On the dimensional stabilization of woods with treatment of Polyethylene Glycol-400 (폴리에치렌 글리콜-400에 의한 목재(木材)의 칫수안정화(安定化))

  • How to stabilize wood against shrinking and swelling in variable atmospheric moisture conditions is important to the wood-using industry and a challenge to research. Polyethylene glycol stabilize wood by bulking the fiber. PEG also serve as a chemical seasoning agent, suppress decay in high concentrations, and have slight effect on physical properties, gluing or finishing. The study designed to determine the effect of PEG-400 on the dimensional stabilization of local hardwoods for wood carvings that could supply a greatly expanding tourist trade and making curved furniture parts, lamp stands and other decorative objects, and possible gunstock. The species examined were 6 species, Seo-Namoo (Carpinus laxiflora), Cheungcheung-Namoo (Cornus controversa), Gorosae-Namoo (Acer mono), Karae-Namoo (Juglans mandshurica), Jolcham-Namoo (Quercusserrata) and Sanbud-Namoo (Prunus sargentii), used as block of 5cm thick radially to the grain, 7cm wide tangentially, and 70cm long parallel to the wood grain. All these test piecies were conditioned above the fiber saturation point before impregnation. The stabilization effects were determined for PEG-400 treated woods in a 50 percent solution for 20 days. The following conclusions were obtained. PEG retentions increased with treating time. It was more effective to treat at 60$^{\circ}C$ than at room temperature. In degree of PEG-400 impregnation on species, Cheungcheung-Namoo havinglow specific gravity had the highest retentions, 68.77% but the lowest, 56.33% was shown in Jolcham-Namoo with high specific gravity. Specific gravity of treated wood increased considerably with effectiveness of polymer loading. The increases in specific gravity were 5.36 to 13.16 percent. The highest was Jolcham-Namoo, the lowest Karae-Namoo. On the dimensional stability, a 40 percent of effectiveness of polymer loading was just as effective as 60 percent in reduction in water absorptivity (RWA), antishrinkage efficiency (ASE) and antiswelling efficiency (AE), and from over 60 percent they increased more rapidly. Also species response varied considerably. ASE was 30.12 to 69.97 percent tangentially and 27.86 to 56.37 percent radially, AE 34.06 to 73.76 percent tangentially and 30.11 to 70.12 percent radially, and RWA 42.31 to 65.32 percent. No differences in volume swelling among the 6 species were observed. Its values were ranged from 14.98 to 19.55 percent and also increased with PEG retentions. On the mechanical properties, the strengths very much decreased with PEG-400 loadings as shown in Figure 12; that were 11.41 to 22.90 percent in compression, 21.61 to 34.35 percent in bending and 22.83 to 36.83 percent in tensile strength. PEG retention in cell wall was less than 1 percent and the most of PEG were immersed in cell lumen. Except for Korae-Namoo, effectivenesses of polymer loading were as much high as 61.58 to 75.02 percent. This is believed to be due to the effect of PEG-400 on excellant dimensional stability of treated woods.

  • In this study, styrene butadiene rubber(SBR)/organoclay nanocomposites were manufactured using the latex method with 3-aminopropyltriethoxysilane(APTES) as a modifier. The X-ray diffraction(XRD), transmission electron microscopy(TEM) images, Fourier transform infrared(FTIR) spectroscopy, swelling ratio and mechanical properties were measured in order to study the interaction between filler and rubber according to the mixing temperature in the internal mixer. In the case of SBR/APTES-MMT compounds, the dispersion of the silicates within the rubber matrix was enhanced, and thereby, the mechanical properties were improved. The characteristic bands of Si-O-C in APTES disappeared after hydrolysis reaction in the MMT-suspension solution and the peak of hydroxyl group was increased. Therefore the formation of chemical bonds between the hydroxyl group generated from APTES on the silicate surface and the ethoxy group of bis(triethoxysilylpropyl) tetrasulfide(TESPT) was possible. Consequently, the 300% modulus of SBR/APTES-MMT compounds was further improved in the case of using TESPT as a coupling agent. However, the silanization reaction between APTES and TESPT was not affected significantly according to the increase of mixing temperature in the internal mixer.

