Saxifraga cortusifolia (Saxifragaceae): A new record for the Korean flora from Jejudo Island

  • Saxifraga cortusifolia Siebold & Zucc. (Saxifragaceae, sect. Irregulares, ser. Rufescentes), a species considered endemic along mountain streams throughout Honshu, Shikoku, and Kyushu in Japan, was discovered as a wild population in the Cheona Valley in Haean-dong, Jeju-si on Jeju-do Island of Korea. The population consisted of three subpopulations occupying narrow areas 10 × 10 m2, 5 × 5 m2, and 10 × 5 m2 in size. The number of individuals in the three subpopulations was confirmed to exceed 100. Most of the individuals were on or below rocks at the edge of the valley. A morphological description, detailed photos, and a key to related taxa in Korea are presented.

The Effect of Ecklonia stolonifera Extracts on Blood Flow and Serum Lipid Level in Ovariectomized Rats (곰피 추출물이 난소를 절제한 흰쥐의 혈액 유동성 및 혈중 지질함량에 미치는 영향)

  • Estrogen deficiency in peri- and postmenopausal women results in variety of neurovegetative, psychic and somatic symptoms, and may contribute to severe diseases within the aged female population. Ecklonia stolonifera (ES) is an edible brown algae traditionally used in fishery towns in Far East Asia. This study was performed to investigate the effects of ES extracts on blood flow and serum lipid concentration in ovariectomized rats. Weight-matched female Sprague-Dawley strain rats were assigned to four groups. Three groups were surgically ovariectomized (OVX). The fourth group was sham operated. Rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-CON), ovariectomized rats supplemented with ES extract at 50 mg/kg bw/day (OVX-ES50) and ovariectomized rats supplemented with ES extract at 200 mg/kg bw/day (OVX-ES200). Serum total cholesterol and triglyceride contents decreased in the sham group compared to the OVX-CON group. Six weeks feeding of ES extract resulted in a significant (p<0.05) lowering of serum triglyceride and a lowering tendency of total cholesterol level. The level of Hdl-cholesterol in serum increased by supplementation of ES extracts at 50 mg/kg and 200 mg/kg bw/day (p<0.05). Blood passage time of the ES extracts-supplemented group was higher than the OVX-CON group. The ability of platelet aggregation of groups treated with ES extracts was less than that of the OVX-CON group. These results suggest that the beneficial effects of ES extract may be used to possibly improve on the metabolic syndrome of menopausal women.

Effect of Ecklonia stolonifera Extracts on Bone Turnover Markers in Ovariectomized Rats (곰피가 갱년기 장애 유도 흰쥐의 골 대사 지표물질의 변화에 미치는 영향)

  • Menopause is often associated with the incidence of several chronic diseases including osteoporosis, cardiovascular disease, and obesity. The purpose of this study was designed to evaluate the possibility of osteoporosis prevention in postmenopausal women. In this study, we investigated the effects of Ecklonia stolonifera (ES) extracts on bone turnover markers in ovariectomized rats. For this study, the following four groups of 9-week-old Sprague-Dawley rats were evaluated over 6 weeks: normal rats (SHAM), ovariectomized rats (OVX-CON) and ovariectomized rats that were treated with ES extracts. We measured the osteocalcin and C-telopeptide of collagen cross-links (CTx) content, enzyme ALP activity in serum and collagen content in the cartilage, bone, skin and lungs. We found that the levels of indicators of bone metabolism such as alkaline phosphatase (ALP), osteocalcin and CTx were lower in rats in the ES extract group than the OVX-CON group. In addition, the collagen contents in the bone, cartilage, skin and lungs decreased in response to ovariectomy, but the levels of collagen were greater in the bone of rats that were treated with ES extract than in the bone of rats in the OVX-CON group. These results suggest that the ES may be an effective functional food to prevent osteoporosis in postmenopausal women.

Development of Domestic Cultivation Kit using Water Celery (Oenanthe stolonifera DC.) Seed (미나리 종자를 이용한 가정용 재배 키트 개발)

