The purpose of this study is to investigate the optimum conditions of biomass pretreatment with potassium hydroxide (KOH) for efficient utilization of cellulose, hemicellulose and lignin from Miscanthus. The optimization of variables was performed by response surface methodology (RSM). The variation ranges of the parameters for the RSM were potassium hydroxide 0.2~0.8 M, reaction temperature 110~190℃ and reaction time 10~90 min. The optimum conditions of alkali pretreatment from Miscanthus were determined as follows: concentration of KOH 0.47 M, reaction temperature 134℃ and reaction time 65 min. At the optimum conditions, the yield of cellulose from the solid fraction after pretreatment was predicted to be 95% by model prediction. Finally, 66.1 ± 1.1% of cellulose were obtained by verification experiment under the optimum conditions. The order contents of solid extraction were hemicellulose 26.4 ± 0.4%, lignin 3.7 ± 0.1% and ash 0.5 ± 0.04%. The yield of ethanol concentration of 96% was obtained using separated saccharification and fermentation.

This study was conducted to optimize the processing conditions of seasoned laver Pyropia yezoensis with conger eel Conger myriaster seasoning sauce (CES) using response surface methodology (RSM). The RSM program results for bonesoftness showed that the optimum independent variables based on the dependent variables (Y1, lipid removal rate; Y2, texture; and Y3, sensory fish odor score) were 431.0% for X1 (water amount), 115.6℃ for X2 (retort-operated temperature), and 50.1 min for X3 (retort-operated time). The RSM program results for the CES blend showed that the optimum independent variables (X1, amount of bone-softened conger eel by-products; X2, mixed sauce amount; and X3, starch amount) based on the dependent variables (Y1, amino-N; Y2, Hunter redness; and Y3, drying time) were 44.8% for A (pre-treated conger eel by-product), 36.0% for B (mixed sauce), and 19.2% for C (starch). The RSM program results for seasoned laver with CES showed that the optimum independent variables based on the dependent variables (Y1, water activity; Y2, Hunter yellowness; and Y3, overall acceptance) were 5.0% for X1, (CES amount), 313.8℃ for X2 (roasting temperature), and 6.0 s for X3 (roasting time). The seasoned laver with CES prepared under the optimum conditions was superior to commercial seasoned laver in terms of overall acceptance.

The objective of this study was to develop and validate secondary models that can predict growth parameters of L. monocytogenes Scott A as a function of concentrations (0-3%) of a commercial potassium lactate (PL) and sodium diacetate (SDA) mixture, pH (5.5-7.0), and temperature (4-37DC). A total of 120 growth curves were fitted to the Baranyi primary model that directly estimates lag time (LT) and specific growth rate (SGR). The effects of the variables on L. monocytogenes Scott A growth kinetics were modeled by response surface analysis using quadratic and cubic polynomial models of the natural logarithm transformation of both LT and SGR. Model performance was evaluated with dependent data and independent data using the prediction bias ($B_f$) and accuracy factors ($A_f$) as well as the acceptable prediction zone method [percentage of relative errors (%RE)]. Comparison of predicted versus observed values of SGR indicated that the cubic model fits better than the quadratic model, particularly at 4 and $10^{\circ}C$. The $B_f$and $A_f$for independent SGR were 1.00 and 1.08 for the cubic model and 1.08 and 1.16 for the quadratic model, respectively. For cubic and quadratic models, the %REs for the independent SGR data were 92.6 and 85.7, respectively. Both quadratic and cubic polynomial models for SGR and LT provided acceptable predictions of L. monocytogenes Scott A growth in the matrix of conditions described in the present study. Model performance can be more accurately evaluated with $B_f$and $A_f$and % RE together.
For the maximal production of aphicidal metabolites produced by the Beauveria bassiana Bb08, statistical methods such as the Box-Behnken experimental design and response surface methodology were used. The fungal culture filtrate was sprayed towards 3-star aphids and the mortality was examined. After the statistical analysis of the aphid mortality, the optimal culture conditions were found to be a culture temperature of $26.2^{\circ}C$, medium pH 5.9, flask shaking speed of 209.0 rpm, and culture time of 5.9 days. The expected mortality on days 4, 5, and 6 after spraying the filtrate on to the aphids were 76.8%, 84.9%, and 89.4%, respectively. All 4 factors of the culture conditions significantly affected the production of the aphicidal metabolites, and the order of significance was temperature, pH, culture time and shaking speed.

