The Preparation and Identification of Hydrolysis Oligosaccharide from White Copra Meal by Yeast Fermentation and Sunflower Seed Enzymes

  • $\beta$-1,4-Mannotriose was prepared b he enzymatic hydrolysis of white copra meal (WCM) and the subsequent elimination of monosaccharides from the resultant hydrolysate with a yeast. The enzyme system from sunflower seed hydrolyzed WCM and produced monosaccharides and $\beta$-1,4-mannotriose without other oligomers at the final stage of the reaction. WCM(50g) was hydrolyzed at 5$0^{\circ}C$ and pH 4.5 for 24 hr with crude enzyme solution (500 mL) from sunflower seed. By the elimination of monosaccharides from the hydrolysis products with a yeast (Candida glaebosa), 8.1 g of crystalline mannotriose was obtained without the use of chromatographic techniques. After 48hr of yeast cultivation, the total sugar content decreased from 4.6% to 3.5%, whereas the average degree of polymerization increased from 2.3 to 3.1.

Comparative Evaluation of Cotton Seed Meal and Sunflower Seed Meal in Urea Molasses Blocks Versus Commercial Concentrate as Supplement to Basal Ration of Wheat Straw with Stall-fed Buffalo Calves

  • Eighteen water buffalo calves of Nili-Ravi breed (about 15 months age and of $147{\pm}12$ kg average body weight and mixed sex) were used with six animals on each treatment. All the animals were fed long wheat straw for ad libitum intake as the basal ration. Animals in group-I were supplemented with ad libitum amount of urea molasses block having cotton seed meal (CSMB) while the animals in group-II were supplemented with ad libitum urea molasses block containing sunflower seed meal (SFMB) and group III animals were supplemented with a fixed amount of commercial concentrate feed (CCF). The experiment lasted for a period of 80 days (April to June). Results revealed a significantly decreased total feed intake (wheat straw+supplements) in group-III ($1,666{\pm}52$ g/h/d) as compared to group I ($2,299{\pm}194$ g/h/d) and group-II ($2,193{\pm}230$ g/h/d). Average daily supplement intakes were $891{\pm}87$; $666{\pm}104$ and $593{\pm}0$ grams per head in group I, II and III, respectively. Supplement intakes among groups were different (p<0.05). Average daily body weight gains (g/h) were $214{\pm}25$, $174{\pm}23 $and $183{\pm}24$ for group I, II and III, respectively. Feed conversion ratio (FCR) was found to be $10.74{\pm}1.12$, $12.60{\pm}0.88$ and $9.90{\pm}1.33$ grams for group No. I, II and III, respectively. The economic net benefit of live weight gain of calves were 7.63, 6.11 and 7.33 rupees/h/d for group No. I, II and III, respectively. Thus SFM can replace CSM and urea molasses blocks can replace commercial concentrates as supplement to basal ration of wheat straw.

Response of Growing and Laying Japanese Quails (Coturnix japonica) to Dietary Sunflower Seed Meal

  • High fibre (CF, 242 g/kg) sunflower seed meal (SSM) was incorporated in standard grower and layer quail diets at 0, 50, 75 or 100 g/kg replacing part of the soybean meal (SBM) and deoiled rice bran (DORB). Each grower diet was offered to 30 quails housed in battery cages from day one to 5 weeks of age. At the end of the growing period, Twelve female quails from each of the four respective dietary groups were transferred to the individual laying cages and their laying performance from 7 to 20 weeks of age were evaluated on diets containing SSM at similar levels as that during the growing period. The results of growth bioassay revealed that the live weight gain, feed intake, nutrient retention and carcass characteristics of quails in different dietary groups did not vary significantly (p>0.05), although the quails fed SSM based diets utilized the feed less (p<0.05) efficiently than those fed the control diet. During the laying period, the egg production and egg quality traits remained unaffected due to dietary treatments but feed required per unit egg production increased (p<0.05) as the level of SSM in diet was increased to 100 g/kg. Although, the cost of feed per kg decreased due to the incorporation of SSM in diet but the same was not reflected in feed cost per unit gain, meat or egg production as such parameters were comparable on all the diets. It could thus be concluded that the SSM can be incorporated in quail diets upto 100 g/kg for part of SBM, an item usually expensive and also in short supply in developing countries including India.

