Structural Characteristics on InAs Quantum Dots multi-stacked on GaAs(100) Substrates

  • The InAs self-assembled quantun dots (SAQDS) were grown on a GaAs(100) substrate using a molecular beam epitaxy (MBE) technique. The InAs QDs were multi-stacked to have various layer structures of 1, 3, 6, 10, 15 and 20 layers, where the thickness of the GaAs spacer and InAs QD layer were 20 monolayers (MLs) and 2 MLs, respectively. The nanostructured feature was characterized by photoluminescence (PL) and scanning transmission electron microscopy (STEM). It was found that the highest PL intensity was obtained from the specimen with 6 stacking layers and the energy of the PL peak was split with increasing the number of stacking layers. The STEM investigation exhibited that the quantum dots in the 6 stacking layer structure were well aligned in vertical columns without any deflect generation, whereas the volcano-like deflects were formed vertically along the growth direction over 10 periods of InAs stacking layers.

EFFECTS OF bFGF AND PDGF-BB ON OSTEOBLAST DIFFERENTIATION OF BONE MARROW-DERIVED MESENCHYMAL STEM CELL IN RAT (bFGF, PDGF-BB가 백서 골수기원 간엽 줄기세포의 조직골세포 분화에 미치는 영향에 관한 연구)

  • In this study we evaluate the effects of bFGF-BB and PDGF on in vitro proliferation, differentiation and mineralization of mesenchymal stem cells (MSCs) from rat. MSCs were prepared from the bone marrow of 6 or 7-week-old male rats with a technique previously described by Maniatopoulos et al. in 1988. Lineage differentiation to osteogenesis, chondrogenesis and adipogenesis were performed. At first, we characterized the cultured cell on passage 1, 3, 5, 7 with immunocytochemical staining using CD29, 44, 34, 45, ${\alpha}$-SMA and type I collagen. And to study the effects of bFGF and PDGF-BB on proliferation, differentiation and mineralization, we seeded the expanded cell at a density of 6 $6{\times}10^3\;cells/cm^2$ to 100-mm dish for evaluation of cell proliferation and MTT assay was carried out on day 2, 4, 7, 9. We also resuspended the cells with same density $(6{\times}10^3\;cells/cm^2)$ to 24 well plates for subculture. On the following day, the attached cells were exposed to 2.5ng/ml bFGF and/or 25ng/ml PDGF-BB daily during 5 days. The osteocalcin (OC) level was assessed and mineral contents were evaluated with alizarin red S staining on subculture day 2, 7, 14, 21. We identified the mesenchymal stem cell from the bone marrow derived cells of rat through their successful multi-differentiation and stable display of its phenotype. And bFGF and PDGF-BB showed the effect that inhibited osteoblastic differentiation and mineralization mildly in above concentration at in vitro culture. Protective Effects of the Methanol Extracts of Acanthopanax koreanum against Oxidative Stress (산화적 스트레스에 대한 섬오갈피 메탄올 추출물의 보호 효과)

  • The protective effects of the aqueous MeOH extracts of stem and root Acanthopanax koreanum Nakai against oxidative stress were investigated. Anti-oxidant activity of the stem extract of A. koreanum was observed in the DPPH free radical scavenging $(IC_{50}=58.7\;{\mu}g/ml)$ and the SOD test $(IC_{50}=17.52\;{\mu}g/ml)$. According to data analysis of cell survival ratio of normal fibroblasts, the skin irritation by both extracts from A. koreanum was concerned. However, in the skin primary parch test, both extracts obtained Grade I, which means that there was no skin irritation. After induction of oxidative irritation, cell survival ratio of normal fibroblasts was also monitored and it turned out that stress-inducing group with both extracts had more increased cell survival ratio. The cell extension of the stress-inducing group treated with the stem extract was most dominant in morphological study. Based on these results, the stem extract of A. koreanum showed the protective effect against oxidative stress on normal fibroblast.