Over 12 years have passed since the first commercial operation of a Korean high-speed train. Since then, the transport capacity of the high-speed lines has become almost saturated. Therefore, studies have been carried out to increase the operating speed of the trains in order to increase their transportation capacity. This study was carried out to improve the critical speed of the KTX-Sancheon, Korean high-speed train, in order to increase its operating speed. A dynamic analysis of the KTX-Sancheon train was performed using the contact data obtained from the wheel wear profiles that were measured from a KTX-Sancheon train in commercial operation. The analysis results were verified by comparing them with the measurement acceleration data obtained from KTX-Sancheon. The suspension parameters were optimized to improve the operation speed. The critical speed of KTX-Sancheon was increased by 9.4% after the optimization by the response surface method. The optimized suspension parameters are expected to be used for the new bogie design to increase the operating speed of KTX-Sancheon from 300km/h to 350km/h.

The effects of rainfall input uncertainty on predictions of stream flow are studied based extended GLUE (Generalized Likelihood Uncertainty Estimation) approach. The uncertainty in the rainfall data is implemented by systematic/non-systematic rainfall measurement analysis in Weida catchment, Germany. PDM (Probability Distribution Model) rainfall runoff model is selected for hydrological representation of the catchment. Using general correction procedure and DUE(Data Uncertainty Engine), feasible rainfall time series are generated. These series are applied to PDM in MC(Monte Carlo) and GLUE method; Posterior distributions of the model parameters are examined and behavioural model parameters are selected for simplified GLUE prediction of stream flow. All predictions are combined to develop ensemble prediction and 90 percentile of ensemble prediction, which are used to show the effects of uncertainty sources of input data and model parameters. The results show acceptable performances in all flow regime, except underestimation of the peak flows. These results are not definite proof of the effects of rainfall uncertainty on parameter estimation; however, extended GLUE approach in this study is a potential method which can include major uncertainty in the rainfall-runoff modelling.
This research was carried out to investigate the effect of installation spacing of subsurface drip irrigation pipe on the mineral content, nutrient uptake, yield of lettuce, water requirement for irrigation, and soil chemical properties in greenhouse cultivation. Semi-forcing and retarding culture were implemented in this experiment, with four treatments containing overhead spray irrigation and three subsurface irrigation lateral spacing intervals of 30, 40, 50 cm at a depth of 30 cm from soil surface, respectively. Each mineral content of lettuce grown under subirrigation system did not show significant difference between treatments, however the uptake of nutrients was lower at 50 cm-distance. The yield was largest in 30 cm-subirrigation (SI), followed by 40 cm-SI, overhead spray, and 50 cm-treatment. Water requirement for irrigation was highest in overhead spray, and it was in reverse proportion to the distance of irrigation pipes. $NO_3$-N content in the soil, at a depth of 10 cm, showed a higher value in 50 cm-SI, followed by 40 cm-SI, overhead spray and 30 cm-SI. Exchangeable K content was highest in 50 cm-SI, Mg was highest in 40 cm-SI, and Ca was lowest in 30 cm-SI. In conclusion, the lettuce yield was not different between 30 and 40 cm-SI, but water requirement for irrigation was lower as the distance of irrigation pipes was further. And it seems to be needed more precise research on this theme, because crop yield and the dynamics of soil minerals in subsurface irrigation can vary with the depth and distance of irrigation pipes, dripper, water flow depending on the soil texture, and plant response to soil minerals.
Relation between soil salinity and forage yield of five upland crops such as sudan grass, sesbania, barnyard grass, corn and soybean was studied in the three reclaimed lands of Korea during two years from 2007 to 2008. Although plant number emerged was obtained satisfactory by desalting treatment, further growth of crops was sharply affected by re-salting process according to soil drying. Soil salinity varied place by place and changed constantly, crop growth was differently responded to soil salinity according to the specific conditions of the reclaimed lands. In the Iweon reclaimed land with higher soil water conductivity and sandy soil texture, crop growth and yield sharply decrease with increase of soil salinity. Relation between soil salinity and crop growth and yield was well expressed as logarithmic function. Surface soil EC to reach at 50% of growth reduction to the tallest height of crops was $5dS\;m^{-1}$ for soybean, $6dS\;m^{-1}$ for corn and $7dS\;m^{-1}$ for sudan grass, sesbania, and barnyard grass by logarithmic function. In the Hwaong and Yeongsangang reclaimed lands with low soil conductivity and finer soil texture, plant growth response to salt stress was statistically vague by mixing of harmful influence from flooding and wet injury. However, it is observed that crop growth and yield on the place of lower salinity was better than crop growth on the place of higher salinity. It is accordingly concluded that flooding control during summer rainy season is vary important as well as desalting process for good growth of soiling and forage crops in the newly reclaimed land from tidal flat.

