Culture of Endothelial Cells by Transfection with Plasmid Harboring Vascular Endothelial Growth Factor

  • Vascular endothelial cells (EGs) are usually difficult to culture to culture in a large scale because of their complicated requirements for cell growth. As the vascular endothelial growth factor (VEGF) is a key growth factor in the EC culture, we transfected human umbilical vein endothelial cells (HUVEC) using a plasmid containing VEGF gene and let them grow in a culture medium eliminated an important supplement, endothelail cell growth supplement(ECGS). The expression of VEGF by HUVEC tansfected with Vegf GENE was not enough to stimulate the growth of HUVEC, only 40% of maximum cell density obtainable in the presence of ECGS. However, when the culture medium was supplied with 2.5 ng/ml of basic fibroblast growth factor (bFGF), a synergistic effect effect of VEGE and bFGF was observed. In this case, the final cell density was recovered was recovered up to about 78% of maxium value.

Evaluation of Mulberry (Morus alba) as Potential Feed Supplement for Ruminants: The Effect of Plant Maturity on In situ Disappearance and In vitro Intestinal Digestibility of Plant Fractions

  • The in situ nylon bag degradation and in vitro intestinal digestibility of dry matter (DM), and crude protein (CP) of mulberry (Morus alba) plant fractions was studied at four harvest stages, 3 (W3), 5 (W5), 7 (W7) and 9 (W9) weeks. Degradability of DM and CP of the whole plant and stem fractions declined significantly (p<0.01) with advancing plant maturity in the order W3>W5 and W7>W9 and W3>W5>W7>W9, respectively. The degradation of DM and CP of the leaf fraction was also influenced by plant maturity but no trend was observed. The degradation of DM and CP of the whole plant and leaves increased rapidly during the first 48 and 24 h of incubation, respectively, when maximum degradation was reached. In vitro intestinal digestibility of CP was more influenced by the residence time in the rumen than by plant maturity. This study showed that mulberry is suitable as a supplement, particularly to low-quality roughages, in providing a source of rapidly available nitrogen to the rumen microbes, hence improving the roughage degradability and intake.

  • Internet is deeply rooted in everyday life and many things are performed using internet in real-world, therefore internet users increased because of convenience. Also internet accident is on the increase rapidly. The security vendor developed security system to protect network and system from intruder. Many hackings can be prevented and detected by using these security solutions. However, the new hacking methods and tools that can detour or defeat these solutions have been emerging and even script kids using these methods and tools can easily hack the systems. In consequence, system has gone through various difficulties. So, Necessity of intruder trace-back technology is increased gradually. Trace-back technology is tracing back a malicious hacker to his real location. trace-back technology is largely divided into TCP connection trace-back and IP packet trace-back to trace spoofed IP of form denial-of-service attacks. TCP connection trace-back technology that autonomously traces back the real location of hacker who attacks system using stepping stone at real time. In this paper, We will describe watermark trace-back system using TCP hijacking technique to supplement difficult problem of connection maintenance happened at watermark insertion. Through proposed result, we may search attacker's real location which attempt attack through multiple connection by real time.

Stimulation of Dendritic Cell Maturation and Induction of Apoptosis in Leukemia Cells by a Heat-stable Extract from Azuki bean (Vigna angularis), a Promising Immunopotentiating Food and Dietary Supplement for Cancer Prevention

  • Non-toxic stimulation of dendritic cells (DCs), which are central immunomodulators, may aid the prevention of cancer. Furthermore, induction of apoptosis in cancer cells by anticancer agents contributes to the induction of DC maturation. We previously reported that extracts from $Pinus$ $parviflora$ Sieb. et Zucc pine cone and $Mucuna$ seed induce differentiation of mouse bone marrow cells into mature dendritic cells and also induce apoptosis in various human cancer cell lines. In the present study, we screened 31 kinds of edible beans with biological activity similar to that of extracts from pine cone and $Mucuna$ and found that the heat-stable extract from azuki bean ($Vigna$ $angula$) stimulated differentiation of bone marrow cells into immature DCs with the greatest efficacy. The level of IL-6 produced by sequential treatment of DCs with azuki extract and lipopolysaccharide was the highest among the examined beans. Azuki extract also inhibited the growth of human leukemia U937 cells, leading to induction of apoptosis. These results suggest that azuki bean and its extract are immunopotentiating foods that can be used as a dietary supplement for cancer prevention and immunotherapy.

