Detection of estrogenic hormone 17β-estradiol in soil samples by a recombinant yeast bioassay and supercritical fluid extraction

  • Recombinant yeast estrogenicity (YES) assay was used as a bioanalytical tool in order to screen $17{\beta}$-estradiol in the soil samples collected from different sites of South Korea. Solvent extraction and supercritical fluid extraction (SFE) methods were compared for the extraction of the estradiol from the soils. Most high detection of the estradiol based on YES assay was observed in the soils extracted with methanol. Different types of estrogenic hormones including $17{\beta}$-estradiol were suggested to be possibly exiting in the soils, since the methanol extracts of the soils showed an estrogenic activity that was not observed in the hexane extracts of the soil. SFE extracts showed estrogenic activity in some of the samples but methanol extract showed best activity.

  • Investigations were performed on the effects of extraction temperature, pressure, time on solubility and selectivity of egg yolk lipid and cholesterol, and color and fatty acid composition of the residue in supercritical carbon dioxide(SC-Co$_2$) extraction. Lipid and cholesterol solubility increased as the increase of Co$_2$ density and was found to strongly depend on the extraction pressure rather than the extraction temperature. The relative concentration of cholesterol in the extract increased with an increase in temperature and decreased with an increase in pressure and extraction time. extraction of dried eg yolk for 3hr at 4$0^{\circ}C$/276 bar removed 46.1% of cholesterol from the residual egg yolk with a yield of 63.2%. SC-Co$_2$ extraction produced a lighter color egg yolk with less redness and yellowness. As the extraction time increased, the resultant residual egg yolk became more saturated with fatty acids. SC-$CO_2$ extraction offers a safe, natural method for removing cholesterol from dried egg yolk.

  • 초임계 유체 이산화탄소를 이용하여 그 온도와 밀도가 물고기 기름의 구성성분인 각종 지방산 에스터의 분배계수에 미치는 영향을 규명하였다. 각 지방산 에스터의 분배계수는 초임계 유체의 온도와 밀도에 따라서 상당한 차이를 나타내어 이산화탄소유체에 의한 지방산의 초임계 유체 추출분리 가능성을 확인할 수 있었다. 각 지방산의 분배계수 차이가 크게 나타나는 초임계 유체의 밀도는 0.3 g/mL 내지 0.4 /mL사이의 범위에 분포되어 있었다. 또한 각 지방산은 모두 초임계 유체의 밀도가 낮은 쪽보다 높은 쪽에서 온도에 따른 역행응축현상을 나타내었다.

  • 토종콩으로부터 비배당체 이소플라본을 추출하기 위한 다양한 기계적 및 화학적 방법을 적용하였다. 용매추출법의 침적과 교반, 초음파에너지와 아임계/초임계 수 추출이 포함되었다. 용매의 조성에 의한 추출효율의 증가, 교반에 의한 특정성분의 추출효과의 증대, 초음파에너지에 의한 추출성능의 개선, 용매보다 용해력이 좋은 초임계유체의 적용에 관한 실험을 수행한 결과에 의하면 비배당체 이소플라본의 추출은 초음파에너지에 의한 것이 가장 우수하였다.

  • 본 연구에서는 에탄올추출과 초임계추출(SFE) 조건에 따른 생강 oleresin(GO) 추출물의 Gram 양성균(Bacillus subtilis, Staphylococcus aureus, Listeria monocytogenes)과 Gram 음성균(Pseudomonas aeruginosa, Escherichia coli)에 대한 항균활성을 살펴보았다. GO 에탄올추출은 추출물의 농도가 증가할수록 clear zone이 비례적으로 증가하였다. 이의 항균성은 Gram 양성균이 Gram 음성균보다 크게 나타났으며, 특히 Listeria monocytogenes에서 항균활성이 크게 나타났다. 생강분말과 에탄올의 추출비율이 1:6인 E-III 처리구가 항균활성이 높았다. GO 에탄올추출물의 항균성은$80^{\circ}C$ 조건에서 처리하는 것이 효과적이었고, 추출시간 (1시간~7시간)은 처리구 간에 큰 차이가 없었다. 초임계추출물에 대한 항균성은 Listeria monocytogenes에서 가장 크게 나타났다. 초임계추출물에서는 100 bar $35^{\circ}C$, 250 bar $35^{\circ}C$, 250 bar $65^{\circ}C$ 조건에서 강한 항균력을 보였다. 그리고 용매추출물의 경우 추출조건에 따라서 항균활성에 큰 차이를 나타내지 않았으나, 초임계추출물의 경우 추출압력 100 bar, 500 bar 처리구와 추출온도 $35^{\circ}C$, $65^{\circ}C$ 처리구에서 항균활성이 크게 나타났다.

