A Study on Shaft Alignment of the Rotating Machinery by Using Strain Gages (스트레인게이지를 이용한 회전체의 축정렬 연구)

  • Misaligned shafts of the rotating machinery have caused noise, vibration. bearing failures, and stress concentration of coupling parts which decrease the efficiency and life of shaft systems. Therefore the proper shaft alignment of those system should be monitored continuously in dynamic condition. To solve these problems under dynamic condition a telemetry system is used. In this study, the condition of the least bending moment which is known by analyzing the structure and stress induced by misalignment is found. After the shaft is aligned by dial gage, a telemetry system with strain gages is installed on shaft. The relationship between bearing displacement and moment of coupling part influenced by misalignment is investigated. The moment derived from two shaft strain at the nearby coupling is measured. The bending strain is measured 5 times for average in static state as well as in dynamic state with 100∼700 rpm.

Isolation and Characterization of an Extremely Thermophilic Sulfur-metabolizing Bacterium from a Deep-sea Hydrothermal Vent System

  • A water sample was taken from a black smoker chimney of a deep-sea hydrothermal vent by using an unmanned submersible "Dolphin 3K". The temperature of the hydrothermal fluid from the black smoker was $276^{\circ}C$. After isolation by repeated serial dilutions, An extremely thermophilic bacterial strain was selected. The strain designated as DT1331, was an anaerobic, non-motile, coccoid shaped bacterium with about 0.5 to $1.0\;\mu\textrm{m}$ in diameter. The strain DT1331 could grow up to $93^{\circ}C$, but the optimum temperature of this strain was $80^{\circ}C$. The growth occurred in the pH range of 4.5 to 8.5 and the optimum pH was 6.0. The strain DT1331 required 1% to 5% NaCl for growth and cell lysis was observed below 1% NaCl concentration. The bacterium could grow on polypeptides such as tryptone, peptone, soytone and on proteins such as casein or gelatin. However, no growth was observed on single amino acids, sugar and organic acids. Hydrogen gas was detected slightly during growth. This bacterium obligately required elemental sulfur and hydrogen sulfide gas was produced during growth.

Development of Bifidobacterium bifidum Strains Resistant to Rifampicin and Ofloxacin (Rifampicin과 Ofloxacin에 내성인 Bifidobacterium bifidum 균주의 개발)

  • Bifidobacterium bifidum, one strain of medical preparation being on the market for human intestinal disorders, was sensitive to rifampicin and fluoroquinolones. If this preparation is taken with rifampicin and fluoroquinolones, its therapeutic effect can't be expected. Serial passage of B. bifidum RFR61, which was obtained by MNNG mutation method, on agar with 2-fold minimal inhibitory concentration of ofloxacin produced B. bifidum OFR9 with minimal inhibitory contentrations of fluoroquinolones up to $4{\sim}256-fold$ higher than that for the original strain. B. bifidum OFR9 produced almost the same amount of organic acid as parental strain. This strain showed growth inhibitory activity against E. coli NM522, Shigella dysenteriae ATCC9752 and E. coli 078. No inactivations of rifampicin and ofloxacin by this resistant mutant strain were found.

Stress-strain distribution at bone-implant interface of two splinted overdenture systems using 3D finite element analysis

  • PURPOSE. This study was accomplished to assess the biomechanical state of different retaining methods of bar implant-overdenture. MATERIALS AND METHODS. Two 3D finite element models were designed. The first model included implant overdenture retained by Hader-clip attachment, while the second model included two extracoronal resilient attachment (ERA) studs added distally to Hader splint bar. A non-linear frictional contact type was assumed between overdentures and mucosa to represent sliding and rotational movements among different attachment components. A 200 N was applied at the molar region unilaterally and perpendicular to the occlusal plane. Additionally, the mandible was restrained at their ramus ends. The maximum equivalent stress and strain (von Mises) were recorded and analyzed at the bone-implant interface level. RESULTS. The values of von Mises stress and strain of the first model at bone-implant interface were higher than their counterparts of the second model. Stress concentration and high value of strain were recognized surrounding implant of the unloaded side in both models. CONCLUSION. There were different patterns of stress-strain distribution at bone-implant interface between the studied attachment designs. Hader bar-clip attachment showed better biomechanical behavior than adding ERA studs distal to hader bar.

