Annotation and Expression Profile Analysis of cDNAs from the Antarctic Diatom Chaetoceros neogracile

  • To better understand the gene expression of the cold-adapted polar diatom, we conducted a survey of the Chaetoceros neogracile transcriptome by cDNA sequencing and expression of interested cDNAs from the Antarctic diatom. A non-normalized cDNA library was constructed from the C. neogracile, and a total of 2,500 cDNAs were sequenced to generate 1,881 high-quality expressed sequence tags (ESTs) (accession numbers EL620615-EL622495). Based on their clustering, we identified 154 unique clusters comprising 342 ESTs. The remaining 1,540 ESTs did not cluster. The number of unique genes identified in the data set is thus estimated to be 1,694. Taking advantage of various tools and databases, putative functions were assigned to 939 (55.4%) of these genes. Of the remaining 540 (31.9%) unknown sequences, 215 (12.7%) appeared to be C. neogracile-specific since they lacked any significant sequence similarity to any sequence available in the public databases. C. neogracile consisted of a relatively high percentage of genes involved in metabolism, genetic information processing, cellular processes, defense or stress resistance, photosynthesis, structure, and signal transduction. From the ESTs, the expression of these putative C. neogracile genes was investigated: fucoxanthin chlorophyll (chl) a,c-binding protein (FCP), ascorbate peroxidase (ASP), and heat-shock protein 90 (HSP90). The abundance of ASP and HSP90 changed substantially in response to different culture conditions, indicating the possible regulation of these genes in C. neogracile.

Psychopharmacological Profile of the Water Extract of Gardenia jasminoides and Its Constituents, Genipin and Geniposide, in Mice

  • Gardenia jasminoides (G. jasminoides) is traditionally used to treat insomnia, jaundice, emotional disorders, hepatic disease, and inflammatory disease. Previously, we found that geniposide and the water extract of G. jasminoides increased $Cl^-$ influx in neuroblastoma. Here we examined the sychopharmacological activities of G. jasminoides and its constituents. G. jasminoides extract was orally administered at 100 and 200 mg/kg, and genipin and geniposide were intraperitoneally injected at 2, 10, and 20 mg/kg. G. jasminoides extract (200 mg/kg) significantly decreased total open field activity but increased rearing activity in the center of the open field, suggesting an increase in exploratory activity. Genipin and geniposide did not change open field activity, but geniposide (20 mg/kg) increased rearing activity in the central area. The extract (200 mg/kg) significantly decreased rotarod and wire-balancing activity, but genipin and geniposide did not. No compounds influenced thiopental-induced sleeping or electroshock-induced seizures. The extract (200 mg/kg) significantly increased staying time in the open arms of the elevated plus maze and the entry ratio into the open arms, and geniposide (20 mg/kg) also increased open arm entry. Electroshock stress decreased open arm activity, but the extract and geniposide (20 mg/kg) significantly reversed that effect. This results indicate that G. jasminoides extract and geniposide alleviated anxiety with greater efficacy in stressed animals than normal animals.

Dynamic response of functionally gradient austenitic-ferritic steel composite panels under thermo-mechanical loadings

  • In this paper, the dynamic response of functionally gradient steel (FGS) composite cylindrical panels in steady-state thermal environments subjected to impulsive loads is investigated for the first time. FGSs composed of graded ferritic and austenitic regions together with bainite and martensite intermediate layers are analyzed. Thermo-mechanical material properties of FGS composites are predicted according to the microhardness profile of FGS composites and approximated with appropriate functions. Based on the three-dimensional theory of thermo-elasticity, the governing equations of motionare derived in spatial and time domains. These equations are solved using the hybrid Fourier series expansion-Galerkin finite element method-Newmark approach for simply supported boundary conditions. The present solution is then applied to the thermo-elastic dynamic analysis of cylindrical panels with three different arrangements of material compositions of FGSs including ${\alpha}{\beta}{\gamma}M{\gamma}$, ${\alpha}{\beta}{\gamma}{\beta}{\alpha}$ and ${\gamma}{\beta}{\alpha}{\beta}{\gamma}$ composites. Benchmark results on the displacement and stress time-histories of FGS cylindrical panels in thermal environments under various pulse loads are presented and discussed in detail. Due to the absence of similar results in the specialized literature, this paper is likely to fill a gap in the state of the art of this problem, and provide pertinent results that are instrumental in the design of FGS structures under time-dependent mechanical loadings.

