Detection of Escherichia coli O157:H7 Using Combined Procedure of Immunomagnetic Separation and Test Strip Liposome Immunoassay

  • A model system for the immnunochemical detection of Escherichia coli O157:H7 using a combined immunomagnetic separation (IMS) and test-strip liposome immunoassay (LIA) procedure was developed. Immunomagnetic beads coated with anti-E. coli O157 IgG antibodies were used to separate the E. coli O157 (including the H7 serotype) from culture. Immunoliposomes, whose surface was conjugated to goat anti-E. coli O157:H7 IgG and which encapsulated the marker dye, sulforhodamine B, were used as a detection label. The test strip, onto which antibodies to goat IgG were immobilized, was the immunosensor capturing immunoliposomes that did not bind to E. coli O157:H7 on the immunomagnetic bead-E. coli O157:H7 complexes. In experiments, pure cell culture suspensions of $10^5 E.$ coli O157:H7 organisms per ml produced a measurable signal inhibition, whereas a weak yet detectable signal inhibition occurred with $10^3CFU/ml$. The inhibition signals increased, when the incubation time for IMS was extended to 90 min and higher IgG-tag density (0.4mol%) was used on the liposomes. With 0.2 and 0.4mol% IgG-tagged liposomes, the IMS-LIA procedure showed more improved signal inhibitions than those of a direct (no IMS) LIA. The combined assay, which measures the instantaneous signal from immunoliposomes, can be completed within 90 min, making it significantly faster than conventional plating methods and enzyme-linked immunosorbent assay (ELISA). Accordingly, it is quite feasible to use the combined immunoassay format of IMS and dye-loaded immunoliposomes for the detection of E. coli O157:H7.

Two-dimensional deformation measurement in the centrifuge model test using particle image velocimetry

  • The centrifuge model test is usually used for two-dimensional deformation and instability study of the soil slopes. As a typical loose slope, the municipal solid waste (MSW) landfill is easy to slide with large deformation, under high water levels or large earthquakes. A series of centrifuge model tests of landfill slide induced by rising water level and earthquake were carried out. The particle image velocimetry (PIV), laser displacement transducer (LDT) and marker tracer (MT) methods were used to measure the deformation of the landfill under different centrifugal accelerations, water levels and earthquake magnitudes. The PIV method realized the observation of continuous deformation of the landfill model, and its results were consistent with those by LDT, which had higher precision than the MT method. The deformation of the landfill was mainly vertically downward and increased linearly with the rising centrifugal acceleration. When the water level rose, the horizontal deformation of the landfill developed gradually due to the seepage, and a global slide surface formed when the critical water level was reached. The seismic deformation of the landfill was mainly vertical at a low water level, but significant horizontal deformation occurred under a high water level. The results of the tests and analyses verified the applicability of PIV in the two-dimensional deformation measurement in the centrifuge model tests of the MSW landfill, and provide an important basis for revealing the instability mechanism of landfills under extreme hydraulic and seismic conditions.

Permeability of a Capsaicin Derivative $[{14}^C]DA-5018$ to Blood-Brain Barrier Corrected with HPLC Method

  • In the present work , the transport mechanism of a capsaicin derivative, DA-5018, through blood-brain barrier (BBB) has been investigated to evaluate the feasibility of potential drug development. The result of pharmacokinetic parameters obtained from the intravenous injection of plasma volume marker,$[3^H]RSA$ and $[{14}^C]DA-5018$, indicated that both AUC, area under the plasma concentration curve and VD, volume of distribution in brain of $[3^H]RSA$ agreed with those reported ($1620{\pm}10 $percentage injected dose minute per milliliter (%IDmin/ml) and $12.0{\pm}0.1{\mu}l/g$, respectively). Elimination half-life and AUC of $[{14}^C]DA-5018$is corrected by the PHLC analysis, 19.6$\pm$1.2 min and 7.69$\pm$0.85% IDmin/ml, respectively. The metabolic rate of $[{14}^C]DA-5018$was very rapid. The blood-brain barrier permeability surface area (PS) product of $[{14}^C]DA-5018$ was calculated to be 0.24$\pm$0.05 $\mu$l/min/g. The result of internal carotid artery perfusion and capillary depletion suggested that [14C]DA-5018 pass through BBB with the time increasingly. Investigation of transport mechanism of $[{14}^C]DA-5018$ using agonist and antagonist suggested that vanilloid (capsaicin) receptor did not exist in the BBB, and nutrient carrier system in the BBB has no effect on the transport of DA-5018. In conclusion, despite the fact that penetration of DA-5018 through BBB is significant, the intact drug found in the brain tissue is small because of a rapid metabolism. Therefore, for the central analgesic effect of DA-5018, the method to increase the metabolic stability in plasma and the brain permeability should be considered.