생지황(生地黃) 증류 추출 약침액과 초임계 유체 추출물의 성분 연구(硏究) (The Study on the Composition of Rehmanniae Radix Extracts by Supercritical Carbon Dioxide Extraction and by Hydrodistillation Extraction)

  • Objectives : The purpose of this study was to investigate the composition of Rehmannia glutinosa's essential oils with Rehmanniae Radix herbal acupuncture Methods : I obtained the essential oils of Rehmannia Radix by hydrodistillation extraction method and supercritical fluid extraction(SFE) method, and then I analyzed those by GC/MS(gas chromatography/mass spectrum). Results : 1. With GC(gas chromatography) and GC/MS(gas chromatography/mass spectrum) analysis. I identified 9 compounds in the Rehmanniae Radix's essential oil obtained through the SFE method. The main compounds were as follows : Hexachloroethane(2.24%), N-Butyl-benzenesulfonamide(2.05%), hexadecanoic acid(1.93%), hexadecanoic acid, ethyl ester(3.49%), 9,12-Octadecadienoic acid(z,z)(2.70%), (9E)-9-Octadecenoic acid(6.14%), ethyl linoleate(4.43%), ethyl oleate(5.80%). 2. I failed to get Rehmanniae Radix's essential oil obtained through the hydrodistillation method. 3. With GC(gas chromatography) and GC/MS(gas chromatography/mass spectrum) analysis. I identified 4 compounds in the Rehmanniae Radix's essential oil obtained through the hydrodistillation method. The main compounds were as follows : Ethylbis(trimethylsilyl)amine(1.04%), 2-(Trimethylsiloxy)benzoic methyl ester(2.65%), Hexadecanoic acid trimethylsilyl ester(12.61%), octadecanoic acid, trimethylsilyl ester(6.28%). Conclusions : The substances by hydrodistillation method may not perfectly match with the substances by supercritical fluid extraction(SFE) method in essential oils extracted form Rehmanniae Radix. But, the main substances was assumed Hexadecanoic acid and octadecanoic acid.

초임계 유체 추출법을 이용한 용담사간탕(龍膽瀉肝湯) 약침액(藥鍼液)의 정유(精油)성분에 대한 연구(硏究) (The Study on the Composition of Yongdamsagan-tang(龍膽瀉肝湯)'s Essential Oil Obtained by Supercritical Carbon Dioxide Extraction)

  • Objectives : This study was performed to analyze the effective components of essential oil obtained from Yongdamsagantang, which has been efficacious against leukorrhea in gynecologic diseases. Methods : I obtained the essential oils of Yongdamsagan-tang by hydrodistillation extraction method and supercritical fluid extraction(SFE) method, and then I analyzed those by GC/MS(Gas Chromatography/Mass Spectrum). Results : 1. The optimum SFE(Supercritical Fluid Extraction) condition was obtained in the following experiment conditions: pressure 200atm, temperature $45^{\circ}C$, duration of extraction 25minutes. 2. With GC(Gas Chromatography) and GC/MS(Gas Chromato- graphy/Mass Spectrum) analysis, I identified 37 compounds in the Yongdamsagan-tang's essential oil obtained through the SFE method. The main compounds were as follows : 3-Methyl-but-2-enoic acid,2,2-dimethyl-8-oxo-3,4-dihydro-2H,8H -pyrano[3,2-g]chromen-3-yl ester(49.81%), (Z)-6-Pentadecen-1 -ol(3.19%), (-)-Spathulenol(2.40%). 3. I identified 4 compounds in the Yongdamsagan-tang's essential oil obtained through the hydrodistillation method. The main compounds were as follows : 3-Methyl-but-2-enoic acid, 2,2-dimethyl-8-oxo-3,4-dihydro-2H,8H-pyrano [3,2-g]chromen-3-yl ester(2.61%). 4. 3-Methy I-but-2-enoic acid, 2,2-dimethyl-8-oxo-3,4-dihydro-2H,8H-pyrano[3,2-g] chromen-3-yl ester, all were identified in both the SFE method and the hydrodistillation method, but the others were not identified in common. 5. I also conducted an additional test in order to examine the essential oil's antimicrobial action against bacteria. Both MIC(Minimum Inhibitory Concentrations) and MBC(Minimum Bactericidal Concentrations) were $0.125mg/m{\ell}$ against N. meningitidis, however MIC and MBC were $1.0mg/m{\ell}$ in antimicrobial action against 12 different genera of bacteria.

