Isolation and Structural Determination of Free Radical Scavenging Compounds from Korean Fermented Red Pepper Paste (Kochujang)

  • Sixteen antioxidative active compounds isolated from the EtOAc layer of MeOH extracts of kochujang, Korean fermented red pepper paste, were structurally elucidated as fumaric acid, methyl succinate, succinic acid furan-2-yl ester methyl ester (gochujangate, a novel compound), 2-hydroxy-3-phenylpropanoic acid, 3,4-dihydroxybenzoic acid, 2,3-dihydroxybenzoic acid, 2,4-dihydroxybenzoic acid, 6,7-dihydroxy-2H-chromen-2-one (esculetin), caffeic acid, cis-p-coumaric acid, trans-p-coumaric acid, daidzin, genistin, apigenin 7-O-$\beta$-D-apiofuranosyl($1{\rightarrow}2$)-$\beta$-D-glucopyranoside, apigenin 7-O-$\beta$-Dglucopyranoside, and quercetin 3-O-$\alpha$-L-rhamnopyranoside by mass spectrometry (MS) and nuclear magnetic resonance (NMR) experiments. These compounds were analyzed for the first time as antioxidants from kochujang.

The Changes in Organic Acids and Fatty Acids in Kochujang Prepared with Different Mashing Methods (담금방법을 달리한 고추장의 유기산 및 지방산의 변화)

  • Organic acids and fatty acids of four different Kochujang prepared with different mashing methods were determined by HPLC and GC. Citrate, malate, lactate, succinate, formate, oxalate and acetate were identified. Citrate was found as a major organic acid ($450{\sim}565$ mg%) followed in decreasing order by lactate and malate. Total organic acid content of Meju Kochujang was the highest at the initial fermentation time, while that of koji Kochujang was higher than any other one after 90 days of fermentation. There was a little change of total acid among the treatments. Oleic acid was a major fatty acid ($68.59{\sim}75.38%$) during the fermentation of Kochujang. There was no significant change of fatty acids during the fermentation and among the treatments.

Dynamic Mechanical and Morphological Studies of Styrene-co-Methacrylate and Sulfonated Polystyrene Ionomers Containing Aliphatic Dicarboxylate Salts

  • This study examined the effects of the sodium salts of aliphatic dicarboxylic acids (DCAs) on the dynamic mechanical properties and morphology of two sets of poly(styrene-co-sodium methacrylate) (MNa) and poly(styrene-co-sodium styrenesulfonate) (SNa) ionomers. When the DCA content was relatively low, the ionic moduli of the MNa and SNa ionomers increased but the matrix and cluster glass transition temperature ($T_g$) did not change significantly. The increasing ionic modulus was almost independent of the type of the ionic groups of the ionomer, and the chain length of DCAs. When a large amount of the sodium succinate (DCA4) was added to the MNa and SNa ionomers, the ionic moduli of the two ionomers increased strongly but the matrix and cluster $T_g's$ increased slightly and significantly, respectively. In the case of sodium hexadecanedioate (DCA 16), DCA 16 increased the ionic moduli of the two ionomers. The addition of DCA16 changed the matrix and cluster $T_g's$ of the MNa ionomer slightly, but decreased the cluster $T_g$ of the SNa ionomer significantly with no change in the matrix $T_g$. In addition, the DCA-containing ionomers showed an X-ray diffraction peak indicating the presence of ordered domains of DC As in the ionomers. Hence, DCA4 acts mainly as a reinforcing filler in MNa and SNa systems. In the case of DCA 16, it initially behaved like a filler but also functioned as a preferential plasticizer for the clusters at high content.

Geometrical Characteristics and Reactivities of Tetracoordinated Pd Complexes: Mono- and Bidentate Ligands and Charged and Uncharged Ligands