무선 인터넷 공유기에서 사물 인터넷 장치 제어를 위한 가상 오브젝트 호스팅 기술 연구 (A Virtual Object Hosting Technology for IoT Device Controlling on Wireless AP's)

  • IoT(Internet of Things)로 대표되는 사물 인터넷 환경에서 최근 개인 건강 및 홈 자동화 관련 센서류 장치들이 급격히 보급되고 있다. 설치된 장치의 수가 증가함에 따라 사용자 입장에서는 이용하고자 하는 장치마다 개별 서비스 또는 어플리케이션을 찾아 실행해야 하는 번거로움이 생기고, 서비스 공급자 입장에서는 장치간의 정보 공유나 연동 및 통합 제어 기능을 제공하기 어려운 문제가 심화된다. 본 논문에서는 이러한 문제점을 해결하기 위해 댁내 인터넷 서비스의 거점이 되는 무선 인터넷 공유기를 이용, 다양한 IoT 장치들을 연동하고 통합 제어하는 방법을 제시 한다. 이를 위하여, 먼저 IoT 장치를 가상 오브젝트(Virtual Object)화하는 방법을 제시하고, 생성된 가상 오브젝트들을 무선 인터넷 공유기상에서 실행, 연동 및 관리하기 위한 서비스 구조를 제안한다. 또한 이를 직접 홈 환경의 무선 인터넷 공유기상에 구현해 다수의 IoT 장치들을 연결한 상황에서 안정적인 서비스 제공 여부와 무선 라우팅 기능과 병행 동작시의 성능에 대해 시험함으로써, 실제 환경에서의 이용 가능성을 검증한다.

유방암 생존자의 생활습관, 우울, 부부친밀도가 삶의 질에 미치는 영향 (Influence of lifestyle, depression, and marital intimacy on quality of life in breast cancer survivors)

  • Purpose: This study investigated lifestyle, depression, marital intimacy, and quality of life (QoL) in breast cancer survivors, with the goal of identifying the impacts of these factors on QoL. Methods: A sample of 146 breast cancer survivors was surveyed in this cross-sectional study. Data were collected from March 20 to May 30, 2019, using self-report structured questionnaires at a hospital located in Jeonju, Korea. Data were analyzed using the independent t-test, analysis of variance, Pearson correlation coefficients, and hierarchical regression analysis. Participants agreed to complete a face-to-face interview, including administration of the Health Promoting Lifestyle Profile II, Depression Anxiety Stress Scale 21-Depression Scale, Marital Intimacy Scale, and Functional Assessment Cancer Therapy-Breast Cancer tool. Results: QoL was positively correlated with lifestyle (r=.49, p<.001) and marital intimacy (r=.45, p<.001) and negatively correlated with depression (r=-.72, p<.001). Hierarchical multiple regression analysis showed that depression (β=-0.63, p<.001), marital intimacy (β=0.19, p=.001), and lifestyle (β=0.13, p=.031) had significant effects on the QoL of breast cancer survivors, accounting for 63.3% of variance in related QoL. Conclusion: This study provides insights into how breast cancer survivors' QoL was influenced by depression, marital intimacy, and lifestyle. To improve the QoL of breast cancer survivors, healthcare providers should consider developing strategies to decrease depression, to increase marital intimacy, and to improve lifestyle.

