Cloning, expression, and activity of type IV antifreeze protein from cultured subtropical olive flounder (Paralichthys olivaceus)

  • Antifreeze proteins (AFPs) lower the freezing point but not the melting point of aqueous solutions by inhibiting the growth of ice crystals via an adsorption-inhibition mechanism. However, the function of type IV AFP (AFP IV) is questionable, as its antifreeze activity is on the verge of detectable limits, its physiological concentration in adult fish blood is too low to function as a biological antifreeze, and its homologues are present even in fish from tropic oceans as well as freshwater. Therefore, we speculated that AFP IV may have gained antifreeze activity not by selective pressure but by chance. To test this hypothesis, we cloned, expressed, and assayed AFP IV from cultured subtropical olive flounder (Paralichthys olivaceus), which do not require antifreeze protein for survival. Among the identified expressed sequence tags of the flounder liver sample, a 5'-deleted complementary DNA (cDNA) sequence similar to the afp4 gene of the longhorn sculpin was identified, and its full-length cDNA and genome structure were examined. The deduced amino acid sequence of flounder AFP IV shared 55, 53, 52, and 49 % identity with those of Pleuragramma antarcticum, Myoxocephalus octodecemspinosus, Myoxocephalus scorpius, and Notothenia coriiceps, respectively. Furthermore, the genomic structure of this gene was conserved with those of other known AFP IVs. Notably, the recombinant AFP IV showed a weak but distinct thermal hysteresis of $0.07{\pm}0.01^{\circ}C$ at the concentration of 0.5 mg/mL, and ice crystals in an AFP IV solution grew star-shaped, which are very similar to those obtained from other polar AFP IVs. Taken together, our results do not support the hypothesis of evolution of AFP IV by selective pressure, suggesting that the antifreeze activity of AFP IV may have been gained by chance.

KRM 모델을 이용한 제주바다목장 해역 주요 우점종의 음향반사강도 추정 (Target strength estimation of dominant species in marine ranching ground of Jeju coastal water by KRM model)

  • The indirect target strength (TS) estimation method which uses the Kirchhoff ray mode model (KRM model) was discussed to apply for a biomass estimation in the water of mixed species. TS of 25 live scorpion fishes for 120kHz were measured by a tethered method and of others dominant 5 species in the marine ranching ground of Jeju coastal water including a scorpion fish were also estimated by KRM model. The measurement TS of scorpion fish well agreed with the theoretical values and the standard formula of scorpion fish was estimated as $TS_{120kHz}=20Log\;(L)-72.9$ ($r^2=0.67$). TScm values estimated on trial to each sample of dominant 5 species were from -69.3dB to -75.1dB at 120kHz and they were in the general range of swimbladdered fish. It was clarified that TS by KRM model can be used to estimate fish biomass estimation by increasing a sample number and is more effective under the condition that there is rare TS information for inhabiting species in mixed-species area.

경남 통영 풍화리 잘피밭의 어류 종조성 변동 (Variations in Species Composition of Fishes in the Eelgrass Beds of Punghwa-ri, Tongyeong, Korea)

  • 경남 통영시 풍화리 잘피밭에서 어류의 종조성 변동을 파악하기 위해 2021년 4월부터 22년 3월까지 지인망(surf net)을 이용하여 조사하였다. 10년전 수행된 선행연구 결과와의 비교에서 종조성과 우점종에 유의한 차이를 나타냈다. 또한, 선행연구에서 출현하지 않았던 아열대성 어종인 독가시치(Siganus fuscescens), 쏠종개(Plotosus lineatus), 두줄베도라치(Petroscirtes breviceps)가 출현하였다.

Response of appetite-related genes in relation to the rearing water temperature in red spotted grouper (Epinephelus akaara)

  • Growth of fish is controlled by various environmental factors, including water temperature (WT). WT is also a major factor that affects the eating behavior of fish. In this study, we studied the relationship between WT and expression profiles of factors involved in appetite regulation and digestion in red spotted grouper. Fish were divided into three groups with 30 fish per group and reared for 2 weeks in a recirculating system at three different WT (natural WT, 20 ℃, and 25 ℃). After 2 weeks, they were sacrificed immediately after food intake (+ 0 h), 3 h after (+ 3 h), 6 h after (+ 6 h), and 3 h before being fed the next day (- 3 h). Fish samples were obtained for analysis of mRNA expression levels of cholecystokinin (CCK), leptin A (LepA), leptin B (LepB), and neuropeptide Y (NPY) in the brain, liver, and intestine. Expression of CCK and NPY mRNA was significantly higher at 25 ℃ compared to that at natural WT and 20 ℃, but no difference was observed for leptin. This suggests that the red spotted grouper shows characteristics typical of subtropical fish and has higher levels of feeding and digestion when reared at 25 ℃ water temperature condition.

