Effects of Coptidis Rhizoma Herbal Acupuncture Extract on the Acute Gastric Mucosal Lesion Progression Induced by Compound 48/80 in Rats

  • Objectives : Coptidis Rhizoma has been used for stomach disease. However, its property is so cold that it might be avoided to prescribe for the elderly and the infirm having indigestion or diarrhea. Accordingly, the present study was designed to investigate the protective effects of Coptidis Rhizoma herbal acupuncture extract against acute gastric mucosal lesions induced by compound 48/80 in rats. Methods : The Coptidis Rhizoma herbal acupuncture (CRHA) was injected in Choksamni and Chungwan 1 h before compound 48/80 treatment. The animals were sacrificed under anesthesia 3 h after compound 48/80 treatment. The stomachs were removed and the amount of gastric adherent mucus, gastric mucosal hexosamine, SOD, XO, TBARS and histological examination were performed. Results : The decline of gastric adherent mucus, gastric mucosal hexosamine and the histological defects of gastric mucus were significantly protected by CRHA treatment. Gastric adherent mucus in control group was reduced to $38.2{\pm}5.0%$. CRHA groups significantly protected the loss of mucus to $77.5{\pm}4.9%$. Mucosal hexosamine content showed similar patterns. Mucosal hexosamine content in control group was reduced to $45.2{\pm}6.2%$. CRHA groups significantly protected the loss of mucus to $83.0{\pm}7.0%$. The changes of gastric mucosal SOD and TBARS were recovered by CRHA treatment as well. Conclusions : CRHA showed the protective effects on the acute gastric mucosal lesions induced by compound 48/80 in rats. These results suggest that CRHA may have protective effects on the gastritis.

  • Feeding habits of released Sebastes inermis obtained in daily and monthly collections from August 2007 to December 2007 in coastal waters off Jam Island in Jinhae Bay, Korea were assessed. A total 151 individuals of S. inermis (6.3-9.7 cm TL) were recaptured during the study period. Initial food intake of released S. inermis occurred within three days of released. Stomach content index increased with time, however, the percentage of stomachs devoid of contents decreased. Sebastes inermis mainly consumed gammarid and caprellid amphipods, gastropods and mysids. Diet also included small quantities of bivalves, shrimps, copepods and ostracods. Amphipods were mainly consumed at all size groups and consumption of gastropods, bivalves and shrimps increased with increasing fish size.

  • The natural high productivity of temperate coastal waters in Korea can be used in advantage to reduce the feed costs for the cage-cultured marine carnivorous fish species. By using the night-lights methods an alternative of supplementary feeding strategy can be offered to the cage farmers and maintain sound environmental conditions that could enhance maximum sustainable yields. The aim of this study was to investigate the effects of night-lights which shown positive results on feeding and growth in sea cage cultured rockfish, Sebastes schlegeli. The study showed that for the water around overhead illuminated sea cage, higher zooplankton density was observed at night than during the daytime. Increased amounts of for-age, evidenced by stomach content analyses were observed in the early evening, but decreased amounts were observed in the morning and afternoon. That is, feeding activities of the fish were most intensive from midnight to dawn. In a three month feeding experiment, the results showed that night-lighted groups were superior to groups with the highest feeding efficient. This study suggested that the evidence that night-lights superimposed on only day-lights enhance growth of rockfish in sea cages during summer and winter, with timing of exposure affecting growth of juvenile fish. The capability to control the feeding behavior of marine life via manipulation and external stimuli could considerably benefit the advancement of sea cage aquaculture in coastal areas.