Augmented Reality to Localize Individual Organ in Surgical Procedure

  • Objectives: Augmented reality (AR) technology has become rapidly available and is suitable for various medical applications since it can provide effective visualization of intricate anatomical structures inside the human body. This paper describes the procedure to develop an AR app with Unity3D and Vuforia software development kit and publish it to a smartphone for the localization of critical tissues or organs that cannot be seen easily by the naked eye during surgery. Methods: In this study, Vuforia version 6.5 integrated with the Unity Editor was installed on a desktop computer and configured to develop the Android AR app for the visualization of internal organs. Three-dimensional segmented human organs were extracted from a computerized tomography file using Seg3D software, and overlaid on a target body surface through the developed app with an artificial marker. Results: To aid beginners in using the AR technology for medical applications, a 3D model of the thyroid and surrounding structures was created from a thyroid cancer patient's DICOM file, and was visualized on the neck of a medical training mannequin through the developed AR app. The individual organs, including the thyroid, trachea, carotid artery, jugular vein, and esophagus were localized by the surgeon's Android smartphone. Conclusions: Vuforia software can help even researchers, students, or surgeons who do not possess computer vision expertise to easily develop an AR app in a user-friendly manner and use it to visualize and localize critical internal organs without incision. It could allow AR technology to be extensively utilized for various medical applications.

Alteration of Apoptosis during Differentiation in Human Dental Pulp-Derived Mesenchymal Stem Cell

  • Because mesenchymal stem cells (MSCs) maintain distinct capacities with respect to self-renewal, differentiation ability and immunomodulatory function, they have been highly considered as the therapeutic agents for cell-based clinical application. Of particular, differentiation condition alters characteristics of MSCs, including cellular morphology, expression of gene/protein and cell surface molecule, immunological property and apoptosis. However, the previous results for differentiation-related apoptosis in MSCs have still remained controversial due to varied outcomes. Therefore, the present study aimed to disclose periodical alterations of pro- and anti-apoptosis in MSCs under differentiation inductions. The human dental pulp-derived MSCs (DP-MSCs) were differentiated into adipocytes and osteoblasts during early (1 week), middle (2 weeks) and late (3 weeks) stages, and were investigated on their apoptosis-related changes by Annexin V assay, qRT-PCR and western blotting. The ratio of apoptotic cell population was significantly (p < 0.05) elevated during the early to middle stages of differentiations but recovered up to the similar level of undifferentiated state at the late stage of differentiation. In the expression of mRNA and protein, whereas expressions of pro-apoptosis-related makers (BAX and BAK) were not altered in any kind and duration of differentiation inductions, anti-apoptosis marker (BCL2) was significantly (p < 0.05) elevated even at the early stage of differentiations. The recovery of apoptotic cell population at the late stage of differentiation is expected to be associated with the response by elevation of anti-apoptotic molecules. The present study may contribute on understanding for cellular mechanism in differentiation of MSCs and provide background data in clinical application of MSCs in the animal biotechnology to develop effective and safe therapeutic strategy.

The Expression of Immunomodulation-Related Cytokines and Genes of Adipose- and Bone Marrow-Derived Human Mesenchymal Stromal Cells from Early to Late Passages