제주 연안에 서식하는 쏨뱅이 (Sebastiscus marmoratus)의 식성 (Feeding Habits of Sebastiscus marmoratus in the Coastal Waters of Jeju Island, Korea)

  • The feeding habits of Sebastiscus marmoratus collected from the Western Coastal Waters of Jeju Island between June 2008 and July 2009 were studied based on examination of the stomach contents. A total 254 individuals of S. marmoratus ranged from 13.2 to 29.6cm (mean 20.7cm) in Standard length (SL). S. marmoratus consumed mainly teleosts such as Chromis notata and Sebastes inermis. Its diet also includes shrimps and crabs. The individuals smaller than 15cm SL consumed mainly shrimps, but on the other hand the individuals larger than 25cm SL consumed mainly fishes. The proportion of fish consumption increased with increasing size of S. marmoratus.

독도 주변해약에서 삼중자망으로 어획한 어류의 종조성 및 계절변동 (Seasonal Variation in Species Composition of Fish Collected by Trammel Net Around Dokdo, East Sea of Korea)

  • Seasonal variation in species composition around Dokdo, East Sea of Korea, was investigated using trammel-net catches, from 2006 to 2009. A total of 53 fish species belonging to 23 families in 12 orders were captured; the orders Perciformes (12 families, 22 species) and Scorpaeniformes (four families, 22 species) were dominant. Between 2008 and 2009, 43 species were collected by trammel net. The number of species was highest in August 2009 (25 species) and lowest in February 2009 (11 species). The number of individuals and total biomass peaked in November 2009. Diversity indices for fish catches were highest in August 2008 (2.4368) and lowest in November 2009 (0.4253). The dominant species were Thamnaconus modestus and Sebastes schlegeli. Hierarchical clustering analysis showed five fish groups, with frequency and number of individuals similar to results of correspondence analysis (CA), which showed a closer relationship to the year term than the season term. CA showed that temperature was an important factor influencing fish species richness and abundance. Three main fish assemblage types coexisted around Dokdo: an East Sea coastal fish assemblage, a subtropical fish assemblage, and a cold water fish assemblage.

잠수관찰을 통한 통영 영운리 연안의 어류 종조성 및 계절변동 (Species Composition and Seasonal Variation of Fish by SCUBA Observation in the Coastal Water off Tongyeong, Korea)

  • 이번 연구는 경남 통영시 영운리의 어류 종조성 및 계절변동에 대한 연구를 2016년 10월부터 2017년 9월까지 잠수관찰기법으로 실시하였다. 조사기간 동안 총 5목 35과 45종 5,358개체가 관찰이 되었다. 그 중 농어목(Perciformes)이 29종으로 64%, 쏨뱅이목 (Scorpaeniformes)이 8종 18%, 복어목(Tetraodontiformes) 3종 7%, 차지하였다. 우점종으로는 볼락 (Sebastes inermis)이 차지하였고 아우점종으로는 그물코쥐치 (Rudarius ercodes)였다. 관찰된 종수는 10월에 33종으로 가장 많았고 2월에 4종으로 가장 적었으며 수온이 상승하는 3월 이후에 종수가 증가하였다. 개체수는 2016년 10월에 950개체로 가장 많았으며 이 시기는 주요 출현어종 인상어(Neoditrema ransonnetii), 그물코쥐치(Rudarius ercodes), 흰줄망둑(Pterogobius zonoleucus), 볼락(Sebastes inermis)이 대부분 차지하였다. 특히 아열대어류인 청줄돔 (Chaetodontoplus septentrionalis), 청줄청소놀래기(Labroides dimidiatus), 노랑거북복(Ostracion cubicus) 세 종이 출현하였고 노랑거북복과 청줄청소놀래기는 통영에서 처음 출현하였다.

