Inhibition of Growth and Microcystin Toxicity, and Characterization of Algicidal Substances from Lactobacillus graminis against Microcystis aeruginosa (Microcystis aeruginosa에 대한 Lactobacillus graminis의 성장 억제능, microcystin 분해 및 살조 물질의 특성)

  • For several decades, lactic acid bacterium (Lactobacillus graminis: LAB) has been generally recognized as safe. To develop the pan-environmental bio-control agent, algicidal activity of the live LAB cell and its culture filtrate (CF) was examined against Microcystis aeruginosa. LAB cells perfectly lysed M. aeruginosa within 3 days, while the CF had a less effect than the live cells, approximately 78% inhibition of algal growth during a same culture period. The concentration of microcystin in alone culture of M. aeruginosa was $7.1{\mu}gL^{-1}$, but gradually increased and leach $158.5{\mu}gL^{-1}$ on 10 days. However, LAB cells clearly decreased the microcystin by $10.3{\mu}gL^{-1}$ in the same period, approximately 93.5%. CF of LAB showed a strong algicidal activity over 75% between pH 2-7, 91.3% by the treatment of proteinase K, 87.8% by below 3 kDa in particle size, and 75.3% by heat treatment, respectively. Of five solvents, fractions of CF passed through solvents diethyl ether and ethyl acetate showed an obvious algicidal activity in the algal-lawn test. Among 5 fractions purified by silica-gel TLC plate, two spots showed a most strong removal activity on M. aeruginosa. Another analysis of GC indicate that CF contained six representative fatty acids. Even though most of these substance have been known as an anti-algal substance against M. aeruginosa, oleic acid is the most effective. These results suggested that the culture filtrate or specific substances, like a fatty acids, in comparison with live L. graminis can be a successful and eco-friendly agent to control Microcystis bloom.

Flavor Characteristics of Volatile Compounds from Shrimp by GC Olfactometry (GCO) (GC Olfactometry를 이용한 새우의 휘발성성분 특성평가)

  • Volatile compounds from shrimp whole body (SWB) and shrimp shell waste (SSW) were isolated, and identified by the combination of SDE (simultaneous steam distillation and solvent extraction), GC (gas chromatography, HP-5890 plus)and MSD (mass selective detecter) or olfactometry. The peak numbers isolated from SWB and SSW were 20 and 46, respectively. The amounts of the volatile compounds isolated from SSW were higher than those of SWB. SWB produced more low-boiling compounds below 7$0^{\circ}C$ and SSW did more high boiling compounds over 10$0^{\circ}C$. The volatile compounds identified from SSW were 9 pyrazines,5 acids,4 aldehydes, and 4 alcohols. These volatile compounds were evaluated by aroma extraction dilution analysis and gas chromatography olfactometry (GCO). Some compounds which were not detected by GC-FID and GC-MSD were found to be a strong shrimp flavor of log$_3$ FD 3 value by GCO. Strong shrimp odors were detected in low temperature while nutty aromatic odors and unpleasant oily smells were found in high temperature.

Preparation of Organic-inorganic Hybrid PES Membranes using Fe(II) Clathrochelate (Fe(II) clathrochelate을 이용한 유.무기 PES 복합막의 제조)

  • Metal-templated condensation of cyclohexanedione dioxime and phenylboronic acid in the presence of Fe(II) sulfate heptahydrate proceeds cleanly in methanol to furnish the Fe(II) clathrochelate. An organic/inorganic hybrid membranes composed of Fe(II) clathrochelate and polyethersulfone was prepared by using phase inversion method. For membrane preparation, the Fe(II) clathrochelate was highly soluble (3~5 g/L) in DMF, NMP, and DMAc, which meets the requirements for the solubility of metal complexes in polar aprotic solvent used in membrane preparation. It was stable even in the presence of strong acids, such as trifluorosactic acid (pKa = 0.3). It was characterized by UV-vis spectroscopy, and their stability in solution phase studied in the presence of (i) strong acids or (ii) competing chelates. Organic/inorganic hybrid membranes were prepared with polyethersulfone, polyvinylpyrrolidone, p-toluenesulfonic acid, Fe(II) clathrochelate and DMF by using nonsolvent induced phase inversion method. The addition of Fe(II) clathrochelate leads increase of surface pore density, mean pore size and flux. We can obtain highly asymmetric membranes by addition of Fe(II) clathrochelate.

