In this paper we describe the cloning and expression of the genes encoding the flavonoid-biosynthetic enzyme dihydroflavonol 4-reductase (DFR) in Matthiola incana R. Br. A heterologous cDNA probe from Zea mays was used to isolate full-size DFR cDNA clone from a corolla-specific cDNA library. Comparison of the coding region of this DFR cDNA sequence including the sequences of Zea mays, Anthirrinum majus, Petunia hybrida, Callistephus chinensis, Dianthus caryophyllus and Rosa hybrida reveals a identity higher than 61% at the nucleotide level. The DFR transcript is G/C rich in monocotyledonous plants show a strong codon bias preferring codons with a G or C in the third position. The function of this nucleotide sequences were verified by comparison with amino acid sequences of the amino-terminus and tryptic peptides from purified plant enzyme, by northern blotting with mRNA from wild type and mutant plants and by in vitro expression yielding an enzymatically active reductase. Genomic southern blot analysis showed the presence of one gene for DFR in Matthiola incana. Northern blot analysis of the DFR wild type and mutant lines showed that the lack of DFR activity in the stable acyanic mutant k17b is clearly by a transcriptional block of the DFR gene.
In vitro proliferation system was achieved by using nodal segment excised from greenhouse grown juvenile stock plants of Alnus japonica. Stem explants were cultured on MS medium supplemented with different plant growth regulators of cytokinin and/or their combinations. The most effective cytokinin source was the combination of zeatin 2.0 mg/L and TDZ 0.05 mg/L producing the average number of shoots (16.8 $\pm$ 3.6). In addition, healthy roots were formed after small clumps of shoots were transferred to half strength of MS medium containing IBA 0.02 mg/L with optimal rooting capacity. Soil acclimatization was successfully conducted in cell tray containing artificially mixed soil with 92 % survival rate.
To obtain the basic information of high yielding ability for varietal improvement and cultural practice, reciprocal grafts were made analyzed among five sweet potato cultivars. Five cultivars were characterized with the different in whose source and sink capacities. Grafting decreased main vine length, number of tuberous roots per plant, but its effects were different among grafting combinations. The growth of vines was affected more by scion, while growth and yield of under-ground were influenced more by stock. Grafting effects, interactions between stock and scion for main vine length and number of tuberous roots were highly significant in diallel grafting. The percentage of marketable tuberous roots was higher in the order of stem cutting>self grafting>reciprocal grafting.
New types of cytoplasmic male sterility in Brassica species would be very useful for the production of F$_1$, hybrid seeds. Leaves and stems of rapid cycling stock of B.juncea (CrGC4-3) containing Anand CMS were used as experimental materials for plant regeneration from protoplast culture. Very high plant regeneration rate (85%) was found in the Kao & Michayluk medium supplemented with 2 mg/L zeatin, 0.5 mg/L BAP, and 1 mg/L NAA when only leaf, not stem, segments were cultured. Protoplasts were isolated from leaves using mixtures of enzymes (1% Cellulycin, 0.5% Macerozyme) in 0.4 M mannitol and 50 mM $CaCl_2$.$2H_2$O. Mcrocalli induced from protoplasts were transferred to the shoot regeneration medium containing 2 mg/L BAP, 2 mg/L zeatin, and 0.5 mg/L NAA. After 60 days of initial protoplast culture, regenerated plantlets were obtained, acclimatized, transplanted into the pots, and grown up to the flowering stage.
In Ethiopia, dry land vegetation including the fairly intact lowland and western escarpment woodlands occupy the largest vegetation resource of the country. These forests play a central role in environmental regulation and socio-economic assets, yet they received less scientific attention than the moist forests. This study evaluated the woody plant species composition, population structure and carbon sequestration potential of the A. senegal woodland across three distance gradients from the settlements. A total of 45 sample quadrants were laid along a systematically established nine parallel transect lines to collect vegetation and soil data across distance gradients from settlement. Mature tree dry biomass with DBH>2.5 cm was estimated using allometric equations. A total of 41 woody plant species that belong to 20 families were recorded and A. senegal was the dominant species with 56.4 IVI value. Woody plant species diversity, density and richness were significantly higher in the distant plots compared to the nearest plots to settlement (p<0.05). The cumulative DBH class distribution of all individuals had showed an interrupted inverted J-shape population pattern. There were 19 species without seedlings, 15 species without saplings and 14 species without both seedlings and saplings. A significant above ground carbon (5.3 to 12.7 ton ha-1), root carbon (1.6 to 3.6 ton ha-1), soil organic carbon (35.6 to 44.5 ton ha-1), total carbon stock (42.5 to 60.7 ton ha-1) and total carbon dioxide equivalent (157.7 to 222.8 ton ha-1) was observed consistently with an increasing of distance from settlement (p<0.05). Distance from settlement had significant and positive correlation with species diversity and carbon stock at 0.64⁎⁎ and 0.78⁎⁎. Disturbance intensity may directly influence the variation of species composition, richness and density along the A. senegal woodland. The sustainability of the A. senegal woodland needs urgent protection, conservation and restoration.
Grapevines will experience various types of winter damage. Some winter damages are caused by mechanical injury, freezing temperatures or poor vine vigor. This research was conducted to find out the appropriate control methods through selection of resistant rootstocks and improvement of overwintering methods for the control of crown gall disease on 'Kyoho' grape. The crown gall symptoms were not found when three stock plants of grapevine SO4, 5BB and 3306 were inoculated with $10^4cfu/ml$ of Agrobacterium vitis strains (YK2823, YK3312, LMG259, HKA234). But when they were inoculated with higher concentration $(10^6 cfu/ml)$ of A. vitis, irrespective of stocks plants, crown galls were formed all of them and the gall size was much smaller than that of kyoho. Three stock plants were selected as resistant based on above mentioned. Covering trunks and branches with rice straw and insulating coverlet was the most effective method for prevention of crown gall disease. This treatment minimized the ambient temperature changes on grapevine trees during winter season to $9.6^{\circ}C$ and the normal plant growth was due to the absence of freezing injury.