당근 쥬스가 감자 쥬스의 효소적 갈변 반응에 미치는 영향 (Effect of Carrot Juice on Enzymatic Browning of Potato Juice)

  • Potato and carrot are the main sourses of vitamin C and vitamin A each. As a folk ramedy, potato-carrot mixtures have been used to cure a stomach ulcer in Korea. To investigate the effects of carrot juice on the browning of potato juice, we examined the brewing in various ratio of two juices. We also investigated the role of potato juice in the carotene oxidation of carrot juice. Delta "L" values of potato juice were abruptly decreased after 5 min. reaction and they were very different from the juice mixture of potato and carrot. Those containing higher ratio of potato were decreased greatly. In blanching treatment to eliminate the effect of the enzymes in potato and carrot, delta "L" values of cooked potato were decreased a little but those of cooked carrot were decreased greatly. To investigate the fact that the inhibitory effect of carrot juice in potato browning was due to the dilution of polyphenolics of potato juices by carrot juice mixing, we added H20 equivalent to carrot water content to potato juice. The diluted sample showed less decreasing pattern than nondiluted sample. We also added appropriate amount of ${\beta}$-carotene to the same samples. Delta "L" values of with added B-carotene were more slowly decreased than those of without added B-carotene.

흰쥐에서 미주신경자극이 위장관 장크롬친화성세포의 미세구조와 세로토닌 함량에 미치는 영향 (Effect of Vagus Nerve Stimulation on the Ultrastructure and the Serotonin Content of Enterochromaffin Cells in the Gastrointestinal Tract of Rats)

  • The present study was performed to clarify the effect of vagus nerve stimulation on the enterochromaffin(EC) cells in the body of the stomach, the first part of the duodenum and the ceceum of rats by using routine electron microscopy and immunogold labelling. The changes in the ultrastructure and in the labelling density of the gold particles of the EC cells were investigated after vagus nerve stimulation. The vagus nerve was electrically stimulated with a square wave pulse generator for a duration of 5 minutes each, a total of 8 times at 2 minute intervals. Immunogold labelling demonstrated that the epithelial serotonin immunoreactive cells of the gastrointestinal tract are EC cells containing characteristic pleomorphic granules. Immunocytochemically labelled gold particles were largely concentrated in the dense matrix of the granules of the EC cell, and the labelling density of the gold particles considerably increased after the vagus nerve stimulation. Except for a slight activation of Golgi complexes, no remarkable changes in the ultrastructures of the EC cells were noted after the vagus nerve stimulation. The above results suggest that vagus nerve stimulation may activate serotonin biosynthesis in EC cells.

Antioxidative Changes of Blueberry Leaf Extracts in Emulsion-Type Sausage during In Vitro Digestion

  • This study was conducted to investigate the effects of in vitro human digestion on the antioxidant activity of blueberry leaf extracts (BLE) in emulsion-type sausages (ETS). Leaves from four cultivars of blueberries (Bluecrop, Bluegold, Duke, and Northland) collected from a wild blueberry farm were extracted with 80% ethanol. ETS were prepared with 0.2% BLE. The samples were then passed through an in vitro human digestion system which simulates the composition of the mouth, stomach, and small intestine juice. Only one phenolic compound (chlorogenic acid) was detected in the BLE. Northland BLE had appreciably higher amounts of chlorogenic acid than that of other BLE, both before and after in vitro human digestion. Antioxidant activity of any BLE was not influenced by in vitro human digestion, whereas the antioxidant activity of chlorogenic acid standard increased in response to in vitro human digestion in both 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and ferric-reducing ability of plasma (FRAP). In the present study, the antioxidant activities of the BLE were not strongly influenced by in vitro human digestion, and the antioxidant activity depended on the chlorogenic acid content of ETS. Thus, compounds from blueberry leaves may have important applications in the future as natural antioxidants for meat products.

In Vitro Stability of β-galactosidase Microcapsules

  • The present study was carried out to examine the efficiency of microcapsules and a stability of lactase in vitro in the simulated gastric and intestinal conditions. As a coating materials, medium-chain triacylglycerol (MCT) and polyglycerol monostearate (PGMS) were used. The highest efficiency of microencapsulation was found in the ratio of 15:1 as coating to core material with both MCT (91.5%) and PGMS (75.4%). In a subsequent experiment, lactose content was measured to study a microcapsule stability. Lysis of microcapsules made by MCT in simulated gastric fluid was proportionally increased such as 3% in pH 5 and 11% in pH 2 for 20 min incubation. In the case of PGMS microcapsulation, 11-13% of lactose was hydrolyzed at 20 min in all pHs and also very little amount (less than 3%) of lactose was hydrolyzed after 20 min in all pHs. The highest percentages of lactose hydrolysis in MCT and PGMS microcapsules were 68.8 and 60.8% in pHs 7 and 8 during 60 min, respectively. Based on our data, the lactase microcapsules seemed to be stable when they stay in the stomach, and hydrolyzed rapidly in small intestine where the bile acid was excreted.

