부하유무에 따른 수영운동이 중추신경계 손상 흰쥐의 혈청 BDNF 농도 및 행동변화에 미치는 영향 (The Effect of the Swimming Exercise by Load on Concentration of BDNF in Serum and Behavioral Change of CNS Injury in the Rats)

  • 본 연구의 목적은 중추신경계 손상의 쥐에 운동강도에 따른 신경계 회복을 알아보기 위해 부하수영과 무부하수영이 혈청 BDNF 농도와 행동학적 변화에 어떠한 영향을 미치는가를 확인하고자 하였다. 실험동물로는 Sprague-Dawley 수컷 흰쥐를 사용하였으며, Ketamine을 복강 내에 주사하여 마취한 뒤 제 1-2요추의 척수에 6-OHDA를 $100{\mu}{\ell}$ 주입하여 척수손상을 유발시켰다. 척수 손상으로 유발된 행동학적 변화를 알아보기 위해 BBB와 경사판 검사를 기록하였다. 혈청 BDNF는 수술 후 14일에 혈액을 채취해 흡광도를 측정하였다. 척수 손상 후 무부하군에서는 행동변화가 대조군에 비해 유의하게 증가하였고, 혈청 BDNF 농도는 다른 군에 비해 무부하군에서 증가하였다. 이러한 결과로 보아 저강도 수영운동을 중추신경 손상 후 초기에 적용하면 신경조직의 회복에 영향을 주리라 생각된다.

수양경(手陽經) 경혈(經穴) 자침(刺鍼)이 백서(白鼠)의 족과 염좌(捻挫) 통증(痛症)에 미치는 영향(影響) (Effect of Acupuncture applied to Hand Yang Meridian on the Rat Model of Ankle Sprain Pain)

  • Objectives : In the present study, the effect of electroacupuncture (EA) applied to hand yang meridian on the ankle sprain model was examined. Methods & Results : A common source of persistent pain in humans is the lateral ankle sprain. To model this condition, the rat's right ankle was bent repeatedly, overextending lateral ligaments, for 4 min under halothane anesthesia. The rat subsequently showed swelling of the ankle and a reduced stepping force of the affected limb for the next several days. The reduced stepping force of the limb was presumably due to a painful ankle. EA was applied to the several acupuncture point on the contralateral forelimb for 30 min under gaseous anesthesia. After the termination of EA, behavioral tests measuring stepping force were periodically conducted during the next 4 h. EA applied to SI-6 point produced a significant improvement of stepping force of the sprained foot lasting for at least 2 h. However, neigher LI-4 point nor TE-3 point produced any significant increase of weight bearing force. The improvement of stepping pressure was interpreted as an analgesic effect. The analgesic effect was specific to the acupuncture point since the analgesic effect on the ankle sprain pain model could not be mimicked by EA applied to a nearby point, LI-4 or TE-3. The analgesic effect of EA applied to SI-6 was more powerful when EA was applied by low-frequency and high-intensity stimulation. In addition, this effect need to be stimulated more than 15 min. Conclusions : These data suggest that EA produces a potent analgesic effect on the ankle sprain pain model in the rat. This analgesic effect is produced by applying EA to a Tae-Yang meridian at opposite side from the painful area in a stimulus point-specific way.

국소 허혈성 뇌손상 흰쥐 모델에서 경두개직류전기자극이 앞다리 운동감각 기능 증진에 미치는 효과 (Effect of Improved Forelimb Sensorimotor Function on the Transcranial Direct Current Stimulation in a Focal Ischemic Brain Injury Rat Model)

  • 본 연구는 국소 허혈성 뇌손상 흰쥐 모델에서 tDCS의 자극 적용시간을 달리하였을 때, 앞다리 운동감각 기능변화와 신경영양인자(GAP-43)발현에 미치는 영향을 알아보고자 하였다. 뇌손상 모델은 Sprague -Dawley계 흰쥐 80마리를 'Longa'방법을 이용하여 중대뇌동맥(middle cerebral artery)을 폐색하여 유발하였고, 실험군을 4개로 나누었다; 실험군I은 허혈성 뇌손상 유발군(n=20), 실험군II는 허혈성 뇌손상 유발 후 tDCS(10분) 적용군(n=20), 실험군III은 허혈성 뇌손상 유발 후 tDCS(20분) 적용군(n=20), 실험군IV는 허혈성 뇌손상 유발 후 tDCS(30분) 적용군(n=20)으로 나누었다. 앞다리운동감각 기능검사를 위해 수정된 앞다리배치 검사와 단일 팰릿 닿기 검사를 실시하였으며, 신경가소성에 대한 면역조직화학적 검사로 운동감각 영역에서의 GAP-43 단백질 발현을 관찰하였다. 앞다리운동감각 검사는 14일에서 실험군III (p<0.05)이 다른 군들에 비해 유의한 차이를 보였으며, 단일 팰릿 닿기 검사는 14일에서 실험군III(p<0.01)과 실험군IV(p<0.05)에서 유의한 차이를 보였다. 또한, 면역조직학적 검사는 14일에 실험군III이 다른 군들에 비해 현저한 면역양성반응의 증가를 보였다. 따라서, 0.1 mA의 강도로 20분간 적용했을 때가 앞다리운동감각 기능과 신경가역성 인자 GAP-43 발현에 가장 좋은 조건임을 알 수 있었다.