In this paper, we applied the Utility Interactive Photovoltaic-Wind Hybrid Generation System for stock raising waste disposal system. For the power consumption reduction of proposed system, we used the electromotive control system. So as to verify the propriety of proposed system, we made a Pilot Plant and examined the system.

The recycling rate of recovered paper in Korea is the highest in the world, 92%, but remanufacturing yield is low due to the extremely poor quality of the paper. The poor quality, in turn, influences to the reject amount in deinking process. To increase the yield of old newspaper recycling process, hydrophobic degree of inorganic pigments of deinking stock must be reduced. To determine the hydrophobicity, Pitch Potential Deposit Tester (PDT) was newly designed and applied with respect to the SB latex property of various quality used in Korea; its hydrophobic degree according to Tg, gel content, charge and particle size of latex and optimum designing condition of SB latex. And below are the conclusions: 1. The reason of excessive reject from old newspaper deinking process for total amount of printed ink is loss of inorganic pigments. When lipase, a biochemical catalyst, was applied with the purpose of preventing inorganic pigments loss about more than 70% of total reject weight and promoting hydration of pulp for deinking, deinking process yield of pre flotation secondary stage increased remarkably without any changes of deinking efficiency. 2. Lipase improved deinking stock by cutting ester linkage on surface of hydrophobic materials to promote its hydration. From this, it reached the conclusion that hydration degree of stock exercises significant effect on flotation deinking process yield. 3. Inorganic alkali promotes hydration of deinking stock. But there have been needs for more fundamental measures other than inorganic alkali of promoting hydration for yield improvement. For this, this study intended to find out reasons of chemical properties change on surface of hydrophobic material by change of pH. 4. Pitch Deposit Test (PDT) was performed for understanding principle of why surface of coating flake from OMG is hydrophobic and why it becomes hydrophilic when pH of stock is alkaline. As a result of this test, it is determined that swelling property by change of pH of latex film, which were used as coating adhesive is the reason for hydrophobic change. 5. Hydrophilicity of coating flake increased with hydrophilic pigments. And as more of SB Latex adhesive was used and higher of calcium hardness of stock became, its hydrophilicity decreased. SB Latex adhesive film is reformed by mechanical friction. For having hydrophilicity under neutral pH, strong bruising action such as kneading is required. 6. Because swelling of adhesive film decreases as Tg of SB latex gets lower and mean diameter gets smaller, it shows hydrophobicity under neutral pH. This lowers hydrophilicity of coating flake, which leads to easy elimination with flotation reject on DIP process. Therefore, for improving future flotation yield, it is necessary to develop to use eco-friendly clean SB latex by raising Tg and increasing mean diameter for recycling, and as a result, to reduce excessive loss of coating flake as a reject from deinking process.

Paratuberculosis (Johne's disease), a chronic wasting disease caused by Mycobacterium avium subspecies paratuberculosis (MAP), is a major cause of economic loss in the cattle industry. In Korea, national monitoring of breeding stock for MAP has been implemented. In this study, we report the results of serological testing to determine the prevalence of MAP in breeding stock of Korean native and dairy cattle during 2008 and 2009. A total of 3,927 serum samples were submitted (3,692 Korean native cattle and 235 dairy cattle) to Animal Disease Diagnostic Division, Animal, Plant and Fisheries Quarantine and Inspection Agency. The samples were classified into four different age groups for MAP; group 1 (${\leq}2$ year, n = 1,509), group 2 (>2 years to ${\leq}3$ years, n = 486), group 3 (>3 years to ${\leq}4$ years, n = 441), and group 4 (>4 years, n = 1,491). Overall seroprevalence of MAP in this study was 0.5% (21/3,927), which was much lower than that of conventional cattle (1.2-16.4%) in Korea. Also, the seroprevalence was determined by age groups: three of group 1 (0.2%), two of group 2 (0.4%), three of group 3 (0.7%), and 13 of group 4 (0.9%) were seropositive for MAP, respectively. Although seropositive samples were found in all age groups, the seroprevalence tended to increase with age. Our study showed that the seroprevalence of MAP in pure-bred breeding dairy cattle (0%) was lower than that in pure-bred breeding Korean native cattle (0.6%).
