Perforation Adjustment of Unit Package for 'Fuji' Apples during Short-term Cold Storage and Export Simulation ('후지' 사과의 단기 저온저장 및 모의수출 과정에서 소포장의 천공도 조절 효과)

  • Various types of unit packaging methods were applied for 'Fuji' apples during short-term cold storage and export simulation. Gas tightness of the package was controlled stepwise in the successive two-year experiments using different perforation treatments (none, punch hole, or pinhole) and sealing methods (tie v s. heat seal). Risk of tight packaging and effectiveness of macroperforation on weight loss and quality maintenance were analyzed as related to changes in gas concentration inside the packages. Immediately after harvest, each 5 apple units were packaged in $40{\mu}m$ polypropylene (PP) film bags, stored 4 weeks at $0^{\circ}C$, and then put on the shelf for one week at ambient temperature in the preliminary experiment, In the main experiment, export process was imposed after storage simulating 2 week refrigerated container shipment at $0^{\circ}C$ plus one week local marketing at ambient temperature. Non-perforated film packaging with relatively high gas tightness induced flesh browning caused by carbon dioxide accumulation regardless of the sealing methods. Among perforated film packaging, in contrast, atmospheric modification was partly established only in the pinhole treatment and flesh browning symptom was not observed in all the treatments. Even the punch hole perforated film packaging without gas tightness effectively reduced the weight loss, whereas had slight benefits for quality maintenance. Reduced perforation using pinhole treatment seemed to improve sensory texture, while effects on physicochemical quality were insignificant. Overall results suggest the need of more minute perforation treatments on the packaging film to ensure modified atmosphere effects on quality maintenance.

  • Monitoring of soil fertility and crop productivity in long-term application of silicate fertilizers is necessary to use fertilizers efficiently. This study was conducted to investigate effects of continuous application of silicate fertilizer for rice cultivation from 1969 to 2014. The treatments were no silicate fertilizer treatments (N, NC, NPK, and NPKC) and silicate fertilizer treatments (N+S, NC+S, NPK+S, and NPKC+S). The 46-yr input of $2\;ton\;ha^{-1}yr^{-1}$ of silicate fertilizer increased pH 0.6 ~ 1.1 and exchangeable Ca $2.0{\sim}2.4cmol_c\;kg^{-1}$ in silicate fertilizer treatments (N+S, NC+S, NPK+S, and NPKC+S) compared with no silicate fertilizer treatments (N, NC, NPK, and NPKC) because silicate fertilizer included Ca component. Also, available silicate concentrations of silicate fertilizer treatments (N+S, NC+S, NPK+S, and NPKC+S) increased $169mg\;kg^{-1}$ compared to no silicate fertilizer treatments. In Period II ('90~'14), the mean annual Si field balance varied from 62 to $175kg\;ha^{-1}yr^{-1}$ in silicate fertilizer treatments, indicating continuous accumulation of soil Si. Silicon uptake and grain yield of rice had greater differences between N treatment and N+S treatment than other treatments. This showed that the application of silicate fertilizer had greater effect in nutrient-poor soils than in proper nutrient soils. Thus the application of silicate fertilizer led to improvement the fertility of soil and increasement of rice production for the lack of soil nutrients.

Distinct sets of lysosomal genes define synucleinopathy and tauopathy

  • Neurodegenerative diseases are characterized by distinct protein aggregates, such as those of α-synuclein and tau. Lysosomal defect is a key contributor to the accumulation and propagation of aberrant protein aggregates in these diseases. The discoveries of common proteinopathies in multiple forms of lysosomal storage diseases (LSDs) and the identification of some LSD genes as susceptible genes for those proteinopathies suggest causative links between LSDs and the proteinopathies. The present study hypothesized that defects in lysosomal genes will differentially affect the propagation of α-synuclein and tau proteins, thereby determining the progression of a specific proteinopathy. We established an imaging-based high-contents screening (HCS) system in Caenorhabditis elegans (C. elegans) model, by which the propagation of α-synuclein or tau is measured by fluorescence intensity. Using this system, we performed RNA interference (RNAi) screening to induce a wide range of lysosomal malfunction through knock down of 79 LSD genes, and to obtain the candidate genes with significant change in protein propagation. While some LSD genes commonly affected both α-synuclein and tau propagation, our study identified the distinct sets of LSD genes that differentially regulate the propagation of either α-synuclein or tau. The specificity and efficacy of these LSD genes were retained in the disease-related phenotypes, such as pharyngeal pumping behavior and life span. This study suggests that distinct lysosomal genes differentially regulate the propagation of α-synuclein and tau, and offer a steppingstone to understanding disease specificity.

