  • A human Cu, Zn-superoxide dismutase (Cu, Zn-SOD) was fused with a Tat PTD of HIV-1 to produce a novel anti-aging ingredient, Tat-SOD for cosmeceuticals. Test of stability and evaluation of transduction efficacy and enzymatic activity suggest Tat-SOD is an effective active ingredient for anti-aging treatment.

생쥐에서 N-Nitrosodiethylamine에 의한 산화성 스트레스에 대한 Lutein의 항산화효과 (Antioxidant Effect of Lutein on N-Nitrosodiethylamine-induced Oxidative Stress in Mice)

  • To investigate the antioxidant effect of lutein on N-nitrosodimethylamine (NDEA)-induced oxidative stress in mice, we measured lipid peroxidation, superoxide dismutase (SOD) and catalase of various tissues. Body weight was almost similar in lutein and control groups during 3 weeks. NDEA increased significantly the activities of typical marker enzymes of liver function (AST, ALT and ALP) in both groups. However, the increase of plasma aminotransferase activity significantly decreased in lutein group. Lipid peroxidation and SOD in various tissues, such as heart, lung, liver, kidney, spleen and plasma were significantly increased by NDEA, which were significantly reduced by lutein at a dose of 50 mg/kg. Catalase activity decreased significantly in control and lutein groups treated with NDEA, the effect being less in lutein group. Lesser effect on SOD and catalase in NDEA-treated lutein group indicates the improvement of protective mechanisms by lutein. Thus, it can be concluded from the present study that lutein can offer a useful protection against NDEA-induced oxidative stress.

  • 본 연구는 염화니켈 급성노출에 따른 북방전복의 산화스트레스효소 활성에 대한 변화를 알아보고자 하였다. 실험은 1개의 대조구와 5개의 농도구 (5.9, 8.8, 13.3, 20.0, 30.0 mg/L)에서 4일간 진행되었다. superoxide dismutase (SOD) 활성은 발과 간췌장의 경우 모든 니켈 노출구에서 증가하였으나 아가미에서는 감소하는 결과를 보였다. catalase (CAT) 활성은 발의 경우 20.0 mg/L 와 30.0 mg/L 니켈 노출구를 제외하고 증가하였으나 아가미에서는 대조구와 유사한 결과를 보였다. 간췌장에서는 모든 니켈 노출구에서 대조구에 비해 증가하는 결과를 보였다. 하지만 SOD와 CAT 활성 모두 대조구에 비해 유의적인 차이는 보이지 않았다 (P > 0.05).

생맥산가미방 추출물이 멜라닌 생합성 저해 효과와 SOD 활성에 미치는 연구 (Study of ShengmaisanJiaweifang Extracts on the Inhibitory Effects of Melanin Synthesis and Superoxide Dismutase Activity)

  • This study aims to evaluate the effects of Shengmaisan (SMS) and three types of ShengmaisanJiaweifang on the inhibitory effect of melanin synthesis in B16F10 cells, the mechanism of action through tyrosinase, and the antioxidant effect through superoxide dismutase (SOD) activity. In this study, we used ShengmaisanJiaweifangs (SMS, SMSRR, SMSAD, SMSAR) to research the whitening effects in B16F10 cell lines. Shengmaisan (SMS) was a herbal medicine composed of Ginseng Radix, Liriopis Tuber, and Schisandrae Fructus. ShengmaisanJiaweifangs included SMSRR (SMS added with Rehmanniae Radix), SMSAD (SMS added with Asparagi Radix) and SMSAR (SMS added with Astragali Radix). We measured the cell viability, the inhibition rate of the melanin biosynthesis, and the activity of tyrosinase and SOD in malignant melanoma, B16F10 cells, to survey the whitening effect and the mechanism of the impact on the sample. As a result, SMSRR significantly suppressed the cell viability of B16F10 at more than $500{\mu}g/m{\ell}$ and significantly inhibited the generation of melanin induced by ${\alpha}$-MSH at more than $250{\mu}g/m{\ell}$. SMSRR ($500{\mu}g/m{\ell}$) decreased the activity of tyrosinase while increased the activity of SOD. Therefore, we considered that the SMSRR would be able to produce high value-added products more SMS if used as a commercial.

