Voltage Dependent N Type Calcium Channel in Mouse Egg Fertilization

  • Repetitive changes in the intracellular calcium concentration ([Ca2+]i) triggers egg activation, including cortical granule exocytosis, resumption of second meiosis, block to polyspermy, and initiating embryonic development. [Ca2+]i oscillations that continue for several hours, are required for the early events of egg activation and possibly connected to further development to the blastocyst stage. The sources of Ca2+ ion elevation during [Ca2+]i oscillations are Ca2+ release from endoplasmic reticulum through inositol 1,4,5 tri-phosphate receptor and Ca2+ ion influx through Ca2+ channel on the plasma membrane. Ca2+ channels have been characterized into voltage-dependent Ca2+ channels (VDCCs), ligand-gated Ca2+ channel, and leak-channel. VDCCs expressed on muscle cell or neuron is specified into L, T, N, P, Q, and R type VDCs by their activation threshold or their sensitivity to peptide toxins isolated from cone snails and spiders. The present study was aimed to investigate the localization pattern of N and P/Q type voltage-dependent calcium channels in mouse eggs and the role in fertilization. [Ca2+]i oscillation was observed in a Ca2+ contained medium with sperm factor or adenophostin A injection but disappeared in Ca2+ free medium. Ca2+ influx was decreased by Lat A. N-VDCC specific inhibitor, ω-Conotoxin CVIIA induced abnormal [Ca2+]i oscillation profiles in SrCl2 treatment. N or P/Q type VDC were distributed on the plasma membrane in cortical cluster form, not in the cytoplasm. Ca2+ influx is essential for [Ca2+]i oscillation during mammalian fertilization. This Ca2+ influx might be controlled through the N or P/Q type VDCCs. Abnormal VDCCs expression of eggs could be tested in fertilization failure or low fertilization eggs in subfertility women.

Carbon monoxide releasing molecule-2 suppresses stretchactivated atrial natriuretic peptide secretion by activating largeconductance calcium-activated potassium channels

  • Carbon monoxide (CO) is a known gaseous bioactive substance found across a wide array of body systems. The administration of low concentrations of CO has been found to exert an anti-inflammatory, anti-apoptotic, anti-hypertensive, and vaso-dilatory effect. To date, however, it has remained unknown whether CO influences atrial natriuretic peptide (ANP) secretion. This study explores the effect of CO on ANP secretion and its associated signaling pathway using isolated beating rat atria. Atrial perfusate was collected for 10 min for use as a control, after which high atrial stretch was induced by increasing the height of the outflow catheter. Carbon monoxide releasing molecule-2 (CORM-2; 10, 50, 100 μM) and hemin (HO-1 inducer; 0.1, 1, 50 μM), but not CORM-3 (10, 50, 100 μM), decreased high stretch-induced ANP secretion. However, zinc porphyrin (HO-1 inhibitor) did not affect ANP secretion. The order of potency for the suppression of ANP secretion was found to be hemin > CORM-2 >> CORM-3. The suppression of ANP secretion by CORM-2 was attenuated by pretreatment with 5-hydroxydecanoic acid, paxilline, and 1H-[1,2,4] oxadiazolo [4,3-a] quinoxalin-1-one, but not by diltiazem, wortmannin, LY-294002, or NG-nitro-L-arginine methyl ester. Hypoxic conditions attenuated the suppressive effect of CORM-2 on ANP secretion. In sum, these results suggest that CORM-2 suppresses ANP secretion via mitochondrial KATP channels and large conductance Ca2+-activated K+ channels.

Analysis on the Marketing Efficieny of Paprika (착색단고추의 유통효율성 분석)

  • The purpose of this study is to analyse current marketing practices and marketing efficiency using operational efficiency. Through field survey on marketing situations and channels, we found that major marketing channels of paprika were classified into six types : channels in exporting market included (1) producer's organization leading channel (type I), (2) regional cooperative leading channel (type II), (3) individual producer leading channel (type III), and channels in domestic market included (4) wholesale markets targeting channel (type IV), (5) distribution agents targeting channel (type V), (6) cooperative system selling channel (type VI). According to the estimated result of operational efficiency of marketing channels, marketing efficiency of type III and IV was relatively higher than type I,II and type V, VI respectively.

The agonistic action of URO-K10 on Kv7.4 and 7.5 channels is attenuated by co-expression of KCNE4 ancillary subunit

  • KCNQ family constitutes slowly-activating potassium channels among voltage-gated potassium channel superfamily. Recent studies suggested that KCNQ4 and 5 channels are abundantly expressed in smooth muscle cells, especially in lower urinary tract including corpus cavernosum and that both channels can exert membrane stabilizing effect in the tissues. In this article, we examined the electrophysiological characteristics of overexpressed KCNQ4, 5 channels in HEK293 cells with recently developed KCNQ-specific agonist. With submicromolar EC50, the drug not only increased the open probability of KCNQ4 channel but also increased slope conductance of the channel. The overall effect of the drug in whole-cell configuration was to increase maximal whole-cell conductance, to prolongate the activation process, and left-shift of the activation curve. The agonistic action of the drug, however, was highly attenuated by the co-expression of one of the β ancillary subunits of KCNQ family, KCNE4. Strong in vitro interactions between KCNQ4, 5 and KCNE4 were found through Foster Resonance Energy Transfer and co-immunoprecipitation. Although the expression levels of both KCNQ4 and KCNE4 are high in mesenteric arterial smooth muscle cells, we found that 1 μM of the agonist was sufficient to almost completely relax phenylephrine-induced contraction of the muscle strip. Significant expression of KCNQ4 and KCNE4 in corpus cavernosum together with high tonic contractility of the tissue grants highly promising relaxational effect of the KCNQ-specific agonist in the tissue.