  • Hunter syndrome or mucopolysaccharidosis type II (MPS-II) (OMIM 309900) is a rare lysosomal storage disorder caused by deficiency in the activity of the enzyme iduronate-2-sulfatase. This enzyme is responsible for the catabolism of the following two different glycosaminoglycans (GAGs): dermatan sulfate and heparan sulfate. The lysosomal accumulation of these GAG molecules results in cell, tissue, and organ dysfunction. Patients can be broadly classified as having one of the following two forms of MPS II: a severe form and an attenuated form. In the severe form of the disease, signs and symptoms (including neurological impairment) develop in early childhood, whereas in the attenuated form, signs and symptoms develop in adolescence or early adulthood, and patients do not experience significant cognitive impairment. The involvement of the skeletal-muscle system is because of essential accumulated GAGs in joints and connective tissue. MPS II has many clinical features and includes two recognized clinical entities (mild and severe) that represent two ends of a wide spectrum of clinical severities. However, enzyme replacement therapy is likely to have only a limited impact on bone and joint disease based on the results of MPS II studies. The aim of this study was to review the involvement of joints in MPS II.

Lyso-globotriaosylsphingosine induces endothelial dysfunction via autophagy-dependent regulation of necroptosis

  • Fabry disease is a lysosomal storage disorder characterized by the lysosomal accumulations of glycosphingolipids in a variety of cytotypes, which include endothelial cells. The disease is inherited and originates from an error in glycosphingolipid catabolism caused by insufficient α-galactosidase A activity, which causes uncontrolled progressive storage of intracellular globotriaosylceramide (Gb3) in the vasculature and extracellular accumulation of lyso-Gb3 (a deacetylated soluble form of Gb3). Necrosis can lead to inflammation, which exacerbates necrosis and creates a positive feedback loop that triggers necroinflammation. However, the role played by necroptosis, a form of programmed necrotic cell death, in the cell-to-cell inflammatory reaction between epithelial and endothelial cells is unclear. Thus, the present study was undertaken to determine whether lyso-Gb3 induces necroptosis and whether necroptosis inhibition protects endothelial dysfunction against lyso-Gb3 inflamed retinal pigment epithelial cells. We found lyso-Gb3 induced necroptosis of a retinal pigment epithelial cell line (ARPE-19) in an autophagy-dependent manner and that conditioned media (CM) from ARPE-19 cells treated with lyso-Gb3 induced the necroptosis, inflammation, and senescence of human umbilical vein endothelial cells. In addition, a pharmacological study showed CM from lyso-Gb3 treated ARPE-19 cells induced endothelial necroptosis, inflammation, and senescence were significantly inhibited by an autophagy inhibitor (3-MA) and by two necroptosis inhibitors (necrostatin and GSK-872), respectively. These results demonstrate lyso-Gb3 induces necroptosis via autophagy and suggest that lyso-Gb3 inflamed retinal pigment epithelial cells trigger endothelial dysfunction via the autophagy-dependent necroptosis pathway. This study suggests the involvement of a novel autophagy-dependent necroptosis pathway in the regulation of endothelial dysfunction in Fabry disease.

Inhibitory Effect of Garlic Extract on Histamine Accumulation in Mackerel Meat (마늘 추출물의 고등어육에서의 히스타민 생성 억제)

  • This study was carried out to investigate the effects of garlic extract on histamine reduction in mackerel meat stored at $4^{\circ}C$ and $15^{\circ}C$, respectively. The number of total bacteria in mackerel meat treated with 7.5% garlic extract was ten times lower than that of the control stored for 6 days of $4^{\circ}C$. However, there was no difference among the samples after 9 days of storage. Reduction of eicosapentaenoic acid (C20:5) and docosahexaenoic acid (C22:6) contents was observed in the normal control group at both temperatures. However, contents of these two polyunsaturated fatty acids slightly increased in garlic extract-treated fish. Amounts of volatile basic nitrogen increased over time in the control group, but decreased in the garlic extract-treated meat, indicating that garlic extract might suppress decomposition. No histamine was detected initially in all samples. However, a small amount of histamine (42.87 mg/kg) was detected in the control at 3 days after storage at $4^{\circ}C$. Histamine content increased continuously with storage period but was lower in mackerel meat containing garlic extract, in which histamine suppression was proportional to the concentration of garlic extract. It can be concluded that garlic extract could be utilized to extend the storage period of mackerel.

