In Vitro Neural Cell Differentiation Derived from Human Embryonic Stem Cells: I. Effect of Neurotrophic Factors on Neural Progenitor Cells

  • This study was to investigate the effect of neurotrophic factors on neural cell differentiation in vitro derived from human embryonic stem (hES, MB03) cells. For neural progenitor cell formation derived from hES cells, we produced embryoid bodies (EB: for 5 days, without mitogen) from hES cells and then neurospheres (for 7 - 10 days, 20 ng/㎖ of bFGF added N2 medium) from EB. And then finally for the differentiation into mature neuron cells, neural progenitor cells were cultured in ⅰ) N2 medium (without bFGF), ⅱ) N2 supplemented with brain derived neurotrophic factor (BDNF, 5ng/㎖) or ⅲ) N2 supplemented with platelet derived growth factor-bb (PDGF-bb, 20ng/㎖) for 2 weeks. Effect of Physical Properties of Soil on Ginseng Seedling Growth in Nursery Bed (양식묘단 토양의 물리성이 묘삼생육 및 수량에 미치는 영향)

  • This study was conducted to elucidate the effect of physical properties of soil in nursery bed with different densities on growth of ginseng seedling. Stem length, leaf length and leaf width of ginseng seedling showed the decreasing tendency with increasing the hardness of the nursery soil. Fresh root weight per seedling and number of available seedlings were increased significantly with decrease of the soil hardness. For solid, liquid phases, bulk density and hardness of soil, negative correlations were shown in stem length, leaf length, leaf width, root weight per seeding, and number t of available seedlings. On the other hand, gas phase, air permeability and porosity of soil had positive correlations with stem length, leaf length, leaf width, root weight per seedling and number of available seedlings.   • To study antimicrobial activities of Impatiens balsamina, its leaf and stem was extracted by methanol and water. The extracts were tested their microbial inhibition activities against several microorganisms, staphylococcus aureus KCTC 1928, Escherichia coli KCTC 1116, Klebsiella pneumoniae KCTC 2690, Trichophyton mentagrophytes KCTC 6316. The methanol extract and water extract of the leaf and stem didn't show the inhibition effect on E. coli but showed the inhibition effect against the ohter 3 species of the microorganisms tested. The extract of leaf showed higher inhibition than the extract of stem. And the methanol extract showed higher inhibition than the water extract.

Carnosol induces the osteogenic differentiation of bone marrow-derived mesenchymal stem cells via activating BMP-signaling pathway

  • Carnosol is a phenolic diterpene phytochemical found in rosemary and sage with reported anti-microbial, anti-oxidant, anti-inflammatory, and anti-carcinogenic activities. This study aimed to investigate the effect of carnosol on the lineage commitment of mouse bone marrow-derived mesenchymal stem cells (mBMSCs) into osteoblasts and adipocytes. Interestingly, carnosol stimulated the early commitment of mBMSCs into osteoblasts in dose-dependent manner as demonstrated by increased levels of alkaline phosphatase activity and Alizarin red staining for matrix mineralization. On the other hand, carnosol significantly suppressed adipogenesis of mBMSCs and downregulated both early and late markers of adipogenesis. Carnosol showed to induce osteogenesis in a mechanism mediated by activating BMP signaling pathway and subsequently upregulating the expression of BMPs downstream osteogenic target genes. In this context, treatment of mBMSCs with LDN-193189, BMPR1 selective inhibitor showed to abolish the stimulatory effect of carnosol on BMP2-induced osteogenesis. In conclusion, our data identified carnosol as a novel osteoanabolic phytochemical that can promote the differentiation of mBMSCs into osteoblasts versus adipocytes by activating BMP-signaling.

STK899704 inhibits stemness of cancer stem cells and migration via the FAK-MEK-ERK pathway in HT29 cells

  • Colon cancer is one of the most lethal and common malignancies worldwide. STK899704, a novel synthetic agent, has been reported to exhibit anticancer effects towards numerous cancer cells. However, the effect of STK899704 on the biological properties of colon cancer, including cancer cell migration and cancer stem cells (CSCs), remains unknown. Here, we examined the inhibitory effect of STK899704 on cell migration and CSC stemness. In the wound healing assay, STK899704 significantly inhibited the motility of colon cancer cells. Furthermore, STK899704 downregulated the mRNA expression levels of the cell migration mediator focal adhesion kinase (FAK). STK899704 also suppressed mitogen-activated protein kinase kinase and extracellular signal-regulated kinase, which are downstream signaling molecules of FAK. Additionally, STK899704 inhibited stemness gene expression and sphere formation in colon cancer stem cells. These results suggest that STK899704 can be used to treat human colon cancer.

