탁주(濁酒) 양조중(釀造中) 유기산(有機酸) 및 당류(糖類)의 소장(消長)에 관(關)한 연구(硏究) (Studies on the quantitative changes of Organic acid and Sugars during the fermentation of Takju)

  • 1. 탁주(濁酒)의 발효율(醱酵率)을 국자단용법(麴子單用法)($S_1$)과 국자(麴子) 및 미국혼용법(米麴混用法)($S_2$)으로 담금하여 비교(比較)하고 또한 탁주원료(濁酒原料)와 그 술덧중(中)의 일반성분(一般性分)의 소장(消長)을 분석(分析)하였다. 그 결과(結果) i. $S_2$$S_1$에 비(比)하여 다소(多少) 완만(緩慢)한 발효(醱酵)를 하였으나 잔당량(殘糖量)이 훨신 적고 또 총당량(總糖量)이 약(約) 10% 적은 담금이였는데도 주정생성량(酒精生成量)은 거이 같았다. ii. 담금후(後) $3{\sim}4$일(日만)에 $S_1$, $S_2$에서 모두 전주정생성량(全酒精生成量)의 80% 이상(以上)이 생성(生成)되었다. iii. pH는 담금시(時)에 $S_1$$6.0{\sim}6.2,\;S_2$$4.8{\sim}5.2$에서 $1{\sim}2$일후(日後)부터는 양자(兩者)가 다같이 $4.0{\sim}4.2$로 되어 이값을 계속유지(繼續維持)하였다. 2. Paper partition chromatography법(法)에 의(依)하여 탁주원료중(濁酒原料中)의 유기산(有機酸) 및 당류(糖類)를 검색(檢索)하였다. 그 결과(結果) i. 검출(檢出)된 주요(主要)한 유기산(有機酸)으로서는 백미(白米)에서 Fumaric, Malic, Succinie, Citric, Acetic acid등(等)과 국자(麴子)에서 Fumaric, Succinie, Acetic, Citric, malic, oxalic acid등(等)이고 미국(米麴)에서는 백미(白米)에서와 같았으나 특(特)히 Citric acid가 현저(顯著)히 증가(增加)되어 있었다. ii. 당류(糖類)로서는 백미(白米)에서 Glucose, Suorose, Fructose, 및 Raffinose, 미국(米麴)에서는 Glucose와 그리고 백미중(白米中)에 있었던 Sucrose가 없어지고 Isomaltose, Kojibiose및 Sakebiose라고 추측(推測)되는 3개(個)의 당(糖)이 검출(檢出)되었으며 국자(麴子)에서는 Xylose, Glucose, Fructose 및 Kojibiose라고 생각되는 당(糖)이 검출(檢出)되었다. 3. 술덧에서 p.p.c,법(法)에 의(依)하여 검출(檢出)된 주요(主要)한 유기산(有機酸)으로는 Lactic, Succinic, Acetic acid이었고 Citric acid는 $S_2$에서는 담금초(初)부터 검출(檢出)되었으나 $S_1$에서는 후기(後期)에가서 검출(檢出)되었으며 당(糖)은 원료중(原料中)에 없었던 것으로서 Isomaltotriose와 Panose라고 추측(推測)되는 2개(個)의 당(糖)이 검출(檢出)되고 Maltose는 담금후(後) 1일(日)째에서는 검출(檢出)되었으나 그후(後)부터는 검출(檢出)되지 않았으며 검출(檢出)된 당(糖)들 중(中)에서 Isomaltose라고 추측(推測)되는 것이 숙성시(熟成時)까지 가장 뚜렷이 나타났었다. 4. Somogyi법(法)에 의(依)하여 탁주원료중(濁酒原料中)의 당함량(糖含量)과 술덧중(中)의 당소장(糖消長)을 정량(定量)한 바 백미중(白米中)에서는 Sucrose Glucose, Raffinose Fructose, 의 순(順)으로 함유(含有)되여있었고, 국자(麴子)에서는, Fructose,와 Glucose가 거이 동량(同量)이었으며 미국(米麴)에서는 Glucose가 재래미중(在來米中)의 약(約) $25{\sim}30$배(倍)로 증가(增加)되었고 숙성말기(熟成末期)의 술덧중(中)에는 극소량(極少量)의 유리당(遊離糖)이 함유(含有)되여 있을뿐이였다.

한국전통간장의 맛과 향에 관여하는 주요 향미인자의 분석(I) - 일반특성 및 당류와 유기산 분석 - (Analysis of Significant Factors in the Flavor of Traditional Korean Soy Sauce (I) - Analysis of General Characteristics, Sugars and Organic Acids Contents -)

