Structure-based Functional Discovery of Proteins: Structural Proteomics

  • The discovery of biochemical and cellular functions of unannotated gene products begins with a database search of proteins with structure/sequence homologues based on known genes. Very recently, a number of frontier groups in structural biology proposed a new paradigm to predict biological functions of an unknown protein on the basis of its three-dimensional structure on a genomic scale. Structural proteomics (genomics), a research area for structure-based functional discovery, aims to complete the protein-folding universe of all gene products in a cell. It would lead us to a complete understanding of a living organism from protein structure. Two major complementary experimental techniques, X-ray crystallography and NMR spectroscopy, combined with recently developed high throughput methods have played a central role in structural proteomics research; however, an integration of these methodologies together with comparative modeling and electron microscopy would speed up the goal for completing a full dictionary of protein folding space in the near future.

  • Since most engineering problems have had open-ended and ill-defined characteristics, design process is in advance attended with determination of alternatives based on realistic constraints after definition of appropriate problem. And it is completed with selection of best alternative through their comparison and investigation, and with performance of selected-alternative's detail design. As the process of structural design compared with that of general design, this paper presents a paradigm which can generate structural design alternatives, select optimum structure among them and simultaneously set its optimum design variables in reference of several objective as a result in more extended design region. For this purpose, specialized genetic algorithms which can handle design alternatives and multicriteria problems is used.

  • The purpose of this study is to show how to gain the morphology of the Hybrid Spatial Structures and to get the geometrical data such as node coordinates, member relationships and graphic images. To form spatial structures, we have developed morphological aspects of general spatial structures, programming process and techniques. Structural design has many processes. Especially, it is very important to consider the determination of structural configuration. Regular Hybrid Spatial Structures have complex configuration, so we need to make use of automated computer process to determine structural shape in Hybrid Spatial Structures. We have applied morphological aspects to double layer plate, single layer dome, double layer dome and tensegrity structure.

  • In this paper, the effects of vehicle loads on flat slab system are investigated on the basis of the previous studies for beam-girder parking structural system. The influence surfaces of flat slab for typical design section are developed for the purpose of obtaining maximum member forces under vehicle loads. In addition, the equivalent vehicle load factors for flat slab parking structures are suggested using artificial neural network. The network responses are compared with the results by numerical analyses to verify the validation of Levenberg-Marquardt algorithm adopted as training method in this paper. Many parameter studies fur the flat slab structural system show dominant vehicle load effects at the center positive moments in both column and middle strips, like the beam-girder parking structural system.

Fracture energy and tension softening relation for nano-modified concrete

  • This paper presents the details of size independent fracture energy and bi-linear tension softening relation for nano modified high strength concrete. Nano silica in powder form has been used as partial replacement of cement by 2 wt%. Two popular methods, namely, simplified boundary effect method of Karihaloo et al. (2003) and RILEM (1985) fracture energy with P-${\delta}$ tail correction have been employed for estimation of size independent fracture energy for nano modified high strength concrete (compressive strength ranges from 55 MPa to 72 MPa). It is found that both the methods gave nearly same values, which is an additional evidence that either of them can be employed for determination of size independent fracture energy. Bi-linear tension softening relation corresponding to their size independent fracture energy has been constructed in an inverse manner based on the concept of non-linear hinge from the load-crack mouth opening plots of notched three-point bend beams.

Diagnosis and recovering on spatially distributed acceleration using consensus data fusion

  • The acceleration information is significant for the structural health monitoring, which is the basic measurement to identify structural dynamic characteristics and structural vibration. The efficiency of the accelerometer is subsequently important for the structural health monitoring. In this paper, the distance measure matrix and the support level matrix are constructed firstly and the synthesized support level and the fusion method are given subsequently. Furthermore, the synthesized support level can be served as the determination for diagnosis on accelerometers, while the consensus data fusion method can be used to recover the acceleration information in frequency domain. The acceleration acquisition measurements from the accelerometers located on the real structure National Aquatics Center are used to be the basic simulation data here. By calculating two groups of accelerometers, the validation and stability of diagnosis and recovering on acceleration based on the data fusion are proofed in the paper.

  • The purpose of this research is to identify the factors of self-determination theory (e.g., autonomy, competence, and relatedness) affecting flow, satisfaction, and loyalty. This research uses structural equation modeling (SEM) to confirm the validity and analyzes the causal relationship of the suggested model. The results indicate that competence and relatedness positively influence flow and that autonomy positively influences satisfaction. First, autonomy is significantly shown to influences satisfaction and does not significantly influence flow. Second, that competence significantly influenced flow and indirectly influenced satisfaction. Third, relatedness significantly influenced flow and indirectly influenced satisfaction. Finally, flow influenced satisfaction, and flow and satisfaction directly and indirectly influenced loyalty.

  • This paper proposes a new method for the preliminary design of cable-stayed bridges that belong to the radial system subjected to static loads (self weight, traffic loads, concentrated loads, etc). The method is based on the determination of the each time existing relation between the tension forces of the cables and the corresponding bridge-deck deformations, and can be extended on any type of cable layout (fan, parallel, or mixed system). Galerkin's method is used for the final determination of the cable stresses and the bridge deformation. The determination of the equation, which gives the forces of the cables in relation to the deck's configurations, permits us to convert the problem to the solving of a continuous beam without cables.

Procedures for determination of elastic curve of simply and multiple supported beams

  • In this paper two procedures for determination of the elastic curve of the simply and multiple supported beams are developed. Determination of the elastic curve is complex as it requires to solve a strong nonlinear differential equation with given boundary conditions. For numerical solution the initial guess of the slope at the end of the beam is necessary. Two procedures for obtaining of the initial guess are developed: one, based on transformation of the supported beam into a clamped-free one, and second, on the linearization of the problem. Procedures are applied for calculating of elastic curve of a simply supported beam and a beam with three supports. Obtained results are compared. Advantages and disadvantages of both methods are discussed. It is proved that both suggested procedures give us technically accurate results.

  • Safety and serviceability of a planar steel frame are assessed. Attention is turned to the individual main steps in the assessment procedure, i.e., to the definition of loads, selection of transformation model, determination of the response of the structure to the loading, and to the definition of the limiting values (considering safely and serviceability of the structure). The potential of the method using direct Monte Carlo technique as a powerful tool is emphasized.
