Antimicrobial Activity of Trifoliate Orange (Poncirus trifoliate) Seed Extracts on Gram-Positive Foodborne Pathogens (탱자씨 추출물의 그람양성 식중독균에 대한 항균효과)

  • 물(water, TW), 에탄올(ethanol, TE) 및 핵산(n-hexane, TH)을 이용하여 탱자씨 추출물(trifoliate orange seed extracts, TSEs)을 각각 제조한 후 그람양성 식중독균 6종(Bacillus cereus KCCM 11341, Bacillus subtilis KCTC 1022, Listeria monocytogenes ATCC 12692, Staphylococcus aureus ATCC 19111, Streptococcus mutans KCTC 3065 및 Yersinia enterocolitica KCCM 41657)에 대한 항균활성과 Lactobacillus acidophilus IFO 3025에 대한 프레바이오틱 효과(prebiotic effect)를 측정하였다. 그 결과, 탱자씨 핵산추출물(TH)은 S. aureus ATCC 19111에 대해 배양시간이 증가함에 따라 대조군에 비해 강한 증식저해활성을 보였으며, 탱자씨 에탄올 추출물(TE)은 약간의 증식저해활성을 보였다. 특히, 배양 81시간 후 대조군은 90.31%의 증식활성을 보인 것에 반해 탱자씨 핵산추출물과 에탄올추출물은 64.95%와 75.50%의 증식활성을 각각 보였다. 탱자씨 추출물의 Lb. acidophilus IFO 3025에 대한 프레바이오틱 효과는 대조군에 비해 증식활성을 보이지는 않았으나, 적어도 생육저해효과를 보이지는 않았다. 따라서 탱자씨의 핵산 및 에탄올 추출물은 S. aureus ATCC 19111에 대한 항균활성물질로서의 가능성을 제시하였다.

Effect of a Tooth-brushing Education Program on Oral Health of Preschool Children (잇솔질 교육 프로그램이 학령전기 아동의 구강건강에 미치는 효과)

  • Purpose: To examine the effect of tooth-brushing education on the oral health of preschoolers. Methods: A quasi-experimental design with a non-equivalent control group was used. Two kindergartens were selected and 39 preschoolers from one kindergarten were assigned to the experimental group with tooth-brushing education and 39 from the other kindergarten to the control group. The tooth-brushing education program included 1 session on oral health education, individual tooth-brushing instruction for 1 week and supervised tooth-brushing after lunch for 4 weeks. Oral health behavior including use of tooth paste, tooth-brushing time and method of tooth-brushing, plague, streptococcus mutans, lactobacillus and dental caries were measured before and after the education. Fisher's exact test, t-test and paired t-test with the Window SAS 9.1 program were used to analyze the data. Results: A significant increase in the use of tooth paste, tooth-brushing time and the practice of correct tooth-brushing and a decrease in plague and development of dental caries were observed in the experimental group. Conclusion: This tooth-brushing education was partially effective in improving oral health of preschoolers.

Relationship of Oral Bacterial Load Over One Year of Smoking Cessation

  • Background: Smoking exerts an adverse effect on the periodontal tissue by reorganizing the ecosystem of oral microorganisms and is considered to be an important factor in the development of periodontal disease. Although cross-sectional studies on smokers and non-smokers have been attempted to investigate the microbial differences in periodontal oral cavity, only few studies have been conducted to investigate the changes in oral microorganisms during smoking cessation. The purpose of this study was to investigate the changes of bacteria in saliva and gingival crevicular fluid (GCF) over a period of one year among 11 smokers trying to quit smoking. Methods: Eleven smokers trying to quit smoking visited the clinic at baseline, two weeks, two months, four months, six months, and 12 months to give saliva and GCF samples. The amounts of 16S rRNA, Porphyromonas gingivalis, Treponema denticola, Prevotella intermedia, Fusobacterium nucleatum subsp. nucleatum, Streptococcus mutans, and Streptococcus sobrinus in saliva and GCF were quantified using real-time polymerase chain reaction TaqMan probe assay. The results were analyzed by nonparametric statistical analysis using Friedman test and Spearman correlation coefficient. Results: After cessation of smoking, the amounts of 16S rRNA corresponding to P. gingivalis, F. nucleatum, P. intermedia, and T. denticola in saliva decreased and then again increased significantly. The amount of F. nucleatum 16S rRNA in GCF decreased significantly after smoking cessation. Positive correlations were observed between 16S rRNA and F. nucleatum and between F. nucleatum and T. denticola in saliva and GCF. Conclusion: Even if the number of subjects in this study was small, we suggest that smoking cessation may reduce the total bacterial amount and F. nucleatum in GCF. However, the results regarding changes in the microbial ecosystem due to smoking or smoking cessation were inconsistent. Therefore, further in-depth studies need to be carried out.

