Development of Solar Warehouse for Drying and Storing the Agricultural Products (농산물(農産物) 건조(乾燥) 및 저장(貯藏)을 위(爲)한 태양열(太陽熱) 저장고(貯藏庫)의 개발(開發)에 관(關)한 연구(硏究))

  • Recent concern regarding price and availability of fossil fuels has spurred the interest in alternative sources for farm crop drying. Among the available options such as biomass energy, wind power, nuclear energy and solar energy etc., the increasing attention is being directed to the utilization of heat from solar energy especially for farm crop drying. Even though solar energy is dispersed over a large land area and only a relatively small amount of energy can be simply collected, the advantages of solar energy is that the energy is free, non-polluting. The study reported here was designed to help supply the informations for the development of simple and relatively inexpensive solar warehouse for farm crop drying and storage. Specifically, the objectives of this study were to determine the performance of the solar collector fabricated, to compare solar supplemented heat drying with natural air drying and to develop a simulation model of temperature in stored grain, which can be used to study the effects due to changes in ambient air temperature. For those above objectives, solar collector was fabricated from available materials. Corrugated steel galvanized sheet, painted flat black, was used as absorbers and clear 0.2mm polyethylene sheet was the cover material. The warehouse for rough rice drying and storage was constructed with concrete block, and the solar collector was used as the roof of warehouse instead of original roofing system of it. The results obtained in this study were as follows: 1. The thermal efficiency of the solar collector was average 26 percent and the overall heat transfer coefficient of the collector was approximately $25kJ/hr.m^2\;^{\circ}K$. 2. Solar heated air was sufficient to dry one cubic meter of rough rice from 23.5 to 15.0 percent in 7 days and natural air was able to dry the same amount of rough rice from 20.0 to 5 percent in l2 days. 3. Drying with solar heat reduced the required drying time to dry the same amount of rough rice into a half compared to natural air drying, but overdrying problems of the bottom layer were so severe that these problems should be thoroughly analyzed. 4. Simulation model of temperature in stored grain was developed and the results of predicted temperature agreed well with test results. 5. Based on those simulated temperature, changes in the grain-temperature were a large at the points of the wallside and the damage of the grain would be severe at the contact area of wall.

Effect of Storage Method on Seed Chemical and Germination Traits in Barley (보리 저장방법에 따른 주요 성분 및 발아력 변화)

  • This study was carried out to investigate storability and develop suitable storage method for barley. Rough and milled barley such as Olbori(covered barley), Ssalbori(naked barley) and Chalssalbori (waxy naked barley) were packaged in 3P bag and OPP film bag, respectively, and stored at ambient and low temperature warehouse conditions. The weight loss was higher in milled barley than that of rough barley during barley storage. Acid value, one of the most important factor for grain quality evaluation, was increased from 4.9-9.0 mg KOH/100g to 6.5-19.9mg KOH/100g in milled barley at 16 months after storage. Reducing sugar content was increased while germination and water absorption rates were decreased during barley storage period. In terms of penal taste of boiled barley, rough barley could be stored safely more than one year regardless of storage methods where-ase milled barley stored in ambient temperature ware-house produced nasty flavor at 8 to 10 months after storage.

Changes in Physicochemical Properties of Rice Grain during Long-Term Storage (장기저장한 벼 종실의 이화학적 특성 변화)

  • The experiment was carried out to investigate the changes in physicochemical properties of milled rice harvested in different year and stored for four to sixty four months. The fat acidity of milled rice increased sharply from four to sixteen months storage, and after then it was increased slowly to sixty four months. Amylose, magnesium, potassium content, and K/Mg ratio of milled rice were not changed by storage duration. Water absorption rates of milled rice at 21$^{\circ}C$ and 77$^{\circ}C$, and alkali digestion value were increased by longer storage duration. Difference of water absorption rate between rice samples was greater during initial forty minutes after soaking at 21$^{\circ}C$ and with longer the soaking time at 77$^{\circ}C$. Shorter gel length of rice flour was found with prolonged storage duration, while peak, minimum, cool, breakdown and setback viscosity of gelatinized rice flour on amylograph were increased along with increasing the storage duration. The volume expansion rate during cooking and degree of iodine coloration of cooking water were higher in the longer stored rice compared with shorter one, and the amount of soluble solid in cooking water was significantly decreased in rice stored longer period.