야채 soup mix의 제조조건이 야채죽의 물리적 및 관능적 특성에 대한 영향을 조사하였다. Soup mix의 제조조건인 주입액의 양 (쌀 무게에 대한 물의 체적 비), 버섯, 당근 그리고 대파의 첨가량(쌀 무게에 대한 잣의 무게 비)에 따른 물리적 특성과 관능적 특성의 변화를 모니터링 하고자 반응 표면분석법 (response surface methodology, RSM)을 사용하였으며, 이때 실험계획은 중심합성계획법을 적용하였다. 요인변수(Xn)를 중심합성계획에 따라 17실험구로 구분하여 조리실험을 실시하였고, 이들 요인변수에 의해 영향을 받는 반응변수(Yn)는 야채죽의 물리적 특성으로 하여 회귀분석에 사용하였다. 회귀분석에 의한 모델식의 예측에는 SAS (statistical analysis system) program을 사용하였으며, 야채죽의 조리조건이 물리적 및 관능적 특성에 미치는 영향은 SAS program을 이용한 3차원 반응표면분석법으로 해석하였다. 야채의 배합비를 달리한 야채죽의 물리적 특성인 색도 L, a 및 b값에 대한 F-value는 자각 1.50, 11.75 및 5.58이고,유의수준이 각각 0.3044, 0.0019와 0.0169로서 a값과 b값의 유의성이 1% 수준에서 인정되어 이들 제조조건이 a값과 b값에 큰 영향을 미치는 것으로 나타났다. 점도와 퍼짐성에 대한 유의수준은 각각 0.6920과 0.7528이고, 반응표면회귀식의 $R^2$은 각각 0.4766과 0.4436으로 유의성이 인정되지 않았다. 고형분에 대한 유의수준은 0.2026이고 회귀식의 $R^2$는 0.7107으로서 버섯, 당근 및 대파 첨가량이 고형분의 변화에 영향을 미치지 않은 것을 알 수 있었다. 관능적 특성인 색상에 대하여 soup mix의 제조조건이 야채죽에 미치는 영향은 F-value는 6.23이고, 유의수준이 0.0124으로서 1%수준에서 유의성이 인정되었으며, 회귀식의 $R^2$은 0.8890이다. 향에 대한 유의확률은 0.4555이고 0.05이상이므로 유의성이 인정되지 않아 설정된 범위내에서 야채죽의 향에 대하여 크게 영향을 미치지 않는 것으로 나타났다. 점성에 대한 반응표면회귀식의 $R^2$은 0.8134로서 그 유의성이 5%수준에서 인정되었다. 맛과 전반적 기호도에 대하여 야채의 배합비에 따른 반응표면회귀식의 $R^2$은 각각 0.7374와 0.8651이며, 유의화률은 0.1578과 0.0228으로 나타나 전반적인 기호도에 대한 영향은 10% 유의수준에서 영향을 주었다. 결론적으로, 물리적 특성인 색도, 점성, 퍼짐성과 고형분의 함량은 관능적 특성인 색과 비교적 높은 정(正)의 상관을 나타내었으며, 관능적 특성인 향과의 상관은 유의성이 인정되지 않았다. 야채죽 제조를 위한 soup mix의 제조조건에 있어서 야채의 배합비는 색과 점성에 영향을 미치는 가장 주요한 조건이라고 생각된다.
Objectives : The purpose of this study is to observe the inhibitory effects of $Chrthami$ semen oil pharmacopuncture(CSOP) on CIA (collagen-induced arthritis) mice. Materials and Methods : Two types of experiments were conducted: $in$$vitro$ assay, inhibition of MIF mRNA and TNF-${\alpha}$ mRNA expressions in synovial membranes was observed, and $in$$vivo$ assay, $1{\mu}{\ell}/kg$ CSOP was injected every day to the left $Weizhong$ ($BL_{40}$) from day 3 to 21 after induction of CIA, and changes in paw edema, apical surface morphology, neovascularization in synovial membranes, fibrosis, pro-inflammatory cytokines production, Th-1 differentiation, and anti-inflammatory effect were investigated. Results : 1. In synoviocytes of the CIA mice treated with CSOP, MIF mRNA and TNF-${\alpha}$ mRNA expressions were down-regulated in a dose-dependent manner. 2. Paw edema of the CIA mice treated with CSOP was diminished. 3. Tissue injury in the synovial membranes, capillary distribution and fibrosis were reduced in CSOP-treated mice. 4. MIF, TNF-${\alpha}$, IL-6, MMP-9 expressions were repressed in CSOP-treated mice during the experiment to observe the inhibitory effect on cytokines production in early stage RA. 5. IL-12 and CD28 were reduced in CSOP-treated mice during the observation of inhibitory effect on Th 1 differentiation. 6. PPAR-${\gamma}$ was increased during the experiment to observe the anti-inflammatory effect of CSOP. Conclusions : The results may suggest that administration treatment using $Chrthami$ semen oil pharmacopuncture decreases the inflammatory response on an Animal Model with CIA.