Exploiting W. Ellison model for seawater communication at gigahertz frequencies based on world ocean atlas data

  • Electromagnetic (EM) waves used to send signals under seawater are normally restricted to low frequencies (f) because of sudden exponential increases of attenuation (𝛼) at higher f. The mathematics of EM wave propagation in seawater demonstrate dependence on relative permeability (𝜇r), relative permittivity (𝜀r), conductivity (𝜎), and f of transmission. Estimation of 𝜀r and 𝜎 based on the W. Ellison interpolation model was performed for averaged real-time data of temperature (T) and salinity (S) from 1955 to 2012 for all oceans with 41 088 latitude/longitude points and 101 depth points up to 5500 m. Estimation of parameters such as real and imaginary parts of 𝜀r, 𝜀r', 𝜀r", 𝜎, loss tangent (tan 𝛿), propagation velocity (Vp), phase constant (𝛽), and α contributes to absorption loss (La) for seawater channels carried out by using normal distribution fit in the 3 GHz-40 GHz f range. We also estimated total path loss (LPL) in seawater for given transmission power Pt and antenna (dipole) gain. MATLAB is the simulation tool used for analysis.

Inhibitory effects of isoscopoletin on thrombus formation via regulation of cyclic nucleotides in collagen-induced platelets

  • An essential component of the hemostatic process during vascular damage is platelet activation. However, many cardiovascular diseases, such as atherosclerosis, thrombosis, and myocardial infarction, can develop due to excessive platelet activation. Isoscopoletin, found primarily in plant roots of the genus Artemisia or Scopolia, has been studied to demonstrate potential pharmacological effects on Alzheimer's disease and anticancer, but its mechanisms and role in relation to thrombus formation and platelet aggregation have not yet been discovered. This research investigated the effect of isoscopoletin on collagen-induced human platelet activation. As a result, isoscopoletin strongly increased cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) levels in a concentration-dependent manner. In addition, isoscopoletin greatly phosphorylated inositol 1,4,5-triphosphate receptor (IP3R) and vasodilator-stimulated phosphoprotein (VASP), known substrates of cAMP-dependent kinase and cGMP dependent kinase. Phosphorylation of IP3R by isoscopoletin induced Ca2+ inhibition from the dense tubular system Ca2+ channels, and VASP phosphorylation was involved in fibrinogen binding inhibition by inactivating αIIb/β3 in the platelet membrane. Isoscopoletin finally reduced thrombin-induced fibrin clot production and finally reduced thrombus formation. Therefore, this research suggests that isoscopoletin has strong antiplatelet effects and is likely to be helpful for thrombotic diseases involving platelets by acting as a prophylactic and therapeutic agent.

Improvement of the subcooled boiling model for the prediction of the onset of flow instability in an upward rectangular channel

  • The MARS code has been assessed for the prediction of onset of flow instability (OFI) in a vertical channel. For assessment, we built an experiment database that consists of experiments under various geometry and thermal-hydraulic condition. It covers pressure from 0.12 to 1.73 MPa; heat flux from 0.67 to 3.48 MW/m2; inlet sub-cooling from 39 to 166 ℃; hydraulic diameters between 2.37 and 6.45 mm of rectangular channels and pipes. It was shown that the MARS code can predict the OFI mass flux for pipes reasonably well. However, it could not predict the OFI in a rectangular channel well with a mean absolute percentage error of 8.77%. In the cases of rectangular channels, the error tends to depend on the hydraulic diameter. Because the OFI is directly related to the subcooled boiling in a flow channel, we suggest a modified subcooled boiling model for better prediction of OFI in a rectangular channel; the net vapor generation (NVG) model and the modified wall evaporation model were modified so that the effect of hydraulic diameter and heat flux can be accurately considered. The assessment of the modified model shows the prediction of OFI mass flux for rectangular channels is greatly improved.

Heat Flux Evaluation of KSR-III Sub-scale Chamber (KSR-III 축소형 연소기의 열유속 평가)

  • A water-cooled calorimeter chamber with 8 cooling channels based on KSR-III sub-scale chamber has been designed and manufactured. One dimensional empirical correlation has been used at the design stage and full three-dimensional CFD analysis has been conducted to confirm the cooling condition for hot fire test is safe. Predicted heat flux is accurate around the nozzle throat when the thermal resistance of carbon deposit is considered. However relatively large difference is observed in chamber part.

Current-Integrating DFE with Sub-UI ISI Cancellation for Multi-Drop Channels

  • This paper presents a half-rate current-integrating DFE receiver with sub-unit interval (sub-UI) inter-symbol interference (ISI) cancellation. By having a single additional DFE tap in each data path, the proposed DFE receiver can minimize BER degradation due to input pattern dependency and feedback tap latency problems in conventional current-integrating DFE receivers. The proposed DFE receiver was designed and fabricated in a 45 nm CMOS process, whose measurement results indicated that the BER bathtub width is increased from 0.235 UI to 0.315 UI (34% improvement) at $10^{-12}$ BER level.

  • Multiple Description Coding(MDC) is a technique used to obtain two or more (often correlated) descriptions of a source, which are transmitted over different channels to receiver. Two descriptions of the source support two levels of reconstruction quality. When all the descriptions are received and used in the reconstruction, the source should be reconstructed with acceptable quality. In this work, we consider Multiple Description Scalar Quantizer(MDSQ) to wavelet transform domain. Conventional MDSQ schemes in wavelet domain considered description with equal weights at each sub-bands after quantization. But each sub-bands is unequal contribution to whole image quality. Therefore, we experiment the multiple description with unequal weight in each sub-bands.