Inheritence, linkage and Possible Use of Fractured Starch Mutant in Barley (Hordeum Vulga L.)

  • In order to breed barley lines with reduced viscosity of wort, a fractured starch mutant of naked barley cultivar, Nubet, was obtained from the M2 seeds mutated by the diethyl sulfate treatment. Seeds of this fractured starch mutant were opaque and the endosperm consists of angular, irregular and fractured starch. The mutant was caused by single recessive mutation and assigned by the symbol fra. The gene was located on chromosome 4, distal in long arm by linkage recombinations using translocation homozygote lethal test set. The linkage value between the fractured starch mutant and 72-4a, 72-4d were 26.0$\pm$4.9, 34.2$\pm$3.1 percent respectively. In addition to the reduced seed size, fewer kernels per spike and higher tillering ability, lower $\beta$-glucan viscosity and higher lysine content of the grain were associated with this mutant. $\beta$-glucan viscosity of the Nubet grains increased from 3 weeks after anthesis to matury and most of the viscose substances appeared to be stored in the middle of the endosperm tissue. Since the mutant grains showed better milling property as compared to Nubet, it can be used as breeding resources to develope new barley cultivars for maltins and milling purpose.

SIMULATION OF HIGH BURNUP STRUCTURE IN UO2 USING POTTS MODEL

  • The evolution of a high burnup structure (HBS) in a light water reactor (LWR) $UO_2$ fuel was simulated using the Potts model. A simulation system for the Potts model was defined as a two-dimensional triangular lattice, for which the stored energy was calculated from both the irradiation damage of the $UO_2$ matrix and the formation of a grain boundary in the newly recrystallized small HBS grains. In the simulation, the evolution probability of the HBS is calculated by the system energy difference between before and after the Monte Carlo simulation step. The simulated local threshold burnup for the HBS formation was 62 MWd/kgU, consistent with the observed threshold burnup range of 60-80 MWd/kgU. The simulation revealed that the HBS was heterogeneously nucleated on the intergranular bubbles in the proximity of the threshold burnup and then additionally on the intragranular bubbles for a burnup above 86 MWd/kgU. In addition, the simulation carried out under a condition of no bubbles indicated that the bubbles played an important role in lowering the threshold burnup for the HBS formation, thereby enabling the HBS to be observed in the burnup range of conventional high burnup fuels.

Starch Phosphorylase and its Inhibitor from Sweet Potato Root

  • Based on a tracer study, starch phosphorylase was implicated as an agent in the starch synthesis in sweet potato roots. The enzyme was purified from the tissue as a cluster of isozymes with an average mw of 205K (fresh roots) or 159K (roots stored for 3 mon.). On SDS polyacrylamide gel electrophoresis, one large subunit of 98K mw and several small ones of 47${\sim}57K mw were observed. From the mw data and the results of peptide mapping and immunoelectrophoretic blotting using mono- and polyclonal antibodies, it was deduced that a large part of the large subunit was cleaved at the middle part of the peptide chain to give rise to the small subunits, and on storage, the enzyme molecules were further modified by proteolysis. During the course of phosphorylase purification, a proteinaceous inhibitor of the enzyme was isolated. It had a mw of 250K and was composed of 5 identical subunits of 51K mw. In the direction of starch synthesis, the inhibitor showed a noncompetitive kinetics with a Ki of $1.3{\times}10^{-6}\;M$. By immunohistochemical methods, both the enzyme and the inhibitor were located on the cell wall and amyloplast. Crossreacting materials of the inhibitor were present in spinach leaf, potato tuber and rice grain. These findings indicate the wide occurrence of the inhibitor and also imply its possible participation in regulating starch phosphorylase activity in vivo.

