  • The purpose of this study was carried out to investigate the cause of sulfur dioxide occurrence, general element composition, sulfur compounds, heavy metals, macro- & micro-minerals, and oxidation-reduction potential (ORP) following baking time course of RS (RS1, RS2, RS3, and RS4) and mudflat solar salts (MSS). Sulfur dioxide ($SO_2$) and sulfite ($SO{_3}^{2-}$) were not detected in MSS or RS. However, sulfate ($SO{_4}^{2-}$) content significantly decreased in RS (29,878.15~36,097.45 ppm) compared to that in MSS (35,601.65 ppm). ORP was 181.15 mV in MSS, and 58.55 mV in RS1. Moisture content was 9.34% in MSS and 0.00% in RS with increased NaCl (94.77~95.77%). Moisture and NaCl contents showed no significant difference in RS. Insoluble and sandy residues were higher in RS than in MSS, whereas Ca and K showed no significant difference. Mg and Cl contents were higher in RS than in MSS. Br level was higher in MSS (628.1 ppm) than in RS (512.72~586.62 ppm), but there was no significant difference in $NO_3$. Heavy metals (Pb, As, and Hg) were more abundant in RS than in MSS, but levels were still safe. These results suggest that MSS and RS may increase protection against from $SO_2$ and $SO{_3}^{2-}$.

  • Periodontal ligament(PDL) cells and human gingival fibroblasts(HGFs) play important roles in development, regeneration, normal function, and pathologic alteration. PDL cells and HGFs have the similarity related with general characteristics of fibroblast such as spindle shaped morphology, the presence of vimentin intermediate filament and the synthesis of interstitial collagens and fibronectin. There were many studies about the differences between PDL cells and HGFs, but they were not about whole gene level. In this study, we tried to explain the differences of gene expression profiles between PDL cells and HGFs, and the differences among three individuals by screening gene expression patterns of PDL cells and HGFs, using cDNA microarray. Although there were some variants among three experiments, a set of genes were consistentely and differentially expressed in one cell type. Among 3,063 genes, 49 genes were more highly expressed in PDL cells and 12 genes were more highly expressed in HGFs. The genes related with cell structure and motility were expressed more highly in PDL cells. These are cofilin 1, proteoglycan 1 secretory granule, collagen type I(${\alpha}$ 1), adducin gamma subunit, collagen type III(${\alpha}$ 1), fibronectin, lumican(keratan sulfate proteoglycan), and ${\alpha}$ -smooth muscle actin. Tissue inhibitor of metalloproteinase known as the enzyme controlling extracellular matrix with matrix metalloproteinase is more highly expressed in PDL cells, osteoprotegerin known as osteoclastogenesis inhibitory factor is more highly expressed in HGFs. We performed northern blot to verify cDNA microarray results on selected genes such as tissue inhibitor of metalloproteinase, fibronectin, osteoprogeterin. The result of northern blot analysis showed that each cell expressed the genes in similar pattern with cDNA microarray result. This result indicates that cDNA microarray is a reliable method in screening of gene expression profiles.

  • Asparagine biosynthesis by soybean sprouts grown under the dark conditions has been demonstrated. The amount of free asparagine synthesized in ten day-old soybean sprouts increases to 22.7% on the dry weight base. The effects of nitrogen compounds such as $NH_4Cl,\;(NH_4)_2SO_4$ and urea on asparagine synthesis during the sprouting were examined and the results showed that urea was more effective than other two compounds. Glutamine-dependent asparagine synthetase was partially purified (8.6 folds) through ammonium sulfate fractionation, followed by Sephadex G-150 gel filtration. The enzyme was very labile and required protection by thiol groups or high level of glycerol. The mixture of ATP and $Mg^{++}$ ion also stabilized the enzyme activity. The enzyme utilized glutamine more effectively than ${NH_4}^+$ as an amide donor for the formation of asparagine. The enzyme required L-aspartate (Km=3.1 mM), L-glutamine, ATP and $Mg^{++}$. It showed pH optimum of 7.5 and catalyzed the formation of ${\beta}-aspartyl$ hydroxamate in the presence of L-aspartate, ATP, $Mg^{++}$ and $NH_2OH$ in the reaction mixture.