  • BACKGROUND: Mesenchymal stromal cells (MSCs) are multipotent stem cells that can differentiate into several cell types. In addition, many studies have shown that MSCs modulate the immune response. However, little information is currently available regarding the maintenance of immunomodulatory characteristics of MSCs through passages. Therefore, we investigated and compared cytokine and gene expression levels from adipose (AD) and bone marrow (BM)-derived MSCs relevant to immune modulation from early to late passages. METHODS: MSC immunophenotype, growth characteristics, cytokine expressions, and gene expressions were analyzed. RESULTS: AD-MSCs and BM-MSCs had similar cell morphologies and surface marker expressions from passage 4 to passage 10. Cytokines secreted by AD-MSCs and BM-MSCs were similar from early to late passages. AD-MSCs and BM-MSCs showed similar immunomodulatory properties in terms of cytokine secretion levels. However, the gene expressions of tumor necrosis factor-stimulated gene (TSG)-6 and human leukocyte antigen (HLA)-G were decreased and gene expressions of galectin-1 and -3 were increased in both AD- and BM-MSCs with repeated passages. CONCLUSION: Our study showed that the immunophenotype and expression of immunomodulation-related cytokines of AD-MSCs and BM-MSCs immunomodulation through the passages were not significantly different, even though the gene expressions of both MSCs were different.

Monoclonal antibody K312-based depletion of pluripotent cells from differentiated stem cell progeny prevents teratoma formation

  • Human pluripotent stem cells (PSCs) have been utilized as a promising source in regenerative medicine. However, the risk of teratoma formation that comes with residual undifferentiated PSCs in differentiated cell populations is most concerning in the clinical use of PSC derivatives. Here, we report that a monoclonal antibody (mAb) targeting PSCs could distinguish undifferentiated PSCs, with potential teratoma-forming activity, from differentiated PSC progeny. A panel of hybridomas generated from mouse immunization with H9 human embryonic stem cells (hESCs) was screened for ESC-specific binding using flow cytometry. A novel mAb, K312, was selected considering its high stem cell-binding activity, and this mAb could bind to several human induced pluripotent stem cells and PSC lines. Cell-binding activity of K312 was markedly decreased as hESCs were differentiated into embryoid bodies or by retinoic acid treatment. In addition, a cell population negatively isolated from undifferentiated or differentiated H9 hESCs via K312 targeting showed a significantly reduced expression of pluripotency markers, including Oct4 and Nanog. Furthermore, K312-based depletion of pluripotent cells from differentiated PSC progeny completely prevented teratoma formation. Therefore, our findings suggest that K312 is utilizable in improving stem cell transplantation safety by specifically distinguishing residual undifferentiated PSCs.

Evaluation of the Potential Effects of Retinol and Alginate/Gelatin-Based Scaffolds on Differentiation Capacity of Mouse Mesenchymal Stem Cells (MSCs) into Retinal Cells

  • Background and Objectives: Retinal stem cells (RSCs) resided in ciliary epithelium have shown to possess a high capacity to self-renew and differentiate into retinal cells. RSCs could be induced to differentiate when they are exposed to stimuli like natural compounds and suitable contexts such as biomaterials. The aim of this study was to examine the effects of Retinol and alginate/gelatin-based scaffolds on differentiation potential of mesenchymal stem cells (MSCs) originated from mouse ciliary epithelium. Methods and Results: MSCs were extracted from mouse ciliary epithelium, and their identity was verified by detecting specific surface antigens. To provide a three-dimensional in vitro culture system, 2% alginate, 0.5% gelatin and the mixed alginate-gelatin hydrogels were fabricated and checked by SEM. Retinol treatment was performed on MSCs expanded on alginate/gelatin hydrogels and the survival rate and the ability of MSCs to differentiate were examined through measuring expression alterations of retina-specific genes by ICC and qPCR. The cell population isolated from ciliary epithelium contained more than 93.4% cells positive for MSC-specific marker CD105. Alginate/gelatin scaffolds showed to provide an acceptable viability (over 70%) for MSC cultures. Retinol treatment could induce a high expression of rhodopsin protein in MSCs expanded in alginate and alginate-gelatin mixtures. An elevated presentation of Nestin, RPE65 and Rhodopsin genes was detected in retinol-treated cultures expanded on alginate and alginate-gelatin scaffolds. Conclusions: The results presented here elucidate that retinol treatment of MSCs grown on alginate scaffolds would promote the mouse ciliary epithelium-derived MSCs to differentiate towards retinal neurons.

