Forskolin Effect on the Lineage Specification of Trunk Neural Crest Cells in vitro

  • Recent evidence has suggested that trunk neural crest cell generally assumed to have equivalent differentiation potentials, demonstrate differentiation bias along the anterior/posterior axis. In amphibian and fish, neural crest cells give rise to three chromatophore types, melanophores, xantho-phores, and iridophores. Each pigment cell type has distinct characteristics but there is speculation about the cellular plasticity that exists among them. Neural crest cells migrate along specific routes, ventromedially and dorsolaterally. Neural crest cells that travel dorsolaterally are the first cells to begin migration in the axolotl and are the major contributors to the visible pigment pattern. Many factors and mechanisms that are responsible for guiding migratory neural crest cells along potential pathways or determining their fate remain unknown. A single lineage of the crest, which becomes restricted to one of the three pigment cell types, gives us the opportunity to examine the existence of neural crest stem cell populations and cellular plasticity. Study presented here showed results from recent in vitro studies designed to identify parameters influencing differentiation events of individual neural crest-derived pigment cell lineages. Melanophore production from neural crest explants originating from different levels along the anterior/posterior axis of wild type-axolotl embryos were compared and demonstrate that the differentiation of melanophores is enhanced in subpopulation of neural crest treated with forskolin. Forskolin (an adenylate cyclase activator) increases intracellular CAMP concentration and eventually activates the protein kinase-A signaling pathway. Melanophore number, melanin content, and tyrosinase activity in explants taken from the anterior-most region of the crest increased significantly in response to forskolin treatment. This study suggests implications of region specific influences and developmental regulation in the development of pigment pattern.

  • Atypical teratoid rhabdoid tumours (ATRTs) are the most common malignant central nervous system tumours in children ${\leq}1year$ of age and represent approximately 1-2% of all pediatric brain tumours. ATRT is a primarily monogenic disease characterized by the bi-allelic loss of the SMARCB1 gene, which encodes the hSNF5 subunit of the SWI/SNF chromatin remodeling complex. Though conventional dose chemotherapy is not effective in most ATRT patients, high dose chemotherapy with autologous stem cell transplant, radiotherapy and/or intrathecal chemotherapy all show significant potential to improve patient survival. Recent epigenetic and transcriptional studies highlight three subgroups of ATRT, each with distinct clinical and molecular characteristics with corresponding therapeutic sensitivities, including epigenetic targeting, and inhibition of tyrosine kinases or growth/lineage specific pathways.

  • Ultrastructural characteristics of hemocytic differentiation pathway in Sericinus montetla Grey were observed with transmission electron microscope. Hemocytic differentiation took place in loose islets of hemopoietic organs as indicated by the presence of differentiating hemocytes; prohemocytes, plasmatocytes, granular cells and spherule cells. They are differentiatied from the stem cells through individual cell lineages. However, differentiating aspects of the oenocytoids were not observed. According to Brehelin and Zachary (1986), the premature hemocytes around the hemopoietic organs were classified into five types; prohemocytes, plasmatocytes, granular cells, spherule cells and oenocytoids.

  • Objective : The purpose of this study is to analyse research trend on alopecia including androgenetic alopecia, alopecia areata, etc.Methods : We searched Pubmed, CNKI and OASIS on the title "alopecia" in recent 5 years.Results : We found 23 studies on Pubmed, 20 on CNKI, and 3 on OASIS about treatments of alopecia. The subjects of them were about 5α-reductase inhibitor, Minoxidil, steroids, immunosuppression, laser therapy, PRP, microneedling, stem cell, herbal medicine, integrative medicine, external treatments and syndrome differentiation.Conclusions : In recent studies in western medicine were about combination therapy or safety and presented validities and superiorities about new treatments. Integrative medicine, external treatment and researches about patients' characteristics were reported in TCM. Korean medicine has advantages of considering whole body and phychological problems.

