  • The durability and anti-microbial stability of plastics, which have been thought to be favorable characteristics, cause ecoloical problems due to non-degradation after disposal. For a possible solution of these ecological and environmental problems, the biodegradable polymers. especially aliphatic polyesters, have been widely investigated.   • Lee, Yung Nok and Chin, Pyung (Dept. of Biology, Korea University, Seoul, Korea) : Action of ascorbic acid indoleacetic acid on the oxidation of succinate and coupled phosphorylation in Chlorella mitochondria. Kor. Jour. Microbiol., Vol.2, No.1, p12-16 (1964) Mitochondria were isolated from Chlorella ellipsoidea and the action of ascorbic acid and indoleacetic acid on the succinate oxidation and coupled phosphorylation in mitochondria suspension were examined. Oxidation of succinate used as substrate, and phosphorylation coupled to oxidation were strikingly enhanced by the addition of ascorbic acid, while in case of indoleacetic acid it were a little. In a view of phosphorylative efficiency, P/O ratio resulting from the addition of ascorbic acid was decreased and it may be considered as the result of a partial oxidation of ascorbate in mitochondria.

  • The quaternary random copolyesters of poly (butylene glutarate-co-adipate-co-succinate-co-terephthalate)(PBGAST) were synthesized and charaterized by $^1H-NMR$ spectrometry, DSC method, and X-ray diffractometry. Thus the melting point trends and crystallization behaviors of PBGAST copolyesters were obviously depended on terephthalate content in copolymers as well as reaction condition.

Histochemical studies on effect of low concentrated carbon monoxide on the caudate nucleus in rat (저농도 일산화탄소가 흰쥐 미상핵에 미치는 영향에 관한 조직화학적 연구)

  • This study was undertaken to investigate the changes of enzyme activities resulted from low concentrated carbon monoxide poisoning on the caudate nucleus in rat. The activities of cytochrome oxidase, succinate dehydrogenase and lactate dehydragenase were observed histochemically, after the experimental animals were poisoned to 100ppm carbon monoxide for 8 hours every day from one day to 16 days. The materials were sliced from coronal section at the level of the optic chiasm and immediately frozen sections of $10{\mu}m$ thickness were cut on the cryostat at $-15^{\circ}C$ and incubated in the medium containing substrate for histochemical detection of cytochrome oxidase, succinate dehydrogenase and lactate dehydrogenase. The sections were mounted in glycerol gelatin and observed under light microscope. It was obtained that cytochrome oxidase activity decreased moderately and succinate dehydrogenase activity showed marked or moderate activity during entire poisoning period and lactate dehydrogenase activity showed marked or moderate activity from one to 8 days but recovered to normal condition at 16th day.

Mechanism of Enzymatic Degradation of Poly(butylene succinate)

  • Poly(butylene succiate) (PBS), poly(butylene succinate-co-L-lactate) (PBSL), and poly(butylene succinate-co-6-hydroxycaproate) (PBSCL) polymers were degraded by lipase $PS^{(R)}$, and the enzymatic degradation mechanism of PBS was analyzed in detail. The enzymatic degradation of PBS gave 4-hydroxybutyl succinate (4HBS) as the main product. An exo-type hydrolysis mechanism was proposed based on this observation. The terminal chain of PBS had conformational similarity to ordinary tri- and diglycerides and could be incorporated as a substrate in the active site of this lipase. The surface adsorption of the lipase was much larger on PBS and its copolymer films than on the other polyester films because the lipase adhered quite strongly to the polymer terminal through a specific adsorption mechanism. Kinetic analysis showed that the total number of surface adsorption points per unit area of PBSL and PBSCL copolymers was larger than that of the PBS homopolymer.

Effects of Polyacetylenes from Panax ginseng on Some Microsomal and Mitochondrial Enzymes

  • Effects of panaxydol, panaxynol and panaxytriol isolated from Panax ginseng C.A. Meyer on some enzyme activities were determined. Activities of ATPase, membrane-bound enzyme from Sarcoma 180 and rat liver were slightly inhibited by panaxydol. Activities of 5'-nucleotidase, membrane-bound enzyme and succinate cytochrome c reductase in mitochonidria from sarcoma 180 and rat livers were significantly inhibited in a dose-dependent manner by panaxynol. The inhibitory effects of panaxydol and panaxynol on succinate cytochrome c reductase activities were more potent than those on 5'-nucleotidase activities and panaxynol was found to be a very potent inhibitor of succinate cytochrome c reductase. Activities of glucose-6-phosphatase in endoplasmic reticulum from Sarcoma 180 and rat livers were not affected by all three polyacetylenes. These results suggested that the inhibitory effects of panaxydol and panaxynol on enzyme activities might contribute to their biological activities.

  • The biodegradable Properties of various polyester resins with different chemical structures have been studied by applying the controlled compost test and soil burial test. Celluose was taken as a fully biodegradable reference resin while PVC and PE were empolyed as non-biodegradable reference chains or ester group were rather easily degraded by hydrolase, meanwhile copolymer type polyesters which contain aromatic rings showed relatively low biodegradability. According to the results from controlled compost test, cellulose(the positive reference) showed 70.6% degradation after 45 days, whereas synthetic poly(butylene adipate-co-succinate), poly(butylene succinate), poly(butylene adipate-co-succinate-co-terephthalate) showed 44.0%, 32.0% and 23.4% degradation respectively. In this regard, it was concluded that biodegradable properties of polymers are largely dependant on the chemical structures constituting the polymers.

  • The separation and purification of succinate are necessary for the succinic acid production by a fermentation process. Among the purification processes, desalination of succinate is inevitable. In this work, electrodialysis was selected as a desalination method and its operating parameters affecting the degree of desalination and energy consumption were examined. Commercialized electrodialysis apparatus was used in this work and its optimum operating parameters were determined by using design of experiment (DOE) method. Voltage, concentration of succinate, and pH were selected as main parameters. Among them, voltage seemed to be the most important one. The final conversion of succinate to succinic acid was calculated when the operating parameters were optimized. Finally, the effect of impurities, such as corn steep oil, yeast extract, and dextrose on the electrodialysis efficiency was also studied.