Antiplatelet Activity of [5-(2-Methoxy-5-chlorophenyl)furan-2-ylcarbonyl]guanidine (KR-32570), a Novel Sodium/hydrogen Exchanger-1 and Its Mechanism of Action

  • The anti platelet effects of a novel guanidine derivative, KR-32570 ([5-(2-methoxy-5-chlorophenyl) furan-2-ylcarbonyl]guanidine), were investigated with an emphasis on the mechanisms underlying its inhibition of collagen-induced platelet aggregation. KR-32570 significantly inhibited the aggregation of washed rabbit platelets induced by collagen $(10{\mu}g/mL)$, thrombin (0.05 U/mL), arachidonic acid $(100{\mu}M)$, a thromboxane (TX) $A_2$ mimetic agent U46619 (9,11-dideoxy-9,11-methanoepoxy-prostaglandin $F_2,\;1{\mu}M$) and a $Ca^{2+}$ ATPase inhibitor thapsigargin $(0.5{\mu}M)$ ($IC_{50}$ values: $13.8{\pm}1.8,\;26.3{\pm}1.2,\;8.5{\pm}0.9,\;4.3{\pm}1.7\;and\;49.8{\pm}1.4{\mu}M$, respectively). KR-32570 inhibited the collagen-induced liberation of $[^3H]$arachidonic acid from the platelets in a concentration dependent manner with complete inhibition being observed at $50{\mu}M$. The $TXA_2$ synthase assay showed that KR-32570 also inhibited the conversion of the substrate $PGH_2$ to $TXB_2$ at all concentrations. Furthermore, KR-32570 significantly inhibited the $[Ca^{2+}]_i$ mobilization induced by collagen at $50{\mu}M$, which is the concentration that completely inhibits platelet aggregation. KR-32570 also decreased the level of collagen $(10{\mu}g/mL)$induced secretion of serotonin from the dense-granule contents of platelets, and inhibited the NHE-1-mediated rabbit platelet swelling induced by intracellular acidification. These results suggest that the antiplatelet activity of KR-32570 against collagen-induced platelet aggregation is mediated mainly by inhibiting the release of arachidonic acid, $TXA_2$ synthase, the mobilization of cytosolic $Ca^{2+}$ and NHE-1.

A Pilot Test for the Utilization of Road Subsoil of the Tertiary Mudstone in Pohang Basin (포항분지 제3기 이암의 도로 노체 활용을 위한 현장시험)

  • The purpose of this study is to present the possibility a utilization of the tertiary mudstone in Pohang as road subsoil material through pilot experiments on the road embankment structure. This mudstone is an unconsolidated rock that is distributed in the soft rock sedimentary layer, the tertiary layer of the Cenozoic, and causes physical problems such as slaking, swelling, and reduced shear strength and chemical problem like acid drainage. In order to solve various complex problems, an laboratory mixing test was conducted, and the optimal mixing conditions of the tertiary mudstone (90%), composite slag (steel making 70%, blast furnace 30%), and neutralization and coating agent treatment were derived. In order to prove its utilization, a real-scale road embankment structure was constructed and tests were conducted for each section. The pre-processing section is stable due to the design of optimal mixing conditions, while in post-processing section, natural weathering proceeded rapidly, and structural problems were concerned. Since the effect of neutralizing and coating agents was confirmed in temporary-staking section, the neutralizing and coating agents can be applied during the temporary storage period.

May-Thurner Syndrome with Deep Vein Trombosis after Total Knee Arthroplasty (인공 슬관절 전치환술 후 발생한 메이-터너 증후군 및 심부정맥 혈전증)

  • May-Thurner syndrome (MTS), also known as iliac vein compression syndrome, is a condition, in which compression of the common venous outflow tract of the left lower extremity can cause discomfort, swelling, pain or blood clots in the iliofemoral veins. The problem is due to left common iliac vein compression by the overlying right common iliac artery. This paper describes the case of a 75-year-old female with MTS after performing right total knee replacement arthroplasty. The authors diagnosed MTS through intravenous angiography and angiographic computed tomography on swelling and pain of the left lower extremities after performing right total knee replacement arthroplasty. The thrombus was removed using a thrombolytic agent and mechanical thrombectomy, and an intravenous stent then inserted after angioplasty. No case of MTS after performing total knee replacement arthroplasty has been reported in Korea. Therefore, this case is reported along with review of the relevant literature.

