Synthesis of Sodium ${\alpha}$-Sulfo Fatty Acid Allyl Ester Oligomer and Didpersion Stability of Pigments in Aqueous Solution ((${\alpha}$-술폰 지방산 알릴에스테르 올리고머의 합성 및 안료 분산성)

  • Allylaliphatic carboxylate oligomers were prepared from polymerization giving allyl aliphatic carboxylates in the presence of potassium persulfate in methanol and the ${\alpha}-sulfonation$ of these allyl aliphatic carboxylates oligomers were carried by direct addition of dry sulfur trioxide. The dispersing performance of oligomer type anionic surfactants and SDS in the aqueous suspension of $Fe_{2}O_{3}$ and $Tio_{2}$ particles were evaluated by particle size distribution and ${\zeta}-potential$ measurement. As results, the particles of $Fe_{2}O_{3}$ and $Tio_{2}$ were flocculated by addition of small amount of oligomer type anionic surfactants and SDS, then the flocks redispersed by more addition oligomer type anionic surfactants and SDS. The dispersion and flocculation were observed in lower concentration range of oligomer type anionic surfactants than SDS.

Extraction and Characterization of Aloe Glucomannan: Assessing Its Flocculation Capability

  • Acemannan, a highly acetylated glucomannan, was extracted from fresh Aloe vera leaves by ethanol fractionation, resulting in a concentration increase of more than threefold. The presence of acemannan was confirmed using FTIR and 1H NMR analysis, revealing an average molecular weight of 780 kDa. The flocculating activity of the fractionated aloe gel polysaccharide was assessed through settling tests in a 1% (w/v) bentonite suspension. The results demonstrated that the aloe polysaccharide exhibited remarkable stability within a temperature range of 20~70 ℃. The maximal flocculation rate at different pH levels ranged from 93% to 97%, with an optimal dose for maximum flocculation rate between 0.25 mg/mL. Notably, the minimum dose required for flocculation was achieved at a pH of 3, attributed to the compression of electrostatic repulsion on the surface of bentonite particles. However, the flocs obtained under acidic conditions were less dense and compact, exhibiting lower sedimentation velocity compared to those formed under neutral and alkaline pH conditions. Additionally, the addition of salt showed a slight synergistic effect on flocculation, significantly enhancing the sedimentation velocity. This investigation highlights the potential of Aloe vera polysaccharide as a natural and edible flocculant, offering promising applications in various industries.

The Stability Assessment of an Aquifer in Pohang Yeongil Bay due to CO2 Injection (이산화탄소 주입에 따른 포항 영일만 대수층 안정성평가)

  • It is imperative to inject carbon dioxide($CO_2$) into an aquifer for alleviating the emission of $CO_2$. However, faults in the aquifer can be reactivated due to pressure increasement. Analyses of pressure change of the aquifer is necessary to prevent the fault reactivation. In this research, we assess the stability of an aquifer in Pohang Yeongil bay by investigating the pressure variation of faults EF1 and EF2. Two scenarios, which repeat $CO_2$ injection and suspension during two years, are simulated. Each scenario includes cases of injection rates of 20, 40, and 100 tons/day. In addition, we analyze planned and predicted injection rates for each case. In case of 20 tons/day, the maximum pressure of faults is 65% of the reactivation pressure. Even if daily injection rates are increased to 40 and 100 tons/day, the maximum pressures are 71% and 80% of the reactivation pressures, respectively. For 20 and 40 tons/day cases, planned injection rates almost accord with predicted injection rates during whole simulation period. On the other hand, predicted injection rates are smaller than planned injection rates for the 100 tons/day case due to bottom-hole pressure limit of the injection well.