촉각과 온각 자극에 따른 악력, 상지 기민성, 그리고 손 기능의 차이 (The Difference of Grip Strength, Upper Limb Dexterity, and Hand Function according to Light Touch and Thermal Sensory Stimulus)

  • 본 연구의 목적은 촉각과 온각 자극에 따른 악력, 상지 기민성, 그리고 손 기능 변화의 차이 를 비교 분석하여 움직임에 가장 영향력 있는 자극의 종류를 규명하는 것이었다. 건강한 성인 남성 40명을 대상으로 하였고 신체적 특성을 파악한 뒤 악력과 상지 기민성 및 손 기능을 측정하였다. 이 후 부드러운 솔을 이용하여 촉각 입력을 발현한 뒤 재측정 하였으며 다음날 핫팩을 이용하여 온각 자극 발현 뒤 재측정하였다. 연구 결과 악력과 상지 기민성 검사의 모든 변인에서 통계적으로 유의한 차이가 나타났고(p<.001), Lifting small objects를 제외한 모든 손 기능 변인에서 통계적으로 유의한 차이가 나타났다(p<.01). 결론적으로 자극의 입력은 악력, 기민성, 그리고 손 기능을 증가시킨다. 또한 자극의 종류에 따른 영향에서 온각 자극 입력은 촉각 자극 입력보다 악력, 기민성, 그리고 손 기능을 더욱 증가시킨다. 따라서 재활치료 및 산업 현장에서 상지와 손의 기능 촉진을 위해서는 적절한 자극의 적용이 도움이 될 것이며 앞으로 자극의 빈도와 강도에 대한 연구가 더욱 필요할 것으로 생각된다.

동맥경화 예방과 치료를 위한 연구시도: Nitric Oxide의 역활 -광 유도 nitric oxide(PIANO)의 혈관이완에 따른 cyclic GMP의 증가 (Possible Role of Nitric Oxide in Prevention of Atherosclerosis: Photo-induced adequate nitric oxide (PIANO)-mediated relaxation involves cyclic GMP increment)

  • 본 연구의 목적은 광 유도에 의한 nitric oxide (PIANO)유리가 혈관이완에 대해 cyclic GMP (cGMP)가 관여하는 지의 여부와 아울러 ${\alpha}$-수용체를 통한 수축에 PIANO가 어떻게 작용하는 지를 파악하고자 하였다. In vitro 실험에서 흰쥐의 대동맥을 준 최고농도의 phenylephrine (PE)으로 수축시킨 후 nitric oxide 생성을 변화시키는 약물이나 광민감성 (photosensitizing) 약물에 대한 반응을 등장력 변화로 기록하였다. PIANO에 의한 혈관이완은 광노출 강도와 기간 및 광민감성 약물농도에 비례하여 증가하였고 cGMP의 증가를 수반하였다. PE에 의해 증대되는 phosphatidylinositide(PI) 전환은 PIANO에 의해 억제되었다. 이상의 결과는 cGMP의 증가로 인해 PIANO에 의한 혈관이완이 일어나며 ${\alpha}$-아드레날성 수용체 자극에 의한 PI 전환의 억제현상은 cGMP 증가의 결과로 생각할 수 있다. 결론적으로 PIANO에 의한 혈관이완은 cGMP의 증가로 인함을 확인할 수 있었다.