  • The "Ginseng Pork" produced by feeding ginseng by-products can be a compatible product in the sense of increasing pork consumption and developing functional food in the international pork market. This experiment was conducted to produce "Ginseng Pork" with emphasis on growth performance and meat quality. Experiments were conducted in which 30 Landrace heads were fed with bark of ginseng root(BGR) or heating extracts ginseng leaves and stem(HEG). WB-shear force was not different among the treatment groups until 15 days of ageing, but pork fed with the 6% BGR showed a higher shear force at 20 day of storage at 4$^{\circ}C$. Cooking loss showed lower value for the 9% BGR group compared with the control group. At 15 day, the 3% and 9% BGR groups showed lower cooking losses than control. Pork groups fed HEG showed a significantly(p<0.05) lower TBARS values after 5 days of storage. As for VBN analysis, the feeding groups of 9% BGR and 5.5% HEG had significantly lower values at 5 and 20 days when compared to the other treatment groups. It might be concluded that the accumulation of ginseng saponin in the pork resulted in retarding the ageing and inhibiting the oxidation.

Effects of Different Carbon Dioxide and Oxygen Concentration on the Quality of Oriental Melon during CA Storage (참외 CA저장시 $CO_2$$O_2$농도에 따른 품질 변화)

  • Oriental melon was stored in controlled atmospheres(5, 10 or 15% CO$_2$in combination with 3 or 10% O$_2$) and air as control, and was analyzed half monthly during a 45-day storage for various quality Parameters. Oriental melon which stored in CA condition was lower in weight loss and decay rate than that stored in air. There was no significant difference in firmness by CO$_2$or O$_2$concentration during a month of storage. Fruits stored in 10% CO$_2$+3% O$_2$showed higher level of soluble solids, Vitamin C content and L value than other condition. And also comparatively lower endogenous ethanol accumulation and better state of quality was observed in that condition.

Soil Carbon Storage in Upland Soils by Biochar Application in East Asia: Review and Data Analysis (바이오차를 이용한 밭 토양 탄소 저장: 동아시아 지역 연구 리뷰 및 데이터 분석)

  • BACKGROUND: Biochar is a solid material converted from agricultural biomass such as crop residues and pruning branch through pyrolysis under limited oxygen supply. Biochar consists of non-degradable carbon (C) double bonds and aromatic ring that are not readily broken down by microbial degradation in the soils. Due to the recalcitrancy of C in biochar, biochar application to the soils is of help in enhancing soil carbon sequestration in arable lands that might be a strategy of agricultural sector to mitigate climate change. METHODS AND RESULTS: Data were collected from studies on the effect of biochar application on soil C content conducted in East Asian countries including China, Japan and Korea under different experimental conditions (incubation, column, pot, and field). The magnitude of soil C storage was positively correlated (p < 0.001) with biochar application rate under field conditions, reflecting accumulation of recalcitrant black C in the biochar. However, The changes in soil C contents per C input from biochar (% per t/ha) were 6.80 in field condition, and 12.58 in laboratory condition. The magnitude of increment of soil C was lower in field than in laboratory conditions due to potential loss of C through weathering of biochar under field conditions. Biochar production condition also affected soil C increment; more C increment was found with biochar produced at a high temperature (over 450℃). CONCLUSION: This review suggests that biochar application is a potential measures of C sequestration in agricultural soils. However, as the increment of soil C biochar was affected by biochar types, further studies are necessary to find better biochar types for enhanced soil C storage.

Effects of Dietary Conjugated Linoleic Acid Levels and Periods on Meat Quality in Breast and Thigh Muscles of Broiler (Conjugated Linoleic Acid 급여기간과 첨가수준이 저장기간 중 육계의 부위별 품질특성에 미치는 영향)

  • A total of 180 Arbor Acre broilers (35 days of age) were fed a diet containing 0%, 0.6%, 1.2%, 1.8%, 2.4% or 4.8% conjugated linoleic acid (CLA) for 3 weeks then slaughtered at week 1, 2 and 3. Thigh and breast muscles were seamed out, vaccum-packed prior to storage in a chill for days 7 to analyse thiobarbituric acid reactive substances (TBARS). The accumulation of CLA in breast and thigh muscles was increased significantly (P<0.05) as the CLA level was increased, but no significant difference amongst the treatments. The contents of saturated fatty acids were significantly increased(P<0.05) with an increase of dietary CLA level, but those of unsaturated fatty acids were significantly decreased (P<0.05). The content of mono unsaturated fatty acid (MUFA) was decreased as dietary CLA level increased, even though that of poly unsaturated fatty acid (PUFA) had a little difference between dietary CLA level. Dietary CLA reduced the degree of lipid oxidation in raw chicken meat during storage.
