Glycerol이 흰쥐 신장에서의 Malondialdehyde 함량과 Superoxide Dismutase 활성도 및 요중 단백질 배설량과 $N-acetyl-{\beta}-D-glucosaminidase$ 활성도에 미치는 영향 (Effects of Glycerol on the Malondialdehyde Level and Superoxide Dismutase Activity in the Kidney and Urinary Protein Excretion and $N-acetyl-{\beta}-D-glucosaminidase$ Activity of the Rats)

  • In an attempt to dofine the early biochemical determinants that participate in the pathogenesis of glycerol-induced nephrotoxicity, especially focusing on oxygen free radicals and $N-acetyl-{\beta}-D-glucosaminidase$ (NAG) activity, we studied 24-hours urine outflow, 24-hours urinary protein excretion and urinary NAG activity after the injection of glycerol and also we studied malondialdehyde(MDA) level and superoxide dismutase(SOD) activity in the kidney of rats at 24hr after the injection of glycerol. Sprague-Dawley albino rats weighing 240 to 260 gm were injected intramuscularly with a 50% solution of glycerol(2ml/kg, 4ml/kg and 8ml/kg). The group treated with glycerol showed significantly lower urine outflow level and urinary protein excretion level and higher urinary NAG activity after the injection as compared to those of control group. Also the group treated with glycerol showed significantly higher MDA level and lower SOD activity at 24hr after the injection as compared to those of control group. These results suggest that the excessive oxygen free radicals resulting from the depression of SOD activity is an important determinant in the pathogenesis of glycerol-induced nephrotoxicity and higher urinary NAG activity is an index of renal tubular cell damage in the glycerol-induced nephrotoxicity.

흰쥐에 있어 방사선에 대한 셀레늄의 보호효과 (The Protective Effect of Selenium on Radiation in Rat)

  • 본 연구에서는 강력한 항산화제로 알려진 셀레늄의 방사선방호효과를 연구하였다. 렛드에 셀레늄을 14일간 경구투여 후 10 Gy의 방사선을 전신조사하였다. 그리고 1일, 3.5일, 7일, 21일 후에 혈구 성분 및 SOD(Superoxide Dismutase)와 소장의 변화를 관찰하였다. 방사선조사군에 비해 셀레늄 투여 후 방사선조사군에서 조혈면역계의 손상을 경감시키는 유의한 방호 효과가 있었다(p<0.05). 그리고 셀레늄이 항산화 효소인 Superoxide Dismutase(SOD)의 활성을 증가시키는 유효한 성분이며, 방사선 조사에 의한 소장움 세포의 apoptosis 발현을 억제하는 효과가 있음을 확인하였다. 이 결과를 바탕으로 셀레늄은 방사선으로부터 생명체를 보호하는 필수 성분이라 생각한다.

  • 유해산소제거효소는 세포내에서 생성되는 유해산소를 산소와 과산화 수소로 바꿈으로서 유해산소의 농도를 낮은 수준으로 유지하여 세포를 유해산소의 독성으로부터 보호하는 기능을 담당하고 있다. 이전의 연구에서 파낙사다이올(PD)와 진세노사이드 $Rb_2$가 전사조절인자 AP2를 유도하여 유해산소 제거효소의 전사조절부위 내의 AP2결합부위를 통해 유해산소제거효소의 함량증대를 유도함을 보고한 바 있다. 이를 토대로 본 연구에서는 인삼의 각부위에서 추출된 조사포닌으로 panaxadiol(PD)와 panaxatriol(PT)의 성분함유비가 다른 시료를 이용하여 이들이 유해산소제거효소의 발현 유도성에 미치는 영향을 조사하였다. 이를 조사하기위해 유해산소제거효소의 전사조절부위를 클로람페니콜 아세틸트란스퍼라제의 구조유전자와 융합시킨 벡터를 인간의 간세포에 도입하여 활성도를 측정하였다. 그 결과, PD 성분의 함량비증가에 비례적으로 유해산소제거효소의전사가 증대 되었다. 또한 동일한 결과로서, PD 대 PT의 함량비가 약 2.6으로 PD의 함량이 가장높은 세세미 (finely-hairy root) 추출분획에서 유해산소제거 효소의 전사촉진이 대조군에 비해 3배이상 촉진됨을 관찰할수 있었다. 이상의 결과는 PD계의 분획이 유해산소제거효소의 유도성효과를 나타냄을 시사하고 있으며, 유해산소제거효소의 유도물질로서 PD분획과 세세미 추출물이 유용하게 이용될수있음을 제시하고 있다.