Iron supplement tablet embedded in the oral cavity mimicking neoplasm: a case report

  • The detection of foreign bodies in the upper-aerodigestive tract is a fairly frequent event and can occur in various areas and for various reasons. In rare cases, foreign bodies can simulate a neoplasia. We evaluated similar cases during emergency regimen with an oral cavity mucosal lesion, causing lockjaw, sore throat, dysphagia, and swelling of the submandibular and laterocervical region. Physical examination revealed an extensive mucosal ulceration in the floor of the mouth and the lateral surface of the tongue, comparable to oral cancer. During a second, more accurate assessment, a partially deteriorated iron supplement tablet was found embedded in a mucosal pocket. After removing the tablet, gradual normalization of the tissue was observed without any sequelae. This is one of the many reasons why it is advisable and useful in cases of oral lesions to collect a detailed medical history and to perform an accurate clinical evaluation, including inspection and palpation of the lesion, before proceeding to further diagnostic assessments, especially in elderly patients taking many medications. However unlikely, it is possible that difficulty in swallowing pills or tablets could generate tumorlike lesions.

The Effect of Rice Germ Oil Supplement on Serum and Hepatic Lipid Levels of Streptozotocin-Induced Diabetic Mice (쌀눈기름의 급여가 Streptozotocin으로 유발한 당뇨 마우스의 혈액 및 간조직 중의 지질 수준에 미치는 영향)

  • This study was carried out to investigate the effects of rice germ oil supplement on the lipid metabolism of insulin-dependent diabetic mice. Streptozotocin-induced diabetic mice were fed three kinds of experimental diets with 20% lipid, composed of 20% lard (L) : 10% lard and 10% rice germ oil (LRGO) ; and 20% rice germ oil (RBO), respectively, for 7 weeks. Diet intake, body weight, organ weight and lipid levels of serum, liver and feces were measured. There was no significant difference in diet intake, body weight and organ weight among the experimental groups. But the concentrations of serum triglyceride in the LRGO and RGO groups, and of serum total cholesterol in the RGO group, were significantly lower than those of the L group fed the 20% lard diet. The levels of hepatic total lipid of the RGO group, and of hepatic total cholesterol of the LRGO and RGO groups were significantly lower than those of the L group. The contents of total lipid and total cholesterol excreted in the feces of the LRGO and RGO groups were higher than those of the L group. These results suggest that rice germ oil can reduce the levels of total cholesterol concentrations in the serum or livers of insulin-dependent diabetic mice, and that the hypolipidemic effect of rice germ oil may be due to increasing fecal lipid excretion and decreasing lipid absorptivity.

Effect of Fiber on Serum Lipid Metabolism in Rats with Diet-Induced Cholesterolemia (식이섬유가 고콜레스테롤혈증 흰쥐의 혈청 지질대사에 미치는 영향)

  • The study was conducted to investigate the effect of cholesterol and fiber on serum lipid metabolism in rats. Male Sprague Dawley rats were administrated 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia and were fed on diet containing three concentrations(0%, 5%, 10%) of cellulose and pectin, respectively. The rats were sacrified after 5 weeks of feeding periods. Serum triglyceride concentration was significantly decreased in proportion to level of fiber. Phospholipid concentration was more decreased in 10% fiber groups than 5% fiber groups but did not show any difference by fiber types. Free cholesterol and cholesteryl ester concentrations in serum were significantly decreased by fiber supplement and pectin was more effective than cellulose in cholesterol lowering effect. Whereas HDL cholesterol concentration and HDL C/TC were increased in proportion to level of fiber and the effect of pectin was not more significant than that of cellulose. Serum chylomicron cholesterol concentration also showed significant decrease by pectin supplement and LDL , VLDL cholesterol concentrations were more significantly decreased in 10% fiber groups than those of 5% fiber groups. The results indicate that 10% pectin may have beneficial roles in hypercholesterolemia.
