ANALYSIS OF MIXING EFFICIENCY OF A TUBULAR HEAT-EXCHANGER REACTOR USING CFD (CFD를 이용한 관상 열교환기형 반응기의 mixing 효율 분석)

  • We have investigated the mixing behavior of a tubular heat exchanger reactor using CFD and compared its mixing performance with different type of reactors such as jet mixer and continuous stirred tank reactor (CSTR). The mixedness in each reactor was quantified introducing a statistical average value, the coefficient of variation (CoV), which is a normalized standard deviation of concentration of a component over the whole fluid domain. Through the analysis of the flow pattern and turbulent energy distribution, we suggested a simple but effective way to improve the mixing performance of the tubular heat-exchanger reactor, which include the addition of the internals and/or the increase of the recycle flow rate. It was found that the CoV value of the tubular reactor could be nearly equivalent to that of CSTR by applying those two alternatives suggested here.

  • Thermal decomposition characteristics of ethyl methacrylate (EMA) and Styrene (St.) copolymer was investigated with synthesis at 8$0^{\circ}C$ in a continuous stirred tank reacto (CSTR) using toluene and benzoyl peroxide(BPO) as solvent and initiator, respectively. The thermal decomposition was considered to be side scission at below 30$0^{\circ}C$ and estimated 2nd-order reaction kinetics of EMA/St. copolymer. The activation energies of decomposition on this copolymers were in the ranges of 38-43 kcal/mol for EMA/St. and a good additivity rule was observed in each composition. The thermogravimetric trace curves agreed well with the theoretical calculation.

  • The goal of this paper is to develop a nonlinear observer-based control strategy for a multi-variables continuous stirred tank reactor (CSTR). A new robust nonlinear observer is constructed to estimate the whole process state variables. The observer is coupled with a nonlinear controller, designed based on the input-output linearization for controlling the concentration and reactor temperature. The closed loop system is shown to be globally asymptotically stable based on Lyapunov arguments. Finally, computer simulations are developed for showing the performance of the proposed controller.

Perfusion Cultivation of Transgenic Nicotiana tabacum Suspensions in Bioreactor for Recombinant Protein Production

  • A perfusion culture of transgenic Nicotiana tabacum cell suspensions, transformed to express recombinant glucuronidase (GUS), was successfully performed in a 5-1 stirred tank bioreactor. With 0.1 $day^{-1}$ of perfusion rate, the maximum dry cell weight (DCW) reached to 29.5 g/l in 16 days, which was 2.1-fold higher than the obtained in batch culture (14.3 g/l). In terms of the production of GUS, the volumetric activity could be increased up to 12.8 U/ml by using perfusion, compared with 4.9 U/ml in batch culture. The specific GUS activities in both perfusion and batch cultures were maintained at similar levels, 200-400 U/g DCW. Consequently, a perfusion culture could be a good strategy for the enhanced production of recombinant proteins in a plant cell culture system.

  • Thermal decomposition of the copolymer of butyl methacylate(BMA) with styrene(St) was investigated. The copolymer Was obtained at 80 $^{\circ}C$ in a continuous stirred tank reactor(CSTR) using toluene and benzoyl peroxide(BPO), as solvent and initiator, respectively. The reactor volume was 0.3 liters and residence time was 3 hours. The thermal decomposition followed the second order kinetics for BMA/St copolymer. The activation energies of thermal decompositon were in the ranges of 38 ${\sim}43$ kcal/mol for BMA with St copolymer and a good additivity rule was observed with the composition of copolymer. The thermogravimetric trace curve agreed well with the theoretical calculation.