  • Rectovaginal fistula, which can arise after an injury to the vaginal canal or rectum, is a troublesome obstacle for patients' everyday life. In most cases, it can be covered with a local flap, but previous radiation therapy increases the recurrence rate, making it especially difficult to cure. As the application of stromal vascular fraction (SVF) obtained from enzymatically digested autologous adipose tissue has become increasingly common, several reports have advocated its effectiveness for the treatment of refractory wounds. In light of the angiogenic, regenerative characteristics of SVF, it was incorporated as a treatment option in two cases of rectovaginal fistula discussed here. As described in this report, irradiated rectovaginal fistulas in rectal cancer patients were successfully treated with SVF injection, and we suggest SVF as a feasible treatment option for cases of rectovaginal fistula that would otherwise be very difficult to cure.

17β-estradiol mediated effects on pluripotency transcription factors and differentiation capacity in mesenchymal stem cells derived porcine from newborns as steroid hormones non-functional donors

  • The estrogen-mediated effect of mesenchymal stem cells (MSCs) is a highly critical factor for the clinical application of MSCs. However, the present study is conducted on MSCs derived from adult donors, which have different physiological status with steroid hormonal changes. Therefore, we explores the important role of $17{\beta}$-estradiol (E2) in MSCs derived from female and male newborn piglets (NF- and NM-pBMSCs), which are non-sexually matured donors with steroid hormones. The results revealed that in vitro treatment of MSCs with E2 improved cell proliferation, but the rates varied according to the gender of the newborn donors. Following in vitro treatment of newborn MSCs with E2, mRNA levels of Oct3/4 and Sox2 increased in both genders of MSCs and they may be correlated with both estrogen receptor ${\alpha}$ ($ER{\alpha}$) and $ER{\beta}$ in NF-pBMSCs, but NM-pBMSCs were only correlated with $ER{\alpha}$. Moreover, E2-treated NF-pBMSCs decreased in ${\beta}$-galactosidase activity but no influence on NM-pBMSCs. In E2-mediated differentiation capacity, E2 induced an increase in the osteogenic and chondrogenic abilities of both pBMSCs, but adipogenic ability may increased only in NF-pBMSCs. These results demonstrate that E2 could affect both genders of newborn donor-derived MSCs, but the regulatory role of E2 varies depending on gender-dependent characteristics even though the original newborn donors had not been affected by functional steroid hormones.

Characterization of Odontoblasts in Supernumerary Tooth-derived Dental Pulp Stem Cells between Passages by Real-Time PCR (과잉치 치수유래 줄기세포의 Real-time PCR에 의한 계대간 상아질모세포 발현 특성)

  • The aim of this study is to compare the properties of odontoblast gene of early passage cells and late passage cells derived from impacted maxillary supernumerary teeth. Impacted supernumerary teeth with maxilla were extracted from 12 patients (8 males, 4 females) between 6 - 9 years old without medical history. Real-time polymerase chain reaction (PCR) was conducted to compare characterization of odontoblast cell in the 3rd and 10th passage, and between with bone inducing additive group and without additive group. Genes for odontoblasts characteristics are osteonectin (ONT), alkaline phosphatase (ALP), osteocalcin (OCN), dentin matrix protein 1 (DMP-1) and dentin sialophosphoprotein (DSPP). The level of gene expression was in a decreasing order of ONT, ALP, OCN, DMP-1 and DSPP in the 3rd passage, and in decreasing order of ONT, DMP-1, OCN, ALP, and DSPP in the 10th passage in the undifferentiation and differentiation group. The order of ONT, DMP-1, and OCN did not changed. ALP and DMP-1 were switched in order. ALP and DMP-1 may be used as important markers for differentiating between the 3rd passage and 10th passage cells. Considering that supernumerary tooth was extracted young age and the time required to cultured 10th passage was short, supernumerary tooth can be considered a useful donor site of dental pulp stem cells.