Partial Characterization of Allelopathic Substances in Sorghum Stem by Different Organic Solvents and pH (수수 줄기에 함유(含有)된 타감물질(他感物質)의 용매(溶媒)와 pH에 따른 특성구명(特性究明))

  • The toxic compounds of sorghum stem extracts were isolated by different organic solvents and pH, and characterized and quantified in terms of their inhibition of seed germination and seedling growth in Echinochloa colona(L.) Link and radish(Raphanus sativus L.). Sequential partitioning of stem extract with various organic solvents with increasing polarity showed that all fractions of hexane, ethyl ether, methylene chloride, ethyl acetate, and the aqueous remainder inhibited germination and seedling growth in E. colona. Of the five fractions, the ethyl ether fraction had the greatest inhibitory effect on E. colona. Further separation of the ethyl ether fraction at different pH(pH 2-11) showed that phytotoxic compounds were acidic. The result indicates that the phytotoxin present in the stem extract may be nonpolar and acidic.

Stem Rot of Lily(Lilium L.) Caused by Phytophthora cactorum in Korea (Phytophthora cactorum에 의한 백합 역병)

  • Stem rot symptomof lily caused by Phytophthora was observed in Chilkok, Korea in 1997. Oogonia of Phytophthora were abundantly developed on epidermal tissues of stem and leaves of the diseased plants. The casual fungus was identified as Phytophthora cactorum based on their cultural and morphological characteristics. Sporangia were markedly papillate and the shape was broadly spherical to ovoid both on agar and in water. The size was 28~44$\times$20~28 ${\mu}{\textrm}{m}$ (av. 36~24) and a short pedicel was attached to each sporangium. The fungus was homothallic and oogonia were 28~32 ${\mu}{\textrm}{m}$ (av. 30) in size and slightly yellow. Antheridia were paragynous and spherical or irregularly club shaped. Optimal temperature for mycelial growth was 25~27$^{\circ}C$. The fungus did not grow under 6$^{\circ}C$ and over 34$^{\circ}C$. The fungus was highly pathogenic to three cultivars of lily. Symptom was developed at 4-day after artificial inoculation with zoospores (103 cell/ml) on the stem of lily. The symptom was identical to those produced by natural infection in the field and the plant was blighted within 14 days after inoculation.

  • The corneal tissue consists of three layers : epithelium, stroma, and endothelium. Central cornea is a highly differentiated tissue whereas the limbus contains the epithelial stem cell. In the present study. we report the engineering of the three-dimensional reconstructed cornea derived from rabbit limbal epithelial and stromal cells. The differentiation degree of corneal stem cells were assessed in serum concentration and inoculation density of stromal cells. Optimal condition differentiation of corneal stem cells is achieved when 5% FBS was supplemented to culture medium and $1-2{\times}10^5$ cells/ml inoculation density of stromal cells.

Antioxidative Compounds isolated from the Stem Bark of Eucalyptus globulus (유칼리나무의 수피로부터 분리한 항산화활성 물질)

  • Seven antioxidative compounds were isolated from chloroform and ethyl acetate extracts of the stem bark of Eucalyptus globulus (Myrtaceae). They were identified as rhamnazin (1), rhamnetin (2), naringenin (3), eriodictyol (4), quercetin (5), taxifolin (6) and dihydrokaempferol-3-rhamnoside (7) on the basis of various spectroscopic analyses. These compounds inhibited lipid peroxidation with $IC_{50}$ values of 0.08-30 ${\mu}g/ml$.

Tumor cell growth inhibition and antioxydative activity of flavonoids from the stem bark of Cudrania tricuspidata (꾸지 뽕나무로부터 분리한 flavonoid계 화합물의 암세포성장 저해 및 항산화 활성)

  • Five cytotoxic and antioxidative flavonoids were isolated from the stem bark of Cudrania tricuspidata by consecutive purification using HP-20, silicagel and prep-HPLC. They were identified as taxifolin, orobol, eriodictyol, dihydrokaempferol and steppogenin by means of spectral studies. The antioxidative activities $(IC_{50})$ assayed by TBA method of these compound $1{\sim}5$ to were 6, 3, 3, >50, and $10\;{\mu}g/ml$, respectively. The effect on the growth inhibition $(IC_{50})$ of these compounds against P388 cell line were found to be 0.18, 3.3, 15 and $6.2\;{\mu}g/ml$, respectively in the order of compound 2 to 5.

