 4 mA f3r 15 minutes. All subject was measured the duration of local anesthesia, pressure pain threshold, pain perception threshold using rectangular wave at 0.2 ms, 1 ms, 50 ms of rectangular current stimulation after procaine iontophoresis. For comparisons of the sensory characteristics and efficacy of iontophoresis between the groups, an one-way ANOVA and Kruskal-Wallis were used. The significant difference the duration of local anesthesia were found between the groups (p<0.001). The local anesthetic duration of IT, HVS+IT were significantly longer than TA. Meanwhile, the local anesthetic duration of US+IT was significantly longer than HVS+IT, IT and TA group (p<0.05). Also, the pressure pain threshold, pain perception threshold at 0.2 ms, 1 ms, 50 ms were significant difference between the groups (p<0.001). All sensory characteristics including pressure pain threshold, pain perception threshold of IT, HVS+IT was significantly increased than TA, whereas, US+1T was significantly increased HVS+1T, IT and TA (p<0.05). This study showed that the procaine iontophoresis have increase the duration of local anesthesia concomitantly pressure pain threshold and pain perception threshold of sensory nerve fibers such as $A-{\beta}$, $A-{\delta}$ and C fiber. This findings suggest that the iontophoresis enhanced the transdermal delivery of drug ions in vivo. The combination of ultrasound application and iontophoresis synergized the transdermal delivery of drug ions. It is suggests that an electric field, mechanical and heating property of ultrasound may contribute to synergic effect due to temporary changes of structure in the stratum corneum.

대전방지제에 의한 폴리에스테르 수지의 표면저항과 인장강도 특성 (Surface Resistance and Tensile Strength of Polyester Resin by Anti-static Agents)

  • 선박의 엔진실 등 유증기가 많이 발생하는 위험한 환경에도 적용할 수 있는 대전방지용 폴리에스테르 수지를 개발할 목적으로, 폴리에스테르 수지의 표면저항과 인장강도에 미치는 대전방지제의 영향을 조사하였다. 대전방지제로는 3종의 유기성 대전방지제와 전도성 카본블랙을 단독 혹은 혼합 사용하였다. 이들 대전방지제들의 종류와 첨가량, 그리고 혼합 사용하였을 때의 효과 등에 대한 실험을 수행하였다. 대전방지제의 첨가량이 증가함에 따라 생성되는 폴리에스테르 수지의 표면저항과 인장강도는 전반적으로 감소하였으나, 유기성 대전방지제와 카본블랙을 혼합 사용하여 인장강도가 비교적 우수하면서도 표면저항이 낮은 폴리에스테르 수지를 제조할 수 있었다.

  • In order to evaluate the anti-tumor and synergic effect of BoJungIkKeeHapDaeChilKi-Tang on doxorubicin, the inhibitory concentration(IC), IC50 and IC90 of single use of doxorubicin and BoJungIkKeeHapDaeChilKi-Tang with their concomitant treatment against MKN-45(Human stomach carcinoma) was observed using MTT(Microculture Tetrazolium test) assay. In addition, their anti-tumor effects were also observed in the xenograft nude mice models agianst MKN-45 cell lines. BoJungIkKeeHapDaeChilKi-Tang has only mimic direct anti-tumor effect against to MKN-45 cell lines but they were decreased general depressed signs induced by implantation of tumor cell lines and increased the total WBC and lymphocyte numbers. So, it is considered or expected that BoJungIkKeeHapDaeChilKi-Tang extracts were reduced by the critical toxicity of doxorubicin and shows favorable synergic effect with doxorubicin and BoJungIkKeeHapDaeChilKi-Tang extracts.

So, it is considered or expected that Soonkiwhajungtang extracts were reduced the critical toxicity of doxorubicin and shows favorable synergic effect with doxorubidn and Soonkiwhajungtang extracts.