  • The goal of this study is to provide basic data to establish a foundation for the provision of safe drinkable water. The raw water of natural mineral water was analyzed to determine the quantities of anions (F-, Cl-, NO3-N-, and SO42- ) and cations (Ca2+, K+, Mg2+, and Na+) during the former and latter half of 2016. Analysis of the current quality of the raw water of natural mineral water among domestic manufacturers showed average anions contents of 0.46mg/L of fluorine, 8mg/L of chlorine ion, 1.5mg/L of nitrate nitrogen, and 12mg/L of sulfate ion. While the fluorine content was greater than the water quality criterion of 2.0mg/L at four points, the fluorine level was overall stable. The average cations contents included 21.3mg/L of calcium, 1.0mg/L of potassium, 3.4mg/L of magnesium, and 9.6mg/L of sodium. The chemical characteristics were compared among the major ions, and the results are presented in a piper diagram. The content ratio of cations was in the order of Ca2+> Na+>Mg2+>K+, whereas that of anions was in the order of SO42->Cl->NO3-N->F-. While the cations were slightly scattered, the anions were generally concentrated except for at a few points. The Ca-Na-HCO3 type was dominant overall in water sources from diorite, gneiss, and granite, while the Na-Mg-Ca-HCO3-Cl type was dominant in basalt sources. Mineral water manufacturers source their water under various conditions, including in-hole casing, excavation depth, and contact state of bedrock; even within the same rocky area, some differences in the water quality type can occur. When the depth of the water source was taken into account, the mean anions contents of F-, Cl-, NO3-N-, and SO42- were similar, with no significant differences according to depth. Of the cations, K+ and Na+ showed no significant differences across all the tubular wells, whereas Ca2+ and Mg2+ decreased in content with depth.

  • The potato proteinase inhibitor II (PI-II) protein contains chymotrypsin and trypsin inhibitory site. Among several PI-II genes isolated from genomic library, amino acid sequence deduced from PI-IIT gene has 84% identity with that of the polypeptide chymotrypsin inhibitor (PCI). Therefore a gene fragment having homology with the PCI was cloned into a vector using polymerase chain reaction(PCR) from the potato proteinase inhibitor IIT gene. Two different primers were utilized for cloning; primer A contains NdeI restriction site and 30 nucleotides, which has AUG N-terminal methionine codon, primer B contains BclI restriction site and 28 nucleotides, which has TAG translation stop codon. After PCR, about 160 bp-long DNA fragment was cloned into pRT146, derivative of pUC118, and sequenced. The sequenced NdeI/BclI fragment was moved to pET3a, containing bacteriophage T7 promoter and terminator. The expressed proteins in E. coli BL2l(DE3) were determined on a polyacrylamide gel containing sodium dodecyl sulfate. The expected size of protein deduced from the sequenced gene fragment is about 6,500 dalton whose size was similar to the IPTG-induced protein (6,000 dalton) on a gel. However the expression level was much lower than expected.

  • This investigation was carried out to clarify the changes of pigments and soluble protein, and photosystem II activity in the leaves of barley (${SO}_2$-sensitive) and corn (${SO}_2$-resistant) seedlings induced by the ${SO}_2$ fumigation (10, 50ppm). The pH changes of the leaf extract, the content of sulfite and sulfate, the activities of catalase, peroxidase, and polyphenoloxidase were compared in the leaves of barley and corn seedlings induced by ${SO}_2$ fumigation. The results are summarized as follows: An appreciable effect of pH change of leaf extract by ${SO}_2$ fumigation was observed in barley leaves (pH 6.10 to 5.18), but only a small change occurred in corn leaves (pH 5.66 to 5.50). The same pattern of pH changes was recorded when the solution of 0.2N HCl was added to leaf extract, providing lower buffering capacity of the barley leaves than corn leaves. After 2 hours of exposure to 10 ppm ${SO}_2$, the contents of ${SO}^{2-}_3$ and ${SO}^{2-}_4$ were increased in barley leaves, while only ${SO}^{2-}_4$ increased in corn leaves. After fumigation with 10ppm ${SO}_2$ for 2 hours, barley leaves showed significant decreases in activities of catalase, to 17% peroxidase, to 58%, and polyphenoloxidase, to 88%. Corn leaves showed increases in activities of peroxidase, to 136%, and polyphenoloxidase, to 128%. Absorption spectra of pigments obtained from ${SO}_2$-fumigated leaves were gradually decreased with the fumigation time increases, but the decrease was more significant in barley leaves. Fumigation with 50ppm ${SO}_2$ for 2 hours induced the greatest decomposition in carotenoid, followed by chlorophyll a and then chlorophyll b in barley leaves. The ratio of chlorophyll a/b was decreased from 4.1 to 3.6 in barley leaves, but in corn leaves it was maintained almost a constant level(4.9-4.8). The rate of decomposition of chlorophyll and carotenoid in corn leaves was very slow than those in the barley leaves. Fumigation with 50 ppm ${SO}_2$ for 2 hous, decreased the protein content of barley leaves to 59%, and that of corn leaves to 89%, and the extent of decrease in protein content was greater than that of pigments in barley and corn leaves. The rate of DCIP9dichlorophenol indophenol) photoreduction in ${SO}_2$-fumigated leaves was decreased to 18 and 67% in barley and corn leaves, respectively. However, DCIP photoreduction was considerably recovered about 32 and 92% with the addition of DPC(diphenylcarbazide) as an exogenous electron donor in barley and corn leaves, respectively.