Antibacterial Effect of Bacteria Isolated from the Korean Traditional Foods against Pathogenic Bacteria (한국전통식품으로부터 분리 된 세균의 항균활성 효과)

  • Aquaculture continues to be an ever-growing sector. However, high-density farming increases disease outbreaks due to deteriorating water quality and internal stress. To prevent disease, the most common method chemotherapy is using antibiotic administration. In this study, probiotic bacteria were isolated from Korean traditional foods, such a Gochu pickle and cutlassfish salted seafood. Various bacteria were isolated, and their 16S rDNA sequences were analyzed. The antimicrobial activities of four isolates from Gochu pickle and seven isolates from cutlassfish salted seafood were assayed, in addition to the antibacterial activity of culture pellet and supernatant. The antibacterial activity of the pellet was higher than that of the supernatant. Isolate JKM-2 showed the highest antibacterial activity against Streptococcus iniae (43 mm), S. parauberis (40 mm), S. mutans (35 mm), and Vibrio vuinificus (26.5 mm). The sequences of the isolated strains were compared with those of Bacillus subtilis (97.71%), B. tequilensis (97.71%), Brevibacterium halotolerans (97.71%), B. subtilis (97.63%), B. subtilis (97.63%), B. mojavensis (97.54%), B. vallismortis (97.46%), B. nanillea (97.45%), B. methylotrophicus (97.37%), and B. ssiamensis (97.37%). Future through analysis and new strains confirmed the bacterial cell material investigation of JKM-3, and to ensure sufficient stability, it is desired to verify the utility value as a substitute material for antibiotics by application to the form of the industry.

  • In our search for bioactive phenolics from plants, the culms of Phyllostachys bambusoides has been selected for investigation of anti-cariogenic and 1,1-diphenylpicrylhydrazyl (DPPH) radical scavenging agents based on the initial screening results. Fractionation process of n-hexane and $CHCl_3$ extracts afforded four phenolic constituents, ferulic acid (1), vanillin (2), coniferaldehyde (3), and coniferyl alcohol (4) as guided by their DPPH free radical scavenging activities. Additionally, activity-guided fractionation of EtOAc extract with anti-cariogenic activity has resulted in the isolation of coniferaldehyde (3), 2,6-dimethoxy-p-benzoquinone (5), p-methoxycinnamic acid (6), (${\pm}$)-balanophonin (7), and 6-methoxychromanone (8). The structures of 1 - 8 were determined by spectroscopic data interpretation, and also by comparison of their data with the published values. Phenolic compounds 1 - 4 exhibited similar DPPH radical scavenging activities compared with the synthetic antioxidant, butylated hydroxytoluene (BHT), and compounds 3 and 5 - 8 showed significant antibacterial activity against cariogenic oral streptococci, Streptococcus mutans and S. sobrinus.

Antibacterial Effect of Amentoflavone and Its Synergistic Effect with Antibiotics

  • Selaginella tamariscina is a traditional herb used in medicine. Phytochemical amentoflavone, a biflavonoid class of flavonoids, was isolated from the plant of Selaginella tamariscina. In this study, the antibacterial effects and combination effects of amentoflavone and conventional antibiotics such as ampicillin, cefotaxime, and chloramphenicol were investigated. These results showed that amentoflavone had a considerable antibacterial effect and synergistic interaction with antibiotics against various bacterial strains (fractional inhibitory concentration index ${\leq}$ 0.5), except for Streptococcus mutans. To study the mechanism(s) involved in the synergistic activities between amentoflavone and antibiotics, we detected hydroxyl radical formation using 3'-(p-hydroxyphenyl) fluorescein and measured the $NAD^+/NADH$ ratio by $NAD^+$ cycling assay. The results indicated that the formation of hydroxyl radical would be a cause of the synergistic effect and that this oxidative stress originated from a transient NADH depletion. This study suggests that amentoflavone synergizes with antibiotics and has potential as a therapeutic agent for antimicrobial chemotherapy.

Synthesis, Antibacterial, Docking and Anticancer Evaluation of N-Substituted Benzoyl Derivatives

  • A series of N-benzoylated ligands incorporating three different benzoyl groups 2,2'-(benzoyliminodiethylene)-4-substituted phenols ($L^{1,4,7}$), 2,2'-(4-nitrobenzoyliminodiethylene)-4-substituted phenols ($L^{2,5,8}$) and 2,2'-(3,5-dinitrobenzoyliminodiethylene)-4-substituted phenols ($L^{3,6,9}$) were synthesized and characterized by IR, $^1H$ NMR, $^{13}C$ NMR and mass spectroscopy. The In vitro antibacterial activity of investigated ligands were tested against human pathogenic bacteria such as four Gram (-) Enterococcus faecalis, Pseudomonas aeruginosa, Staphylococcus aureus, Vibrio cholera, Vibrio harveyi and two Gram (+) Staphylococcus aureus, Streptococcus mutans. Furthermore, docking studies were undertaken to gain insight into the possible binding mode of these compounds with the binding site of the topoisomerase II (PDB: 4FM9) enzyme which is involved in DNA superhelicity and chromosome seggregation. The N-benzoylated derivatives $L^{5,7,8}$ have significant anticancer activity as Topoisomerase inhibitors. The ligands $L^5$ and $L^8$ were tested for their anticancer activity against human liver adenocarcinoma (HepG2) cell line with the MTT assay.