Bag-1L is a Stress-withstand Molecule Prevents the Downregulation of Mcl-1 and c-Raf Under Control of Heat Shock Proteins in Cisplatin Treated HeLa Cervix Cancer Cells

  • Background: Cisplatin, a DNA damaging agent, induces apoptosis through increasing DNA fragmentation. However, identification of intrinsic resistance molecules against Cisplatin is vital to estimate the success of therapy. Bag-1 (Bcl-2-associated anthanogene) is one anti-apoptotic protein involved in drug resistance impacting on therapeutic efficiency. Elevated levels of this protein are related with increase cell proliferation rates, motility and also cancer development. For this reason, we aimed to understand the role of Bag-1 expression in Cisplatin-induced apoptosis in HeLa cervix cancer cells. Cisplatin decreased cell viability in time- and dose-dependent manner in wt and Bag-1L+HeLa cells. Although, $10{\mu}M$ Cisplatin treatment induced cell death within 24h by activating caspases in wt cells, Bag-1L stable transfection protected cells against Cisplatin treatment. To assess the potential protective role of Bag-1, we first checked the expression profile of interacting anti-apoptotic partners of Bag-1. We found that forced Bag-1L expression prevented Cisplatin-induced apoptosis through acting on Mcl-1 expression, which was reduced after Cisplatin treatment in wt HeLa cells. This mechanism was also supported by the regulation of heat shock protein (Hsp) family members, Hsp90 and Hsp40, which were involved in the regulation Bag-1 interactome including several anti-apoptotic Bcl-2 family members and c-Raf.

Mechanisms of Chilling Tolerance in Relation to Antioxidative Enzymes in Rice

  • In order to examine the mechanistic basis for differential sensitivities to chilling and subsequent recovery between two rice (Oryza sativa L.) cutivars, a chilling-tolerant japonica type (Ilpumbyeo) and a chilling-susceptible indica type (Taebaekbyeo), changes of physiological responses and antioxidant enzymes were investigated. Both cultivars at 3 leaf stage were exposed at a low temperature of $5^{\circ}C$ for 3 days and subsequently recovered in a growth chamber at a $25^{\circ}C$ for 5 days with 250 mmol $m^{-2}$ $s^{-1}$. Physiological parameters such as leaf fresh weight, relative water content, cellular leakage, lipid peroxidation, and chlorophyll a fluorescence showed that the chilling tolerant cultivar had a high tolerance during chilling. However, the chilling-susceptible cultivar revealed severe chilling damages. The chilling-tolerant cultivar was also faster in recovery than the chilling-susceptible cultivar in all parameters examined. We analyzed the activity and isozyme profiles of four antioxidant enzymes which are: superoxide dismutase (SOD), caltalase (CAT), ascorbate peroxidase (APX), and glutation reductase (GR). We observed that chilling-tolerance was due to a result of the induced or higher antioxidant enzyme system, CAT and APX in leaves and SOD, CAT, APX, and GR in roots. Especially, we observed the most significant differences between the chilling-tolerant cultivar and -susceptible cultivar in CAT and APX activity. Also in isozyme profiles, CAT and APX band intensity in the chilling-tolerant cultivar was distinctively higher than in the chilling-susceptible cultivars during chilling and recovery. Thus, the cold stability of CAT and APX are expected to contribute to a tolerance mechanism of chilling in rice plants. In addition, the antioxidative enzymes activity in roots may be more important than in that of leaves to protect chilling damage on rice plants.

효소처리가 대두단백질의 기능특성과 두부의 품질에 미치는 영향 (Effects of Functional Properties of Soy Protein Isolate and Qualities of Soybean Curd upon Proteolytic Hydrolysis)