Generation of single stranded DNA with selective affinity to bovine spermatozoa

  • Objective: This study was conducted to generate single stranded DNA oligonucleotides with selective affinity to bovine spermatozoa, assess its binding potential and explore its potential utility in trapping spermatozoa from suspensions. Methods: A combinatorial library of 94 mer long oligonucleotide was used for systematic evolution of ligands by exponential enrichment (SELEX) with bovine spermatozoa. The amplicons from sixth and seventh rounds of SELEX were sequenced, and the reads were clustered employing cluster database at high identity with tolerance (CD-HIT) and FASTAptamer. The enriched nucleotides were predicted for secondary structures by Mfold, motifs by Multiple Em for Motif Elicitation and 5' labelled with biotin/6-FAM to determine the binding potential and binding pattern. Results: We generated 14.1 and 17.7 million reads from sixth and seventh rounds of SELEX respectively to bovine spermatozoa. The CD-HIT clustered 78,098 and 21,196 reads in the top ten clusters and FASTAptamer identified 2,195 and 4,405 unique sequences in the top three clusters from the sixth and seventh rounds, respectively. The identified oligonucleotides formed secondary structures with delta G values between -1.17 to -26.18 kcal/mol indicating varied stability. Confocal imaging with the oligonucleotides from the seventh round revealed different patterns of binding to bovine spermatozoa (fluorescence of the whole head, spot of fluorescence in head and mid- piece and tail). Use of a 5'-biotin tagged oligonucleotide from the sixth round at 100 pmol with 4×106 spermatozoa could trap almost 80% from the suspension. Conclusion: The binding patterns and ability of the identified oligonucleotides confirms successful optimization of the SELEX process and generation of aptamers to bovine spermatozoa. These oligonucleotides provide a quick approach for selective capture of spermatozoa from complex samples. Future SELEX rounds with X- or Y- enriched sperm suspension will be used to generate oligonucleotides that bind to spermatozoa of a specific sex type.

Determination of the Accurate Effective Length for Buckling Design of Cable-Supported Bridges (케이블지지교량의 좌굴설계를 위한 유효좌굴길이 산정)

  • In order to obtain the effective length factor of beam-column members of plane frames, this paper extensively used an alignment chart approach, based on the nomograph given in LRFD-AISC specification commentaries. However, it should be noted that various simplifications and assumptions were introduced in constructing the alignment chart. To overcome the practical limitations of the alignment chart, this paper proposes a simple but accurate procedure that determined the effective buckling length for stability design of main members of cable-supported bridges. This method requires the full system buckling analysis. The numerical examples showing the suitability of the present scheme are discussed and some conclusions are drawn.

  • For the high center of gravity vehicles the roll stiffness of their suspensions is arranged to be very high because such vehicles are in some danger of tipping over in cornering. In some cases, the effective roll stiffness is determined significantly by the compliance of the tires because of the very stiff anti-roll members incorporated in the suspension. In such cases, it is clear that the shock absorbers which may be effective in damping heave oscillations have little effect on roll oscillations. Therefore, wind gusts and roadway unevenness may cause large swaying oscillations. In this paper, to improve the stability for the high center of gravity vehicles a control scheme to augment the damping of the roll mode is proposed. As the feedback signals needed to provide damping of the roll motion, the front or rear steer angles or both are chosen because they are very related to roll motion. The scheme is effective from moderate to high speeds and stabilizes the roll mode without introducing disturbance moments from roadway unevenness as shock absorbers do. The validity on the proposed method is verified through the computer simulation.

  • Regeneration of protoplast was effective by preincubating spore suspension containing 30$\mu g$/ml of 2-DG for 4 hours, and CBE medium containing casamino acid, bovine serum albumin, ergosterol and myoinositol was found to be more efficient than any other regeneration medium tested in this experiment. The regeneration frequency was about 30%. Optimal conditions for conidial protoplast fusion were obtained by treatment of protoplasts with 10 mM $CaCl_2$ and 30% polyethylene glycol 4000 (pH 7.5) as fusogenic agent at $37^{\circ}C$ for 10 minutes. The fusion frequency was $8.2\times 10^{-4}$. The higher productivity of enzyme of fusant FWN-56 was achived: 2.3-fold for CMCase, 1.5-fold for avicelase, 1.8-fold for $\beta$-glucosidase and 2.5-fold for xylanase compared to that obtained in two parental strains. The genetic stability of fusant after maintenance on minimal medium for more than 4 weeks was high because segregant rate was below 1%. The conidial DNA content of fusant was 1.4-1.6 times higher than that of the parental strains, The nucleus size of fusants were also higher than that of each parental strains.