Effects of different culture systems on the culture of prepuberal buffalo (Bubalus bubalis) spermatogonial stem cell-like cells in vitro

  • Currently, the systems for culturing buffalo spermatogonial stem cells (SSCs) in vitro are varied, and their effects are still inconclusive. In this study, we compared the effects of culture systems with undefined (foetal bovine serum) and defined (KnockOut Serum Replacement) materials on the in vitro culture of buffalo SSC-like cells. Significantly more DDX4- and UCHL1-positive cells (cultured for 2 days at passage 2) were observed in the defined materials culture system than in the undefined materials system (p < 0.01), and these cells were maintained for a longer period than those in the culture system with undefined materials (10 days vs. 6 days). Furthermore, NANOS2 (p < 0.05), DDX4 (p < 0.01) and UCHL1 (p < 0.05) were expressed at significantly higher levels in the culture system with defined materials than in that with undefined materials. Induction with retinoic acid was used to verify that the cultured cells maintained SSC characteristics, revealing an SCP3+ subset in the cells cultured in the defined materials system. The expression levels of Stra8 (p < 0.05) and Rec8 (p < 0.01) were significantly increased, and the expression levels of ZBTB16 (p < 0.01) and DDX4 (p < 0.05) were significantly decreased. These findings provided a clearer research platform for exploring the mechanism of buffalo SSCs in vitro.

Comparison of Plant Growth and Morphological Characteristics Among the Korean Ginseng, the American Ginseng and the Bamboo Ginseng (고려인삼, 미국삼 및 죽절삼의 생육 및 형태적 특성 비교)

  • An investigation was conducted to ascertain the basic information on characteristics of growth and morphological characters among the Korean (Panax. ginseng), the American (Panax. quinquefolium) and the Bamboo (Panax. japonicus) ginseng. In aerial parts growth of the ginseng species by age, The Korean ginseng and American ginseng's stem and leaf growth was alike in 2-4 years old, but growth cycle changed in 6 years old. The Korean ginseng was more vigorous than the American ginseng. The Korean ginseng roots were highly observed in ratio of red skin roots among three species, whereas The American ginseng roots were highly infected by root rot. It seems to be variable depending on growing stage and species. The Korean ginseng flowered about the middle of May, the American ginseng early June, and the Bamboo ginseng was late of May, The berry color of the ginseng species was observed, The Korean and American ginseng's mature berry color was red, The Bamboo ginseng's berry was three type of color and shape. In root characteristics of the seedling, Korean (p. ginseng), American (p. quinquefolium) ginseng's root shape was similarity in type, the bamboo ginseng showed different type, which root length and root weight was smaller than those of ginseng. In morphological characters of Leaf surface, pollen, and stoma, the Korean ginseng and American ginseng had crystal rosette on epidermis cell, but the Bamboo ginseng didn't has crystal rosette. Pollen shape observed tricolpate pollen and size was media type among the ginseng species, and also guard cell was anomocytic type, which were observed by scanning electronic microscope.

Transduction of eGFP Gene to Human Embryonic Stem Cells and Their Characterization (인간 배아줄기세포로의 eGFP 유전자 도입 및 특성 분석)

  • Objective: Human embryonic stem cells (hESCs) can proliferate indefinitely and differentiate into all kinds of cell types in vitro. Therefore, hESCs can be used as a cell source for cell-based therapy. Transduction of foreign genes to hESCs could be useful for tracing differentiation processes of hESCs and elucidation of gene function. Thus, we tried to introduce enhanced green fluorescent protein (eGFP) gene to hESCs, XX and XY cell lines in this study. Methods: Lentivirus containing eGFP was packaged in 293T cells and applied to hESCs to transduce eGFP. Expression of transduced eGFP was evaluated under the fluorescence microscope and eGFP positive population was analyzed by FACS. Expression of undifferentiation state markers such as Oct4, Nanog, SSEA4 and Tra-1-81 was examined by RT-PCR and/or immunofluorescence in eGFP-hESCs after transduction. In addition, the ability of eGFP-hESCs to form embryoid bodies (EBs) was tested. Results: eGFP was successfully transduced to hESCs by lentivirus. eGFP expression was stably maintained up to more than 40 passages. eGFP-hESCs retained expression patterns of undifferentiation state markers after transduction. Interestingly, disappearance of transduced eGFP was notably observed during spontaneous differentiation of eGFP-hESCs. Conclusion: We established eGFP expressing hESC lines using lentivirus and showed the maintenance of undifferentiation characteristics of these eGFP-hESCs. This reporter-containing hESCs could be useful for tracing the processes of differentiation of hESCs and other studies.