Sustained release of alginate hydrogel containing antimicrobial peptide Chol-37(F34-R) in vitro and its effect on wound healing in murine model of Pseudomonas aeruginosa infection

  • Background: Antibiotic resistance is a significant public health concern around the globe. Antimicrobial peptides exhibit broad-spectrum and efficient antibacterial activity with an added advantage of low drug resistance. The higher water content and 3D network structure of the hydrogels are beneficial for maintaining antimicrobial peptide activity and help to prevent degradation. The antimicrobial peptide released from hydrogels also hasten the local wound healing by promoting epithelial tissue regeneration and granulation tissue formation. Objective: This study aimed at developing sodium alginate based hydrogel loaded with a novel antimicrobial peptide Chol-37(F34-R) and to investigate the characteristics in vitro and in vivo as an alternative antibacterial wound dressing to treat infectious wounds. Methods: Hydrogels were developed and optimized by varying the concentrations of crosslinkers and subjected to various characterization tests like cross-sectional morphology, swelling index, percent water contents, water retention ratio, drug release and antibacterial activity in vitro, and Pseudomonas aeruginosa infected wound mice model in vivo. Results: The results indicated that the hydrogel C proved superior in terms of cross-sectional morphology having uniformly sized interconnected pores, a good swelling index, with the capacity to retain a higher quantity of water. Furthermore, the optimized hydrogel has been found to exert a significant antimicrobial activity against bacteria and was also found to prevent bacterial infiltration into the wound site due to forming an impermeable barrier between the wound bed and external environment. The optimized hydrogel was found to significantly hasten skin regeneration in animal models when compared to other treatments in addition to strong inhibitory effect on the release of pro-inflammatory cytokines (interleukin-1β and tumor necrosis factor-α). Conclusions: Our results suggest that sodium alginate -based hydrogels loaded with Chol-37(F34-R) hold the potential to be used as an alternative to conventional antibiotics in treating infectious skin wounds.

Characterization of Lactide/Hyaluronic Acid Polymer Cross-Linked by 1,3-Butadiene Diepoxide (1,3-Butadiene Diepoxide에 의해 가교된 락타이드/히아루론산 고분자의 특성)

  • The hyaluronic acid (HA) polymers cross-linked with lactide (LA) using the crosslinking agent, 1,3-butadiene diepoxide (BD), were prepared in order to develop a biomedical material for tissue engineering. The degree of lactide and BD reaction of the crosslinked polymer was determined by the analysis of nuclear magnetic resonance spectroscopy. Both degree of reaction and swelling ratio increased with BD concentration or LA/HA mole ratio. Tensile modulus decreased with increasing BD concentration or decreasing LA/HA mole ratio. Degradation was shown to be progressed at two different stages and became slow with increasing BD concentration. It was shown that the first stage degradation was mainly due to the decomposition of ester linkage in the crosslinked structure. The cell growth inhibition increased with BD concentration. Although cytotoxicity was slightly observed in the high BD concentration, the value was very low (below 6%) enough not to affect the cell growth.