  • This present study was conducted to develop the domestic cultivation kit using water celery(Oenanthe Stolonifera DC.) seed. As the result of germination rates in 3 type inbred lines, the IT 232354 had the highest initial germination rate and final germination rate, and was selected as the inbred line to be used in the cultivation kit. The development of the domestic cultivation kit was carried out using the IT 232354 seeds. It was possible to cultivate up to 3 times harvest using the same root in this cultivation kit, though the growth decreased rapidly in the $4^{th}$ cultivation. As a result of investigating the effects of the type of nutrient solution on growth of water celery, the overall growth was the lowest in the nutrient solution for Oenanthe Stolonifera DC.(N.S.D.). The shoot growth was similar to that of nutrient solution for leaves and stem vegetables (N.S.L.S.) and amino acid fermentation by-product liquid (A.F.B.L.), however in the root growth, the N.S.L.S. was more effective than A.F.B.L. When it was harvested 4 times consecutively after 1 time of planting, the last survival ratio of A.F.B.L. was 100% while their ratios were 96.4% and 49.8% each for N.S.L.S. and N.S.D. For the growth by cultivation kit type, the hole type cultivation kit increased slightly compared to the 3 hole type cultivation kit in the $1^{st}$ and $2^{nd}$ harvest, but there was no difference in the $3^{rd}$ and $4^{th}$ harvest. Total yield of one cultivation kits showed the 3 hole type cultivation kit is much higher than the 2 hole type cultivation kit. According to the results of this experiment, it is possible to harvest three times by planting one times if it was cultivated using N.S.L.S. and A.F.B.L. in the 3 hole type cultivation kit.

Uptake of Heavy Metal Ions by Water Dropwort (Oenanthe stolonifera DC.) and Identification of Its Heavy Metal-Binding Protein (미나리의 중금속 흡수량 측정 및 중금속 결합단백질의 동정)

  • Uptake of hen metal ions by water dropwort (Oenanthe stolonifera DC.) and its cadmium-binding protein were studied to probe for good method to remove heavy metal contaminants from environments. The plant was cultured in the culture medium (pH 7.0) containing the various concentrations of $Cd^{2+}$, $Cr^{3+}$ or $Pb^{2+}$, for 3 and 7 days. The residual heavy metals deposited in roots linearly increased as the metal ions concentration increased up to 17 ppm for $Cd^{2+}$, 20 ppm for $Cr^{3+}$ and 50 ppm for $Pb^{2+}$. Above these concentrations, the plant growth was inhibited and the uptake rates of the metal ions decreased. The heavy metals absorbed by the plant were mostly deposited in roots. In particular, the residual concentration of lead in roots was about four times higher than those of cadmium and chromium. When cultured in the medium containing 20 ppm of each metal ion, 80% of cadmium, 90% of cromium and 96% of lead were deposited in roots out of the total residual metal ions in the plant. These values correspond to 6.1 mg of cadmium, 5.2 mg of chromium and 23.6 mg of lead per one gram of roots tissue on a dry weight basis. A cadmium-binding protein was partially purified by extraction, gel filtration and DEAE-Cellulose chromatography from water dropworts that was grown in the medium containing 20 ppm $Cd^{2+}$. The purified protein was a single band on SDS- and non-denaturing- polyacrylamide gel electrophoresis. Its molecular mass was estimated to be ca. 5,000 dalton by gel filteration. Analysis of amino acid composition of the protein indicated that it had a typical amino acid composition of heavy metal-binding protein in that it contained 27% of acidic amino acids and 9.9% of cysteine. However, it is likely that the protein is a new plant metal-binding protein, since its amino acid composition is somewhat different from those of phytochelatins that have been known so far.

Verification of the Fractions with Strong Estrogenic Activities from Brown Algae (갈조류로부터 에스트로겐 고활성 분획의 검증)

  • In order to evaluate estrogenic compounds in brown algae, an in vitro test system for the verification of estrogenic activity was applied. Fractions from ethanol extracts of each brown alga were prepared by a systematic fractionation procedure with solvents such as $H_2O$, hexane, butanol and methanol. Aqueous fractions of brown algae showed the highest estrogenic activities. Estrogenic activities of $500\;{\mu}g/ml$ aqueous fractions of Undaria pinnatifida and Laminaria japonica showed almost the same strength as that of $10^{-7}\;M$ standard solution ($17{\beta}$-estradiol). Furthermore, estrogenic activities of $500\;{\mu}g/ml$ aqueous fractions of Ecklonia stolonifera and Porphyra suborbiculate represented higher activities than that of $10^{-8}\;M$ $17{\beta}$-estradiol. These observations suggest that aqueous fractions of all these brown algae are expected to possess estrogenic compounds and could be developed as estrogenic agents for postmenopausal disorder.

