감미단백질 관련 브라제인, 타우마틴 및 미라쿨린 유전자를 이용한 형질전환 상추 육성 및 발현분석 (Stable expression and characterization of brazzein, thaumatin and miraculin genes related to sweet protein in transgenic lettuce)

  • 감미료(sweetener)는 단맛을 느끼게 하는 첨가물 중 하나로 인공감미료와 설탕이 대표적이며, 단맛의 특성을 지닌 감미 단백질도 잘 알려져 있다. 본 연구는 천연 감미 단백질, brazzein, thaumatin 및 miraculin의 안정적인 생산을 위해 Agrobacterium방법으로 상추 세포를 형질 전환시켰다. 감미 단백질을 코딩하는 합성 유전자들은 상시적으로 발현하는 프로모터의 제어 하에 상추에 형질전환 하였다. 형질전환한 잎을 이용하여 RT-PCR 및 Western blot 분석을 실시한 결과 감미 단백질이 안정적으로 발현하는 것을 확인하였다. 형질전환 상추에서 발현한 단백질은 단맛을 유발하는 단백질 활성을 가지고 있었다. 이러한 결과는 유전자 재조합 감미 단백질들은 형질전환한 상추에 잘 발현된 것을 보여 주며, 이들 생산 시스템은 식물로부터 감미단백질 생산을 위한 좋은 대안이 될 수 있을 것으로 생각된다.

Developing a Descriptive Analysis Procedure for Korean Pumpkin Gruel (Hobakjuk)

  • The objective of this study was to develop a reliable and reproducible descriptive analysis procedure for Korean style sweet pumpkin gruel (Hobakjuk). The sensory attributes of the sweet pumpkin gruel were developed and defined, the sample preparation method was standardized, and the sensory evaluation procedure for a sample was established. Seven types of sweet pumpkin gruel (five ready-to-eat type vs. two ready-to-heat type) were selected to be analyzed. Panel training and descriptive analysis were carried out with these 7 samples. A total of 12 sensory attributes (2 aroma/odor, 5 taste/flavor, 4 texture/mouthfeel, and 1 aftertaste attributes) were developed to describe the sensory characteristics of the sweet pumpkin gruel. The definition and reference standards for each sensory attribute were determined to clearly understand each attribute. In the main experiment, trained panelists evaluated the sensory characteristics of the 7 gruel samples based on a fifteen-point intensity scale using the developed attributes. The results were statistically analyzed by analysis of variance, principal component analysis, and cluster analysis. The results showed that the 7 sweet pumpkin gruel samples significantly differed in their intensities of all attributes except for sweet pumpkin aroma and viscosity. The ready-to-eat style samples were distinctly characterized by their sweet pumpkin aroma and flavor, whereas the ready-to-heat style samples were markedly characterized by their low intensity of gelatinized starch and pumpkin flavor retention.

동결 건조 자색고구마 가루를 첨가한 국수의 품질특성 및 항산화성 (Quality Characteristics of Wet Noodles Added with Freeze-dried Purple Sweet Potato Powder)

  • 본 연구는 동결로 건조한 자색고구마 가루를 첨가하여 국수를 제조하고 안토시아닌 함량과 항산화 활성도 및 품질특성을 검토하여 국수 제조 시 최적의 비율을 연구하고자 하였다. 색도는 자색고구마 분말 첨가량이 증가 할수록 명도(L)와 황색도(b)는 대조군에 비해 점차 감소하는 경향이 나타났고 적색도(a)는 첨가량이 증가 할수록 확연히 증가하였다. 안토시아닌 함량은 99.62, 항산화 활성도는 94.60로 비교적 우수한 것으로 나타났다. 조리한 국수의 기계적 성질은 견고성, 응집성 및 씹힘성은 첨가량이 증가 할수록 감소하는 경향을 보였으며 탄력성과 점착성에서 유의적 차이를 확인할 수 있었다. 관능적 품질 평가에서는 색, 향미, 맛과 씹힘성 및 전반적 기호도에서 6% 첨가군이 가장 높게 평가되었으며 안토시아닌 함량과 DPPH 라디컬 소거 활성에서 우수한 결과로 나타나 품질 특성과 관능검사에서 긍정적으로 평가된 6% 첨가군이 국수 제조 시 가장 알맞은 첨가 비율로 사료된다.

반응표면분석법을 이용한 연자육 분말 첨가 찹쌀머핀의 품질 특성 및 최적화 (Quality Characteristics and Optimal Conditions for Sweet Rice Muffin Lotus (Nelumbo nucifera Gaertn) Seed Powder, Applying the Response Surface Method)

  • This study was undertaken to optimize preparation of sweet rice muffins using lotus (Nelumbo nucifera Gaertn) seed powder. The experimental conditions used in the preparation included lotus seed powder (X1) and soybean oil (X2). The muffin formulation was optimized using rheology. Lightness (P<0.01) displayed a quadratic model pattern, whereas yellowness (P<0.01) and redness (P<0.05) was presented a linear model. Texture properties significantly differed in cohesiveness (P<0.05). Evaluating the sensory characteristics of muffins, except for flavor and texture, the remaining properties of color (P<0.01), taste, overall quality, and appearance (P<0.05) were found to be significantly different. The overall quality was also affected by the proportion of lotus seed powder, rather than the amount of soybean oil. The optimal ratio for palatability of muffins was determined to be 47.80 g lotus seed powder and 95.28 g soybean oil. Taken together, our results indicate that sweet rice muffins prepared using lotus seed powder are sufficiently competitive in terms of function and quality.