Extracellular Superoxide Dismutase (EC-SOD) Transgenic Mice: Possible Animal Model for Various Skin Changes

  • We have generated transgenic mice that expressed mouse extracellular superoxide dismutase (EC-SOD) in their skin. In particular, the expression plasmid DNA containing human keratin K14 promoter was used to direct the keratinocyte-specific transcription of the transgene. To compare intron-dependent and intron-independent gene expression, we constructed two vectors. The vector B, which contains the rabbit -globin intron 2, was not effective for mouse EC-SOD overexpression. The EC-SOD transcript was detected in the skin, as determined by Northern blot analysis. Furthermore, EC-SOD protein was detected in the skin tissue, as demonstrated by Western blot analysis. To evaluate the expression levels of EC-SOD in various tissues, we purified EC-SOD from the skin, lungs, brain, kidneys, livers, and spleen of transgenic mice and measured its activities. EC-SOD activities in the transgenic mice skin were approximately 7 fold higher than in wild-type mice. These results suggest that the mouse overexpressing vector not only induces keratinocyte-specific expression of EC-SOD, but also expresses successfully functional EC-SOD. Thus, these transgenic mice appeared to be useful for the expression of the EC-SOD gene and subsequent analysis of various skin changes, such as erythema, inflamation, photoaging, and skin tumors.

Superoxide dismutase 3 protects mesenchymal stem cells through enhanced autophagy and regulation of FoxO3a trafficking

  • Therapeutic applications of mesenchymal stem cells (MSCs) are limited due to their early death within the first few days of transplantation. Therefore, to improve the efficacy of cell-based therapies, it is necessary to manipulate MSCs so that they can resist various stresses imposed by the microenvironment. Moreover, the role of superoxide dismutase 3 (SOD3) in regulating such survival under different stress conditions remain elusive. In this study, we overexpressed SOD3 in MSCs (SOD3-MSCs) and evaluated its effect under serum starvation conditions. Nutritional limitation can decrease the survival rate of transplanted MSCs and thus can reduce their efficacy during therapy. Interestingly, we found that SOD3-MSCs exhibited reduced reactive oxygen species levels and greater survival rates than normal MSCs under serum-deprived conditions. In addition, overexpression of SOD3 attenuated starvation-induced apoptosis with increased autophagy in MSCs. Moreover, we have demonstrated that SOD3 protects MSCs against the negative effects of serum deprivation via modulation of AMP-activated protein kinase/sirtulin 1, extracellular signal-regulated kinase activation, and promoted Forkhead box O3a trafficking to the nucleus. Taken together, these results demonstrate that SOD3 promotes MSCs survival and add further evidence to the concept that SOD3-MSCs may be a potential therapeutic agent with better outcomes than normal MSCs for various diseases involving oxidative stress and compromised MSCs survival during therapy.

Placental Superoxide Dismutase, Genetic Polymorphism, and Neonatal Birth Weight

  • Background : The roles of antioxidants in the placenta and genetic susceptibility to oxidant chemicals in relation to neonatal birth weight have not been elucidated. We determined whether the level of placental manganese superoxide dismutase (MnSOD) and its genetic polymorphism plays any role in oxidative stress and neonatal birth weight. Methods : We measured placental MnSOD and determined MnSOD genetic polymorphism among 108 pregnant women who were hospitalized for delivery and their singleton live births in Korea. Main outcome measurements are maternal urinary malondialdehyde (MDA) and birth weight. Results : Maternal urinary concentrations of MDA were significantly associated with neonatal birth weight (P=0.04). The enzyme level of placental MnSOD was also significantly associated with MDA concentration (P=0.04) and neonatal birth weight (p<0.01). We observed dose-response relationships between placental MnSOD and maternal urinary MDA, and neonatal birth weight after adjusting for maternal weight, height, age, and neonatal sex. After controlling for covariates, MnSOD variant genotype increased maternal urinary MDA concentrations (p<0.01) and reduced birth weight by 149 gm (P=0.08). Conclusions : This study demonstrates that the placental level of MnSOD during pregnancy significantly affects fetal growth by reducing oxidative stress, and that genetic polymorphism of MnSOD probably modulate the effects of oxidants on fetal growth.