특발성 파킨슨병 환자의 전침치료 효과 (Effect of Electroacupuncture on Patients with Idiopathic Parkinson's Disease)

  • Objective : This study was designed to evaluate the effect of electroacupuncture with various scales on symptoms of idiopathic Parkinson's disease. Methods : Subjects were voluntarily recruited by newspapers and internet. All the subjects are confirmed as idiopathic parkinson's disease by a neurologist. The acupuncture therapy was performed twice a week for 8 weeks. Acupuncture points for EA group were GB34, and LR3, electrical stimulation frequency was 120Hz, duration 15 minutes, and intensity is up to pain threshold according to patients. The patient's symptoms were assessed before, after 4 weeks and after 8 weeks treatments by unified Parkinson's disease rating scale(UPDRS), modified Hoehn-Yahr(H-Y) stage, Schwab & England activity of daily living and freezing of gait questionnaire(FOGQ). Results : UPDRS I, II, III, IV and total UPDRS scores were significantly improved after 4 weeks(p<0.05) and after 8 weeks(p<0.05) compared to the pre-treatment. The scores of ADL were significantly improved after 4 weeks(p<0.05) and after 8 weeks(p<0.05). There were significant changes in H-Y stage after 4 weeks(p<0.05), but There were no significant changes in H-Y stage after 8 weeks. There were significant changes in FOGQ scale after 4 weeks(p<0.05) and after 8 weeks(p<0.05). Conclusion : This study suggests that electroacupuncutre treatments can be applicable to improve symptoms in the patients with idiopathic Parkinson's disease.

족삼리(足三里) 전침(電鍼)이 백서(白鼠)의 슬관절염(膝關節炎) 통증(痛症)에 미치는 영향(影響) (Effects of Acupuncture applied to Food Samli on the Rat Model of Knee Arthritic Pain)

  • Objectives : The usage of acupuncture has gained popularity as an alternative method of treatment for certain chronic pain conditions. However, the efficacy of acupuncture in various diseases has not been fully established and the underlying mechanism is not clearly understood. In the present study, the effect of electroacupuncture (EA) applied to foot samli$(ST_{36})$ on the carrageenan-induced knee arthritic pain was examined. Methods : A common source of persistent pain in humans is the knee arthritis. Knee arthritis was induced by injection of 2 % carrageenan $50\;{\mu}l$ into the knee joint cavity. When rats developed pain behaviors, EA was applied for 30 min. under enflurane anesthesia with repeated train stimuli at the intensity of 10X of muscle twitch threshold. The weight bearing force of the hind limb was measured for an indicator of pain level after each manipulation. Results : The average weight borne by the hind limb during normal gait was 55% of total body weight, which was reduced to less than 10% after knee arthritis. EA improved the weight bearing of the arthritic hind limb significantly for the duration of 4 hr. EA applied to $ST_{36}$ point produced a significant improvement of stepping force of the arthritic foot lasting for at least 4 h. However, $GB_{31}$ point did not produce any significant increase of weight bearing force. The analgesic effect was specific to the acupuncture point since the analgesic effect on the knee arthritis model could not be mimicked by EA applied to a nearby point, $GB_{31}$. The relations between EA-induced analgesia and endogenous nitric oxide(NO) and inducible NO synthase(iNOS)/neuronal NOS was also examined. Results were turned out that both NO production and nNOS/iNOS protein expression which is increased by arthritis were suppressed by EA stimulation applied to $ST_{36}$ point. Conclusions : The data suggest 1) that EA produces a potent analgesic effect on the rat model of chronic knee arthritis pain in a point specific manner and 2) that EA-induced analgesia modulate endogenous NO through the suppression of nNOS/iNOS protein expression.

흰가루병에 대해 성체식물 저항성을 지닌 봄보리에서 $^{14}CO_2$ 동화와 $^{14}C-$동화산물의 대사 ([ $^{14}CO_2$ ] Assimilation and Metabolism of $^{14}C-$Assimilates in Whole Plants of Spring Barley In Relation to Adult-Plant Resistance to Powdery Mildew)