  • This study was carried out in order to investigate general characteristics, sugars and organic acids contents of Korean traditional soy sauce, and to find out possibility of high quality soy sauce production in a short period through high concentration soy sauce making. In this study, we prepared three different types of soy sauce, low concentration soy sauce (Chungjang), high concentration soy sauce and Kyupjang, high quality traditional Korean soy sauce. pH of soy sauce were $4.46{\sim}4.90$ and did not show difference among three samples. Titratable acidity, buffering power and total free acid content were the highest value in Kyupjang. Kyupjang showed the highest contents of salt and pure extract. As the ripening period increased, the salt content increased in Chungjang samples, but decreased in Kyupjang. Reducing sugar contents of Kyupjang, high concentration soy sauce and low concentration soy sauce were 1.13%, 0.76% and 0.53%, respectively. Free sugar in soy sauce were analyzed maltose, glucose, galactose and fructose. Total free sugar content was highest in high concentration soy sauce, however, contents of glucose and fructose were higher in Kyupjang than in Chungjang samples. Galactose was the main free sugar in Chungjang, but glucose was in Kyupjang. Among identified volatile organic acid, acetic acid was present in the highest concentration, and volatile organic acid content was highest in the high concentration soy sauce at 150 days. 20 nonvolatile organic acids were detected in Korean traditional soy sauce. Succinic acid, lactic acid and 2,5-pyridine dicarboxylic acid were the main nonvolatile organic acid in soy sauce.

김치의 혈전용해작용 (Fibrinolysis of Fermented Kimchi)

  • Fibrinolytic activity of fermented kimchi and ingredients used for the preparation were assayed by measuring the lysis area on plasminogen-rich fibrin plate. Fermented kimchi and picked fish sauces from changlan, prwan, and anchovy showed the activity, and the activity of pickled fish sauces were high in the order of pickled changlan, picked anchovy(pickled mulchi), and pickled prawn. However, the activity of kimchi may not be attributed to pickled fish sauce because kimchi containing fish sauce did not possess activities were determined from the samples heated for 30min at 100$\circ$C. There was no changes in activities before and after heat treatment indicating the agents may be components other than protein. Since major changes occur during kimchi fermentation were increased sour taste due to production of organic acids such as lactic, citric, oxalic, and succinic acids, the authentic organic acids present were examined for fibrinolytic activities. The results indicates that the major component posses the activity is lactic acid.

찹쌀의 종류와 전처리를 달리한 약주의 특성 및 휘발성 향기 성분 (Fermentation Characteristics and Volatile Compounds in Yakju Made with Various brewing Conditions; Glutinous Rice and Pre-treatment)

  • 한국의 전통 알코올 음료인 약주를 찹쌀가루, 찹쌀 전분 그리고 누룩을 사용하여 제조하였다. 본 연구에서는 찹쌀의 종류와 전처리의 차이에 따른 발효 특성을 조사하였다. 효모는 Y90-9를 사용하였고 발효제로는 시판되는 개량누룩(SP1800)을 사용하였으며 덧술에 엿기름을 첨가하여 저온에서 발효하였다. 발효기간 중 pH와 당도는 찹쌀가루를 이용한 약주가 찹쌀전분을 이용한 경우보다 높았으며 전처리로 호화를 거친 약주가 높게 나타났고 발효초기 0.3 이상이었던 산도는 발효 종료 후 전처리에 관계없이 비슷한 수준을 나타내였으며, 알코올 함량은 전처리로 호화를 거친 찹쌀가루 약주에서 가장 높게 나타났다. 아미노산 함량은 찹쌀가루를 사용한 경우가 더 높게 나타났고 자외부 흡수와 착색도 또한 찹쌀 가루를 사용한 약주도 같은 결과를 나타내었다. 환원당 함량은 3.5-3.9 mg/mL의 수준이였다. 술에서 맛과 산성도 조정 역할을 하는 유리당의 조성은 glucose > maltose > fructose 순으로 나타났으며 유기산은 검출된 6개 중 succinic acid가 가장 높게 나타났다. 검출된 21개의 휘발성 향기 성분 중 alcohol은 3 종, ester는 17종, 그리고 aldehyde는 1종이 동정되었으며 그 중 부드러운 향을 나타내는 ethyl palmitate가 높게 나타났다. 이상의 결과들로 미루어 호화한 찹쌀가루와 개량누룩을 사용하여 제조한 약주가 알코올의 수율이 높고 풍부한 향미를 갖는 것으로 나타났다.

고추품종을 달리한 전통식 고추장의 숙성중 미생물, 효소활성 및 맛성분의 변화 (Effect of Red Pepper Varieties on the Microflora, Enzyme Activities and Taste Components of Traditional Kochujang during Fermentation)

  • Microbial counts, enzyme activities and taste components of traditional kochujangs prepared with the powders from 4 different varieties of red pepper, were investigated during 90 days fermentation for the industrial production of traditional kochujang. The viable cell counts of anaerobic bacteria in kochujangs did not change remarkably during fermentation, however, aerobic bacterial counts showed a rapid increase up to 90 days of aging. The yeasts in all kochujang samples increased until 60 days of aging and than decreased. After 90 days of aging, the count of aerobic bacteria in Kumtop kochujang was higher than those of others. The activities of liquefying amylase decreased during the aging, but those of saccharogenic amylase increased at 60 days of aging. The activities of neutral protease were higher than those of acidic protease, and increased during the middle and last period of aging. The major free sugars of kochujang were maltose and glucose, and their contents were higher in Hongkwang kochujang. The major organic acids of kochujang were succinic, formic and citric acid, followed by lactic acid. Succinic acid content in kochujang decreased during fermentation, whereas formic and citirc acids were increased. The major free amino acids were serine, proline, glutamic acid, aspartic acid and alanine. Kumtop kochujang contained the highest amount of total free amino acids. Among the nucleotides and related components in kochujang, cytidine-5-monophosphate was the most abundant component at the begining of aging period, while hypoxanthine increased remarkably during fermentation. Hanwang kochujang was higher in the content of nucleotides than others. Capsaicin contents of kochujang decreased during aging and those of Jangter kochujang was higher than that of others. Sensory evaluation showed that Jangter kochujang was significantly better than Kumtop kochujang in overall acceptability, but there were no appreciable differences in color and flavor.