Effect of Rosa rugosa, Thunbergii to the Streptozotocin induced Hyperglycemic Rats (매괴화(玫瑰花) 잎, 줄기, 뿌리가 streptozotocin으로 유발(誘發)된 당뇨(糖尿)흰쥐에 미치는 영향(影響))

  • This paper aims to probe the effect of Rosa rugosa, Thunbergii which streptozotocin induced hyperglycemic rats was examined by glucose contents, body weight, kidney weight and histological observation. The results obtained were as follows: 1. The glucose level is decreased in the adminstered group of the Rosa rugosa compared with control group. These effects are in order named root group, stem group and leaf group group. BUN is increased fold of 1.52 in the control group compared with normal group, but the administered group of Rosa rugosa are decreased., in order named leaf group, root group and stem group. 2. The kidney hypertrophy to body weight is the higest in the control group compared with the other group in rats, and glucose contents are in order named stem group, leaf group and root group. 3. The loss of body weight is the highest in the control group, and the loss is in order named stem group, leaf group and root group in rats. 4. In histological observations, pancreatic pancreatic islets of control group shows disrupted or condensed nuclei, and the intercellular space between acini are enlarged. In the experimental group, histological damages of the acini and pancreatic islet are recovered compared with control group. Thes results suggest that the extracts of Rosa rugosa are effected to diabetes mellitus, root extract correlated with the function of kidney, and leaf extract is decreased plasma glucose.

  • Objectives : The aim of this study was to investigate the effect of various electroacupuncture stimulation on neuronal nitric oxide synthase(nNOS) in cerebral cortex, brain stem, cerebellum of spontaneously hypertensive rats. Methods : We evaluated the changes of nNOS-positive neurons using a immunohistochemical method. The staining intensity of nNOS positive neurons was assessed in a quantitative fashion using a microdensitometrical method based on optical density by means of an image analyzer. Results : The average optical density of nNOS-positive neurons of 100 Hz (bipolar square wave 0.2 ms duration and 100 Hz frequency) electroacupuncture treatment group significantly decreased in most cortical areas comparison between the manual acupuncture and 2 Hz (bipolar square wave 0.2 ms duration and 2 Hz frequency) electroacupuncture groups. In the brain stem, the optical density of nNOS-positive neuron at superficial gray layer of the superior colliculus area, dorsolateral periaqueductal gray area and paralemniscal nucleus were same as cerebral cortex. Conclusion : We conclude that the morphological evidence for nNOS-positive neurons may be have regional change in cerebral cortex brain stem and cerebellum according to various electroacupuncture stimulations.

  • 1) These experiments were couducted to compare the effectiveness of with several new insccticides for the control of the rsce stem borer at Kimhae and Suwon in Korea from 1962 to 1963. 2) Gam ma-dol dusf and gramular were applied on the water surbul by hand and the other insecticides were sprayed on the leves of the paddy rice plaut by knap sack spayer(Table 1). 3) Gamma-dol was for the control of rice stem borers. more effective than other insecticidesused. timing for the applioation of$\gamma-dol$ was 10 days before hatching from the eggs. 4) There were no any significant differences in the effect of$\gamma-dol$ to the rice stem borer in both areas of Kimhae and Suwon.

  • Aldose reductase (AR) has been shown to play an important role in the development of the diabetic complications. To discover novel treatments for diabetic complications from natural sources, 59 Chinese herbal medicines have been investigated for inhibitory activities on AR. Among them, 10 herbal medicines, Catalpa fargesii (stem and leaf), Saussurea Laniceps(whole plant), Alnus nepalensis(stem and leaf), Swertia macrosperma (whole plant), Woodfordia fruticosa (stem and leaf), Elsholtzia bodinieri (whole plant), Elsholtzia fruticosa (whole plant), Rosa multiflora (fruit), Nardostachys chinensis (whole plant), Eurya groffii (stem and leaf) exhibited a significant inhibitory activity compared with 3,3-tetramethyleneglutaric acid (TMG) as positive control. Particularly, 4 herbal medicines, C. fargesii (stem and leaf), S. Laniceps (whole plant), A. nepalensis (stem and leaf), S. macrosperma (whole plant) showed two times more potent inhibitory activity than TMG ($5.37{\mu}g/ml$).

Dlx3 and Dlx5 Inhibit Adipogenic Differentiation of Human Dental Pulp Stem Cells

  • Dlx3 and Dlx5 are homeobox domain proteins and are well-known regulators of osteoblastic differentiation. Since possible reciprocal relationships between osteogenic and adipogenic differentiation in mesenchymal stem cells exist, we examined the regulatory role of Dlx3 and Dlx5 on adipogenic differentiation using human dental pulp stem cells. Over-expression of Dlx3 and Dlx5 stimulated osteogenic differentiation but inhibited adipogenic differentiation of human dental pulp stem cells. Dlx3 and Dlx5 suppressed the expression of adipogenic marker genes such as $C/EBP{\alpha}$, $PPAR{\gamma}$, aP2 and lipoprotein lipase. Adipogenic stimuli suppressed the mRNA levels of Dlx3 and Dlx5, whereas osteogenic stimuli enhanced the expression of Dlx3 and Dlx5 in 3T3-L1 preadipocytes. These results suggest that Dlx3 and Dlx5 exert a stimulatory effect on osteogenic differentiation of stem cells through the inhibition of adipogenic differentiation as well as direct stimulation.