무선 CCTV 시스템을 이용한 환자 고정 보조기술의 개발 (Patient Setup Aid with Wireless CCTV System in Radiation Therapy)

  • 목 적: 본 연구에서는 선형가속기 갠트리 헤드에 부착된 무선 CCTV 카메라를 이용한 영상처리를 통하여 환자 고정과 치료에 있어서의 정확성과 재현성 향상 방안을 개발하고자 하였다. 대상 및 방법: 선형가속기의 유사-빔 방향상(semi-beams eye view, semi-BEV)을 얻기 위하여 무선 CCTV 모듈을 자체 제작된 아크릴 어플리케이터를 이용하여 갠트리 헤드에 부착하였다. CCTV 카메라의 영상은 2.4 GHz의 고주파를 통해 치료실 벽면의 수신기로 전송된다. 선형가속기 작동 시 발생하는 무선 주파수에 의한 간섭현상(RF interference)과 누설 방사선으로 영상에 잡음이 발생하는데, 구리 호일로 카메라를 차폐하고 미디안 필터링과 같은 영상처리 기법을 이용하여 이러한 잡음을 최소화할 수 있었다. 스테레오 정합 기법과 Gauss-Newton 최적화 방법론을 기반으로 자체 제작된 소프트웨어를 통해, 환자의 고정 상태를 나타내는 3차원적 위치, 이동, 회전 정도를 정량적으로 평가하였다. 시스템의 정확도를 평가하기 위하여 팬톰 실험을 수행하였다. 또, 환자 호흡에 대한 실시간 영상분석을 통해 호흡 동기 시스템(respiratory gating system)을 구현하기 위한 방법론을 개발하였다. 결 과: 구리 호일 차폐와 영상처리를 통해 잡음을 80% 이상 줄일 수 있었다. 3차원 위치정보의 오차는 팬톰 실험을 통해 $1.5{\pm}0.7\;mm$로 나타났고, 이동 및 회전량에 대한 오차는 각각, 1 mm, $1^{\circ}$ 미만으로 나타났다. 환자 호흡에 따른 호흡 동기 시스템을 구현한 결과, 0.2초의 오차 범위 내에서 실시간 모니터링이 가능한 것으로 나타났다. 결 론: 선형가속기에 부착된 CCTV를 이용한 환자 고정 보조기술은 기존의 높은 비용을 필요로 하는 타 IGRT 기법에 비하여 설치와 이용이 간편하다. 시스템이 선형가속기와 근접해 있기 때문에 야기되는 문제점은 본 연구에서 제시된 방법을 통해 해결될 수 있었다. 시스템의 정확도를 평가해 볼 때, 임상적으로 적용이 가능할 것으로 판단된다.

증강가소성: 물리적 오브젝트에 형태적 편집가능성 부여하기 (Augmented Plasticity: Giving Morphological Editability to Physical Objects)

  • 디자인과정 후반에서 제품 디자이너는 전경 형태(디테일 디자인)에 대한 다양한 아이디어를 스케치하고 배경형태(기본 조형)와의 수많은 조합을 테스트하게 된다. 이 때 디자이너들은 실제 제품과 거의 동일한 하이피델리티(high fidelity)의 디자인 모형을 제작해 세밀한 부분까지 점검하게 된다. 하지만 디자인 모형 제작에는 적지 않은 시간과 경비가 소요되기 때문에 배경형태와 전경 형태에 대한 다양한 조합을 모두 평가해 보는 것은 사실상 불가능하다고 할 수 있다. 또한 물리적으로 구현된 아이디어는 화면 속의 디지털 모형과는 달리 편집가능성이 부족하기 때문에 '디자인과 평가'라는 순환적 작업과정이 제한적으로 이루어질 수밖에 없다. 이런 문제를 해결하기 위해 본 연구는 증강현실기술을 응용하여 물리적 오브젝트에 디지털적으로 조형적 편집가능성을 증강시킬 수 있는 증가가소성의 개념을 제안하고 이를 디지털스킨으로 구체화하였다. 디지털스킨은 ARToolKit의 비주얼 마커를 이용해 오브젝트 표면의 위치와 방향을 트래 킹하고 차분렌더링기법을 활용하여 변형된 표면을 이음매 없이 덧붙일 수 있다. 본 연구는 구현된 디지털스킨을 제품 디테일과 부분수정 디자인 그리고 디자인 소재탐색 작업에 대한 적용해 보았다. 그 결과 디자인과정 후반에서 효과적으로 디자인 아이디어를 구현하고 테스트하는데 상당한 도움을 줄 수 있을 것으로 평가되었다.