THE EFFECT OF XYLITOL ON THE EXPRESSION OF GTF GENE (gtf 유전자 발현에 대한 xylitol의 영향)

  • Xylitol, a five-carbon natural sugar alcohol, is widely used non-cariogenic sugar substitute. In present study, the effects of xylitol on the expression of mRNA for glucosyltransferase which synthesizes glucan from sucrose were detected by Fluorescent in situ hybridization (FISH) and flow cytometry. FITC fluorescences for mRNA of gtfB, gtfC and gtfD were decreased further with increasing concentration of xylitol from 1% to 10% when detected by FISH. Flow cytometric analysis also showed that the expression of gtfB, gtfC and gtfD was increased by the addition of sucrose and decreased by the addition of xylitol to BHI broth containing 1% sucrose. In conclusion, the expression of gtfB, gtfC and gtfD mRNA was decreased by the addition of xylitol.

Effect of non-thermal atmospheric pressure nitrogen and air plasma on the surface properties and the disinfection of denture base resin (상온대기압 질소 및 공기 플라즈마가 의치상용 레진의 표면 특성과 살균효과에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the effect of non-thermal atmospheric pressure plasma jet(NTAPPJ) on surface properties and Streptococcus mutans disinfection of denture base resin. Methods : Self-cured denture base resin (Jet denture repair resin, Lang dental Mfg, co., USA) was used to make specimen($12mm{\times}2mm$). To observe surface change before and after plasma process, surface roughness and contact angle were measured. For sterilization experiments, the surfaces of specimens were treated with nitrogen and air NTAPPJ for 1 minute after S. mutans was inoculated on the material surfaces. Results : Before plasma process, surface roughness of denture base resin was $0.21{\mu}m{\pm}0.02{\mu}m$. After air and nitrogen NTAPPJ process, surface roughness was $0.19{\mu}m{\pm}0.03{\mu}m$ and $0.18{\mu}m{\pm}0.01{\mu}m$ respectively. There was no significant difference(p>0.05). Contact angle of control group without plasma process was $83.81^{\circ}{\pm}3.14^{\circ}$, while after plasma treatment, contact angles of air NTAPPJ and nitrogen NTAPPJ groups were $63.29^{\circ}{\pm}2.27^{\circ}$ and $46.68^{\circ}{\pm}5.82^{\circ}$ respectively. The result showed a significant decrease in contact angle after plasma process(p<0.05). Compared to the control group 6020.33(CFU/mL) without plasma process, CFU decreased significantly after air NTAPPJ 90.75(CFU/mL) and nitrogen NTAPPJ 80.25(CFU/mL) treatment(p<0.05). Conclusions : It was considered that NTAPPJ can be used for denture disinfection without changing surface properties of materials.

Biological Activities of Solvent Fractions of Capsicum annuum Leaves (고추잎 용매 분획물의 생리활성)

  • Biological activities of solvent fractions obtained from Cnsicum annuum leaves, being used in material of functional food, were examined by the methods of DPPH scavenging activity, reducing power, nitrite scavenging activity, antimicrobial activity and inhibitory effect on tyrosinase activity. The highest yield was obtained from water fraction, where as the lowest yield was obtained from ethyl acetate traction, 16.9% and 0.6%, respectively. Hydrogen donating activity of Capsicum annuum leaves in increased with increasing amount of extract. Reducing power of the ethyl acetate fraction is increased as the amount of extract is increased. Even in the presence of 900 $\mu\textrm{g}$ of ethyl acetate fraction, reducing power was significantly higher than it was fer the control in which there was no extract. Among the various solvent fractions, ethyl acetate fraction showed the strongest scavenging effect on hydrogen peroxide. Nitrite scavenging effects of all concentrations diminished at higher pH, while in the case of pH 1.2, it showed a nitrite scavenging effect of more than 90% at concentration above of ethyl acetate fraction 500 $\mu\textrm{g}$. Among the various solvent fractions from methanol extract of Capsicum annuum leaves, ethyl acetate and butanol fraction showed the strongest antimicrobial activity. Antimicrobial activity of ethyl acetate fraction was 20 mm against Bacillus cereus, 18 mm against Staphylococcus aureus and 17 mm against Streptococcus mutans. Ethyl acetate fraction showed the strongest of inhibitory activity of tyrosinase.