잠재적 미생물 농약으로서 다양한 식물성장 촉진 활성을 가진 siderophore 생산 세균의 분리와 특성 (Isolation and Characterization of Siderophore-Producing Bacteria with Various Plant Growth-Promoting Abilities as a Potential Biocontrol Agent)

  • To develop eco-friendly microbial inoculants, siderophore-producing bacteria were isolated and identified, and their production characteristics and plant growth-promoting abilities were investigated. A strain S21 was isolated from rhizosphere of Korean perilla (Perilla frutescens) and identified as Enterobacter amnigenus by phenotypic properties and 16S rRNA gene sequencing. The highest siderophore production was obtained in a medium containing 0.5% fructose, 0.1% urea, 0.5% K2HPO4 and 0.1% succinic acid. By using this improved medium, siderophore production increased by 2.5 times compared to that of basal medium. The strain S21 showed insoluble phosphate solubilizing, ammonification and antifungal activities, and also produced hydrolytic enzymes (protease and lipase), indoleacetic acid and 1-aminocyclopropane-1-carboxylate deaminase. Our data suggest that E. amnigenus S21 is a potential candidate that can be used as eco-friendly biocontrol agent and biofertilizer.

  • Xanthan gum의 생산력(生産力)이 있는 Xanthomonas SNUF-560 균주(菌株)에서 자외선(紫外線) 조사방법(照射方法)으로 생산능(生産能)이 우수(優秀)한 변이주(變異株)를 선택(選擇)해서 동결건조법으로 보존(保存)했으며 PDA배지(培地)에서 2주(週)마다 이식(移植)하여 생산실험(生産實驗)에 사용(使用)하였다. 최적발효배지(最適醱酵培地)의 검토(檢討)에서 탄소원(炭素源)으로 sucrose 40g/l, 질소원(窒素源)으로 glutamic acid 10mM, 유기산(有機酸)으로 succinic acid 10g/l, $PO_4\;^{-3}\;65mM$일때 각각 좋은 결과(結果)를 얻었으며, 최종적(最終的)으로 배지(培地) 1과 배지(培地) 2에서 각각 1,050cp(12g/l), 900cp(10.5g/l)를 얻었으며 조성(組成)은 다음과 같다. 배지(培地) 1 = sucrose 40g/l, glutamate 10mM, $PO_4\;^{-3}\;65mM(KH_2PO_4\;12.65g/l)$, citrate 2g/l, $MgSO_4{\cdot}7H_2O\;0.2g/l$, $H_3BO_3\;0.006g/l$, ZnO 0.006g/l $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$ 배지(培地) 2 : sucrose 40g/l, $(NH_4)_2SO_4\;2g/l\;PO_4\;^{-3}\;65mM\;(KH_2PO_4\;12.65g/l)$ succinate 10g/l, $MgSO_4{\cdot}7H_2O\;0.02g/l$ $H_3BO_3\;0.006g/l$ $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$.

적변삼의 생물.화학적 특성과 그 발생원인에 관하여 (Studies on the Biological and Chemical Properties of Musty Ginseng Root and its Causal Mechanism)

  • Rusty root of ginseng has been known as one of the limiting factors in ginseng production in Korea. An attempt was, therefore, made to elucidate biological and chemical natures of the rusty root, and the redox Potential of the ginseng cultivated soils were measured and compared with diseased and non-diseased soils. Reddish discoloration was most frequently observed on the epidermis of ginseng root and the pigments were accumulated in all epidermal cells of the diseased lesions. The lower the redox potential of the ginseng cultivated soil was, the more severe the rusty root was observed. Fe content in the diseased epidermis was 3 times higher than that of healthy one. Organic acids such as oxalic, malonic, succinic, and citric acids were also higher in the mss root than in the healthy one. Thin layer chromatogram of phenolic acid fractions obtained from the epidermal cells of the rusty root of ginseng exhibited 3 to 4 unidentified substances not found in the healthy root. Also lignification of the epidermal cells and the activity of phenylalanine ammonia lyase were greater in the rusty root than the healthy root. Colony formation and conidia production of F. solani, And mycelial growth and sclerotium formation of Sclerotinia sp. isolated from ginseng root were suppressed in a nutritionally minimal medium supplemented with water extract of rusty ginseng root epidermis. It is, therefore, suggested that rusty root of ginseng is caused by unfavorable rhizosphere environmental stress or stresses resulting abnormal metabolism in the root as a selfdefence mechanism of non-specific resistance responses.