Antimicrobial Activity of Extracts and Coumaric Acid Isolated from Artemisia princeps var. orientalis (쑥의 추출물 및 Coumaric Acid의 항균활성)

  • Antimicrobial activity of methanol extract and fraction from mugwort leaves(Artemisia princeps val. orientalis) was investigated for the screening of natural antiwucroblal components. By using agar diffusion method, ethyl acetate(EtOAc) layer fractionated from methanol extract of mugwort leaves showed the highest inhibitory effects against tested microorganisms. The ortho-coumaric acid(200∼600ppm) isolated from EtOAc layer showed strong antibacterial activities for Bacillus subtilis and Salmonella typhimurium. As derivatives of o-coumaric acid, antibacterial activity of para-coumaric acid was 1.2∼1.7 fold higher than that of o-coumaric acid. Three types of coumaric acids strong inhibited the growth of B. subtilis in the culture medium. Growth of S. tyhimurium, P. aeruginosa and S. aureus were effectively inhibited by o-, m- and p-coumaric acids, respectively. Minimum inhibitory dose of p-coumaric acid for B. subtilis was $\100∼200mu\textrm{g}$/paper disk.

Inhibitory Effect of Doenjang(fermented Korean soy paste) Extracts and Linoleic Acid on the Growth of Human Cancer Cell Lines

  • The inhibitory effects of doenjang extracts and linoleic acid(LA) which was identified as one of the active compounds in doenjang on the growth of human cancer cells were studied, comparing to the actions on normal cells. Methanol extract and hexane fraction from doenjang exhibited the strong growth inhibitory effect on HT-29 human colon carcinoma cells. Inhibitory effects of chloroform, ethyl acetate, butanol and aqueous fractions on the cancer cells were observed, moderately or weakly. When cell counts of SNU-C$_1$human colon carcinoma cells were determined daily for 6 days, the inhibitory effect of hexane fraction on this cell line was higher than that of the methanol extract from doenjang. LA completely suppressed the growth of SNU-C$_1$cells after 4 days, while conjugated linoleic acid(CLA) resulted in 98% inhibition after 6 days. With the addition of LA and other free fatty acids such as stearic acid, oleic acid, linolenic acid and ${\gamma}$-linolenic acid (${\gamma}$-LnA) to the culture system, the growth of HT-29 cells and SNU-C$_1$cells was greatly suppressed after 6 days. Inhibitory effects of LA ${\gamma}$-LnA on the growth of these cells were stronger than other fatty acids. On the growth of AZ-521 human gastric carcinoma cells, LA and CLA completely cuppressed the growth of the cells after 4 days and 3 days, respectively. At the level of 0.001%~0.01% of LA, there was no cytotoxic effect on normal rat kidney cells and normal intestine human cells. These results showed that LA, a major active compound of doenjang, had strong inhibitory effects on the growth of human cancer cells without damaging normal cells.

Relationships between Fatty Acids and Tocopherols of Conventional and Genetically Modified Peanut Cultivars Grown in the United States (미국산 전통품종과 유전자 재조합 땅콩품종의 지방산과 토코페롤의 상관관계)

  • Relationships between fatty acids and tocopherols in conventional and genetically modified peanut cultivars were studied by gas chromatography with flame ion detector and high performance liquid chromatography with fluorescence detection. Eight fatty acids and four tocopherol isomers in the sample set were identified and quantified. Oleic acid and linoleic acid are major fatty acids and the ratio of oleic and linoleic acids ranged from 1.11 to 16.26. Tocopherols contents were 6.76 to 12.24 for ${\alpha}$-tocopherol (T), 0.08 to 0.39 for ${\beta}$-T, 5.28 to 15.02 for ${\gamma}$-T, and 0.17 to 1.17 mg/100 g for ${\delta}$-T. Correlation coefficient (r) for fatty acids and tocopherols indicated a strong inverse relationship between oleic & linoleic acids (r=-0.97, P<0.05) and positive relationships between palmitic & linoleic acids (r=0.95, P<0.05) and ${\gamma}$-T & ${\delta}$-T (r=0.83, P<0.05). Principal component analysis (PCA) of fatty acids and tocopherols gave four significant principal components (PCs, with eigenvalues>1), which together account for 85.49% of the total variance in the data set with PC1 and PC2 contributing 45.27% and 21.33% of the total variability, respectively. Eigen analysis of the correlation matrix loadings of the four significant PCs revealed that PC1 was mainly contributed by palmitic, oleic, linoleic, and gondoic acids, while PC2 was by behenic acid, ${\beta}$-T, and ${\gamma}$-T. The score plot generated by PC1-PC2 identified sample clusters in the two spatial planes based on the oleic and linoleic acids. The score plot PC3-PC4 didn't separate sample groups.