  • This study investigated the effects of vitamin C supplementation on the nutritional iron status of 31 adolescent girls, aged 12-15 years, with low hemoglobin levels. They were randomly divided into four groups, and for two groups daily150mg or 900mg of L-ascorbic acid(AsA) was given in three equal doses at three meals during 9 weeks. To another group daily 60mg iron as ferrous sulfate was given in the same way as AsA. The control group was given sugar placebo. Body iron status was monitored through the determination of Hb, Hct, MCHC, and serum ferritin concentrations. Dietary AsA and iron intakes were measured from food consumption surveys performed by 3-day 24-hour recalls. The amount of absorbed iron was estimated from the model of Monsen et al. The average amounts of food iron for four groups were 12.3- 15.0mg and 11.1 - 18.9mg at initial and at final period of the supplementation trial, respectively. The tentatively estimated amount of absorbed iron was significantly increased in the 900mg AsA and iron supplementing groups, but not in the 150mg AsA and placebo groups. Both Hb and MCHC were improved to above normal levels in all groups except the placebo group. Hct was elevated only in the AsA 900mg group whose Hct was relatively lower than the other groups. Serum ferritin concentrations of the four groups, which were as low as 8.50 - 14.39ng/mL on average at the intial periods, augmented significantly to 20.18ng/mL and 26.63ng/mL in the 900mg AsA and iron groups, respectively. Serum ferritin was not elevated in either the AsA 300mg group or the placebo group. The above data indicated that the daily supplementaion of 150mg AsA to the meals containing 12-15mg iron per day promoted Hb levels of adolescent girls with low Hb, and the 900mg AsA supplementing improved not only Hb level but also body iron store. A supplementation of 60mg iron per day appeared to be slightly more effective in improving the iron status in comparison to the 900mg AsA supplement. • BACKGROUND: Gypsum($CaSO_4{\cdot}2H_2O$) is known as an ideal amendment to improve soil quality of the reclaimed coastal land. Since gypsum has very high concentration of electron acceptor like ${SO_4}^{2-}$, its application might be effective on reducing $CH_4$ emission during rice cultivation, but its effect has not been studied well. METHODS AND RESULTS: The effect of gypsum on $CH_4$ emission and rice growth characteristics was studied by pot test, which was packed by reclaimed paddy soils collected from Galsa, Hadong, Gyeongnam province. Chemical-grade gypsum was applied in two soils having EC 2.25 and 9.48 dS/m at rates of 0, 0.5, 1.0 and 2.0%(wt/wt). $CH_4$ emission was characterized a week interval by closed chamber method during rice cultivation. $CH_4$ emission rate was significantly decreased with increasing salt accumulation and gypsum application levels. With increasing gypsum application, dissolved ${SO_4}^{2-}$ concentration in the leachate water was significantly increased, which might have suppressed $CH_4$ production in soil. Total $CH_4$ flux was dramatically decreased with increasing gypsum application. In contrast, rice yield was increased with increasing gypsum application and then achieved maximum productivity at 1.0% gypsum application in two soils. CONCLUSION(s): Gypsum is a very good soil amendment to suppress $CH_4$ emission in reclaimed coastal paddy soils, and improve rice productivity and soil properties. The optimum application level of gypsum is assumed at ca. 1% to improve soil productivity with reducing effectively $CH_4$ emission during rice cultivation.