An in vitro study of mesenchymal stem cell proliferation on titanium discs coated with rhTGF-β2/PLGA by electrospray (Electrospray법으로 rhTGF-β2/PLGA 복합체를 코팅한 티타늄에서의 간엽줄기세포 증식에 관한 연구)

  • Purpose: The purpose of this study is to identify the effect of mesenchymal stem cell proliferation on recombinant human transforming growth factor-beta (rhTGF-${\beta}2$) / poly (D,L-lactide-co-glycolide) (PLGA) treated titanium discs by electrospray. Materials and methods: Anodized titanium surface coated with PLGA was used for a control group to compare anodized titanium surface coated with 125 ng/ml and 500 ng/ml rhTGF-${\beta}2$ as test groups. Atomic force microscope (AFM) test was utilized to determine the difference in coating surface roughness, and field-emission scanning electron microscopy (FE-SEM) was taken to visualize even distribution of coating particles on titanium discs. The mesenchymal stem cell proliferation was tested by using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyl-tetrazolium bromide) assay on 1st, 4th, 7th days. Results: According to AFM results, there was no statistically significant difference in titanium discs treated with PLGA and with rhTGF-${\beta}2$/PLGA (P>.05). MTT assay test results showed that there was statistically significant difference in mesenchymal stem cell proliferation on test groups compared to control groups at 7th day, and cell viability on discs coated with rhTGF-${\beta}2$ was significantly higher than control groups (P<.05). Conclusion: Titanium surface coated with rhTGF-${\beta}2$/PLGA shows statistically significant higher cell proliferation and the titanium surface coated with the higher concentration of rhTGF-${\beta}2$ presents faster cell growth activity.

Effects of Tetrapanax papyrifer stem and Akebiae quinata stem on a rat model of monosodium iodoacetate-induced osteoarthritis (통초(通草)와 목통(木通) 추출물이 monosodium iodoacetate(MIA)로 유발된 골관절염 동물 모델에 미치는 효과)

  • Objectives : This study was planned to evaluate the therapeutic effectiveness and possible underlying mechanism of TPE (Tetrapanax papyrifer stem(inner part of the stem Extract) and AQE (Akebiae quinata stem Extract) on osteoarthritis. Methods : Osteoarthritis models were induced through intra-articular injection of MIA (monosodium iodoacetate) 50 μL with 80 mg/ml in rats. Excluding the normal group, Osteoarthritis-induced rats were divided into 4 groups (Control, INDO, TPE, AQE). The drug concentrations were indomethacin 5 mg/kg, TPE 200 mg/kg, and AQE 200 mg/kg, and were orally administered once a day for a couple of weeks. After drug supplementation, the effects of TPE and AQE were measured with serum diagnosis, western blotting, and histopathological staining. Results : It was found that the DPPH and ABTS free radical erasure ability of AQE was better than that of TPE. AQE administration improved rear limb overload and it led to relieving pain. Both PTE and AQE significantly reduced the expression of inflammatory mediators COX-2, iNOS, and inflammatory cytokine IL-1β and IL-6 by inhibiting the phosphorylation of IκBα and deactivating the pathway of NF-κBp65. On the other hand, TNF-α was significantly reduced only by administration of AQE. In addition, histopathological analysis showed that the administration of AQE compared to PTE suppressed cartilage degeneration and effectively suppressed damage to proteoglycan, a component of ECM. Conclusion : Reviewing these experimental results, TPE and AQE possessed the effect of delaying the progress of osteoarthritis and protecting cartilage. In addition, the results of this study show that AQE has a better cartilage protection effect than TPE.