  • A potential biocontrol bacterium, YB-70 was isolated from a rhizosphere in suppressive soil and identified as a strain of Bacillus subtilis. In several biochemical and in vitro antibiosis tests on Fusarium solani with the culture filterates from B. subtilis YB-70, we found that antifungal mechanism of B. subtilis YB-70 was mediated by antibiotic substances produced from the bacterium. These antifungal substances were appeared to be hear-resistant, micromolecular, and ethy alcohol soluble. Antifungal agents produced by B. subtilis YB-70 showed strong inhibified against root-rotting fungi F. solani in in vivo pot test. An antifungal substance. YBS-1s, was purified from the culture broth of B. subtilis YB-70 by isoelectronic precipitation, silica gel column chromatography and Sephadex LH-20 column chromatography analysis by Fab-MASS, $^{1}$H-NMR, $^{13}$C-NMR, DEPT, and amino acid analyzer revealed that the YBS-1A was a peptide antibiotics of iturin class containing seven amino acids from five different groups, and the other(YBS-1B) was an analogue of iturin group composed of 11 amino acids with larher molecular weight of about 1, 500 dalton, which was lager than that of iturin A.

Evaluation of the Apparent Ileal Digestibility (AID) of Protein and Amino Acids in Nursery Diets by In vitro and In vivo Methods

  • The objective was to evaluate in vitro prediction of ileal digestibility of protein and amino acids (AA) for current nursery pig diets (n = 10) by using pepsin and pancreatin incubations. To compare in vivo ileal digestibility, forty nursery pigs (4 pigs per diet) with an initial BW of $12.2{\pm}2.7$ kg were surgically equipped with T-cannula in the distal ileum. In all cases, the values of in vitro digestibility were higher than those of in vivo digestibility (p<0.05). With regard to the relationships of essential and non essential AA (CP), the $r^2$ value was 0.76. With regard to AA, high relationships were observed in Ile, Thr, and Gly (0.85, 0.83, and 0.89, respectively). Also, there was a lower relationship for Arg, Met, Ala, Asp, Glu, Pro, Ser, and Tyr with $R^2$ values of 0.56, 0.54, 0.40, 0.54, 0.45, 0.24, 0.49, and 0.35, respectively between in vitro and in vivo digestibility. The EAA relationship ($R^2$ = 0.71) was generally higher than that of NEAA ($R^2$ = 0.50) numerically. In conclusion, there were strong linear relationships between in vivo and in vitro ileal digestibility (CP, Ile, Thr, and Gly). In vitro prediction of ileal digestibility (CP, Ile, Thr, and Gly) seems to have significant potential for practical application.

Preparation and Properties of Rust-Removing Polymer Gel (녹제거 폴리머겔의 제조 및 특성)

  • The formation of rust on metallic substrate is known to cause the damages and destructions of raw materials, which is one of the leading reasons of sturctural collapses and many kind of hazards in modern industry. Polymer gels with rust removing effects were compounded in this study by employing various kinds of acids like hydrochloric acid, phosphoric acid, gluconic acid, oxalic acid as the rust removing ingredients. TEA(Triethanolamine) as dispersant and hydrophilic chemical were used for effective gelation of acids. Also corrosion inhibitor was added to enable the coating effect and to improve rust removing effect on metallic surface. In order to investigate the rust properites on metallic substrate, artificial rusts were prepared in salt solution, using iron, copper, aluminium and brass as the base metals. The properties of gel compounds were checked by gelation, pH, viscosity, morphology property and rust removing test. Developed gel compounds in this study have a good rust removing property, showing a strong adhesion on horizontal and vertical metallic surface, and can be easily rinsed off by water.

Determination of Phytochemical Contents and Biological Activities from the Fruits of Elaeagnus multiflora

  • The purpose of this study was to investigate the phytochemical contents and biological activities, including $\alpha$-glucosidase inhibitory activity and antioxidant activity, from the fruits of Elaeagnus multiflora using different solvent systems. The 50% ethanol extract of this species showed the most potent $\alpha$-glucosidase inibitory activity (88.5%) at a concentration of 1 mg/mL. Moreover, this extract also displayed the strongest antioxidant activity in each assay, showing 96.3% in DPPH scavenging activity, 98.2% in ABTS scavenging activity, and 2.5% in reducing power. The highest total phytochemical contents, including flavan-3-ols, phenolic acids, and flavonoids, were observed in the 50% ethanol extract. Interestingly, flavan-3-ols (3.150 mg/g) were detected at a significantly higher total content than those of phenolic acids (0.380 mg/g) and flavonoids (not detected). Additionally, the contents of individual phytochemicals showed remarkable differences, especially the epicatechin gallate (2.008 mg/g) and gallic acid (0.099 mg/g), which were the predominant constituents of each phytochemical type. Therefore, our results suggest that the 50% ethanol extract of E. multiflora fruits has strong biological activities, which are correlated with high phytochemical contents.
