Mannan-binding lectin of the sea cucumbers Stichopus japonicus has common antigenic determinants with human serum mannan-binding lectin

  • The host defense system or immune system of all modern animals has their roots in very ancient organisms. After analyzing literature data concerning properties of invertebrates and vertebrates lectins we suggest that mechanism of mannans recognition may exist in marine invertebrates, as a universal mechanism for homeostasis maintenance and host defense, and mannan-binding lectins family of vertebrates has ancient precursor, as was shown for another S-type lectins family. We carried out the screening of mannan-binding type lectin among different species of echinoderms inhabiting in Piter the Grate Bay, the sea of Japan. As a result, the C-type lectins (SJL-32) specific for high mannose glycans was isolated from the coelomic plasma of the sea cucumbers Stichopus japonicus by ion-exchange chromatography on a DEAE-Toyopearl 650M, affinity chromatography on a mannan-Sepharose 6B and gel filtration on a Sephacryl S-200. SJL-32 is homodimer with molecular mass about 32 kDa on SDS-PAGE under non-reducing conditions. Protein part of the lectin has high conteins Asn, Glu, Ser. Hemagglutination of trypsin-treated O blood group human erythrocytes by SJL-32 was competitively inhibited by high-branched -D-mannan composed of -1,2 and -1,6 linked D-mannopyranose residues. In contrast, a variety of mono-, oligo-, and polysaccharides composed of residues of galactose and fucose showed absence or little inhibitory activities. The lectin activity strong depends on Ca2+ concentration, temperature and pH. Monospecific polyclonal antibodies were obtained to the lectin. As was shown by ELISA assay, antibodies to SJL-32 cross-reacted with human serum mannan-binding lectin. This data allows making conclusion about common antigenic determinants and structural homology of both lectins. In our opinion, SJL-32 belongs to evolutionary high conservative mannan-binding lectins (MBLs) family and takes part in the host defense against pathogenic microorganisms.

Study on the Improvement Effect of the Seeds Release of Sea Cucumber, Stichopus Japonicus (돌기해삼(Stichopus japonicus)의 방류효과 향상 연구)

  • Effects of artificial releases of sea cucumber around the marine ranching area in Jeollanam-do were showed very efficient by the released good seeds(total weight, 3.2g) in 'Wolhang', compared with those in 'Hwatae'. The survival rates were the higher in 'Wolhang' by 66.0% than in 'Hwatae' by 27.0%. The results indicates that the releasing effect of sea cucumber depended on the releas size. To analyze the productivity we investigated the growth and survival rates of sea cucumber according to the releasing area. Our result showed that the growth rate and survival rate was higher in 'Hwatae' than other areas. In 'Hwatae', the growth rate and survival rate were 6.2g and 32.1% respective. The results indicates that if the food supply is continued, although proceeding contamination the productivity of sea cucumber increased. Therefore, in order to enhancement and conservation of resource, bottom of fish cage should consider to the utilization.

Screening on Cytotoxicity of Marine Organisms Using Brine Shrimp Bioassay (Brine Shrimp Bioassay를 이용한 해양생물의 세포독성검색)

  • As a part of chemical study on the bioactive metabolites from marine organisms, we have investigated cytotoxicity using brine shrimp bioassay for each solvent fractions of the marine algae(12 species), marine sponges(3 species), coelenterates(2 species), echinoderms(4 species), marine molluscs(17 species), and ascidians(2 species), respectively. As the results, chloroform extract of Stichopus japonicus (LC$_{50}$ : 274 $\mu\textrm{g}$/ml), ethyl acetate extract of Anthocidaris crassispina(LC$_{50}$ : 121 $\mu\textrm{g}$/ml), n-butanol extract of Unda), ia Pinnatifida (LC$_{50}$ : 178 $\mu\textrm{g}$/ml), and water extract of Thais clavigera (LC$_{50}$ : 61 $\mu\textrm{g}$/ml) displayed the most significant cytotoxic activity against brine shrimp. Among the marine organisms tested, echinoderms and marine molluscs were thought to be the most active Phylums on screening of new bioactive compounds.

