NMR Spectroscopy에 의한 Sulfonamide의 정량(定量) (Determination of Sulfonamides by NMR Spectroscopy)

  • A rapid and accurate NMR procedure for the analysis of a mixture of sulfametrol, sulfalene and sulfameter in $CDCl_3$ and $dimethylsulfoxide-d_6$ was developed. Thioacetamide was used as an internal standard. The signals chosen for the analysis were a singlet of 4.03ppm due to the methoxyl group of sulfametrol, a singlet of 3.98ppm due to the methoxyl group of sulfalene and a singlet of 3.83ppm due to the methoxyl group of sulfameter. The quantification of the characteristic peak was done by the integration method and the peak height method. The variation coefficient of sulfametrol, sulfalene and sulfameter were 1.7(n=6), 1.7(n=6) and 1.4%(n=6), respectively in the peak height method and 8.9(n=6), 9.4(n=6) and 7.0(n=6), respectively in the integration method.

  • The European Economic Community four plate test(EEC 4-plate test, FPT, EU) has been used for monitoring antimicrobial drug residues in meat by Local Veterinary Service Center in Korea. This study was performed to evaluate sensitivity and group specificity of some antimicrobial drugs in FPT and to compare FPT with Charm II test. The minimal detectable levels of targeted antimicrobial drugs tested with standard solutions were 0.025∼1.0 ppm for 7 beta-lactams, 0.5∼1.0 ppm for 4 aminoglycosides, 0.05∼0.5 ppm for 5 macrolides, 0.05∼0.25 ppm for 3 tetracyclines and 0.25&1.0 ppm for 6 sulfonamides. In comparison of FPT and Charm II test, the results of FPT were not accord with those of Charm II test having the group specificity of seven families of antimicrobial drugs in meat samples except some families like tetracyclines.

서울에서 도축된 소의 잔류항생물질 비교조사 (Comparison of residual antibiotics in slaughtered cattle in Seoul)

  • This survey was carried out to detect the residual antibiotics in muscles of slaughter cattle from slaughter houses in Seoul from 1991 to 1998 using by EEC-4-plate method. The results were summarized as follows ; 1. Residual materials were detected in 402 samples(3.12%) by EEC-4-plate method. The detection ratios were highest in 1995 (9.51%), autumn (39.8%), Kyeonggi province (54.2%), Holestain(60.4%) and male(50.7%). 2. Residual antibiotics for 18 samples were classified as TCs(72.2%), sulfonamides(38.8% ) and $\beta$-lactams(5.5%) by HPLC 3. The residual concentration of oxytetracycline, sulfonamide and $\beta$-lactams were 0.34~15.93ppm, 0.17~1.18ppm and 0.06ppm, respectively.

  • Sulfonamides, a therapeutically important group of antimicrobial drugs, are widely used to treat and prevent the acute systemic and skin infections in dairy cattle. They also pose an economic hazard through inhibition of growth of dairy starter cultures. This study was carried out to compare four screening methods for detection of sulfamethazine in milk and determine the positive milk sample by HPLC method. Sulfamethazine was used to spike at five levels of sulfamethazine. The Lac-Tek test and CharmII test were also consistent better than TTCII test and Delvo SP test in sulfamethazine detection. Analysis probabilities of obtaining a positive response with TTCII test and Delvo SP test assay at 50 ppb sulfamethazine level in milk samples were only 14%, 42% each. Whereas using the Lac-Tek test and CharmII test would have resulted in 100% identification of the five levels. Determination of sufamethazine using the HPLC method in the spiked milk were 10.64, 19.30, 30.76,38.83 and 50.23 ppb, respectively.

Determination of Sulfur-containing Drugs by Gas Chromatography with Flame Photometric Detector

  • Such sulfur-containing drugs as chlorpromazine, sulfonamides and thiazides were determined by gas chromatography with flame photometric detector (FPD) and flame ionization detector (FID). 0.6% QF-1 or 3% SE-30 on Chromosorb WAWDMCS were found suitable for drugs listed above. In the experimental conditions given maximum response the FPD response and the response ratio of FPD increased with the number of sulfur atom per molecule. It was possible to obtain a linear calibration curve from $10^{-9}$ / mole to $2{\times}10^{-8}$ / mole and to analyze variety of sulfur drugs by the calibration curve of one sulfur standard.