  • Optical Color Corescanner firstly developed by DMT-GeoTec, Germany and further upgraded through the Korea-Germany joint project is capable of duplicating the core surfaces. The tool uses a digital CCD line camera. As the core is rotated by an electric motor, the camera scans the uppermost line, everytime with a circumferential increment of up to 0.05mm(20pixels/mm) and hence a complete 360$^{\circ}$ unwrapped image(core image) is produced. This paper illustrated diverse research benefits of such core images from several test sites in our country. All scanned images could be stored as a data-base one and easily used with software facilities \circled1 to evaluate a percental distribution of mineral components or grain size etc. not only for the rock classification but also for e.g. the assessment of building stones, \circled2 to study potential reservoirs as a hydrocarbon indicator using ultraviolet fluorescence reflection from cores, \circled3 to facilitate the qualitative and quantitative analysis of fractures, \circled4 to evaluate the fractures and thin bedded reservoirs using spectral color responses. Based on abundant scanning experiments, it would seem that this imaging work should lead to reflecting the future trend in underground survey toward a more comprehensive understanding of the properties and behaviors of in situ rocks.

  • Recently, one of the hydrogen production methods has attracted using dense metallic membrane. It has high hydrogen permeation and selectivity which hardly could adopt industrial product because of high cost, hydrogen embrittlment and thermal stability. Meanwhile, vanadium has high hydrogen solubility and it use to instead of Pd-Ag amorphous membrane. Aluminum carried out blocking hydrogen diffusion on grain boundary therefore protecting hydrogen embrittlement. Most of dense metallic membrane is solution diffusion mechanism. The solution diffusion mechanism was very similar hydrogen storing steps such as steps of metal hydride. Thus, V-Al composites were fabricated to use hydrogen induced mechanical alloying. The fabricated V-Al composites were characterized by XRD, SEM, EDS and simultaneous TG/DSC analyses. The hydrogenation behaviors were evaluated using a Sievert's type automatic PCT apparatus. The hydrogenation behaviors of V-Al composites was evaluated too low hydrogen stored capacity and fast hydrogenation kinetics. In PCI results, V-Al composites had low hydrogen solubility, in spite of that, hydrogen kinetics was calculated very fast and hydrogen absorption/desorption contents were same capacity.

Storage of Pollen Biofactory for in vitro Growth and rProtein Synthesis (기내배양과 재조합 단백질 생산을 위한 화분 Biofactory의 저장기술의 개발)

  • A method of collection and long-term storage of viable lily (Lilium longiflorum) pollen grains were developed for their in vitro growth and transformation in consistency. Petroleum ether, n-heptane, cyclohexane and benzene, as pollen collection medium, were determined less toxic to pollen growth in vitro than others tested. Pollen grains, however, lost their growth activity if stored in these solvents more than a week, So, a serial performance, that is, pollen grain collection in these solvents, air-drying and immediate transfer to low temperature condition was determined desirable for keeping the viability much longer. Pollen grains from this storage showed a successful transformation in vitro with a cDNA encoding tissue plasminogen activator (TPA) protein using Agrobacterium via vacuum infiltration according to western blotting analysis.

Fermentative Characteristics of Extruded Meju by the Molding Temperature (메주의 압출성형에서 성형온도에 따른 메주의 발효특성)

  • Effect of molding temperature on the quality changes of extruded meju was studied. Meju was molded at 8$0^{\circ}C$, 6$0^{\circ}C$ and 4$0^{\circ}C$, and then stored at $25^{\circ}C$ with 50% of relative humidity for 30 days. The texture of the cooked soybean grain showed that the firmness and cohesiveness were increased, and consistency was decreased by decrease of molding temperature. The density of the meju molded at 8$0^{\circ}C$, 6$0^{\circ}C$ and 4$0^{\circ}C$ were 1.072g/mL, 1.079g/mL and 1.203g/mL, respectively. The meju molded at 4$0^{\circ}C$ had significantly higher density than those molded at 8$0^{\circ}C$ or 6$0^{\circ}C$. Also, delay of water evaporation, acidification, and rapid growth of fungal mycellium were observed on the sample with molding temperature at 4$0^{\circ}C$ during fermentation. Activity of amylase and protease, contents of total reducing sugar and amino nitrogen of 4$0^{\circ}C$-molded meju were represented lower level than those of 6$0^{\circ}C$ or 8$0^{\circ}C$-molded sample. Therefore, it was considered that the molding temperature was an important factor for meju fermentation and molding temperature of 6$0^{\circ}C$ or over would be acceptable.