Vertical Distribution of Mega-invertebrate and Calculation to the Stock Assessment of Commercial Species Inhibiting Shallow Hard-bottom in Dokdo, Korea (독도 연안 암반에 서식하는 초대형 저서동물의 수직분포와 산업종의 현존량 추정)

  • Distribution and stock assessment of mega-benthos living in the shallow hard bottom around Dokdo were studied in July,2000. Depth and topographical conditions have affected to the densities and biomass of benthic animals. In shallow area, less than 10 m depth, turbo shell Batillus cornutus, mussel, Mytilus corusucs were dominated and showed distinct patterns in vertical distribution. On the other hand, the area over 10m depth, it showed diverse pattern depending on topography. Turbo shell, mussel and sea cucumber Stichopus japonicus were dominated in terms of fishery resources, but abalones were rarely sampled. Stock assessment were estimated to be 6.54 M/T, 3.89 M/T and 8.92 M/T, respectively. Some parts of coastal hard bottom around Dokdo, such as the area between Dongdo and Seodo, seemed to play an Important role as nursery ground. Therefore, it is necessary to the environmental monitoring for coastal fishery managements aspects.

Antimutagenic and Anticancer Effects of Glycoprotein and Chondroitin Sulfates from Sea Cucumber(Stichopus japonicus) (해산 극피동물 중의 당단백질의 특성과 이용 II. 해삼당단백질과 황산콘드로이친의 항돌연변이 및 항암효과)

  • The antimutagenic and anticancer activities of glycoprotein(GP) and chondroitin sulfate(CS) from sea cucumbers were studied using Ames mutagenicity test and human cancer cells culture test. The GP's inhibitory effect toward aflatoxin B1(AFB1) and 3, 2'-dimethyl-4-amino-biphenyl(DMAB) increased with the higher added concentrations up to 5% level(w/w) regardless fractionation methods. The GP from sea cucumbers through DEAE-cellulose column chromatography showed an inhibitory effect ranged from 84 to 98%, and the maximum antimutagenicities resulted in red sea cucumber with 98% (AFB1) and 95% (DMAB). But 5% level of CS from various sea cucumbers had an inhibitory effect toward those both indirect mutagens ranged from 79% to 85%. However, in case of direct mutagens(N-methyl-N'-nitro-N-nitrosoguanidine, MNNG and 4-nitroquinoline-1-oxide, 4-NQO), the GP's inhibitory effect was 55∼78% and the CS had a low inhibitory effect(58∼70%) at the added level of 5%. The GP from sea cucumbers exhibited the strong inhibitory effects with 89∼95% and 82∼92% on the growth of HT-29 human crcinoma cells and AZ-521 human gastric cancer cells (at 5% level).

Review on the Feasibility of Using Red Sea Cucumber (Stichopus japonicus) as a Multi-functional Marine Biomaterial and Comparing the Biochemical Components Collected from Different Regions in Korea (다기능 해양소재로서 홍해삼의 효능과 지역별 성분 비교를 통한 활용 가능성 연구 고찰)

  • This study aims to review the progress of biotechnological applications of sea cucumber and presents fundamental validation data on a range of methods and applications to determine the most promising fields for future exploitation and application. Sea cucumbers from different regions, subjected to numerous industrially applicable drying methods, were evaluated for their biochemical composition. The study highlights the nutritional excellence of Jeju red sea cucumber, particularly the high amino acid and fatty acid composition of its intestinal organs and the possibility of the industrial utilization of these organs. In addition, an efficient extraction method to extract collagen and chondroitin sulfate from red sea cucumber was established. The extracted collagen and chondroitin sulfate can be used as commercially valid biomaterials. Accordingly, the red sea cucumber was confirmed as a valuable source of raw food material of varied functionality and bioactivity. Upon conducting an analysis of patents thus far relating to the sea cucumber, it was found that the associated technology was limited to some parts of primary and secondary industries. Therefore, there is a clear need for the strategic development of technologies to produce specialized functional products from sea cucumber's raw materials, with a view to promote other related industries.

Quality and Preference Changes Red Sea Cucumber (Stichopus japonicus) Kimchi during Storage Period (홍해삼 첨가 김치의 저장기간 중 품질 특성 및 기호도 변화)

  • To diversify usage of aquacultural red sea cucumber, new Kimchi containing red sea cucumber was developed and its quality and consumer preference were studied. The pH and total acidity, index of Kimchi ripening, decreased till 7 days of storage, then pH change became blunt and finally reached optimum pH 4-4.5. The amount of reducing sugar gradually decreased during storage period. The number of lactic acid bacteria increased in early fermentation period in both Kimchi containing boiled and fresh red sea cucumber, however, more lactic acid bacteria found in fresh red sea cucumber containing Kimchi. Preference of Kimchi containing boiled or fresh red sea cucumber compared to control Kimchi containing fermented salted fish was evaluated. The results revealed there were no differences in color, flavor, texture, taste, after taste and overall preference.