  • The geometrical structures, atomic charges, and relative energies of tetracoordinated Pd complexes [PdCl3Z (Z = Cl-, Br-, OH?-, H2O, NH3, PH3), PdCl2Z2 (Z = Br-, OH?-, H2O, NH3, PH3), PdZ?2X (Z = Cl-, OH?-, H2O, NH3, PH3; X = oxalate, O2-?CCO2-), and PdZ2Y (Z = Cl?-, OH?-, H2O, NH3, PH3; Y = succinate, CO2-?CHCHCO2-?)] and the ligand exchange reactions of the Pd complexes were investigated using the ab initio second order Mller-Plesset perturbation (MP2) and Density Functional Theory (DFT) methods. The geometrical characteristics of the tetracoordinated Pd(II) complexes with mono- and bidentate ligands, the effects of the atomic charges for the charged and uncharged ligands, the (dz2-p ) interactions between the dz2-orbital of Pd(II) and the p -orbital of bidentates, and the relative stabilities between the isomers of PdCl2Z2 and PdZ2Y were investigated in detail. The potential energy surfaces for the ligand exchange reactions used for the conversions of {[PdCl2(NH3)2] + H2O} to {[PdCl(NH3)2(H2O)]+ + Cl?-?} and {[PdCl2(PH3)2] + H2O} to {[PdCl(PH3)2(H2O)]+ + Cl?-?]} were investigated. The geometrical structure variations, molecular orbital variations (HOMO and LUMO), and relative stabilities for the ligand exchange processes were also examined quantitatively.

Protective Effects of Ginsenoside Rg3 against Cholesterol Oxide-Induced Neurotoxicity in the Rat

  • Ginsenosides are among the most well-known traditional herbal medicines frequently used for the treatment of various symptoms in South Korea. The neuroprotective effects of ginsenoside $Rg_3$ (G-$Rg_3$) on cholesterol-oxide-(CO)-induced neurotoxicity were investigated through the analyses of rat brains. The recently accumulated reports show that ginseng saponins (GTS), the major active ingredients of Panax ginseng, have protective effects against neurotoxin insults. In the present study, the neuroprotective effects of G-$Rg_3$ on CO-induced hippocampal excitotoxicity were examined in vivo. The in-vitro studies using rat cultured hippocampal neurons revealed that G-$Rg_3$ treatment significantly inhibited CO-induced hippocampal cell death. G-$Rg_3$ treatment not only significantly reduced CO-induced DNA damage but also attenuated CO-induced apoptosis. The in-vivo studies that were conducted revealed that the intracerebroventricular (i.c.v.) pre-administration of G-$Rg_3$ significantly reduced i.c.v. CO-induced hippocampal damage in rats. To examine the mechanisms underlying the in-vitro and in-vivo neuroprotective effects of G-$Rg_3$ against CO-induced hippocampal excitotoxicity, the effect of G-$Rg_3$ on the CO-induced elevations of the apoptotic cells in cultured hippocampal cells was examined, and it was found that G-$Rg_3$ treatment inhibited CO-induced apoptosis. The histopathological evaluation demonstrated that G-$Rg_3$ significantly diminished the apoptosis in the hippocampus and also spared the hippocampal CA1, CA3, and dentate gyrus neurons. G-$Rg_3$ also significantly improved the CO-caused behavioral impairment. G-$Rg_3$ itself had no effect, however, on the CO-induced inhibition of succinate dehydrogenase activity (data not shown). These results collectively indicate the G-$Rg_3$-induced neuroprotection against CO in rat hippocampus. With regard to the wide use of G-$Rg_3$, this agent is potentially beneficial in treating CO-induced brain injury.

A Study on the Preparation of PBAST/PVA Double Layered Hollow Microspheres (PBAST/PVA 이중층 중공미세구의 제조에 관한 연구)

  • In this study, using PBAST (poly(butylene adipate-co-succinate-co-terephthalate)) which was eco-friendly biodegradable aliphatic polyester, PBAST/PVA (poly(vinyl alcohol)) double-layered hollow microspheres were prepared with the water/oil/water multiple emulsion ($W_1/O/W_2$) method. The double-layered hollow microspheres were manufactured with the yield of 30.92% when the concentration of polymer PBAST in organic phase was 5 wt%, the concentration of PVA in inner aqueous phase was 5 wt%, the volume ratio of $W_1/O$ emulsion to outer aqueous phase was 1:4.5, and when co-surfactants that had large gap in HLB (hydrophile-lipophile balance) value were used. The bulk density of prepared hollow microsphere was 0.180 g/ml and particle size was $1.5{\sim}3\;{\mu}m$.

