A New Double-Stranded RNA Mycovirus from Pleurotus ostreatus (ASI 2504)

  • A new virus with a dsRNA genome was isolated and characterized from the Suhan-:neutari strain (ASI 2504) of Pleurotus ostreatus, which was characterized as long and slightly bent with small caps on the stipe of fruit body. Thirty nm isometric viruses with three dsRNA segments (approximately 2.0, 1.84 and 1.82 kb in sizes) were isolated by ultracentrifugation in sucrose gradients. Western analysis of protein extracted purified viruses with anti-virus polyclonal antibody confirmed that viruses have two specific proteins (36 and 68 kDa). Computer analysis of 2.0 kb segment shows that high. sequence identity with RNA-dependent RNA polymerase (RdRp) of partitiviruses, respectively. When compared to other dsRNA mycoviruses in a phylogenetic analysis, OMDV was most related to Pleurotus ostreatus virus 1.

Characteristics and breeding of a new cultivar of Pleurotus ostreatus that is tolerant to envirochanges (느타리 신품종 불량환경내성 '고솔'의 육성 및 자실체 특성)

  • A new commercial strain of oyster mushroom (was developed by hyphal anastomosis, and was improved byhybridization between a monokaryotic strain derived from Pleurotus ostreatus ASI 0635 (Gonji 7ho) and a dikaryotic strain derived from P. ostreatus ASI 0666 (Mongdol). The optimum temperatures for mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $12{\sim}18^{\circ}C$, respectively. When PDA (potato dextrose agar medium) and MCM (mushroom complete medium) were compared, mycelial growth was faster in MCM. Similar results were observed with the control strain P. ostreatus ASI 2504 (Suhan 1ho). Analysis of the genetic characteristics of the new cultivar ('Gosol') showed a different DNA profile from that of the control ASI 2504 strain, when RAPD (raurpDNA) primers URP1, 2, 3, and 7 were used. Fruiting body production per bottle was approximately116 g based on a production performance test. In addition, yields from a farm field trial were stably achieved in an inadequate production enviro. The color of the pileus was blackish gray, and the stipe was long and thick. Therefore, we expect that this new strain will satisfy consumer demand for high quality mushrooms.

Genotypic Identification in Commercial Strains of Pleurotus ostreatus based on AFLP and VCGs (AFLP 분석 및 체세포 불화합성에 의한 느타리 유사품종의 확인)

  • We already reported four groups which contains some similar strains based on URP-PCR in the previous paper. The objective of this study was to confirm those strains by the amplified fragment length polymorphism (AFLP) and vegetative compatibility group (VCG). AFLP analysis showed no difference among these strains except ASI 2595 and 2183 in Weonhyeong group and ASI 2829 in Suhan group. They showed specific DNA bands only in the result of P + AG/M + AAG and P + GT/M + ATG primer combinations out of eight different combinations. The AFLP primers produced a total of 330 fragments between 80 and 1000 bp in length for 31 Pleurotus ostreatus strains. At a genetic similarity of 0.96, the UPGMA analysis separated the isolates into four distinct clusters. Each group was classified by similar strains. Confrontation test by vegetative compatibility groups (VCGs) also showed distinct line between strains from different groups, but no line between similar strains within the cluster. Our results indicate that most of similar strains was not distinctness. Thus, similar strains are considered to be very close on the genealogy of their parent or same strain with different name.

Substitution effect of PEFB(Palm Empty Fruit Bunch) for beet pulp in bottle cultivation of Pleurotus ostreatus (느타리(Pleurotus ostreatus) 병재배시 분쇄 열매탈과팜이삭(PEFB : Palm Empty Fruit Bunch)의 비트펄프 대체효과)

  • This study was carried out to develop the new mushroom cultivation medium for Pleurotus ostreatus using by EFB. Two strains of Pleurotus ostreatus were used for this study and the difference in productivity was observed in each strain. There was not confirmed a mycelial growth inhibition by addition of Empty Fruit Bunch which is palm tree waste. In the productivity test of the mixed medium, In Suhan 1ho, yield of T2 treatment was highest as 90.6 g per bottle, It was 14.9% higher than control. In Kimje variety which is Chunchu strain, Yield of T2 treatment was highest as 139.8 g per bottle. It was 25.2% higher than control. In Kimje variety, as amount of 6mm diameter PEFB pellet is increased in medium, yield of mushroom was decreased. When 8mm diameter EFB was used for 532 medium, yield of both 50% EFB(T3) and 100% EFB (T5) treatment was better than other treatments. Therefore, it is suggested that 8 mm diameter EFB pellet is good material for mushroom cultivation medium and T5 treatment was showed a potential possibility as an alternative medium.

Differentiation Among Commercial Strains of Pleurotus spp. Based on DNA Fingerprinting Using Universal Rice Primer (URP) (Universal Rice Primer(URP)에 의한 DNA 핵산지문법을 이용한 느타리의 유통 품종간 구분)

  • To distinct the commercial strains in Pleurotus spp., 81 strains in eight Pleurotus species were used. DNA fingerprinting using URP-PCR was conducted to determine the phylogenetic relationships among Pleurotus strains. DNA profiles of Pleurotus species obtained by twelve URP primers were analyzed for genetic similarity by NTSYS program. We could divide strains into ten clusters, in which three of them belong to P. ostreatus and the others to the different species, respectively. At the 76% similarity level, 70 P. ostreatus strains were distinguished into three clusters. Cluster I contained 35 strains and some of them showed almost 100% similarity, one strain closely related to Weonhyeong and six strains closely related to Wangheukpyeong. In cluster II, twenty-one out of 23 strains showed 100% to Suhan. Cluster III contained twelve strains, including six strains closely related to Chunchu-2. The results suggested that there are many same strains with different names in mushroom spawn market.

Breeding a new cultivar of Pleurotus ostreatus, 'Otari' and its characteristics (느타리 신품종 '오타리'의 육성 및 특성)

  • Oyster mushroom is one of the most widely cultivated and consumed mushrooms in Korea, and mechanization and automation of cultivation systems have enabled mass production. Many cultivars have been developed to replace the old ones such as 'Suhan' and 'Chunchuneutari 2 ho,' which have been cultivated for over 20 years. Among these, 'Soltari' was developed in 2015. Although it has excellent quality, its cultivation is challenging and the productivity is somewhat lower. To address these issues, the Mushroom Division at the National Institute of Horticultural and Herbal Science selected the genetic resource KMCC05165 and attempted hybridization between monokaryons from KMCC05165 and 'Soltari(KMCC04940)'. Through repeated cultivation tests and evaluation of fruiting body characteristics, the superior strain 'Po-2019-smj22' was selected and finally named 'Otari'. The optimal mycelial growth temperature of 'Otari' was between 25 and 30℃ and optimal fruiting body growth temperature was between 13 and 18℃. Mycelial growth on PDA medium was best at 25℃, and at the same temperature, mycelial growth was similar across four media: PDA, MEA, MCM, and YM. In 1,100 mL bottle cultivation, the yield was approximately 174 g, which is about 5% higher than the control cultivar 'Soltari', and the number of valid individuals was also higher at about 25. The diameter and height of the pileus were 29.8 mm and 17.6 mm, respectively, slightly smaller than 'Soltari', and the stipe was thin and long with a thickness of 12.2 mm. Additionally, the pileus' lightness index (L index) was 30.7, indicating a darker brown color compared to 'Soltari.' With excellent mycelial growth, ease of cultivation, and high yield, the new cultivar 'Otari' is expected to be widely adopted by domestic oyster mushroom farms.