유기 반추동물 전용 수입산 유기사료 및 국내산 유기 부산물의 사료적 가치 평가 (Nutritional Evaluation of Imported Organic Feeds and Locally Produced Agricultural By-products for Organic Ruminant Farming)

  • This study was conducted to evaluate the nutritional value of locally produced organic agricultural by-products to substitute imported organic feeds for organic ruminant farming. Imported organic feeds (corn grain, soybean meal, soybean seed, oat grain, barley grain, wheat grain, buckwheat, sunflower seed meal) and byproducts (rice bran, grape seed meal, rice straw, soybean hull, soybean curd, rice hull, green kernel rice, and crushed rice grain) were analyzed for chemical composition and NDF, ADF, mineral, and amino acid contents and anti-nutritional factors. Dry matter, NDF and ADF contents in organic feeds were higher than those in conventional feeds. Especially, the 9.65% fat content of organic soybean meal was 6 times higher than the 1.95% fat content of conventional soybean meal. Fat contents of rice bran, grape seed meal, green kernel rice, and crushed rice grain were 25.66, 6.09, 3.57 and 1.59%, respectively. Protein contents of soybean hull and soybean curd were 14.68 and 19.87%, respectively, which are highest among organic by-products. Levels of aflatoxin in all feeds were below the safety level. Therefore, organic rice bran, green kernel rice and crushed rice as energy source, and soybean hull and soybean curd as protein source could partial replace imported feeds for organic ruminant farming.

Determination and Prediction of the Amino Acid Digestibility of Sunflower Seed Meals in Growing Pigs

  • This experiment was conducted to evaluate the chemical composition and amino acid (AA) digestibility of sunflower seed meal (SFSM) and to use this data to develop prediction equations for estimating AA digestibility for growing pigs. Ten SFSM were collected from five provinces in China. Twelve barrows ($38.8{\pm}4.6kg$), fitted with ileal T-cannula were allotted into two $6{\times}6$ Latin square designs. Each of six experimental periods comprised a 5-d adaption period followed by a 2-d collection of ileal digesta. The ten test diets contained 50% SFSM as the sole source of AA. Another nitrogen-free diet was used to measure the basal endogenous losses of crude protein (CP) and AA. Chromic oxide (0.3%) was used as an inert marker in each diet. There was considerable variation (CV>10%) among the ten SFSM in chemical composition (dry matter [DM]). The concentration of CP and ether extract (EE) ranged from 29.33% to 39.09% and 0.88% to 11.33%, respectively. Crude fibre (CF), neutral detergent fibre and acid detergent fibre ranged from 21.46% to 36.42%, 38.15% to 55.40%, and 24.59% to 37.34%, respectively. There was variation among the ten SFSM in apparent ileal digestibility (AID) and standardized ileal digestibility (SID) for lysine and threonine, which ranged from 63.16 to 79.21 and 55.19% to 72.04% for AID and 67.03% to 82.07% and 61.97% to 77.01% for SID, respectively. The variation in CP and methionine ranged from 60.13% to 74.72% and 74.79% to 88.60% for AID and 66.70% to 79.31% and 77.16% to 90.27% for SID, respectively. Methionine was a good indicator to predict AA digestibility. These results indicate that conventional chemical composition of SFSM was variable (CV>10%) among the ten SFSM (DM). The results of AID, SID and prediction equations could be used to evaluate the digestibility of SFSM in growing pigs.