경상북도 후포와 강원도 장호에서 정치망으로 채집된 어류 종조성 비교 (Comparison of Fish Species Composition Collected by Set Net at Hupo in Gyeong-Sang-Buk-Do, and Jangho in Gang-Won-Do, Korea)

  • Two major temperature fronts, the Subpolar (Gosung, Gang-won-do; $38^{\circ}-41^{\circ}N$) and Thermal (Jukbyun, Gyeong-sang-buk-do; $36^{\circ}-37^{\circ}N$) fronts, are found in the East Sea along the east coast of Korea. These are located roughly where the Tsushima Warm Current and North Korea Cold Current intersect. To clarify the effect of the Thermal Front, we investigated seasonal variation in fish species composition using set nets in two areas located north (Jangho, Gang-won-do) and south (Hupo, Gyeong-sang-buk-do) of Jukbyun, Gyeong-sang-buk-do, and compared the sea water temperature and salinity. We collected a total of 38 fish species in Hupo and 25 in Jangho. Trachurus japonicus was the most common species at both sites, but the subdominant species differed. At Hupo, the subdominant species were Konosirus punctatus and Diodon holocanthus, whereas Clupea pallasii and Scomber japonicus were subdominant at Jangho. Based on Froese and Pauly (2014), subtropical fishes accounted for 55% of fish in Hupo but only for 33% in Jangho. The difference in fish species composition was most obvious in May and August. According to the Korea Hydrographic and Oceanographic Administration, sea surface temperature and salinity were slightly higher at Hupo than at Jangho. Our findings suggest that the oceanographic boundary resulting from the Thermal Front near Jukbyun, Gyeong-sang-bukdo may have a major effect on the distribution of migratory fish species.

Shoalgrass와 turtlegrass에 棲息하는 小型魚類 個體群들의 季節的 變動에 관한 硏究 (Seasonal Variations in Populations of Small Fishes Concentrated in Shoalgrass and Turtlegrass)

  • 아열대 seagrass 어류군집의 계절적 변동을 결정하기 위해, 미국 Texas 남쪽 해안에 위치한 Redfish Bay의 seagrass meadow에서 서식하는 소형어류개체군들의 밀도를 1982~1983년동안 매달 조사하였다. 23과 40종에 속하는 총 10223개체수가 채집되었으며, 평균밀도는 shoalgrass에서 약 15마리/$m^2$ 이었고 turtlegras 에서 약 6마리/$m^2$ 이었다. Darter goby, pinfish, code goby 그리고 Gulf pipefish가 일년을 통해 양 meadows는 우점종으로 나타났다. 이들 네 어종은 총채집된 개체수의 약 85%를 차지하였다. Darter goby는 얕은곳에 위치한 shoalgrass에서 가장 흔한 어종이었으며 약간,은 곳에 위치한 turtlegrass에서는 pinfish와 code goby가 가장 흔한 어종이었다. 어류의 종구성과 밀도의 계절적 변화는 이들 두 아열대 seagrass meadows에서 뚜렷하게 나타난다. 어류의 최대밀도는 봄에 나타난다. 어류밀도는 여름에 감소하나 가을에 다시 증가한다. 최소밀도는 겨울에 나타난다. 네 우점종은 각기 서로 다른 계절적 밀도변화 양상을 보인다. 이들 사이에는 약간의 중첩이 있지만 일반적으로 다른 종들로부터 1~3개월 분리된 최대밀도 시기를 보인다. 이같이 분리된 최대밀도 시기를 보이는 주요 어종의 계절적 개체수변화 양상은 높은 어류밀도를 지닌 아열대 seagrass meadows에서의 이들 주요 어종의 공존을 가능케하는 것으로 사료된다.

Sexual Maturity and Early Life History of the Mudskipper Scartelaos gigas (Pisces, Gobiidae): Implications for Conservation

  • Scartelaos gigas is an amphibious mudskipper species that inhabits mud flats in Korea, China, and Taiwan. This fish is at risk of extinction because of its very restricted habitat and overexploitation. Information about this fish's reproductive characteristics is needed for species conservation. The sexual maturity and early life history of S. gigas were investigated through histological methods and direct observation of eggs in the wild, respectively. In total, 560 individuals of S. gigas were collected with the aid of fishermen from March 2003 to October 2003 at Jung-do Island, southwest Korea. Through microscopic observations of gonadal development, it was determined that S. gigas of both sexes were immature in April, but began to reach maturity in May, and were then fully mature by June, which was maintained until July. In August, some female fish developed early oocytes, but by September oocytes were observed to have degenerated and had been absorbed. Spawned eggs were elliptical and had an average size of 1.37 mm (long axis) by 0.69 mm (short axis). The newly hatched larvae (3.03 mm total length, TL) had an open mouth and anus, two melanophores near the anus, and one large melanophore between the 18th and 19th myomeres. The larvae (3.18 mm TL) showed absorption of the yolk and oil globule within 5 days after hatching and became prelarvae. This species should be considered vulnerable or conservation-dependent, and thus parental fish need to be protected from fishermen during the main spawning season (June).
