Polycyclic aromatic hydrocarbons (PAHs) are a kind of toxic environmental pollutants and has been accumulated usually in marine sediments. Due to their potential hazardous to human, removal of PAHs from environments has been great concern. In the present study, the effect of microbial inoculation and the supplementation of mixed form cyclodextrin (M-CD) was assessed in the pre-sterilized or nonsterilized microcosms for optimizing operational conditions for ex situ bioremediation of sediments contaminated by PAHs. Activity of electron transport system (ETSA) was increased by the addition of M-CD regardless of inoculation of microorganisms in microcosms without sterilization. The degradation rate of PAHs in sterilized microcosms was app. 9-20% by the inoculation of single strain and 24-37% by the inoculation of microbial consortium supplemented with 1% M-CD, respectively. The degradation was not observed in microcosms without sterilization under the same conditions. The proportion of inoculated microorganisms also decreased in nonsterilized microcosms. Signals of inoculated bacteria were decreased to detection limit after 2 days in the microcosms without M-CD. In conclusion, microbial inoculation with appropriate carbon sources and removal of natural flora and grazers are required for the efficient ex situ bioremediation of sediments contaminated by PAHs in bioslurry reactor.

Saline corrosion is one of the major degradation mechanisms for stainless steel type 304 (SS304) dry storage cask during the spent fuel interim storage period. Slow strain rate test (SSRT) and neutral salt spray test (NSS) were performed at $85^{\circ}C$ and $200^{\circ}C$ with 0.5 wt% sodium chloride mist sprayed on the cold-rolled SS304 specimens of different degrees of reduction in this study. The weight changes of the NSS specimens tested at $85^{\circ}C$ for 2000 hours differed greatly from those at $200^{\circ}C$. The weight loss of NSS specimens was not significant at $85^{\circ}C$ but the weight gain decreased gradually with increasing the cold-rolled reduction. The yield strength (YS) and ultimate tensile stress (UTS) values obtained from the SSRT tests for lightly cold-rolled specimens in the salt spray environment at $85^{\circ}C$ and $200^{\circ}C$ are slightly lower than in air. But for those with 20% reductions, the specimen strengths were no longer changed by the saline corrosion. The preliminary results demonstrated that the quality and performance of cold-rolled SS304 is acceptable for fabrication of dry storage casks. However, more work on the corrosion behavior of cold-rolled stainless steel in the saline atmosphere is needed to better understand its long-term performance.
Salt accumulation in soils of greenhouse due to the massive application of nitrogen fertilizers causes salt stress on the various crops, a serious problem in domestic agriculture. Since the majority of the salinity is nitrate, the excess nitrate should be removed; therefore, a bacterial strain having high capacity of nitrate uptake and identified as Enterobacter amnigenus GG0461 was isolated from the soils of greenhouse. Optimum conditions for the bacterial growth and nitrate uptake were investigated. GG0461 was able to grow without nitrate; however, nitrate facilitated the growth. The rate of nitrate uptake increased at alkaline pH and both growth and nitrate uptake were maximal at pH 8-9. When the initial pH of culture medium was increased to pH 8 or 9, it was decreased to neutral upon bacterial growth and nitrate uptake. These results imply that the major factor mediating bacterial nitrate uptake is a nitrate/proton antiporter. The fact was supported by the effect of nitrate addition in the absence of nitrate, since the addition of nitrate greatly increased the nitrate uptake and rapidly decreased pH of media.