  • This study was designed to obtain the mean values of thirteen soil physico-chemical properties for different paddy soil management groups, and to serve the basic information for improving the soil using amendments. Computerized data on the results of detailed soil survey were used in this study. The clay contents in the B horizon of paddy soil management groups were 28.3% in the well adapted type, 11.8% in the sandy textured type, and 26.8% in the newly reclaimed type. Soil pH of B horizon in the paddy soil management groups except poorly drained type and acid sulfate type were higher than those of A horizon. In the river side paddy soils of well adapted type, the clay contents of A and B horizons were 16.8%, 23.1%, respectively, and soil organic matter contents of those horizons were 42g/kg, 18g/kg, respectively. And also available phosphate content of well adapted type was higher than the other types. Frequency of distribution of soil organic matter content levels in the B horizons of sandy textured type and newly reclaimed type were higher in the organic matter content range of below 10g/kg. And those of well adapted, poorly drained, and saline type were higher in the organic matter content range of 10~20g/kg. Correlation coefficients between 13 variables in the B horizon of well adapted type were all highly significant at 1% or 5% level, respectively.

  • Enviromental geochemisty and heary metal contamination at the Kongjueil mine creek were underaken on the basis of physicohemical properties and mineralogy for various kinds of water (surface, mine and ground water),soil, precipitate and sediment collected of April and December in 1998. Hydrgeochemical composition of the water samples are characterized by relatively significant enricant of Ca+Na, alkiali ions $NO_3$ and Cl inground and surfore water, wheras the mine waters are relatively eneripheral water of the mining creek have the characteristics of the (Ca+Mg)-$(HCO_3+SO_4)$type. The pH of the mine water is high acidity (3.24)and high EC (613$\mu$S/cm)compared with those of surface and ground water. The range of $\delta$D and $\delta^{18}O$ values (relative to SMOW) in the waters are shpwn in -50.2 to -61.6% and -7.0 to -8.6$\textperthousand$(d value=5.8 to 8.7). Using computer program, saturation index of albite, calcite, dolomite in mine water are nearly saturated. The gibbiste, kaolinite and smectite are superaturated in the surface and ground water, respectively. Calculated water-mineral reaction and stabilities suggest that weathing of silicate minerals may be stable kaolinite owing to the continuous water-rock reaction. Geochemical modeling showed that mostly toxic heavy metals may exist larfely in the from of metal-sulfate $(MSO_4\;^2)$and free metal $(M^{2+})$ in nmine water. These metals in the ground and surface water could be formed of $CO_3$ and OH complex ions. The average enrichment indices of water samples are 2.72 of the groundwater, 2.26 of the surface water and 14.15 of the acid mine water, normalizing by surface water composition at the non-mining creek, repectively. Characteristics of some major, minor and rate earth elements (Al/Na, K/Na, V/Ni, Cr/V, Ni/Co, La/Ce, Th/Yb, $La_N/Yb_N$, Co/Th, La/Sc and Sc/Th) in soil and sediment are revealed a narrow range and homogeneous compositions may be explained by acidic to intermediate igneous rocks. And these suggested that sediment source of host granitic gneiss colud be due to rocks of high grade metamorphism originated by sedimentary rocks. Maximum concentrations of environmentally toxic elements in sediment and soil are Fe=53.80 wt.% As=660, Cd=4, Cr=175, Cu=158, Mn=1010, Pb=2933, Sb=4 and Zn=3740 ppm, and extremely high concentrations are found are found in the subsurface soil near the ore dump and precipitates. Normalizing by composition of host granitic gneiss, the average enerichment indices are 3.72 of the sediments, 3.48 of the soils, 10.40 of the precipitates of acid mine drainage and 6.25 of the soils near the main adit. The level of enerichment was very severe in mining drainage sediments, while it was not so great in the soils. mineral composition of soil and sediment near the mining area were partly variable being composed of quartz, mica, feldspar, chlorite, vermiculite, bethierin and clay minerals. reddish variable being composed of quartz, mica, feldspar, chlorite, vermiculite, bethierin and clay minerals. Reddish brown precipitation mineral in the acid mine drainage identifies by schwertmanite. From the separated mineralgy, soil and sediment are composed of some pyrite, arsenopyite, chalcopyrite, sphalerite, galena, malachite, goethite and various kinds of hydroxied minerals.