Steamed Bread Quality as Influenced by the Incorporation of Purple Sweet Potato Powders

  • As an attempt to develop a value-added food product, purple sweet potato powder was added in a model system of steamed bread as a healthy food ingredient and physicochemical properties such as moisture content, specific volume, spread ratio, color, texture as well as consumer preferences on the attributes such as uniformity, color, flavor, elasticity, chewiness, taste, and overall preference were evaluated. Moisture content ranged from 44.16 to 44.55% (wet basis) and appeared independent on the level of purple sweet potato (PSP) powder incorporation. As a result of the addition of PSP powder, the specific volume of steamed bread decreased from 3.22 to 2.55 mL/g, and value of 4.5% sample was significantly lower than other samples (p<0.05). On the other hand, spread ratio ranged from 2.01 to 2.53, and appeared to decrease as the PSP powder concentration increased (p<0.05), indicating a significant improvement. Lightness $(L^*)$ decreased significantly as the PSP powder content increased (p<0.05) for both dough and skin of the steamed bread. In addition, an increasing trend in redness ($a^*$-value) and a decreasing trend in yellowness ($b^*$-value) were noticed. Firmness increased significantly with the addition of PSP powder regardless of concentration (p <0.05); however, firmness was not significantly different among samples containing 1.5-4.5% PSP powder (p>0.05). Consumer acceptance test indicated that incorporation of 3% PSP powder in the formulation of steamed breads would be recommended.

Molecular Basis of Hexanoic Acid Taste in Drosophila melanogaster

  • Animals generally prefer nutrients and avoid toxic and harmful chemicals. Recent behavioral and physiological studies have identified that sweet-sensing gustatory receptor neurons (GRNs) in Drosophila melanogaster mediate appetitive behaviors toward fatty acids. Sweet-sensing GRN activation requires the function of the ionotropic receptors IR25a, IR56d, and IR76b, as well as the gustatory receptor GR64e. However, we reveal that hexanoic acid (HA) is toxic rather than nutritious to D. melanogaster. HA is one of the major components of the fruit Morinda citrifolia (noni). Thus, we analyzed the gustatory responses to one of major noni fatty acids, HA, via electrophysiology and proboscis extension response (PER) assay. Electrophysiological tests show this is reminiscent of arginine-mediated neuronal responses. Here, we determined that a low concentration of HA induced attraction, which was mediated by sweet-sensing GRNs, and a high concentration of HA induced aversion, which was mediated by bitter-sensing GRNs. We also demonstrated that a low concentration of HA elicits attraction mainly mediated by GR64d and IR56d expressed by sweet-sensing GRNs, but a high concentration of HA activates three gustatory receptors (GR32a, GR33a, and GR66a) expressed by bitter-sensing GRNs. The mechanism of sensing HA is biphasic in a dose dependent manner. Furthermore, HA inhibit sugar-mediated activation like other bitter compounds. Taken together, we discovered a binary HA-sensing mechanism that may be evolutionarily meaningful in the foraging niche of insects.

오미자 첨가량에 따른 Demi-Glace 소스의 일반성분과 유리당 함량 및 관능적 특성 (The Proximate Composition, Free Sugars Contents and Sensory Characteristics of Demi-glace Sauce according to the Varying Quantity of Omija Added)

  • This study was conducted to examine the sensory characteristics and the contents of proximate composition and the free sugars of the Demi-glace with varying quantity of Omija extracts added. First, the qualities of Demi-glace sauces with varying quantity of Omija extracts added are as follows: (1) Proximate composition: The moisture content was 66.10~73.50%, and crude ash content was 1.59~1.89%. As the Omija extracts added increased, moisture content increased, whereas crude ash content decreased. The crude protein content was 6.12~7.95%. Among them, the one with the 2% Omija added showed the highest level of the crude protein. (2) Free sugars contents: In terms of total sugar contents, the control showed 1.32% and Omija sauces showed 1.44~1.55%. Major free sugars analyzed in the order of fructose, oligosaccharide, glucose and sucrose. Second, the results of sensory evaluation of Demi-glace sauces with the different levels of Omija added are as follows: (1) The preference of Demi-glace sauces according to occupation: Students and Cooks liked the one with the 2% Omija added in terms of color, flavor, taste, texture and overall acceptability. Cooks showed higher preference than students. (2) The preference of 5 tastes: It was found that Demi-glace sauce with the 2% Omija added was the best. The one with the 2% Omija added was suitable in terms of sweet taste and bitter taste and the one with the 3% Omija added was desirable for hot taste and saline taste. In terms of sour taste, the one with 1% Omija added was regarded as the most desirable one. (3) The preference for authentic tastes for Demi-glace sauce: In terms of simple taste, females showed higher preference than males. Demi-glace sauce with the 2% Omija added was found to be the best for savory taste, flavor enhancer taste, simple taste, after taste and overall acceptability. The one with the 1 % Omija added was regarded as the best for soft taste.

