PLGA/HA composite scaffold fabricated by GF/PL method showed enhanced mechanical property, hydrophilicity and osteoconductivity compared with the SC/PL scaffolds, and this enhancement was most likely due to a higher extent of exposure of HA particles to the scaffold surface. The biodegradable polymer/bioceramic composite scaffolds fabricated by the GF/PL method could enhance bone regeneration efficacy for the treatment of bone defects compared with conventional composite scaffolds.
The purpose of this study is to evaluate histologically the resorption and tissue response of various resorbable collagen membranes used for guided tissue regeneration and guided bone regeneration, using a subcutaneous model on the dorsal surface of the rat. In this study, 10 Sprague-Dawley male rats (mean BW 150gm) were used and the commercially available materials included acellular dermal matrix allograft, porcine collagen membrane, freeze-dried bovine dura mater. Animals were sacrificed at 2,6 and 8 weeks after implantation of various resorbable collagen membranes. Specimens were prepared with Hematoxylin-Eosin stain for light microscopic evaluation. The results of this study were as follows: 1. Resorption : Inner portion of porcine collagen membrane was resorbed a lot at 6 weeks, but its function was being kept for infiltration of another tissues were not observed. Freeze-dried bovine dura mater and acellular dermal allograft were rarely resorbed and kept their structure of outer portion for 8 weeks. 2. Inflammatory reactions : Inflammatory reaction was so mild and foreign body reaction didn't happen in all of resorbable collagen membranes, which showed their biocompatibility. 3. In all of resorbable collagen membranes, multinuclcated giant cells by foreign body reactions were not observed. Barrier membranes have to maintain their function for 4-6 weeks in guided tissue regeneration and at least 8 weeks in guided bone regeneration. According to present study, we can find all of the resorbable collagen membranes kept their function and structure for 8 weeks and were rarely resorbed. Foreign body reaction didn't happen and inflammatory reaction was so mild histologically. Therefore, all of collagen membranes used in this experiment were considered proper resorbable membranes for guided tissue regeneration and guided bone regeneration.
Indica 벼의 원형질체들로 부터 효과적인 식물체 재분화 방법을 개발하였다. 이 방법은 배형성 진탕 세포 배양체로 부터 나출된 원형질체들을 feeder cell들이 agarose에 embedded된 배지 표면에 놓인 filter membrane 위에 배양하는 것이다. Feeder cell로서 Lolium multiflorum 세포배양체를 사용했을 때가 Oryza ridleyi를 사용했을 때보다 효과적이었고, 원형질체 평탄효율은 세포 배양체 age에 따라 달랐지만 최고로 0.68% 까지 증가되었다. Carbohydrate source로서 maltose를 사용하거나 maltose와 sucrose를 1:1로 조합했을 때가 sucrose 단독으로 사용했을 때보다 식물체 재분화율이 증가되었고, 고농도의 agarose를 이용하여 원형질체로부터 유도된 캘러스를 dehydration시켰을 때 또한 재분화율이 괄목하게 증가되었다. 식물체 재분화율은 control 캘러스로부터 3.1∼30.6% 였지만 dehydration처리한 캘러스로부터는 30.7∼70.7%까지 증가되었다. 원형질체로 부터 유도된 식물체들은 형태적으로 정상이며 개화했다.
Purpose: Recently, interest in chitosan has increased due to its excellent biological properties such as biocompatibility, antibacterial effect, and rapid healing capacity. On the other hand, hydroxyapatite is used as a bone substitute in the fields of orthopedics and dentistry. The hydroxyapatite-chitosan (HA-CS) complex containing hydroxyapatite nanoparticles was developed for synergy of both biomaterials. The objective of this study was to evaluate the effect of hydroxyapatite (HA)-chitosan (CS) membrane on bone regeneration in the rat calvarial defect. Methods: Eight-millimeter critical-sized calvarial defects were created in 70 male Sprague-Dawley rats. The animals were divided into 7 groups of 10 animals and received either 1) chitosan (CS) 100% membrane, 2) hydroxyapatite (HA) 30%/CS 70% membrane, 3) HA 30%/CS 70%, pressed membrane, 4) HA 40%/CS 60% membrane, 5) HA 50%/CS 50% membrane, 6) HA 50%/CS 50%, pressed membrane, or 7) a sham . surgery control. The amount of newly formed bone from the surface of the rat calvarial defects was measured using histomorphometry, following 2- or 8- week healing intervals. Results: Surgical implantation of the HA - CS membrane resulted in enhanced local bone formation at both 2 and 8 weeks compared to the control group. The HA - CS membrane would be significantly more effective than the chitosan membrane in early bone formation. Conclusions: Concerning the advantages of biomaterials, the HA-CS membrane would be an effective biomaterial for regeneration of periodontal bone. Further studies will be required to improve the mechanical properties to develop a more rigid scaffold for the HA-CS membrane.
