Upward and Lateral Translocation of $^32 P$ Supplied to Roots of Apple and Citrus Tress

  • $^{32}$ P was supplied to the roots of stem-ringed 1-year-old apple trees and 2-year-old citrus trees on which bark segments were isolated above and below the ring. $^{32}$ P was translocated to shoots and leaves although considerable translocation occurred especially in wood and bark tissues. The accumulation of $^{32}$ P in isolated bark segments indicated that the occurrence of these materials in this tissue was via radial translocation from xylem tissue, and that the main upward translocation pathway of $^{32}$ P supplied to roots is through the xylem.

  • Among the Araliaceae plants indigenous to Korea, those whose medicinal usage are comparatively high have been selected in this serial studies. Chiisanoside and acanthoside D were isolated and identified from the Acanthopanax chinensis leaves and root bark. Chiisanoside and acanthoside D have been found to have the lowering S-GPT, S-GOT value and BSP-retention rate and survival rate, anti-histaminic effect in the toxic state through the bio-pharmacological experiments. ${\alpha}-hederin$, hederagenin pentaglycoside were isolated both stem bark of Kalopanax pictum Nakai var. magnificum and Kalopanax pictum Nakai var. Max. respectively. Syringoside, acanthoside D were also isolated from the root bark of Acanthopanax koreanum. The biological activity of ginsenoside $Rb_1$, $Rg_1$, Re were examined. Ginsenoside $Rb_1$, $Rg_1$, Re promotes the antileaking effect in X-ray (Co 60) irradiated toxic state.

Analgesic, cytotoxic and antioxidant activities of Trewia polycarpa bark

  • The crude ethanol extract of the stem bark of Trewia polycarpa (Family: Euphorbiaceae) was subjected to acetic acid induced writhing inhibition, Brine Shrimp lethality bioassay and 1, 1-diphenyl-2-picryl hydrazyl free radical scavenging assay for screening of analgesic, cytotoxic and antioxidant activity respectively. The extract produced significant (P < 0.001) writhing inhibition in acetic acid induced writhing in mice at the dose of 125, 250 and 500 mg/kg body weight respectively, which were comparable to the standard drug diclofenac sodium. The extract showed significant lethality to Brine Shrimp and the $LC_{50}$ value was $8\;{\mu}g/ml$. The extract showed prominent free radical scavenging activity ($IC_{50}$ about ${\sim}10\;{\mu}g/ml$) compare to standard drug ascorbic acid ($IC_{50}about\;{\sim}15\;{\mu}g/ml$). The results tend to suggest that the crude ethanol extract of the bark might possess analgesic, cytotoxic and antioxidant activities or active constituent(s) responsible for the activities.

Estimation of Aboveground Biomass and Net Primary Production for Pinus rigida in Sancheong, Gyeongnam Province (경남 산청지역 리기다소나무림의 지상부 바이오매스와 순생산량 추정)

  • This study was conducted to estimate the aboveground biomass and net primary production of 40 year old Pinus rigida forest in Sancheong, Gyeongnam province. The aboveground biomass was $127.7Mg\;ha^{-1}$ and $103.1Mg\;ha^{-1}$ of which were found from stem, $17.2Mg\;ha^{-1}$ from branch and $7.4Mg\;ha^{-1}$ from leaf. Biomass distribution ratio of Pinus rigida stands showed the highest in stem wood with 71.1%, followed by the branch with 13.5%, stem bark with 9.6% and lastly the leaf with 5.8%. Net primary production of aboveground biomass was $10.4Mg\;ha^{-1}$ and $3.6Mg\;ha^{-1}$ were found from stem, $2.2Mg\;ha^{-1}$ from branch, $1.3Mg\;ha^{-1}$ from twig and $2.8Mg\;ha^{-1}$ from leaf. Net primary production distribution ratio of Pinus rigida was 34.6% on stem wood, 26.9 on leaf, 21.2% on branch, 12.5% on twig and 4.8% on stem bark.

  • Studies on the stem bark extracts of Garcinia cuneifolia have furnished a new xanthone cuneifolin (1) and the triterpene stigmasterol (2). Structures for these compounds were elucidated based on NMR, 2D NMR, MS and GCMS data. Larvicidal activity screening of the crude bark extract using the larvae of Aedes aegypti indicated the larvae to be susceptible to these extracts. $LC_{50}$ values of the bioassays show the extracts to be moderately toxic to the larvae of Aedes aegypti.

  • This study was conducted to investigate the effects of CEPA(2-chloroethyl phosphonic acid) on bark physiology and anatomy of lactree(Rhus verniciflua Stokes). Sample trees of similar size and growth rate were selected from 7-year-old lactree plantation located in Hyengsung-kun, Kangwon-do. Lanolin pastes containing 0.1, 1, or 10% CEPA were put into the bark-removed hole made by corer(${\phi}1cm$) on the main stem at 1.2m above the ground on June 16, 1995. Five weeks after application of CEPA, bark thickness was markedly increased as a result of the increase in the amount of phloem and intercellular spaces, and correlated with the increased production of urushiol. By the application of 10% CEPA, bark thickness was increased approximately 2.5 times, and the urushiol content within bark was increased 2.8 times compared to that of untreated trees because CEPA stimulated the accumulation of urushiol within bark. Treatment of 10% CEPA also increased the size and the total number of secretory canals, and induced an increase in ray width. The phloem parenchyma cells of CEPA-treated trees were well-developed and closely packed with little intercellular space.