The Fabrication of Thermal Sprayed Photocatalytic $TiO_{2}$ Coating on Bio-degradable Plastic

  • For the production of further functional bio-degradable plastic(polybutylene succinate:PBS) with $TiO_{2}$ as photocatalyst, which shows the decomposition of detrimental organic compound and pollutant under ultraviolet irradiation, we attempted to prepare $TiO_{2}$ coatings on PBS substrate by HVOF and plasma spraying techniques under various conditions. The microstructures of coatings were characterized with SEM and XRD analysis, and the photocatalytic efficiency of coatings was evaluated through the photo degradation of gaseous acetaldehyde. The effects of primary particle size and spraying parameters on the formation behavior, photo catalytic performance and mechanical characteristics of the coatings have been investigated. The results indicated that with respect to both the HVOF sprayed $P_{200}$ and $P_{30}$ coatings, the high anatase ratio off 100% can be achieved regardless of fuel gas pressure. On the other hand, the HVOF sprayed $P_{7}$ coating exhibited largely decreased anatase ratio (from 100% to 49.1%) with increasing the fuel gas pressure, which may be attributed to the much higher susceptibility to heat of 7nm agglomerated powder. In terms of photocatalytic efficiency, HVOF sprayed $P_{200}$ and $P_{30}$ coatings seem to predominate as compared to that of plasma sprayed $P_{200}$ coatings owing to the higher anatase ratio. However, the HVOF sprayed $P_{7}$ coatings didn't show the photo catalytic activity, which may result from the extremely small reaction surface area to the photo-catalytic activity and low anatase ratio. Such functional PBS with new roles is expected to cosiderably contribute to the reduction of aggravated environmel problem.

Preparation of Carrageenan Hydrolysates from Carrageenan with Organic Acid (유기산 처리에 의한 카라기난 가수분해물의 제조)

  • This research was carried out for searching the treatment conditions of organic acid and heating to prepare oligomers from the carrageenan. The applied treatments were autoclaving, micraowaving, and ultrasonicating with acetate, citrate, lactate, malate, and succinate. Among several physical depolymerization methods, auo-claving treatment was the most effective for hydrolyzing the carrageenan to low molecular compounds such as oligosaccharides. Citrate or malate was the most effective catalyst in hydrolyzing carrageenan to some oligo-saccharides among 5~7 different organic acids. An acceptable autoclaving condition for hydrolyzing carrageenan to oligosaccharides was to treat for 120 min at 110~12$0^{\circ}C$. The maximum depolymerization ratio produced by autoclaving was about 23.0%. The depolymerized carrageenan prepared by autoclaving at 12$0^{\circ}C$ had oligo-saccharides of 5~7 species.

IN VITRO STUDY ON THE CYTOTOXICITY OF THE DIFFERENTLY STERILIZED DEMINERALIZED BONE POWDER (멸균방법에 따른 탈회동종골의 세포독성 여부에 관한 실험적 연구)

  • Procurement, cutting, cleansing, freezing, freeze-drying, and demineralization of the allogeneic bone must be made under the germ-free stable condition without bacterial and/or viral contamination. Even thought the bone is procured under the germ free condition, we must have confidence on disinfection of all the solutions that come in contact with tissue during the whole procedure. Lots of antibacterial agents have been introduced for chemical sterilization. Recently ethylene oxide gas sterilization or radiation sterilization is frequently selected as a secendary sterilization procedure. The biological and biochemical response of the graft material differs with the type and concentration of the sterilizing agents, and various toxic reactions have been reported due to the graft material itself and the substance released by the chemicals. The authors conducted the Millipore filter test to observe the toxic effect on L929 fibroblasts according to the effect on activity of succinate dehydrogenase, during the secondary sterilization of the demineralized allogeneic bone powder with irradiation or ethylene oxide gas. The result were as follows : 1. Around the copper disk, positive control group, 10mm diameter discoloration was observed. 2. As same as the negative control group, the disk showed no discoloration. 3. The demineralized allogeneic bone which was sterilized with ethylene oxide gas or irradiation showed no cytotoxicity. 4. From this results, it is suggested that treatment with ethylene oxide gas or irradiation should be effective to sterilize the deminineralized allogeneic bone.

Chemical Transformation of Succinic Acid by Using Amberlite IR-120 and the Reusability of Catalyst (Amberlite IR-120를 사용한 숙신산의 화학적 전환과 촉매의 재 사용성)

  • Dioctyl succinates and succinic anhydride were synthesized from succinic acid in the presence of a cation exchange resin, Amberlite IR-120. The synthesis of dioctyl succinate from succinic acid and octanol was perfomed in 18 h reflux condition with Amberlite IR-120 and the isolated yield of product was over 90%. For the synthesis of succinic anhydride, succinic acid was refluxed in isopropenyl acetate for 18 h with Amberlite IR-120 as a catalyst. The long reaction time was significantly reduced to less than 10 min under microwave irradiation. The catalyst was reusable at least 4 times with keeping product yield of higher than 80%.