  • 흰가루병에 대하여 감수성인 봄보리품종 Peruvian과 성체식물 저항성품종 Asse의 4엽기에서 흰가루병에 감염되었을 때 $^{14}CO_2$동화와 $^{14}C-$동화산물의 대사변화를 연구하였다. 건전식물에서는 Peruvian과 Asse간에 $^{14}CO_2$동화와 $^{14}C-$동화산물 대사에서 차이가 어 없었다. 감염된 엽수가 늘어남에 다라 이들 두 품종에서 $^{14}CO_2$ 동화와 동화산물의 이행량은 감소하였다. 감염정도가 같을때도 Asse보다 Peruvian에서 $^{14}CO_2$동화가 더 감소되었다. 흰가루병이 감염되지 않은 제4엽에서도 $^{14}CO_2$고장이 줄어들었고, Peruvian에서 더 줄었다. 아래 3엽이 감염되었을 때 감염되어 있지 않는 상위 제4엽에서 fructose, glucose등의 탄수화물로 $^{14}C$가 결합되는 것을 억제하였고 Asse보다 Peruvian에서 억제가 더 컸다. 감염된 제3엽에서도 탄수화물내에 $^{14}C$이 줄어들었으나 Peruvian에서 $^{14}C$로 표식된 fructose, glucose함량이 더 높았다. 모든 식물기관에 $^{14}C$-유기산합성이 촉진되었고 Asse보다 Peruvian에서 합성이 더 뚜렷하였다. 감염된 제3엽이나 건전제4엽에서 $^{14}C$이 아미노산으로 결합되는 것이 증가하였으나 엽초로 이동은 감소되었다. 이들 잎에서 어떤 $^{14}C$-아미노산은 Asse보다 Peruvian에 더 많이 축적되었다.

Red Ginseng Ameliorates Place Learning Deficits in Aged Rats Young Rats with Selective Hippocampal Lesions

  • Ameliorating mechanisms of red ginseng on learning deficits were investigated in the following 3 experiments; its effects on 1) place learning deficits in aged rats and in young rats with selective hippocampal lesions (behavioral study), 2) long-term potentiation in the hippocampal formation (neuro- physiological study), and 3) ChAT (choline acetyl transferase) activity in various brain regions of aged rats (pharmacological study). In the behavioral study, first, performance in the place learning tasks were compared among 3 groups of young and aged rats; control young intact rats (10-12 week old) treated with water, aged rats (28-32 month old) treated with water, and aged rats (28-32 month old) treated with red ginseng (100 mghglday) suspended in water. Second, performance in the place learning tasks was compared among 3 groups of young rats; control intact rats treated with water, rats with bilateral hippocampal lesions treated with water, and rats with bilateral hippocampal lesions treated with red ginseng (100 mg/kg/day). Each rat in these 2 behavioral experiments was tested with the 3 types of the place learning tasks in a circular open field using intracranial self-stimulation (ICSS) as reward. The ICSS reward was delivered if the rat (1) moved distance of 100-160 cm (DMT): (2) entered an experiment-determined reward place within the open field, and this place was randomly varied in sequential trials (RRPST); or (3) entered 2 specific places, and did a shuttle behavior between the 2 places (PLT). Performance of the aged rats in the ginseng group was not significantly different from that of control young rats in ICSS (current intensity, bar press rates), DMT and RRPST. However, treatment with red ginseng significantly ameliorated place-navigation learning deficits in aged rats in the PLT. Similarly, red ginseng ameliorated learning and memory deficits in young rats with hippocampal lesions in the same tasks. In the neurophysiological study using young rats, perfusion of hippocampal slices with non-sapon in fraction of red ginseng significantly enhanced magnitudes of the long-term potentiation (LfP) in the CA3 subfield. In the pharmacological study, treatment with red ginseng did not affect ChAT activity in aged rat brain including the hippocampal formation. These results strongly suggest that red ginseng ameliorates learning and memory deficits in aged rats through actions on the CA3 subfield of the hippocampal formation, which were independent of the presynaptic components of the cholinergic system

온침이 만성요통에 미치는 효과에 대한 무작위배정 대조군 예비 임상연구 프로토콜 (Clinical Research of Warm Needle Acupuncture for Chronic Low Back Pain: A Protocol for Randomized, Controlled Pilot Study)

  • Objectives This study is aimed to evaluate the feasibility of further clinical research of warm needle acupuncture as an effective and safe treatment for chronic low back pain. Methods Thirty participants will be randomly assigned to the Warm needle acupuncture group (n=15) and Dry needle acupuncture group (n=15). All patients will receive acupuncture therapy twice a week for 6 weeks, but only the experimental group will receive thermal stimulation. The primary outcome will be measured using the visual analogue scale for bothersomeness of low back pain, and the secondary outcome will be measured using the VAS for pain intensity, Oswestry disability index, 36-Item Short Form Health Survey, Beck's depression inventory, and amount of analgesics used. Conclusions The results of this study will be used to determine the feasibility of a future large-scale randomized controlled trial that provides clinical evidence for the efficacy and safety of warm needle acupuncture in the treatment of patients with chronic low back pain.
