In common globular proteins, the native form is in its most stable state. However, the native form of inhibitory serpins (serine protease inhibitors) and some viral membrane fusion proteins is in a metastable state. Metastability in these proteins is critical to their biological functions. Our previous studies revealed that unusual interactions, such as side-chain overpacking, buried polar groups, surface hydrophobic pockets, and internal cavities are the structural basis of the native metastability. To understand the mechanism by which these structural defects regulate protein functions, cavity-filling mutations of a 1-antitrypsin, a prototype serpin, were characterized. Increasing conformational stability is correlated with decreasing inhibitory activity. Moreover, the activity loss appears to correlate with the decrease in the rate of the conformational switch during complex formation with a target protease. We also increased the stability of a 1-antitrypsin greatly via combining various stabilizing single amino acid substitutions that were distributed throughout the molecule. The results showed that a substantial increase of stability, over 13 kcal/mol, affected the inhibitory activity with a correlation of 11% activity loss per kcal/mol. The results strongly suggest that the native metastability of proteins is indeed a structural design that regulates protein functions and that the native strain of a 1-antitrypsin distributed throughout the molecule regulates the inhibitory function in a concerted manner.
This paper propose a novel scheme, called Mobiel Terminal Initiated Scheme(MTIS), in which mobile terminal initiates the backward handover by sending handover request message with the list of target radio ports. In this scheme, the old ATM switch suporting end-user mobility, denoted by EMAS\ulcorner, checks whether each EMAS\ulcorner, managing the target radio port, has its available resources. If it has, the EMAS\ulcorner performs the path rerouting between CrossOver Switch (COS) and itself after deciding the most suitable target radio port. Therefore, the MT initiates the handover after deciding the most suitable target radio port through the beacon signal of Wireless Access Point (WAP). The EMAS\ulcorner have only to check the resource availability of the target radio port. It is no need to waste time to decide the suitable target radio port. Also, once receiving the request of the resource availability, the EMAS\ulcorner can reduce the rerouting delay time due to perform the path rerouting to the COS. In comparison with that of the ATM-Forum procedure, our proposed MTIS handover delay time reduced 14~21%, and end-to-end transfer delay time reduced 2~9%, as a result of the simulation.

This paper focuses on the actuation system combined with a piezoelectric transducer and an electric circuit, which leads to a new insight; the electric actuation system is equivalent to mechanical variable-stiffness actuation systems. By controlling the switch in the circuit, the electric status of the piezoelectric transducer is changed, and consequently a variable-stiffness mechanism is achieved on the electric actuator. This proposed actuator features a shift in the equilibrium point of force, while conventional electrically-induced variable-stiffness actuators feature the variation of the stiffness value. We intensively focus on the equilibrium shift in the actuation system, which has been neglected. The stiffness of the variable-stiffness actuator is periodically modulated by controlling the switch, to suppress the vibration of the system in an open-loop way. It is proved that this electric actuator is equivalent to its mechanical counterpart, and that the electrical version has some practical advantages over the mechanical one. Furthermore, another kind of electrically-induced variable-stiffness actuator, using an energy-recycling mechanism is also discussed from the viewpoint of open-loop vibration control. Extensive numerical simulations provide comprehensive assessment on both electrically-induced variable-stiffness actuators employed for open-loop vibration control.

In NCW(Network Centric Warfare) environment, mobile nodes communicate through wireless link. But wireless link provides limited networking performance due to signal interferences or mobility of nodes. So it is quite challenge to acquire enough networking resources and use the resources efficiently. In this paper, we have proposed a P2P based tactical information sharing system which provides satisfactory visual information playout for mobile nodes(i.e., military personnel, vehicle,..) in NCW environment. Our proposed system consists of two components. One is caching-enabled switch which stores tactical information segments at its internal storage and then transports them to mobile nodes when require. Another is centralized scheduling algorithm which exploits networking resources more efficiently. To validate performance of proposed system, we performed series of experiments in wireless network testbed. Results show improved performance in terms of segment-missing ratio, networking resources usage, sharing time, and number of simultaneous playout mobile nodes with acceptable playout continuity(i.e., over 95%).

Increasing air pollution levels and the global oil crisis has become a major hindrance in the growth of our automobile sector. Traditional Internal Combustion engines running on non-renewable fuels are proving to be the major culprit for the harmful effects on environment. With few modifications and also with assistance of few additional components current small SI engines can be modified into a pneumatic engine (commonly known as Compressed Air Engines) without much technical complications where the working fluid is compressed air. The working principle is very basic as adiabatic expansion of the compressed air takes place inside the cylinder pushing the piston downwards creating enough MEP to run the crank shaft at decent RPM. With the assistance of new research and development on pneumatic engines can explore the potential of pneumatic engines as a viable option over IC engines. The paper deals with analysis on RPM variation with corresponding compressed air injection at different crank angles from TDC keeping constant injection time period. Similarly RPM variation can also be observed at different injection pressures with similar injection angle variation. A setup employing a combination of magnetic switch (reed switch), magnets and solenoid valve is used in order to injection timing control. A conclusive data is obtained after detailed analysis of RPM variation that can be employed in newly modified pneumatic engines in order to enhance the running performance. With a number of benefits offered by pneumatic engine over IC engines such as no emissions, better efficiency, low running cost, light weight accompanied by optimized injection conditions can cause a significant development in pneumatic engines without any major alteration.
The purpose of this experiment was to develope Triggered Vacuum Switch(TVS) and to improve understanding of the high current vacuum arc phenomena in the TVS. The TVS has an array of rods of alternate polarity in which a fixed gap spacing is maintained between the rods. The cross section of each rod has trapezoidal shape. Breakdown of the TVS produced high current vacuum arc plasma. A spectroscopic measurement was performed over 20 kA peak current in the center of electrodes, in the vicinity of cathode, and outside electrodes. The electrode material tested was Fe. Measured Fe spectrum range was from 200 nm to 900 nm. Measurement result showed that over 90 percent of the charge states were FeII and the others were FeI and FeIII. The electron temperature was determined from the relative line intensity ratio methode of FeII system by assuming the local thermal equilibrium(LTE). The electron temperature at the center of electrodes was measured as 1.5 eV at 26 kA peak current. The electron temperature varied with its peak current. Intensity of spectrums is the highest in the vicinity of the cathode. Further we will also present study result of the diode phenomena in the TVS.