Formation of Artificial Lipid Membrane and their Photolysis in Mineral Water including Germanium

  • We have attempted to determine the Germanium ion (G $e^{4+}$) effect on the human body by observing the formation of artificial lipid membrane and photolysis in the mineral water containing G $e^{4+}$ ion. The artificial lipid membrane is prepared by using the phospholipid in the Germanium water and the formation efficiency of the liposomes is compared with those obtained in the plain mineral water without G $e^{4+}$ ion. This work shows that the liposomes are formed in the Germanium water better than in the non-Germanium water. The liposomes can be photolyzed by superoxide anion ( $O_{2-}$$^{.}$) produced in the presence of some peptide such as NAT (N-acethyl-L-tryptophan). However, this is inhibited by superoxide dismutase (SOD), and it was found that the activity of SOD on the inhibition of the liposomes oxidative damage is higher in the Germanium water than in the non-Germanium water. It is concluded that the G $e^{4+}$ ion in mineral water helps the formation of new cell as well as elevation of SOD activity for the lipid oxidation.n.

Zinc(II) ion promotes anti-inflammatory effects of rhSOD3 by increasing cellular association

  • Recently, we demonstrated that superoxide dismutase 3 (SOD3) is a strong candidate for biomedicine. Anti-oxidant function of SOD3 was accomplished without cell penetration, and it inhibited the inflammatory responses via non-enzymatic functions. SOD3 has the heparin binding domain associating cell surface. Interestingly, we found that $Zn^{2+}$ promotes transduction effects of recombinant human SOD3 (rhSOD3) by increasing uptake via the heparin binding domain (HBD). We demonstrated an uptake of rhSOD3 from media to cell lysate via HBD, resulting in an accumulation of rhSOD3 in the nucleus, which was promoted by the presence of $Zn^{2+}$. This resulted in increased inhibitory effects of rhSOD3 on NF-{\kappa}B and STAT3 signals in the presence of $Zn^{2+}$, which shows elevated association of rhSOD3 into the cells. These results suggest that an optimized procedure can help to enhance the inflammatory efficacy of rhSOD3, as a novel biomedicine.

Activities of Oxidative Enzymes Related with Oxygen Tolerance in Bifidobacterium sp.

  • To study the relationship between oxygen tolerance and enzyme activity in the oxygen metabolism of bifidobacteria, the activities of catalase, superoxide dismutase (SOD), NADH oxidase and NADH peroxidase from six typical bifidobacteria and other bacteria were assayed by spectrophotometry. Catalase activity was hardly detected in any of the bifidobacteria tested. SOD activity was detected in every species including the Clostridium species. In particular SOD activity was notably high in the aerosensitive Bifidobacterium adolescentis. This fact indicates that SOD activity is not a critical factor to ensure aerotolerance. Aerosensitive B. adolescentis showed very low NADH oxidative enzyme activity whereas other aerotolerant bifidobacteria exhibited considerable activity for the enzymes. It seems that detoxification of $H_2O_2$ by NADH oxidative enzymes might be an important factor in improving for aerotolerant bifidobacteria survival rates in an oxygen environment.

  • Superoxide dismutase (SOD), one of the essential element of the antioxidant defense system, mainly removes $O^{-10}$ $_2$ and also prevents $O^{-10}$ $_2$ mediated reduction of iron and subsequent OH$^{-10}$ generation, which is highly toxic to the organism. Of these SOD enzymes, Cu, Zn-containing SOD (SODI) is an important component of the antioxidant defense system in eucaryotic cells. The SODI enzyme binds one copper and one zinc ion and displays the Greek Key $\beta$-barrel fuld. (omitted)