Alkaline $\alpha$-amylase Production from Bacillus megaterium

  • The enzyme expressed from strain L-49 was 2.01 times higher than that of original strain. Strain L-49 can grow on culture plate with $50{\mu}g/mL$ ampicillin. The synthesis of $\alpha$-amylase was greatly suppressed when strain L-49 was grown on monosaccharide such as glucose and polysaccharide at the same time cell concentration was low. Amylase production was enhanced when the bacterium was grown on starch and dextrin. Among different nitrogen sources tried, yeast extract was found to be the best followed by panpeptone, peptone, meat extract, bean meal, and corn steep liquor. The average rate of enzyme production was enhanced for 3~4 times in fermentation time from 24h to 44h. The sugar uptake rate has also increased. Low oxygen supply rate enhanced the rate of strain propagation but depressed the enzyme production. Hence it is benefit to obtain high enzyme activity that agitation speed maintained not lower than 400r/min and aeration rate maintained greater than 1:1vvm.

  • When corrugated board is folded at the severely low humidity condition, crack can occur along the scored (or creased) lines of linerboard. This phenomenon is called as score (or crease) crack. It is mainly resulted from the excessive concentration of stress on the outer layer of linerboard. To overcome score crack, many approaches including the installation of constant temperature and humidity system, displacement of low grade raw material by long and strong fibers, or application of water have been tried. We examined the effect of the weight fraction of top layer in two-ply sheet, freeness of top layer stock and wet pressing on strain and elastic modulus of sheet to prevent score crack. Lower freeness and higher press load increased the density and elastic modulus of sheet. Pressing load over the $50kgf/cm^2$, however, decreased the strain of sheet. The weight fraction of top layer had positive effect on strain as well as elastic modulus without increasing the density of sheet.

  • The objective of the current study is to determine cross-sectional profile of intermediate dies in order to improve the plastic strain homogeneity which directly affects not only the dimensional accuracy but also the mechanical properties of final product by redesigning the intermediate dies using the conventional electric field analysis (EFA) method. Initially, the multi-pass shape wire drawing was designed by using the equivalent potential lines from EFA. The area reduction ratio was calculated from the number of passes in multi-pass shape wire drawing but constrained by the capacity of the drawing machine and the drawing force. In order to compensate for a concentration of strain in a region of the cross section of the wire, the process for multi pass wire drawing from initial round material to an intermediate die was redesigned again using the electric field analysis. Both drawing process designs were simulated by the finite element method in which the strain distribution and standard deviation plastic strain of the cross section of drawn wires were examined.

  • The axisymmetric methane-air counterflow flame in microgravity was simulated to investigate effects of the global strain rate on the flame structure. The flame shapes and profiles of temperature and the axial velocity for the mole fraction of methane in the methane-nitrogen fuel stream, Xm= 20, 50, $80\%$, and the global strain rate, ag= 20, 60, 90 $s^{-1}$ each mole fraction were compared. The profiles of the temperature and axial velocity of the axisymmetric simulations were in good agreement with those of OPPDIF, an one-dimensional flamelet code. It was confirmed that the flame is stretched more and the flame radius increases and the flame thickness decreases as the global strain rate increases.

  • This is the study on fracture life under the interaction of creep and fatigue. It is difficult to explain the interaction of the creep and fatigue with indication of frequency but the dependency of the time should be considered. The formulation of material varieties causing by interaction of creep and fatigue is required in the accumulative damage method. The strain range partition method requires some of modification corresponding to the changes in temperature and load. All of other method also comprehended with above mentioned problems. Generally, in this field, the variety of stress-strain and suitable parameter is required and connective study between the macro and micro results seems to be insufficient. The linear damage rule is acquiring the support generally but it requires modification in the hgigh temperature instruments. The variety of stress effecting on crack and variety of stress on the metallurgical side are considered to be problems in the future days.

Investigation of the Cultural Characteristics of High Concentration Ehtanol Resistant Acetobacter sp. (고농도 에탄올내성 초산균의 개발 및 배양특성)

  • To increase the yield of acetic acid production, the author developed the bacterial strain which could brow well in high concentration of ehtanol from the seed culture using in conventional vinegar production factory. By attenuation of the isolated strain in the broth media containing 5-10% ethanol, we could get the strain which could grow in the broth medium containing 10% ethanol. This strain was identified and named as Acetobacter sp. FM-10, and it's cultural characteristics were also investigated. The medium containing 10% ethanol, 5% glucose and 1% yest extract was suitable for the acetic acid production with Acetobacter sp. FM-10. Optimum temperature and pH for the growth of Acetobacter sp. FM-10. were $30^{\circ}C$ and 5.0, respectively. The acidity of culture medium was reached to 9.0 % after 20 days static cultivation at $30^{\circ}C$.