Members of Chlorociboria are soft-rot ascomycetes that produce blue-green pigment. We investigated the growth characteristics of two Korean species of Chlorociboria, eight strains of Chlorociboria aeruginascens and Chlorociboria poutoensis, under various culture conditions (solid media, temperature, pH) and screened them for extracellular enzyme activity. Although the growth rate was slow, all tested strains of Chlorociboria spp. grew well on potato dextrose agar (PDA; 16.3~42.6 mm after 60 days) or Sabouraud dextrose agar (SDA), but not on malt extract agar (MEA). Compared with C. aeruginascens strains, C. poutoensis strains exhibited higher expression of blue-green pigments on both PDA and SDA media. The optimal temperature for mycelial growth was $20{\sim}25^{\circ}C$, and mycelial growth was lower at $30^{\circ}C$ than at $10^{\circ}C$. All strains tended to have increased mycelial growth as the incubation temperature increased in the range of 10 to $20^{\circ}C$. The optimal pH of potato dextrose broth (PDB) for mycelial growth varied according to the strain under static culture conditions. Maximum biomass production was obtained at pH 6.0 for NIFoS 579 ($114.3{\pm}5.1mg/60days$), but it maintained a stable pigment expression under a broad pH spectrum. The activities of both cellulase and laccase were observed in all tested strains of Chlorociboria spp. Enzyme activities of NIFoS 579 were remarkably higher than those of the other strains. From these results, we suggest that C. poutoensis NIFoS 579 is a potential candidate for use as a source of natural blue-green dye.
A quantitative assay was performed on the effects of gamma-irradiation (30- 300 Gy) on intracellular proliferation of Toxoplosmn gonnii RH tachyzoites in human leukemic HL-60 cells and murine peritoneal macrophages by means of 3H-uracil uptake assay. Infected non-irradiation group (NI) and uninfected group (incubating only host cells) were prepared. The 3H-uracil uptake by tachyzoites of NI group 12-24 hrs after infection was 2,190-4,787 counts per minute for macrophages and 2,967-8,254 for HL-60 cells, whereas the irradiated tachyzoites revealed only 381-703 (100 Gy) and 218-408 (300 Gy) for macrophages, and 1,911-2,618 (30 Gy), 1,253-1,384 (70 Gyl, 1,013-1,090 (100 Gyl, and 483-588 (300 Gy) for HL-60 cells. The proliferation inhibition rate was similar in macrophages and HL-60 cells, for example, 89-94% and 80-94% respectively by 300 Gy, 12-24 hrs after infection. It is concluded that RH tachyzoites of T gondii are severely affected by gamma-irradiation in their capability of Intracellular proliferation.
The process to acquire intron-GUS gene-expressed transformants from somatic embryos (including embryogenic calli) of Rosa hybrida cv. 'Sweet Yellow' using Agrobacterium-meditated transformation method was reported in this study. Somatic embryos including embryogenic calluses were infected with Agrobacterium tumefaciens AGL1 strain (O.D = 0.7~1.6) including intron-GUS gene for 30 min, and were co-cultured for 3 days. After co-cultivation, they were cultured on embryo germination medium (EGM) supplemented with $250\;mg{\cdot}L^{-1}$ cefotaxim at $4^{\circ}C$ for 7 days. Then, transient GUS gene expression was observed. Shoots were regenerated from the shoot primodia induced from the intron-GUS gene-transferred either somatic embryos or embryogenic calli cultured on EGM supplemented with both cefotaxim $250\;mg{\cdot}L^{-1}$ and ppt $2\;mg{\cdot}L^{-1}$. Before induction of rooting from shoots cultured on shoot growing medium supplemented with both cefotaxim $250\;mg{\cdot}L^{-1}$ and ppt $2\;mg{\cdot}L^{-1}$, the shoots were cultured on multi-shoot induction medium supplemented with both cefotaxim $250\;mg{\cdot}L^{-1}$ and ppt $2\;mg{\cdot}L^{-1}$ to induce multi-shoots. When expression of the gene from a part of the multi-shoots was identified by GUS transient assay, the putative transgenic multishoots were transferred to rooting medium supplemented with cefotaxim $250\;mg{\cdot}L^{-1}$. After the formation of healthy roots, transgenic plantlets were transferred to the greenhouse after acclimatization. The expression rate of the intron-GUS gene in the multi-shoots was 100%.