  • Pressure and flowrate control systems for a supercritical extraction process are designed and analyzed. To do this, the dynamic model was first set up using the performance equations of control valves, CO2 compressor and the state equation of the supercritical fluid. Using this model, optimum pairs of manipulated and controlled variable which give least steady stat interaction are determined though the relative gain analysis.

초임계추출법을 이용한 브롬계 난연제 화합물 환경성 평가 추출효율 분석 연구 (Quantitative Extraction Analysis of Brominated Flame Retardant Substances Using Supercritical-Fluid Method for Environmental Assessment)

  • 전기 전자재료 중 고분자에 포함된 브롬계 난연제의 분석에 있어 용출이 어려운 4가지 재질 PC(polycar-bonate) PP(polypropylene), PET(poly(ethylene terephthalate)), PBT(poly(butylene terephthalate))에 대하여 추출방법 및 헥산/에세톤, THF, 톨루엔, THF/톨루엔에 대한 용매 특이성과 그 특성을 고찰하였다. 각 고분자에서의 deca-BDE(decabromo diphenyl ether)의 함량을 속슬렛, 초음파. 가속용매, 마이크로파, 초임계 유체 추출법을 사용하여 이들 방법의 효율을 정량분석 하였다. 초음파 추출은 낮은 고분자의 용해도 때문에 낮은 추출효율을 보여 사용한 고분자의 경우에 있어서 추출방법으로는 적합하지 않았으며, 나머지 3가지 방법에서는 톨루엔 사용시 약 80% 이상의 높은 추출효율을 보였다. 초임계 유체 추출은 고분자의 난연제 추출에 시도되지 않았던 방법이었으나, 본 실험에서 PP와 PC의 추출에서는 100%에 가까운 매우 높은 추출효율을 보였다.

Characteristics of Purified Horse Oil by Supercritical Fluid Extraction with Different Deodorants Agents

  • This study investigated the impact of activated carbon, palm activated carbon, and zeolite on horse oil (HO) extracted from horse neck fat using supercritical fluid extraction with deodorant-untreated HO (CON) as a comparison. The yield and lipid oxidation of deodorant untreated HO (CON) were not significantly affected by the three deodorants. However, deodorant-treated HOs exhibited significantly elevated levels of α-linolenic acid (C18:3n3) and eicosenoic acid (C20:1n9) compared to CON (p<0.05), while other fatty acids remained consistent. Zeolite-purified HO demonstrated significantly lower levels of volatile organic compounds (VOCs) than other treatments (p<0.05). Remarkably, zeolite decreased the concentration of pentane, 2,3-dimethyl (gasoline odor), by over 90%, from 177.17 A.U. ×106 in CON to 15.91 A.U. ×106. Zeolite also effectively eliminates sec-butylamine (ammonia and fishy odor) as compared to other deodorant-treated HOs (p<0.05). Additionally, zeolite reduced VOCs associated with the fruity citrus flavor, such as nonanal, octanal, and D-limonene in HO (p<0.05). This study suggests that integrating zeolite in supercritical fluid extraction enhances HO purification by effectively eliminating undesirable VOCs, presenting a valuable approach for producing high-quality HO production in the cosmetic and functional food industries.