Growth and Wear Tolerance of Creeping Bentgrass as Influenced by Silica and Potassium Fertilization (규산 및 칼리 시비에 따른 벤트그래스 생육 및 내답압성 반응)

  • The study was conducted to know whether turfgrass wear tolerance, growth, and quality could be improved by the application of silicon and potassium. First, turf responses to silicate and potassium were evaluated by several parameters such as, turf visual quality, root length, shoot density, and dry weight under the field condition. Second, turf responses to traffic frequencies were examined by turf growth (root) length, shoot density and dry weight) and soil hardness under the field condition. Finally, under traffic stress condition, the effects of silica and potassium application on wear tolerance were evaluated through the methods described above. Creeping bentgrass (Agrostis stolonifera) rooting were significantly improved by silica. The root length was enhanced by an increase in potassium silicate application. Certain level of light traffic is beneficial while frequent traffic cause serious adverse effect on visual quality of bentgrass. Under a traffic stress condition with 10 times of footing a day for 30 days, silica and potassium increased turf visual quality by 6.38% and 10.25% respectively when compared to the control. Silica and potassium treatment on trafficked plot increased turf visual quality by 11.4% and 10.2% respectively in comparison with the control with significant reduction of wear injury from the traffic. A co-application of potassium silicate with potassium sulfate provided the enhanced visual quality of turf as compared to application of silica or potassium fertilizer, respectively.

Evaluation of optimal planting combination considering growth characteristics of major landscaping groundcover plants (조경용 주요 지피식물의 생장 특성을 고려한 식재조합 및 혼식 적합성 평가)

  • With the purpose of designing companion planting of groundcover plants for ornamental uses, this study identified the yearly growth characteristics of nine species of different life forms, analyzed the coverage characteristics of individual plants, and suggested combinations of plants suitable for each life form. Polygonatum odoratum var. pluriflorum, Liriope platyphylla and Hosta capitata, as short-grained plants that can grow to more than 20 cm, tended to grow for 60 days after planting in April and maintain their shape thereafter. Their aerial parts started to wither and enter dormancy after September. Saxifraga stolonifera, Dianthus chinensis and Sedum middendorffianum tended to continuously grow until September after planting in April and their growth declined after September. Lysimachia nummularia, as a creeping plant that grows creeping on the ground, started to show a rapid growth three months after planting. Sedum sarmentosum grew slowly until August and the aerial parts started to wither from September when the temperature decreases. The coverage characteristics of these nine species that grow differently after companion planting were surveyed and the growth of Sedum sarmentosum showed the highest number of companions. It was found that Hosta capitata can be companion planted with Sedum middendorffianum, Saxifraga stolonifera, and Lysimachia nummularia. These results indicate that among different shoot growth types species propagated with their stems creeping on the ground or those that can grow vegetatively with non-rhizome parts are more suitable for companion planting with others than those of which rhizomes branch.

First Report of Curvularia Leaf Blight Caused by Curvularia trifolii on Creeping Bentgrass in Korea (Curvularia trifolii에 의한 크리핑벤트그래스 잎마름병 발생)

  • Curvularia leaf blight of creeping bentgrass (Agrostis stolonifera) putting green by caused Curvularia trifolii was observed in Hapcheon, Korea. In July to September 2014, curvularia leaf blight developed on leaf blades of creeping bentgrass as small water-soaked lesions that subsequently turned into dark-colored, necrotic spots. The spots were expanded and became gray, grayish-brown, or light brown, circular to oblong lesions with purple to dark brown borders that often were surrounded by a yellow halo. The necrotic lesions coalesced, became irregular in shape and caused tip or complete blighting of the leaves. Blighted leaf blades appeared grayish-white to tan. The fungus was identified by morphological characters and 16S rDNA sequencing as C. trifolii. Conidia of the pathogen were short, with predominantly 3-septa, straight or often curved, with end cells frequently paler than intermediate cells. Size of the 3-septate conidia in culture are $26{\sim}28{\times}11{\sim}12{\mu}m$. Pathogenicity of the fungus was proved by artificial inoculation on the host. This is the first report of C. trifolii causing leaf blight on creeping bentgrass in Korea.

  • In this study, two species of algae, Ecklonia stolonifera, Sargassum thunbergii and one species of marine plant, Zostera marina(rhizoid and stem) were collected and extamined to determine the extractability of water soluble protein and the influences of various factors including extraction time, temperature, ratio of sample vs solvent and pH upon the extractability were tested. The effects of precipitation treatments for isolation of algal protein from the extracts(TCA treatment, methanol treatment and pH control) were also tested. Amino nitrogen and total nitrogen of purified samples made by obtained optimum conditions were estimated. The effect of the ratio of sample vs solvent on extractability differed from species to species which was enhances at 1:100(w/v) in Sargassum thunbergii and Zostera marina while 1:150(w/v) for Ecklonia stolonifera. The effect of extraction time and temperature was revealed differently in all species which might be considered to be caused by differences in the constitution of algal tissues. But in case of TCA insoluble nitrogen, it was showed the maximum extractability at $40-50^{\circ}C$ for 1 hour extraction. The optimum pH for the ext action of total nitrogen was 9-12 while the optimum pH was 6-7 for TCA insoluble nitrogen. And the pH control appeared to be most effective in the influence of precipitation treatment for isolation of algal protein.