Purpose : It is known that carriage rates of Staphylococcus aureus (S. aureus) are highest in newborns and that the asymptomatic carriage of methicillin-resistant Staphylococcus aureus (MRSA) is associated with invasive MRSA infection with the colonizing strain. This study was carried out to investigate the carriage rates of MRSA in neonates with neonatal jaundice. Methods : We reviewed the medical records of 545 neonates admitted with neonatal jaundice to neonatal intensive care units between January 2006 and December 2010. Nasal and inguinal swab specimens had been taken from them and cultured for the isolation of S. aureus. Antimicrobial susceptibility tests had been done for such isolates to determine methicillinresistance. Results : Out of 545 neonates, 318 (58.3%) were colonized with S. aureus and 214 (39.3%) were colonized with MRSA. Results of the antibiogram analysis showed that 65.7% of MRSA isolates were likely to be community-associated (CA) MRSA. Conclusion : Based on the MRSA carriage rate of 39.3%, a surveillance program for MRSA colonization is considered necessary in neonates transferred from other clinics or hospitals. Out of MRSA isolates, 65.7% were likely to be CA-MRSA. This suggests that CA-MRSA strains were already present in obstetric clinic environments where the neonates were born. It is thought that MRSA surveillance programs in these environments are also necessary.
We compared the transcriptome in response to propanol stress in wild-type and propanol-resistant mutant Escherichia coli using the DNA microarray technique. The correlation value of RNA expression between the propanol-treated wild type and the untreated-one was about 0.949, and 50 genes were differentially expressed by more than twofold in both samples. The correlation value of RNA expression between the propanol-treated mutant and the untreated one was about 0.951, and 71 genes in two samples showed differential expression patterns. However, the values between the wild type and mutant, regardless of propanol addition, were 0.974-0.992 and only 1-2 genes were differentially expressed in the two strains. The representative characteristics among differentially expressed genes in W3110 or P19 treated with propanol compared to untreated samples were up-regulation of hest shock response genes and down-regulation of genes relating to ribosome biosynthesis. In addition, many genes were regulated by transcription regulation factors such as ArcA, CRP, FNR, H-NS, GatR, or PurR and overexpressed by sigma factor RpoH. We confirmed that RpoH mediated an important host defense function in propanol stress in E. coli W3110 and P19 by comparison of cell growth rate among the wild type, rpoH disruptant mutant, and rpoH-complemented strain.

In order to characterize the microscopic fracture behaviour of the weldment din stress corrosion cracking(SCC) phenomena, SCC and acoustic emission(AE) tests were carried out simultaneously and the correlation between mechanical paramenters obtained from SCC and AE tests was investigated. In the case of base metal, much more AE events were produced at -0.5V than at -0.8V because of the dissolution mechanism before the maximum load. Regardless of the applied voltages to the specimens, however, AE events decreased after the maximum load. In the case of weldment, lots of AE events with larger amplitude $range(40{\sim}100dB)$ were produced because of the singularities of weld HAZ in comparision to the base metal and post-weld heat-treated(PWHT) specimens. Numerous and larger cracks for the weldment were observed on the fractured surfaces by SEM examination. From these results, it was concluded that SCC for the weldment appeared most severely in synthetic seawater. Weld HAZ was softened by PWHT which also contributed to the reduced susceptibility to corrosive environment in comparison to the weldment.

This study is to evaluate the characteristics of SCC at the welded region of high strength steel using acoustic emission(AE) method. Specimens were loaded by a slow strain rate method in synthetic seawater and the damage process was monitored simultaneously by AE method. Corrosive environment was controlled using the potentiostat, in which -0.8V and -1.1V were applied to the specimens. In the case of one-pass weldment subjected to -0.8V, much more AE counts were detected compared with the PWHT specimen. It was verified through the cumulative counts that coalescence of micro cracks and cracks for the one pass weldment with -0.8V were mostly detected. In case of the one pass weldment subjected to -1.1V, time to failure became shorter and AE counts were produced considerably as compared with that of the two pass weldment. It was shown that AE counts and range of AE amplitude have close relations with the number and size as well as width of the cracks which were formed during the SCC.