  • Poly(N,N'-Dichloro-N-ethyl-benzene-1,3-disulfonamide) (PCBS) and N,N,N',N'-Tetrachlorobenzene-1,3-disulfonamide (TCBDA) were found to be a mild and efficient reagent for the direct oxidative conversion of sulfur compounds to the corresponding arenesulfonyl chlorides in good to excellent yields through the oxidative chlorination. The overall process is simple, practical, and it provides convenient access to a variety of aryl or heteroarylsulfonyl chlorides. The mild reaction conditions and the broad substrate scope render this method attractive, and complementary to existing syntheses of aryl or heteroarylsulfonyl chlorides.

  • For the development of new antiulcer agents 5, 6-dihydroimidazo[2, 1-b]- thiazoles substituted at the 3-position are sythesized. Thus, the reaction of 3-chloromethyl-5, 6-dihydroimidazo[2, 1-b]thiazole(2) with thiourea and subsequently with 3-chloro-propionitrile gives 3-[3-[5, 6-dihydroimidazo[2, 1-b]thiazolyl]methylthio]propionitrile(4), which by partial alcoholysis with methanol is converted into methyl-3-[3-[5, 6-dihydro-imidazo[2, 1-b]thiazoyl]methylthio]propionimidate(5) . This compound(5) is treated finally with sulfamide or sulfonamides. 3-[3-[5, 6-dihydroimidazo[2, 1-b]thiazoyl]methylthiol-N$^{2}$-sulfamoyl-propionamidine(6) inhibited gastric acid secretion (45%) when administered intraduodenally (100 mg/kg) to pylorus-ligated rats.

Synthesis and Evaluation of the Analgesic and Antiinflammatory Activities of O-Substituted Salicylamides

  • The present investigation deals with the synthesis of some new salicylamidoacetyl sulfonamides 3a,b, salicylamido ethylacetate 4, salicylamido acetic acid hydrazide 5, which is considered as the key intermediate for the synthesis of several series of new compounds such as salicylamido pyrazol 6 and pyrazolone 1. N-imido-derivatives 9, 10, 11, thiadiazole 13, oxadiazole 14, 15, Schiffs bases 16a-f. Cyclocondensation of Schiffs bases with thioglycolic acid gave thiazolidinone 18a-c while with acetylchloride afforded azitidinones 19a-c and with acetic anhydride gave 1,4-benzoxazepine-3,5-dione. Some of the compounds were tested for their analgesic and antiinflammatory activities as well as ulcerogenic effects. Some derivatives were more effective than salicylamide and ulcerogenic activity was variably lowered .

  • This study was undertaken to determined the sulfamethazine residues In urine, serum and muscle of slaughtered pigs(n: 230) by the method of TLC, EEC-4-plate and HPLC. 1. Inhibition diameter characteristics of reference bacterial substance by EEC-4-plate method and antibacterial inhibition zone was appeared sulfonamides in BS pH 7.2 2. Residual sulfamethazine was detceted from 7 serum(3.04%) and 10 urine(4.35%) by TLC test and concentration of residues was higher In urine than serum. 3. Residual sulfamethazine was detected from 9 muscle(3.91%) by EEC-4-plate method. 4. Positive samples by TLC test and EEC-4-plate method were exceed 0.1ppm quantitative analysis from 7 muscle(3.04%).

  • 4-Arylazo-3-phenyl-5-aminopyrazoles (5a, b) and substituted hydroxythiazoles 8a, b were synthesized from the reaction of 4a, b with hydrazine hydrate and mercaptoacetic acid respectively. Compounds 5a, b and 8a, b were also obtained from coupling of 2a, b with 6 and 7, respectively. 4H-1, 4-Benzothiazine 11 was prepared from 1 and 10. The resaction of the diazonium salts 2a-c with diethyl 3-amino-2-cyanopenet-2-en-1, 5-dicarboxylate 12 was also reported.