  • 본 연구에서는 분리대두단백질에 단백질 가수분해효소를 처리하여 대두단백질에 가수분해적 변성을 야기하여 그 기능성을 변화 고찰하고 실제 식품에의 이용방법으로 효소처리된 두유로 두부를 만들어 물성실험과 관능평가를 실시하였다. 장엽에서 SPI를 제조한 후 15분간 bromelain으로 처리하여 2.7% 가수분해된 MSPI를 만들어 기능특성을 측정 비교하였다. MSPI는 전 pH범위에서 용해도가 증가하였고, 특히 등전점에서 15% 정도 증가하였다. 유화형성력과 기포팽창력은 증가하였으나 이들의 안정성은 감소하였는데, 특히 기포안정성은 알칼리쪽에서 급격히 감소하였고, 등전점에서는 가장 큰 것으로 나타났다. 표준두유와 변형두유를 혼합하여 제조한 두부로 물성실험과 관능평가를 실시하였을 때 압착실험에서는 변형두부I이 연하고 탄력성 있는 두부를 형성하는 것으로 나타났다. 각 두부의 물성모형은 spring 한개와 Maxwell 모형 세개를 가진 7요소모형으로 동일한 물성모형을 나타내었다. 관능평가에서는 표준두유와 변형두유를 3 : 1의 비율로 혼합하여 제조한 두부의 품질이 가장 우수한 것으로 평가되었다.

미각 장애 환자 5례를 대상으로 한 대한 한방 치료의 효과 (The Effect of Traditional Korean Medicine Treatment on Patients with Taste Disorders: A Case Series of Five Patients)

  • Objectives: The purpose of this study was to report the effect of traditional Korean medical therapy such as acupuncture, electroacupuncture and herbal medicine in taste disorder patients. Methods: We surveyed 5 taste disorder patients visiting the Oral Diseases Clinic in the Kyung Hee University Oriental Medicine Hospital from January, 2014 to June, 2014. Before starting traditional Korean medical therapy such as acupuncture, electroacupuncture and herbal medicine, the subjects were evaluated on severity of discomfort using visual analogue scale (VAS), salivary flow rate (SFR), quality of life about oral health (based on the 14-item Oral Health Impact Profile-14) and qi-stagnation condition (based on the 23-item qi-stagnation Questionnaire). Visual analogue scale was re-evaluated during the treatment period. Results: There was no relationship between diminished SFR and severity of discomfort. Also change or loss of taste did not influence the quality of life about oral health. However, stress which refers to qi-stagnation could be one of the reasons taste disorder occurs. After receiving traditional Korean medical therapy, all 5 patients' visual analogue scale score decreased. Conclusions: Traditional Korean medical therapy may potentially be an option for taste disorder. Further evaluations including pre-post comparison with larger number of cases will be needed in the future.

금강모치(Rhynchocypris kumgangensis)에서 heat shock protein 70의 클로닝과 수온상승에 의한 발현 변화 분석 (Cloning of Heat Shock Protein 70 and Its Expression Profile under an Increase of Water Temperature in Rhynchocypris kumgangensis)

  • Water temperature is key factor influencing growth and reproduction of fish and its increase give rise to various physiological changes including gene expression. Heat shock protein (Hsp), one of the molecular chaperones, is highly conserved throughout evolution and its expression is induced by various stressors such as temperature, oxidative, physical and chemical stresses. Here, we isolated partial cDNA clones encoding 70-kDa Hsp (Hsp70) and $\beta$-actin using reverse transcriptase-PCR (RT-PCR) from gut of Rhynchocypris kumgangensis, a Korean indigenous species and cold-water fish, and investigated expression profiles of Hsp70 under an increase of water temperature using $\beta$-actin as an internal control for RT-PCR. Cloned Hsp70 cDNA of R. kumgangensis showed homology to Ctenopharyngodon idella (96%), Hypophthalmichthys molitrix (96%), Danio rerio (93%) and Oncorhynchus mykiss (81%) Hsp70. Cloned $\beta$-actin cDNA of R. kumgangensis showed homology to D. rerio (98%), H. molitrix (97%), C. idella (97%) and O. mykiss (90%) $\beta$-actin. Both mRNA of Hsp70 and $\beta$-actin were expressed in gut, brain, and liver in R. kumgangensis. Futhermore, expression of Hsp70, in brain, was highly augmented by an increase of water temperature. These results suggest that Hsp70 mRNA expression level in brain can be used as a biological molecular marker to represent physiological stress against an increase of water temperature.