효소적 가수분해법에 의한 Gum류의 Gel유동특성 변화 (Gel-promoting Ability of the Various Gums by the Purified Enzyme)

  • Affinity chromatography법에 의해 해바라기씨 유래 $\alpha$-galactosidase정제를 수행하여 locust bean gum 증점 대체자원의 개발을 위한 기초연구로서 정제효소처리된 guar gum 과 locust bean gum의 구성당인 galactose와 mannose의 구성비율을 비교 검토하였다. 정제효소처리된 guar gum의 중점효과는 galactose와 mannose비율이 1 : 3.2일 때 가장 증가하였다. 이와같은 비율은 효소적 처리시간을 24시간으로 하였을 때 나타내었다. 해바라기씨 유래 $\alpha$-galactosidase처리에 의한 guar gum으로부터의 galactose의 유리가 증점효과에 영향을 미치는 것으로 사료된다. Xanthan gum과 guar gum의 혼합, xanthan gum과 guar gum 및 정제효소처리된 천연 유래 copra meal과의 혼합물에 대한 증점효과도 비교하였다.

Rumen Degradability and Small Intestinal Digestibility of the Amino Acids in Four Protein Supplements

  • The supplementation of livestock feed with animal protein is a present cause for public concern, and plant protein shortages have become increasingly prominent in China. This conflict may be resolved by fully utilizing currently available sources of plant protein. We estimated the rumen degradability and the small intestinal digestibility of the amino acids (AA) in rapeseed meal (RSM), soybean meal (SBM), sunflower seed meal (SFM) and sesame meal (SSM) using the mobile nylon bag method to determine the absorbable AA content of these protein supplements as a guide towards dietary formulations for the dairy industry. Overall, this study aimed to utilize protein supplements effectively to guide dietary formulations to increase milk yield and save plant protein resources. To this end, we studied four cows with a permanent rumen fistula and duodenal T-shape fistula in a $4{\times}4$ Latin square experimental design. The results showed that the total small intestine absorbable amino acids and small intestine absorbable essential amino acids were higher in the SBM (26.34% and 13.11% dry matter [DM], respectively) than in the SFM (13.97% and 6.89% DM, respectively). The small intestine absorbable Lys contents of the SFM, SSM, RSM and SBM were 0.86%, 0.88%, 1.43%, and 2.12% (DM basis), respectively, and the absorbable Met contents of these meals were 0.28%, 1.03%, 0.52%, and 0.47% (DM basis), respectively. Among the examined food sources, the milk protein score of the SBM (0.181) was highest followed by those of the RSM (0.136), SSM (0.108) and SFM (0.106). The absorbable amino acid contents of the protein supplements accurately reflected protein availability, which is an important indicator of the balance of feed formulation. Therefore, a database detailing the absorbable AA should be established.

Rumen Degradability and Post-ruminal Digestion of Dry Matter, Nitrogen and Amino Acids of Three Protein Supplements

  • This study evaluated the in situ ruminal degradability, and subsequent small intestinal digestibility (SID) of dry matter, crude protein (CP), and amino acids (AA) of cottonseed meal (CSM), sunflower seed meal (SFSM) and distillers dried grains with solubles (DDGS) by using the modified three-step in vitro procedure. The ruminal degradability and subsequent SID of AA in rumen-undegradable protein (RUP-AA) varied among three protein supplements. The result show that the effective degradability of DM for SFSM, CSM, and DDGS was 60.8%, 56.4%, and 41.0% and their ruminal fermentable organic matter was 60.0%, 55.9%, and 39.9%, respectively. The ruminal degradable protein (RDP) content in CP for SFSM, CSM, and DDGS was 68.3%, 39.0%, and 32.9%, respectively, at the ruminal solid passage rate of 1.84%/h. The SFSM is a good source of RDP for rumen micro-organisms; however, the SID of RUP of SFSM was lower. The DDGS and CSM are good sources of RUP for lambs to digest in the small intestine to complement ruminal microbial AA of growing lambs. Individual RUP-AA from each protein source was selectively removed by the rumen microorganisms, especially for Trp, Arg, His, and Lys (p<0.01). The SID of individual RUP-AA was different within specific RUP origin (p<0.01). Limiting amino acid was Leu for RUP of CSM and Lys for both RUP of SFSM and DDGS, respectively. Therefore, different protein supplements with specific limitations should be selected and combined carefully in growing lambs ration to optimize AA balance.