  • 화학 공정에서의 이상 진단 시스템 개발 및 응용에 대한 연구는 지난 5년간 많은 발전이 이루어졌다. 화학 공정의 본질적인 특성으로 대형 시스템, 비선형 특성, 모델링 자체의 어려움, 공정 변수의 large dead time 및 복잡한 인과 관계등을 들수 있으며, 이러한 어려움에도 불구하고 적절한 이상 진단 시스템의 중요성을 인식하여, 초기에는 주로 rule-based approach가 도입되어 현장에서의 조업에 많은 도움을 주었었다. 그러나 개발 기간의 단축화, 개발 과정의 표준화 뿐 아니라 개발된 시스템 자체의 일관성 등을 위하여 체계적인 접근 방법이 필요하게 되었으며, 그중 지식 베이스 합성 문제는 그 동안 활발하게 연구되어 오고 있는 분야이다. 이에 본 연구에서는 기호화된 정성적인 정보를 얻기위한 기존의 실험 방법의 한계를 극복하고자 동적 모사를 이용하여 정량적인 정보로부터 정성적인 정보를 생성시키는 방법론에 대해 연구하였다. CSTR(Continuous Stirred Tank Reactor)에서 나타날수 있는 이상의 종류에 대한 동적 모사를 수행하여 이상 진단 시스템을 위한 지식이 생성되는 과정을 보였다.

Treatment of Wastewater Containing Phenol Using Pseudomonassp. B3 (Pseudomonas sp. B3를 이용한 Phenol 함유 폐수의 처리)

  • Using Pseudomonas sp. B3, identified and isolated from nature, wastewater containing phenol was treated in a continuous stirred tank reactor and its reaction characteristics were studied. Average concentrations of phenol and COD in effluents were 1.5mg/L and 124mg/L at 0.059h-1 dilution rate, respectively. At the dilution rate higher than 0.063h-1, phenol and COD increased abruptly to 19mg/L and 318mg/L. At the dilution rate higher than 0.059h-1, biomass concentration suddenly decreased and was "washed out". Biomass concentration was 150mg/L at a dilution rate of 0.067h-1. Maximum biomass production rate was 15.98mg/L$.$h at a dilution rate of 0.067h-1. When dilution rate increased above 0.059h-1, effluent phenol concentration abruptly increased and biomass production rate decreased. Maximum cell growth rate(${\mu}$max) and Michaelis-Mentens kinetic constant(Ks) were 0.074h-1 and 0.424mg/L, respectively. From the above result low phenol concentration can be expected at a maximum dilution rate, but reactor becomes unstable due to phenol inhibition.

  • Glass particle distribution in a stirred solid/liquid systems was investigated using computational fluid dynamics(CFD). The numerical results were compared to experimental data from the available literature which investigated the local dispersed phase volume fraction by means of an endoscope technique. Eulerian multi-phase model and applications considered high loading of solid particle was used to investigate the influence of the particle concentration and mixing tank size on the solid distribution. A good agreement was obtained between the experimental data and simulation results. The results showed different solid particle distribution in an agitator by particle concentration and mixer size.

Production of Itaconic Acid at Various Bioreactors (다양한 생물반응기에서 이타콘산의 생산)

  • A suitable culture method and bioreactor type for itaconic acid production were chosen by comparing the maximal concentration of itaconic acid produced in various systems. In batch culture, the maximal concentration of itaconic acid produced in a bubble column reactor was about 5% greater than that produced in stirred-tank or external-loop airlift reactor. These results were thought to be due to lower shear force and higher mass transfer efficiency in a bubble column reactor in comparison with other reactors. Moreover, the fed-batch mode in a bubble column was found to be a suitable one, producing about 25% higher concentration of itaconic acid compared to batch mode.

Improved guggulsterone production from sugars, precursors, and morphactin in cell cultures of Commiphora wightii grown in shake flasks and a bioreactor

  • Cell cultures of Commiphora wightii (Arnott.) Bhandari were grown in shake flasks and a bioreactor and an increase in guggulsterone accumulation up to $18{\mu}g\;l^{-1}$ was recorded in cells grown in the production medium containing a combination of sucrose:glucose (4% total), precursors (phenylalanine, pyruvic acid, xylose, and sodium acetate), morphactin, and 2iP. A yield of $10g\;l^{-1}$ biomass and ${\sim}200{\mu}g\;l^{-1}$ guggulsterone was recorded in a 3-l flask and in a 2-l stirred tank bioreactor compared with 6.6 g biomass and $67{\mu}g\;l^{-1}$ guggulsterone in 250-ml flasks. Increased vessel size was correlated with increased biomass and guggulsterone accumulation. 2iP alone was not effective for biomass and guggulsterone accumulation in cell cultures of C. wightii.
