Effects of different creep feed types on pre-weaning and post-weaning performance and gut development

  • Objective: This experiment was carried out to determine the effects of different creep feed types on suckling performance and further adjustments to solid feed after weaning. Methods: A total of 24 multiparous sows and their litters were allotted to one of three treatment groups: i) provided highly digestible creep feed (Creep), ii) provided a pig weaning diet (Weaner), and iii) provided sow feed (Sow) as creep feed until weaning. After weaning, a total of 96 piglets were selected for evaluation of post-weaning performance. Results: For pre-weaning performance, the Creep treatment led to a significantly higher feed intake from 14 to 28 d (p<0.05) and higher body weight gain from 21 to 28 d than piglets that were provided other diets. However, after weaning, the Weaner treatment yielded a significantly higher feed intake and average daily gain than other treatments from 0 to 14 d after weaning (p<0.05); Creep treatment tended to generate lower villus heights in the duodenum than the other treatments (p = 0.07). Conclusion: Highly digestible creep feed improved pre-weaning performance, but feed familiarity and grain-based creep feed improved post-weaning performance.

Experimental infection in guinea pig with foot and mouth disease virus

  • In order to obtain information on murine model for foot and mouth disease virus(FMDV) type Asia 1, we studied whether guinea pig was a suitable model for studying FMDV. Apparently healthy 3 months old albino guinea pigs and unweaned 3 days old Swiss albino mice were used for this study. Total of 8 guinea pigs were divided into the infected(n=5) and control(n=3) groups. The incubation period of FMDV in the guinea pigs were roughly 2 days and the viremia persisted for 3 days in the guinea pigs. Mice inoculated with the plasma from control guinea pigs did not show any sign of viremia. The plasma were titrated by virus neutralization test using suckling mice as an indicator host. The mean virus neutralizing antibody titers of infected guinea pig at 3 DPI, 4 DPI and 5 DPI were log$\_$10/2.16, log$\_$10/ 3.39 and log$\_$10/ 3.44, respectively whereas there was no neutralizing antibody titer in control group. The difference between the mortality pattern and mean virus neutralizing antibody titer of infected and that of control group at day 3, 4, 5 were statistically significant(p<0.0l).

  • A polyvalent live Escherochia coli oral vaccine were prepared by the use of 5 field isolates of enteropathogenic E. coli, serotypes 08 : K87, K88a, b ; 0138 : K81, K88a, c ; 0141 : K85a, b, K88a, b ; 0149 : K91, K88a, c and 0157 : K'V17', K88a, c. Some field experiments were conducted to investigate the effectiveness of the polyvalent live E. coli oral vaccine in the prevention of neonatal colibacillosis in piglets suckling orally vaccinated sows. Seventy-nine pregnant sows in an intensive pig farm were vaccinated with oral vaccine 4 weeks prior to farrowing and 19 sows were chosen for control. The piglets suckling vaccinated sows showed a significant protection against neonatal enteric colibacillosis during the 2 weeks observation period from the farrowing but no significant differences in protection between vaccinated and control group were observed with piglets older than 15 days.

  • Congenital tremor (CT) is a sporadic neurodegenerative disease reported in suckling piglets worldwide. Since atypical porcine pestivirus (APPV) was first identified in US in 2015, it has also subsequently detected in Europe and China as a causative pathogen for CT in suckling piglets. Three new-born piglets died from severe tremor was submitted to Chonbuk National University-Veterinary Diagnostic Center (CBNU-VDC) and various tissues (lung, lymph node, brain, intestine) were tested with panpestivirus RT-PCR and APPV NS5B-specific RT-PCR. All of the samples were positive by both of the PCR tests and the partial NS5B sequences of APPV were confirmed by sequencing on the PCR products of APPV NS5B-specific RT-PCR. Therefore, we report the first identification of APPV from a case of CT in suckling piglets in Korea.

A field approach to eradication of porcine epidemic diarrhoea virus in a breeding pig farm: A case-control study

  • Severe outbreaks of porcine epidemic diarrhoea virus (PEDV) have continued to re-emerge worldwide. Because of the high mortality rate of suckling piglets in PEDV outbreaks, the disease causes significant economic losses in the pig industry. The limited pre-existing immunity against this virus is thought to cause an explosive increase in infection in pig farms. This study aimed to evaluate the clinical symptoms of PEDV after intentional exposure (feedback). During the first few days of the outbreak in a breeding pig farm, 14 sows showed watery diarrhoea, and the disease subsequently spread rapidly throughout the barn. Pigs that were intentionally exposed to PEDV (n=251) showed watery diarrhoea (46.6%), reduced appetite (17.5%), and vomiting (6.0%). However, 75 exposed pigs (29.9%) showed no clinical signs of disease. Four weeks after the feedback challenge, 34 sows gave birth to litters of piglets, which survived with no diarrhoea. Five weeks after the start of the outbreak, PEDV was not detected in any of the examined samples, including environmental swabs. Thus, early diagnosis, prompt establishment of herd immunity, and strict application of biosecurity are good practices to reduce the mortality rates among new-born piglets and control economic losses in pig farms showing PEDV outbreaks.