해바라기박을 이용한 향미유의 변향특성 (Characteristics of Flavor Reversion in Seasoning Oil using Sunflowerseed Meal)

  • 해바라기박을 이용하여 직화법과 autoclaving법에 의하여 2종의 향미유를 제조하고 이의 특성을 살펴보았다. 직화법으로 얻은 원액의 색상, 향미 등이 상대적으로 autoclaving법으로 얻은 원액에 비하여 강하였다. 대조군으로 사용한 참기름에 비하며 향미유의 향미는 낮은 것으로 확인 되었으나 산가는 0.452, 0.463으로 참기름의 1.987보다 월등히 낮은 것으로 측정되었고 Lovibond color는 3종의 시료유에서 거의 유사하게 나타났다. 이들 향미유와 대조군으로 직접 제조한 참기름을 대상으로 정유성분의 휘발성 향기성분조성 및 함량을 측정해 보았다. 그 결과 총 휘발성 성분 함량은 참기름이 1,300.6 ppm, 직화법으로 제조한 향미유(A type)가 697.8 ppm, autoclaving법으로 제조한 향미유(B type)는 648.2 ppm으로 참기름 의 약 53.65% 및 49.84% 수준의 휘발성 성분을 함유한 것으로 확인되었다. 주요 휘발성 성분은 2-butanone, hexanal, methyl pyrazine 등이었으며, 참기름은 pyrazine류가 전체의 50% 이상을 차지하였으나 향미유에서는 이와 같은 특정성분에 의존하기 보다는 전체적으로 분산되는 경향을 나타내었다.

The Effect of Protein Source and Formaldehyde Treatment on Growth and Carcass Composition of Awassi Lambs

  • A trial with twenty-four newly weaned Awassi lambs (initial body weight=21.5$\pm$0.8 kg) was conducted using a 3$\times$2 factorial design to study the effect of feeding three sources of protein supplements (soybean meal (SBM), sunflower seed meal (SSM), and cottonseed meal (CSM)), either untreated or formaldehyde-treated on the growth performance and carcass traits of Awassi lambs. Lambs were randomly assigned to one of the six diets (4 lambs/treatment diet) and were individually fed for a period of 107 days. Experimental diets were isonitrogenous and isocaloric. Final live weight and average daily gain (ADG) were affected by both source of protein and formaldehyde treatment (undegradable protein). Lambs fed untreated diets had better (p<0.01) daily gain compared to those fed formaldehyde-treated diets. Similarly total feed intake per animal was significantly (p<0.05) affected by protein source and formaldehyde treatment. Formaldehyde treatment caused a significant decrease (p<0.01) in feed intake compared to lambs fed untreated diets. Feed requirement per unit of gain was not affected by formaldehyde treatment during all periods of the experiment except for the second period (the second 28 day period), whereby untreated SBM, SSM and CSM had better feed conversion ratio (FCR) than the treated groups. Source of protein had a moderate effect (p<0.10) on FCR but had a significant effect (p<0.05) on hot and cold carcass weight, digestive tract empty weight and liver weight, with lambs fed SBM having higher values than lambs fed SSM and CSM diets. Supplementation with undegradable protein had a significant effect (p<0.05) on dressing-out percentage (p<0.05), final live weight, and hot and cold carcass weight (p<0.01). The lower values pertain to lambs fed treated diets compared to lambs fed untreated diets. In general, there were no significant differences among all carcass linear dimensions, carcass cut weights and dissected loin tissue weights for both treatments (protein source and formaldehyde treatment). Supplementation with undegradable protein but not the source of protein resulted in significantly higher dissected leg total bone weight (p<0.05), tibia and femur weight (p<0.05), and femur length (p<0.01) at the same carcass weight. Results suggest that the treatment of SBM, SSM and CSM with formaldehyde did not improve efficiency of feed utilization, lamb performance or carcass traits and that the SBM diet resulted in an increase in lamb performance compared to other experimental diets.