베르베린 제제의 생체이용율에 관한 연구(II): 황련과 감초 공침물의 항균효과 및 생체이용율 (Studies on the Bioavailability of Berberine Preparations(II) : Antibacterial Activity and Bioavailability of Coprecipitate of Coptidis Rhizoma and Glycyrrhizae Radix)

  • Precipitation reaction occured between berberine in Coptidis Rhizoma and glycyrrhizin in Glycyrrhizae Radix when they were boiled together in aqueous solution and the supernatant solution thus obtained did not show any antibacterial activity which was derived from berberine. The content of berberine in BG and CGP by HPLC analysis were 41.1%, 8.3% respectively. BG was occured mostly at pH 5.0. The solubility of berberine was 0.15%, while that of BG and CGP was 0.07%, 0.12%, respectively. CGP shown more increased antibacterial activity to gram positive bacteria, S. dysenteriae and K. pneumoniae than berberine. The absorption rates of CGP in stomach, duodenum and jejunum of rats were compared with those of Coptidis Rhizoma water extracts (CR), which were increased more than CR. The time required for the maximum serum concentration of berberine from CGP in mice was 90 minutes after oral administration. The maximum serum concentration of berberine from CGP was higher than that from CR. The dissolution of CGP was increased more than berberine and BG in both artificial gastric and intestinal fluids. The dissolution of CGP pill made from gelatin was 63.4% in artificial gastric fluids and that made from CMC was 76.0% in artificial intestinal fluids.

A Study for the isolation of the Berberine-type Alkaloid from Coptidis Rhizoma and for their Antitumor Activities

  • The purpose of this study is the separation of biologically active ingredients from Coptidis Rhizoma which has been widely used as one of oriental herbal medicine for body fever. In this study, berberine-type alkaloids were tested on their biological activities in the aspect of antibacterial, antitumor, anti-herpetic and anti-HIV activity. Contents of five major alkaloids for the various origin of Coptidis Rhizoma were assayed by HPLC. As the results, the content of berberine from Coptis chinensis and Coptis japonica were 6.78% and 7.09%, respectively. The contents of coptisine, jatrorrhizine, berberastine from Coptis chinensis were higher than those of Coptis japonica. The amount of palmatine from both species were almost the same. Surprisingly for antitumor experiment, all compounds have been shown remarkable activity, especially against SNU-l(human stomach cancer) cell line. Among the compounds purified through column chromatography, palmatine, coptisine, and jatrorrhizine inhibited the growth of K-562(human chronic myelogenous leukemia) cell line whereas jatrorrhizine has been shown the effective inhibition of A-549 (human lung) cell line at the same time.

야생식용버섯인 까치버섯(Polyozellus multiplex)의 암예방 효과 (Chemoprevention Effect of Polyozellus multiplex, a Wild and Edible Mushroom)

  • Methanol extract and its fractions of Polyozellus multiplex inhibited the growth of several tumor cell lines and its water fraction showed a higher cytotoxicity effect on the human gastric carcinoma cell, SNU668 than on the other cell lines. The glutathione S-transferase (GSH) content was decreased by MNNG treatment but increased by adding Polyozellus multiplex water fractions. Also the activities of GSH and superoxide dismutase were increased by more the treatment of Polyozellus multiplex water fractions than by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) alone. Polyzellus multiplex water fraction cased significant reduction in the proliferating cell unclear antigen (PCNA) labelling index in the glandular stomach epithelium as compared with the value of MNNG alone.