Retinol에 대한 항산화 연구 (The Study on Antioxidation of Retinal)

  • Retinol에 대한 합성물질과 천연물질의 항산화력을 비교하였다. 수용성 합성 항산화제 tertiary butylhydroquulone(TBHQ)와 수용성 천연 항산화제 $\alpha$-glycosyl rutin($\alpha$-G rutin), licorice, pycnogenol, 지용성 합성 항산화제 butylated hydroxytoluene(BHT)와 지용성 천연 항산화제 $\alpha$-lipoic acid, ferulic acid, natural concentrated tocopherol(no-tocopherol)를 각각 0.0, 0.0l, 0.02, 0.05, 0.l0wt% 사용하여 리포좀에 봉입하였으며, 4$^{\circ}C$, $25^{\circ}C$, 5$0^{\circ}C$에서 8주 동안 retinol의 잔류량 변화를 HPLC로 측정하였다. 사용된 항산화제는 retinol에 대하여 모두 산화억제효과를 나타내었으며, 수용성 항산화제는 licorice>pycnogenol>TBHQ>$\alpha$-G rutin 순으로, 지용성 항산화제는 $\alpha$-lipoic acid>BHT>no-tocopherol>ferulic acid 순으로 나타났다. 또한 $\alpha$-lipolc acid, BHT, licorice, pycnogenol을 흔합하여 retinol에 대한 항산화력 상승효과를 알아보았다. $\alpha$-lipoic acid와 BHT를 혼합하여 사용하였을 때 retinol에 대한 항산화력이 가장 좋게 나타났다.

Differences in Neurotransmitters Level as Biomarker on Sleep Effects in Dementia Patients with Insomnia after Essential Oils Treatment

  • This study aimed to examine the effects of several essential oils on insomnia in dementia patients following transdermal treatment (aromatherapy). The mean change rates (%) of sleep biomarkers were compared between the single essential and jojoba (vehicle) oil massage groups in this study. The lavender (L) essential oil massage group demonstrated a significant decrease in the mean change rate (%) of 24-h urinary free cortisol, whereas the valerian (V) essential oil massage group demonstrated a significant increase in the mean change rate (%) of serum 5-hydroxytryptamine. In addition, a significant increase in the mean change rate (%) of 24-h urinary norepinephrine was observed in the chamomile (C) essential oil massage group only. Based on these results, valerian, lavender, and chamomile oils were mixed in different ratios to produce blending oils A (L:C:V=2:2:1), B (L:C:V=3:1:1) and C (L:C:V=1:3:1). The highest level of serum 5-hydroxytryptamine was observed after administering blending oil A. These results suggest that blending oil A might possess therapeutic effects against insomnia. Overall, it is hypothesized that the optimally blended essential oil will produce synergic effects when combined with hypnotic drugs.

한약을 이용한 항암 실험 연구의 경향과 연구 방향 (An Anti-cancer experimental study using herbs)

  • After examining and analysing the medicinal herbs of fifty-three experimental papers, we studied their effects on immediate tumors in specific cancers. We did not study the influence on the life span of general cancerous cells. We looked to see if the combined usage of medicinal herbs and anticancer agents inhibited the tumor cell's growth. The serum test and blood cell count test showed if the medicinal herbs inhibited the side effects of the anticancer agent. The test showed that more than 80 percent of used medicinal herbs, brought anticancer activities. However, anticancer experimental studies using medicinal herbs have draw-backs. First, it is difficult to choose a prescription using the standards of Oriental Medicine because we are testing a mouse not a man. Second, because we only observed the indirect effect on the whole physiological regulation caused by the synergic effects of the complex prescription, we are not able to understand the detailed mechanism of the herbs. Therefore; if the anticancer effect of the herbs is proved by the experiment, we need to research the concrete medical action of medicinal herbs and the immunological analysis of herbal medicines on the body.

현호색산의 항혈전작용에 대한 연구 (The Experimental Studies on Antithrombotic Effects of Hyunhosaiksan)

  • The purpose of the present study was to investigate the effects of Hyunhosaiksan (HHS) on antithrombotic actions which include blood activation, thrombus removal, warming of circulating blood, and the control of pain on abdomen and lower and upper burning spaces. HHS significantly inhibited platelet aggregation induced by ADP and epinephrine in a HHS dose-dependent manner when analyzed by the Sigmoid Emax model in WinNonlin. EC50 values of HHS were 1.71 ㎍/ml and 0.004 ㎍/ml for ADP and epinephrine respectively. In the vivo study, HHS inhibited pulmonary embolism induced by collagen and epinephrine, which was however statistically insignificant. HHS increased number of platelets, APTT and volume of fibrinogen significantly as compared with the control group in dextran-induced thrombus model. Furthermore, HHS stimulated levels of blood flow in vivo though its effect was not observed in vitro. These results suggest that Hyunhosaiksan (HHS) can be used for treating numerous diseases related with blood aggregation and circulation problems. Further systematic investigations on the synergic effects among drugs used in the oriental medicine as well as in the western medicine in relation to thrombosis therapy would provide an important insight into the potential therapeutic applications.