A variety of titanium (Ti) and its alloys are used in the clinical procedures of bone regeneration for periodontal and dental implant therapies. This study was performed to determine the effect of different surface dental implant materials on biologic responses of a MG-63 human osteoblast-like cell line. MG-63 cells were cultured on Ti coated with hydroxyapatite (HA), calcium metaphosphate (CMP), anodized (A), which compared with non-coated Ti (control). The appearances of surface of dental implant materials and the morphology of these cells were assessed by scanning electron microscopy (SEM). The gene expression profiles of MG-63 cells cultured on Ti were examined by human cDNA microarray (1,152 elements). The expression of several genes was up- and down-regulated by different surfaces of dental implant materials. Interesting, the genes correlated with cellular adhesion and extra cellular matrix (ECM) formation were enhanced, in accordance surface morphology of the dental implant materials used.
The chemical waste treatment, APEG (alkali/polyethylene glycol) process has been shown to be effective for the dechlorination of PCBs in transformer oil. Considerable amount of PCBs, however, still remains in the waste exceeding the 25-50 ppm limit set by regulatory agency. A new thermal regeneration technology has been developed in our laboratory for disposal of hazardous organic wastes. Due to the limited oxidation of carbon surface through the reverse movement of flame front to oxidant flow, this technology was termed counterflow oxidative system (COS). Specially, the oxidant flow in the COS process is a principal parameter which determines the optimum conditions regarding acceptable removal and destruction efficiency of adsorbed organic wastes at minimal carbon loss. The COS process, under optimum conditions, was found to be very effective and the removal and destruction efficiency of 99.99% or better was obtained for residual PCBs in the waste while bulk (≥90%) of carbon was recovered. Any toxic formation of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzo furans (PCDFs) were not detected in the regenerated carbon and impinger traps. The results of surface area measurement showed that the adsorptive property of regenerated carbon is mostly reclaimed during the COS process.
The aim of this study was to achieve the healing of peri-implantitis defects and the hard tissue regeneration using the augmentation of a xenograft on defect site. Two patients were treated with the surgical approach. With a full muco-periosteal flap elevation, the implant surfaces were exposed and taken the debridement of granulation tissue around the abutment. Each surface of the abutments was prepared with the air-abrasive device (PerioFlow$^{(R)}$) for decontamination. Bovine-derived bone mineral (Bio-Oss collagen$^{(R)}$) was then used to fill the defects, and no membrane was placed on the grafting site. Radiographs and clinical photo was taken to compare from baseline status. Within the limits of the present case, this case shows the significance of the surgical treatment of peri-implantitis. And this also verifies the stability of bovine-derived bone mineral and effectiveness of Air-abrasive device (PerioFlow$^{(R)}$).
Computational studies have been performed to analyze the particle loadings onto the filters according to the different inflow pattern of dusty gas and the flow uniformities on the filter surface of back-blown gas at different permeability of porous media setup at the filter outlet. This is preliminary study to find how we can reduce the regeneration time and improve the regeneration efficiency of filters with dust layer.
An efficient plant regeneration system in alfalfa (Medicago sativa L.) through somatic embryogenesis was established. Embryogenic callus was obtained by culture of hypocotyl segments on MS medium with 0.02mg $L^{-1}$ IAA and 1.0mg $L^{-1}$ zeatin after 45 days of culture. Embryogenic calli were converted to the somatic embryos when transferred to either MS medium without plant growth regulators (PGRs) or MS medium containing various cytokinin (BA, kinetin and zeatin). Most of the somatic embryos were developed into plantlets on MS medium supplemented with 0.1 mg $L^{-1}$ kinetin. Also, secondary embryos appeared on the surface of primary embryo but they showed abnormal growth. Regenerated plantlets were transplanted to pots containing vermiculite and perlite for further analysis.

Recently, the barrier membranes have been applied for regenerating bone surrounding peri-implant defects in guided bone regeneration(GBR). GBR membrane should provide mechanical support sufficient to withstand in vivo forces and maintain wound space for bone regeneration. The ability to exclude unwanted tissues of cells(connective tissue and epithelium) is needed. In addition large surface area is conductive to tissue ingrowth. The search for ideal materials that biocompatible, bioresorbable and can support the growth and phenotypic expression of osteoblasts is a major challenge in the biomedical application for the repair of bone defects.