고지방식이에서 서로 다른 불포화 지방산 섭취가 쥐의 혈청과 조직의 Tocopherol함량과 지질과산화물 형성에 미치는 영향 (Effects of Dietary Polyunsaturated Fatty Acids on Tocopherol Contents and Lipid Peroxidation of Plasma and Tissues in Rats Fed High Fat Diet)

  • This study was observed the effect of n6 and n3 polyunsaturated fatty acids of dietary corn oil and fish oil which was supplemented with similar levels of tocopherol in high fat diet on the levels of tocopherol, malondialdehyde ( MDA) productions of plasma and tissues of rats. Also RBC hemolysis, superoxide dismutase (SOD) and glutathione peroxidase (GSH Px) activities In liver were determined. Male Sprague Dawley rats were fed high fat (40%Cal) diet which was different only In fatty acid composition for 6 weeks. Dietary (arts were beef tallow(BT) as a source of saturated fatty acid, corn oil(CO) for n6 linoleic acid (LA) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) + docosahexaenoic acid (DHA). Plasma and liver tocopherol levels were lowered by n3 PUy4 but there was no difference in malondialdehyde(MDA) level by different dietary PUFA. However, MDA content of RBC and hemolysis were increased by n3 PUFA. MDA content, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in liver were increased in more unsaturated dietary fat groups. Especially, SOD activity was increased in proportion to the degree of fat unsaturation.

흰쥐에서 Aminotriazole과 Diethyldithiocarbamate가 Paraquat의 독성에 미치는 영향 (Effects of 3-Amino-1,2,4 Triazole and Diethyldithiocarbamate on Paraquat Toxicity in Rats)

  • The effects of superoxide dismutase(SOD) and catalase on the toxicity of paraquat(PQ) were studied using diethyldithiocarbamate(DDC), 3-amino-1,2,4-triazole(AT) which are inhibitors of Cu, Zn-SOD and catalase in rats. Sprague Dawley rats were divide into 6 groups: control, DDC, PQ, AT, DDC+PQ, and AT+PQ group. The PQ (50 mg/kg body weight(BW); about half dose of $LD_{50}$) was administered with orally, otherwise AT(1.0g/kg BW) and DDC(1.0g/kg BW) were administered by intrperitoneal(iP) injection. The survival rate of rats in PQ+AT group was significantly decreased compared with PQ group while the difference of survival rate between DDC group and DDC+PQ group was not significant. The SOD activity after administration of DDC was decreased in liver, lung and kidney, but catalase activity was not changed. The catalase activity in liver, lung and kidney of AT treated rats was decreased, while SOD activity was not changed in this group. The effects of DDC and AT to the PQ toxicity was also observed in primary cultured rat Skin fibroblasts. The viable cells that was measured with MTT method, was decreased in AT+PQ treated group compared to PQ treated group, but the difference of cell viability between DDC treat group and DDC+PQ treated group was not observed. This result, AT potentlate PQ toxicity while DDC were not affect, suggested that the decreased catalase activity lead to elevation of hydrogen peroxide levels and PQ toxicity may be correlate with the hydrogen peroxide rather than the superoxides.

Antiproliferative Effect of Artemisia argyi Extract against J774A.1 Cells and Subcellular Superoxide Dismutase (SOD) Activity Changes

  • The water and methanol extracts of Artemisia argyi showed significant cytotoxicities against J774A.1 cells but not so much against normal leukocytes. The cytotoxicities were found to be dependent on the extract concentration and the incubation time. The concentration of water and methanol extracts inhibiting 50% of cell proliferation ($IC_{50}$) were estimated to be 44.2 mg/ml and 71.6 mg/ml, respectively. In the presence of Artemisia argyi water extract, total superoxide dismutase (CuZnSOD and MnSOD) activities of media, cytoplasmic and mitochondrial fractions of J774A.1 cells increased in accordance with cytotoxicity. MnSOD was found to be the main component of enhanced total SOD activities, particulary in the mitochondrial fraction. In contrast to SOD, catalase and glutathione peroxidase (GPx) were not found in any instance of the current investigation. In addition, substantial amount of $O_2^-$ appeared to be generated in the mitochondrial fraction under the influence of Artemisia argyi. All data put together, it is postulated that Artemisia argyi extracts seem to stimulate $O_2^-$ generation in mitochondria of J774A.1 cells with concomitant increases of SODs. Since $H_2O_2$, the reaction product of SOD on $O_2^-$, is known to be readily converted to very toxic $OH{\cdot}$ in the absence of catalase and/or GPx cooperation, toxicity derived from ROS such as $O_2^-$, $H_2O_2$, and $OH{\cdot}$ may be the main cause of necrosis and/or apoptosis of J774A.1 cells.