The Protective Effect of Melatonin Administration against Adria-mycin-induced Cardiotoxicity in Rats

  • Adriamycin is a commonly used chemotherapeutic agent for cancer, including acute leukemia, lymphoma, and a number of solid human tumors. However, recent studies have recognized severe cardiotoxicity after an acute dose, which are likely the result of generation of free radicals and lipid peroxidation. Therefore, the clinical uses of adriamycin have been limited. Melatonin, the pineal gland hormone known for its ability to modulate circardian rhythm, has recently been studied in its several functions, including cancer growth inhibition, stimulating the immune system, and acting as an antioxidant and radical scavenging effects. In the present study, we evaluated the effect of melatonin administration on adriamycin-induced cardiotoxicity in rat. Heart slices were prepared using a Stadie-Riggs microtome for the measurement of malondialdehyde (MDA) content used as an index of lipid peroxidation and lactate dehydrogenase (LDH) release as an indicator of lethal cell injury. Serious adriamycin-induced lethality was observed in rat by a single intraperitoneal injection in a dose-dependent manner. A single injection of adriamycin (25 mg/kg, i.p.) induced a lethality rate of 86%, with melatonin (10 mg/kg s.c. for 6 days) treatment reducing the adriamycin-induced lethality rate to 20%. The severe body weight loss caused by adriamycin was also significantly attenuated by melatonin treatment. Treatment of melatonin marked reduced adriamycin-induced the levels of MDA formation and LDH release. A cell damage indicated by the loss of myofibrils, swelling of the mitochondria as well as cytoplasmic vacuolization was seen in adriamycin-treated group. Melatonin attenuated the adriamycin-induced structural alterations. These data provide evidence that melatonin prevents adriamycin-induced cardiotoxicity and might serve as a combination with adriamycin to limit free radical-mediated cardiotoxicity.

Urethroplasty of Urethral Defect with Longitudinal Tubed Flap of Scrotal Skin after Fournier's Gangrene (Necrotizing Fascitis) (회음부 괴사성 근막염후 발생한 요도 결손의 치료를 위한 음낭피부 종축관피판을 이용한 요도성형술)

  • Purpose: The management of urethral defect represents one of the most challenging clinical problems in uroplastic surgery. Especially for defect after Fournier's Gangrene, optimal management is still a hard problem. During extensive urethral reconstruction, to overcome the poor vascularity due to periurethral scarred tissue and limitation of the choice of local flap, we report our experience with one-stage reconstruction of urethral defect using a longitudinal tubed flap of scrotal skin. Methods: A 72-year-old man with several years of diabetes mellitus history visited for swelling and pain of scrotal area. After diagnosis of Fournier's Gangrene, radical debridement was performed and 6 cm of urethral defect on border of penile-scrotal ventral area was made. Rectangular scrotal skin flap ($6{\times}2.5\;cm$) based on external spermatic fascia was elevated and tubed longitudinally. After transfer the flap to the defect area, end-to-end anastomosis was performed bilaterally. Results: 4 weeks after the operation, the patient started voiding him-self and urethrography showed good fluence of contrast agent. Long term evaluation reveals stable performance characteristics without any complications. Conclusion: We suggest a one-stage reconstruction of extensive urethral defect using a longitudinal tubed flap of scrotal skin. Advantages of this procedures are simple, one-stage reconstruction with the reliable scrotal skin flap based on external spermatic fascial vasculature, and no donor morbidity.

Effect of the Structure of the Smallest Poresize Layer on the Permeability of PES Microfiltration Membranes (최소 기공층 구조가 PES계 정밀여과막 투과 성능에 미치는 영향)

  • PES (polyethersulfone) membranes with highly enhanced their asymmetry were prepared by phase inversion process. The membranes were prepared by using PES/DMF (N,N-dimethylformamide)/TSA (p-toluenesulfonic acid)/PVP (polyvinylpyrrolidone) casting solution and water coagulant. The pre-coagulation of membrane surface which was induced by an addition of TSA as a demixing agent and PVP as a swelling polymer in the PES solution and humid exposure time, played a crucial role in determining morphological properties and the PWP (pure water permeation) performance. The PES solution was coated on polyester film under condition of 80% humidity for a while ($72{\sim}144$ sec) before immersing in a coagulation bath. The characterization of membranes was carried out by a capillary flow porometer, a FE-SEM and a permeation test apparatus. As the thickness of the smallest pore size layer (SPL) decreased, the asymmetry of membrane increased under conditions of 20 wt% of TSA and 10 wt% of PVP in 11 wt% of PES solution during longer humid contact time. As a result, the membranes showed a remarkable increase of PWP.