자색고구마 분말과 자색 색소를 이용한 소시지의 아질산염 대체 효과 (Effect of Nitrite Substitution of Sausage with Addition of Purple Sweet Potato Powder and Purple Sweet Potato Pigment)

  • 자색고구마를 이용하여 아질산염에 대한 대체 가능성을 알아보고자 자색고구마 분말과 자색 색소를 혼합하여 첨가하였다. 자색고구마 분말의 함량은 소시지 식품유형을 고려하여 최대 10%까지 사용하였다. 아질산나트륨 0.15% 첨가구를 대조구(F1)로 하고, 자색 색소 0.2%를 첨가한 것을 F2, 자색 색소 0.2%와 자색고구마 분말 5%, 10%를 각각 첨가한 것을 F3, F4로 하였다. 아질산나트륨을 대신하여 첨가한 자색 색소 0.2%는 소시지의 적색도를 감소시키고 황색도를 증가시켜 아질산나트륨에 대한 대체 효과가 미흡하였다. 그러나 자색 색소 0.2%에 자색고구마 분말 5%를 추가함으로써 소시지의 적색도가 높아지고 황색도는 낮아졌다. 자색고구마 분말 함량 증가에 따라 적색도의 증가와 황색도의 감소는 유의적 비례관계를 나타내었다. 자색고구마 분말 5%와 자색 색소 0.2%의 첨가는 아질산나트륨 0.15%를 첨가한 소시지에서 나타나는 색도와 가장 유사해졌다. 자색고구마 분말은 첨가량이 증가함에 따라 칼슘, 칼륨, 마그네슘의 무기질이 증가하였고 경도, 검성, 씹힘성의 조직감이 떨어졌다. 관능평가 결과 자색고구마의 첨가가 소시지의 외관과 향에 영향을 미치지 않았고 자색고구마 분말 5% 이상 첨가에 대한 색의 유의차를 보이지 않았다. 그러나 자색고구마 분말 10%의 첨가는 소시지의 맛과 질감을 떨어뜨렸다. 전체 기호도는 질감(0.901)> 외관(0.895)> 색(0.877)> 맛(0.844)> 향(0.688)의 순서로 유의적으로 양의 상관관계가 있었고 전체 기호도와 색도의 상관성은 밝기에서만 유의적인 양의 상관성을 나타내었다. 적색도와 황색도는 서로에 대해 음의 상관성을 보였다. 자색고구마를 이용한 소시지의 아질산염 대체 효과는 자색고구마 분말 5%와 자색 색소 0.2%를 사용하는 것이 적절한 것으로 나타났다.

발효강황 첨가 머핀의 항산화적·감각적 품질 특성 (Antioxidant and Sensory Quality Characteristic of Muffin with Fermented Turmeric Powder)

  • The antioxidant, physicochemical, and sensory properties of muffins using fermented Curcuma longa L. were investigated. Sensory properties of the samples were examined, and the correlation between the physicochemical, antioxidant, and sensory properties was studied. The pH of the samples was pH 7.26-7.42, which was the optimal pH range for the air formation of the dough, color, or volume. The antioxidant activities of the sample group were significantly higher than the control (p<0.05). In the sensory intensities, the 1.0 and 1.5%-added sample groups showed a similar savory aroma, sweet aroma, savory taste, sweet taste, and texture to those of the control. Samples containing 1.0 and 1.5% showed similar overall acceptance values to the control. The muffin containing 1.0 or 1.5% fermented Curcuma longa L. powder was developed successfully, and it was concluded that the developed muffin with the fermented turmeric powder could be a highly valued product in the increasingly competitive muffin food industry.

여고생의 체형에 따른 식습관 및 식품기호에 관한 연구 - 인천지역 중심으로 - (A Study on the High School Girls′ Food Habit and Food Preference Relating to their Body Weight - in Inchon area -)

  • This study examines the high school girls in Inchon area dividing them into three groups according to their weight, the low-weight group, the normal weight group and the overweight group to find out their food habit and food preference. The result reveals that more students from the low-weight group have breakfast regularly than those from the other two groups, and more students from the low-weight and normal weight groups always have lunch. However, none of the students from those three groups skip their suppers. This study also shows that there are more students from the overweight group having snacks, and more students from the low-weight have the unbalanced diet problem than those from the other two groups. Next, this study examines their attitude toward breakfast. It seems that all the students from three different groups similarly regard breakfast as a very important meal. Additionally in deciding what to eat, the taste of food is considered as the most important factor for them all. In their preference of food taste, there seems to be a slight difference. The students from the low-weight and normal weight groups prefer hot food to sweet food, but those from the overweight group prefer sweet food to hot food. Also in food preference the difference among the groups is that the students from the low-weight and normal groups prefer favorite food and snacks. While those from the overweight groups prefer cereals and noodles.