  • The study of the scars formed on the bark of pine trees damaged by forest fire was carried out in the burned area of Samchuk and Donghae in 2000. Fire scars were formed on the bark of trees when fire passed by the tree. Fire scarring is usually found on the windward and leeward sides of the tree. Fire spread was analyzed topographically using the fire scars formed at the tree stem. Fire spread was closely related to the shape, slope and direction of the forestland. Four fire direction types are classified according to the shapes of the forestland. The height of the fire scar was higher on the leeward side than on the windward side of the trees studied. The burnt area of the total bark of the tree was over 30% in dead trees and below 12% in living trees. The regression model of the burnt stem area using the height of the fire scar, the diameter of breast height and the height of tree was Y=-2.484${\times}$Height+0.04199${\times}$D.B.H-1.686${\times}$Windward+11.172${\times}$Leeward+23.432(r=0.936, F=409.968, P>0.0001).

Quality Properties of Gangjung Added with Detoxified Stem Bark of Rhus verniciflua (RVSB) Extract during Acceleration Storage (무독화 옻 추출물 첨가 강정의 가온저장 중 품질 특성)

  • The quality characteristics of Gangjung added with detoxified Rhus verniciflua stem bark (RVSB) extract, as well as the changes that occurred in the product during accelerated storage ($60^{\circ}C$, 24 hr), were investigated. The expansion rate of Gangjung was not significantly different (p < 0.05). The L-value of the Gangjung with added detoxified RVSB extract was higher than the control, while the a-value decreased as the addition levels of the detoxified RVSB extract increased. The b-value was highest for the Gangjung prepared with 7% (w/w) detoxified RVSB extract. The moisture content of the Gangjung decreased as the storage period increased, but the addition of detoxified RVSB extract (p < 0.05) did not significantly change this effect. We found no significant difference in the hardness of the Gangjung within the concentration range of 0-7% (w/w) detoxified RVSB extract, and the peak number slightly decreased as the storage period increased. After 16 hr of storage, the acid value was significantly lower for the Gangjung containing detoxified RVSB extract at concentration between 5-7%. Furthermore, after 12 hr of accelerated storage, peroxide and TBA values significantly decreased with additions of detoxified RVSB extract that were over 3% (w/w). These results suggest that the physical texture of Gangjung did not improve by the addition of detoxified RVSB extract; however, its rate of lipid oxidation was reduced.

Antifungal Activities of Extracts from the Various Parts of the Genus Pinus Trees (소나무속(屬) 수목의 부위별 추출물의 항균활성)

  • This study was carried out to investigate the antifungal activities of the extracts from various parts of three pinus species, P. densiflora, P. rigida and P. koraiensis to pathogenic fungus Collectotrichum gloeosporioides. The EtOAc fraction from the bark of P. koraiensis stem and root showed 98.8 and 100% of activity, respectively to the fungus. Median effective doses $(ED_{50})$ of above two fractions were 469 and 588 ${\mu}g/ml$, respectively in the bioassay with the fungus. $ED_{50}$ of the EtOAc fraction from the bark of P. koraiensis stem against Alternaria brassicicola and Fusarium oxysporum was 533 and 2,277 ${\mu}g/ml$, respectively. This means that the fraction was more sensitive to the C. gloeosporioides and A. brassicicola than the fungus F. oxysporum. The EtOAc fraction from the leaves of P. densiflord showed 39.6% of activity to C. gloeosporioides, but all the fractions from the leaves of two species showed no activity. The active compounds in the bark of P. koraiensis stem and root are being identified.

Preparation and Characterization of Urushiol Free Fermented Rhus verniciflua Stem Bark (FRVSB) Extracts (Urushiol이 제거된 발효옻 추출물의 제조와 특성)

  • The water extract of the fermented $Rhus$ $verniciflua$ stem bark (FRVSB) was prepared by hot water extracting at $100^{\circ}C$ for 8 h. The urushiol content of the FRVSB water extract was determined by HPLC. The urushiol was not contained in FRVSB water extract, whereas $Rhus$ $verniciflua$ stem bark (RVSB) water extract contained 3.4 mg%. At the lab scale size, suitable water extraction condition for a total solid, polyphenol and flavonoid from FRVSB was at over $100^{\circ}C$ for 6-8 h. The total solid contents was reduced in pilot scale processing system, with 5.7% of the extraction yield. The proximate composition (%) of FRVSB water extract obtained from industrial installation was moisture 4.34, crude fat 1.69, crude protein 10.21, and crude ash 15.80. Gallic acid (1,090.5 mg%) was the most abundant compound in phenolic acids, while fisetin (135.7 mg%) was the predominant flavonoid. The free sugar content was mannitol of 3.48%, glycerol of 0.19%, and glucose of 0.19%. Alanine (244 ppm), serine (231 ppm), and leucine (218 ppm) were predominant amino acids.