가감형방지황탕 열수 추출물이 항산화 작용에 미치는 영향 (Antioxidant Property of the Gagam-Hyungbang-Gihwang-tang Using Biochemical Markers of Carcinogenesis)

  • Objectives : The verity extract of the Gagam-Hyungbang-Gihwang-tang (GHG) was assessed to determine the mechanisms of its antioxidant activity. Methods : The fellowing effects were measured : GHG exhibited a concentration-treatment; scavenging ${\alpha},\;{\alpha}-diphenyl-\beta-picrylhydrazyl$ (DPPH) radical, linoleic acid oxidation in a thiocyanate assay system, and superoxide anion, hydroxyl radical-induced DNA nicking. We investigated mRNA levels such as superoxide-dismutase. Results : The GHG extract showed dose-dependent free radical scavenging activity, including DPPH radicals, hydroxyl radicals, and superoxide anion, using different systems. The GHG was also found to be effective in protecting plasmid DNA against the strand breakage induced by hydroxyl radicals in Fenton's reaction mixture. Furthermore, SOD-1 mRNA expression levels increased in tat hepatoma H4IIE cells Conclusions : We expect that GHG will to helpful to the development of antioxidant activity treatments.

홍삼 투여가 방사선에 조사된 생쥐 신장의 Superoxide Dismutase에 미치는 영향 (The Effect of Red Ginseng Extract on Superoxide Dismutase Activity in the Kidney of Gamma-ray Irradiated Mice)

  • 감마선에 조사된 흰쥐의 신장 내의 효소활성에 홍삼 추출물이 어떠한 영향을 미치는지를 연구하고자 생후 5주령의 생쥐를 4군으로 분류하여 감마선 조사 및 홍삼 추출물 투여 혹은 생리적 식염수를 투여하고 그로부터 1일, 2일, 3일, 4일 및 5일 등 5회에 걸쳐 도살하여 신장을 취하고 신장조직 내의 SOD, peroxidase 및 catalase 등의 활성도를 측정하였다. 그 결과 홍삼 추출물 투여군은 대조군에 비하여 SOD나 catalase 및 peroxidase의 활성이 유의성있게 증가하다가 회복하였으며 감마선 조사 후 생리적 식염수 투여군은 대조군에 비하여 SOD, peroxidase 및 catalase의 활성이 증가하다가 감소하는 경향이었다. 감마선 조사후 홍삼 추출물 투여군은 SOD, peroxidase 및 catalase의 활성도가 감마선 조사 후 생리적 식염수 투여군에 비하여 보다 빠르게 회복되는 경향이었다. 그러므로 홍삼 추출물이 이온화 방사선 조사 후 장해로 일어나는 SOD, peroxidase 및 catalase 활성도의 변화에 회복효과가 있음이 나타났다.

산소 라디칼 관련 효소의 폐흡충 발육 단계별 활성도 변화 (Activities of scavenging enzymes of oxygen radicals in early maturation stages of Paragonimus westermani)

  • Oxygen radical은 생물체의 산소대사 과정의 부산물로 생성되어 세포내 여러 성분을 불환성화시키거나 미생물에 대한 방어기전으로 작용한다. 기생충에 존재하는 antioxidant enzyme은 숙주의 방어기전에서 유리하는 oxygen radical의 독성을 제거하므로 이 효소의 활성도는 기생충의 생존에 영향을 미친다고 생각하고 있다. 폐흡충은 피낭유충이 종숙주에 침입한 다음 숙주내 조직이행 시기를 거쳐서 폐에 도달하여 성충이 되므로 이 과정에서 각 시기별 산소독성과 이에 대항하는 효소의 환성이 다를 것으로 추정하였다. 폐흡충의 피낭유충과 감염후 4주, 8주, 12주에 얻은 충체의 추출물을 조효소(조효소)로 하여 SOD, catalase, peroxidase, glutathione peroxidase의 활성도를 측정하였다. 각 효소의 비환성도(specific activity) 중 catalase는 피낭유충에서 최고치였으며, SOD 와 peroxidase는 4주 충체에서 가장 높았고, glutathione peroxidase는 8주 충체에서 높았다. 이들 4가지 antioxidant효소의 비환성도는 감염 12주인 성충에서 모두 낮게 측정되어 조직 이행시기의 충체에서 더 높은 효 소 활성도를 지니고 있음을 알 수 있었다.