  • This study was based on a survey of 100 sows which were raising in Jeonnam province for 11 months, from February to December 2008 in order to evaluate the medicinal herbs supplementation in sows on the effects of mortality in suckling pigs, and milk quality of sows. The medicinal herbs were supplemented in the feed of sows from 1 weeks before the expected date of delivery to 10 days after the delivery. As comparing stillbirth rate, test group was reduced 9.05%, and control group, 17.37%. It was certain that the medicinal herbs was very effective for reducing stillbirth rate. In comparison of mortality rate of suckling pig, test group was decreased by 1.52% as compared with control group. In analysis of milk components of sows, milk fat was 6.04% in test group and 5.22% in control group. Milk protein contents of test group and control group were 18.06% and 18.36%, respectively, showing no significant differences between two groups. Milk sugar contents showed also no difference between two groups. By milk somatic cell counts, test group ($582{\times}10^3$cells/ml) was found decreased number of milk somatic cell as compared with control group ($688{\times}10^3$cells/ml). These results imply that the medicinal herbs supplementation in sow might be effective for preventing stillbirth and mortality in piglets and mastitis in sow.

Temporal expression profiling of long noncoding RNA and mRNA in the peripheral blood during porcine development

  • Objective: We investigated the temporal expression profiles of long noncoding RNA (lncRNA) and mRNA in the peripheral blood of pigs during development and identified the lncRNAs that are related to the blood-based immune system. Methods: Peripheral blood samples were obtained from the pigs at 0, 7, 28, and 180 days and 2 years of age. RNA sequencing was performed to survey the lncRNA and mRNA transcriptomes in the samples. Short time-series expression miner (STEM) was used to show temporal expression patterns in the mRNAs and lncRNAs. Gene ontology and Kyoto encyclopedia of genes and genomes analyses were performed to assess the genes' biological relevance. To predict the functions of the identified lncRNAs, we extracted mRNAs that were nearby loci and highly correlated with the lncRNAs. Results: In total of 5,946 lncRNA and 12,354 mRNA transcripts were identified among the samples. STEM showed that most lncRNAs and mRNAs had similar temporal expression patterns during development, indicating the expressional correlation and functional relatedness between them. The five stages were divided into two classes: the suckling period and the late developmental stage. Most genes were expressed at low level during the suckling period, but at higher level during the late stages. Expression of several T-cell-related genes increased continuously during the suckling period, indicating that these genes are crucial for establishing the adaptive immune system in piglets at this stage. Notably, lncRNA TCONS-00086451 may promote blood-based immune system development by upregulating nuclear factor of activated T-cells cytoplasmic 2 expression. Conclusion: This study provides a catalog of porcine peripheral blood-related lncRNAs and mRNAs and reveals the characteristics and temporal expression profiles of these lncRNAs and mRNAs during peripheral blood development from the newborn to adult stages in pigs.

부분예산분석을 이용한 오제스키병 발생 농가의 경제적 손실 추정 (Partial Budget Modeling of Economic Losses of Aujeszky's Disease)

  • Aujeszky's disease (AD) is a respiratory, infectious viral illness associated with high mortality, especially in neonatal piglets and has frequently been considered an economically important disease in many endemic countries. Although AD is still occurring in a geographically defined region in Korea, little attention has been paid to the economics of AD. In this study, partial budget technique was used to develop a simulation model to measure financial losses following the disease epidemic in a swine operation utilizing stochastic or deterministic parameters from the literatures and the index case herd of AD occurred in 2005, where available and applicable. For the infected case herd with a 12500-pig, the total economic loss for this operation was estimated to be about 199 million Korean won (95% confidence interval [CI] 148,645,000-250,741,000). Given net loss due to death of a pig at sow level was 119,000 won, total loss for the case herd with 1200 sows accounted for 143 million won (95% CI 92,599,000-193,729,000). The net loss of the death of one pig at growing and fattening level resulted in loss of 46,000 won (95% CI 40,000-53,000) and 126,000 won (95% CI 122,000-131,000), respectively. Taking into account for the number of pigs raised in the case herd, total loss amounted to 8 million won (95% CI 7,167,000-9,347,000) and 12 million won (95% CI 11,959,000-12,891,000), for growers and fatteners, respectively, assuming 63% of saved feed intake when a pig dies halfway through the respective period. Under the model's assumptions, suckling pig mortality was the major factors of loss in estimating the economic consequences (approximately 71.8% of the total loss). The high economic losses of a herd infected with AD suggest that the effective and region-specific control measures should be implemented in disease endemic foci.

우리나라 양돈장에서 Mycoplasma hyopneumoniae 유병율 조사 (Seroprevalence of Mycoplasma hyopneumoniae infection in Korean swine herds)

  • Serum samples of 1,175 pigs from 148 Korean swine farms not using Mycoplasma hyopneumoniae (M. hyo) vaccines were collected for seroepidemiological study of M. hyo infection by indirect ELISA method. Informations of each farm were provided about province where the farm was located and season when blood samples were collected. Then, the selected farms were divided into farm units which had 5 serum samples according to production stages : sow, suckling piglet (<30 days old), nursery pig (30-70 days old), and growing pig (>70 days old). Seroprevalence of M. hyo infection according to production stages, province, and season was investigated by using ELISA-positve rate of the selected samples for each study. This study showed that 85.34% (78.94-91.78%, 95% CI) of farms were positive to M. hyo infection and 34.81% (32.09-37.53%, 95% CI) among pigs were sero-positive to M. hyo infection in Korean swine farms. In the study of seroprevalence by production stage, most farms had sows and growing pigs which were sero-positive to M. hyo infection (sow: 83.05%, growing pigs: 87.72%) and most pigs seemed to be naturally infected by M. hyo at 8-10 weeks of age. Also, M. hyo infection showed seasonal pattern that most pigs were infected in late fall to early winter. However, in the study of seroprevalence by province, there was no significant correlation between province and M. hyo sero-positive rate.