  • The dispersing behaviors of oligomer-type anionic surfactants, cooligomers of diethyl maleate(CmD-Na) or maleic anhydride (CmM-Na) and $C_4{\sim}C_{16}$ alkyl vinylether, which have a different alkyl chain length of the hydrophobic group or degree of polymerization, were studied on the aqueous suspension of ${\alpha}-$ and ${\beta}-$ copper phthalocyanine and carbon black particles. In case of the side alkyl chain length of $C_4{\sim}C_{10}$ of CmD-Na, the dispersing actions were good in the concentration range of 0.01 to 0.1%. Especially, side alkyl groups played an important role in the orientation adsorption on the surface of pigment particles, and oligomers having smaller degrees of polymerization were more effective in the dispersing action, but did not affect the dispersability of carbon black.

Various Types and Manufacturing Techniques of Nano and Micro Capsules for Nanofood

  • Nano and micro capsulation (NM capsulation) involve the incorporation for nanofood materials, enzymes, cells or other materials in small capsules. Since Kim D. M. (2001) showed that a new type of food called firstly the name of nanofood, which means nanotechnology for food, and the encapsulated materials can be protected from moisture, heat or other extreme conditions, thus enhancing their stability and maintaining viability applications for this nanofood technique have increased in the food. NM capsules for nanofood is also utilized to mask odours or tastes. Various techniques are employed to form the capsules, including spray drying, spray chilling or spray cooling, extrusion coating, fluidized bed coating, liposome entrapment, coacervation, inclusion complexation, centrifugal extrusion and rotational suspension separation. Each of these techniques is discussed in this review. A wide variety of nanofood is NM capsulated - flavouring agents, acids, bases, artificial sweeteners, colourants, preservatives, leavening agents, antioxidants, agents with undesirable flavours, odours and nutrients, among others. The use of NM capsulation for sweeteners such as aspartame and flavors in chewing gum is well known. Fats, starches, dextrins, alginates, protein and lipid materials can be employed as encapsulating materials. Various methods exist to release the ingredients from the capsules. Release can be site-specific, stage-specific or signaled by changes in pH, temperature, irradiation or osmotic shock. NM capsulation for the nanofood, the most common method is by solvent-activated release. The addition of water to dry beverages or cake mixes is an example. Liposomes have been applied in cheese-making, and its use in the preparation of nanofood emulsions such as spreads, margarine and mayonnaise is a developing area. Most recent developments include the NM capsulation for nanofood in the areas of controlled release, carrier materials, preparation methods and sweetener immobilization. New markets are being developed and current research is underway to reduce the high production costs and lack of food-grade materials.

  • An improved antibody-coated sensor system based on quartz crystal microbalance was developed for the detection of Salmonella spp. An antibody against Salmonella common structural antigen was immobilized onto one gold electrode of the piezoelectric quartz crystal surface by various immobilization procedures. The best results in sensitivity and stability were obtained with the thin layers of protein A and 3,3'-dithiopropionimidate.2HCI(DTBP), a homobifunctional thiol-cleavable crosslinker. After the addition of a S. typhimurium suspension into a reaction cell with 0.1 M sodium phosphate buffer, pH 7.2, the resonant frequency owing to S. typhimurium adsorption decreased conspicuously. The antibody-immobilized crystals prepared by the gold-protein A complex formation and DTBP thiolation showed the frequency shifts of 80 and 283 Hz, respectively. The time required for maximum frequency shift was about 30~60 min. The antibody-coated crystal could be reused for 6~8 consecutive assays.
