  • Solubility and degree of swelling of the starch were about 15% and 30%, respectively when the starch in agueous solution was heated at 90$^{\circ}C$ for 30minutes. The starch showed no singnificant differences in the degree of swelling and soblubility with a growing period of ginseng. The starch was begun gelatinize at 50-55$^{\circ}C$ and completed at 65-70$^{\circ}C$ by the amy tical methods of X-ray diffraction, disappearance of crystalinity. Brabender angly lographic analysis and amylase digestion test. The maximum and minimum peak viscosities of 10% starch solution were 5.500 B.U and 1960 B.U, respectively. The starch showed no significant differences in pasting temperature, maximum and minimum viscosities on amylogram with a growing period of ginseng. The intrinsic viscosities of the starch and amylose were 0.54 and 1.5-1.9, respectively.

Biochemical Properties of Starch Granule Non-Digestive Enzyme(SGNA) of Bacillus polymyxa No.26

  • A $\alpha$-l, 4-D-glucan maltohydrolase $(\beta$-amylase), secreted by the mesophilic aerobic bacterium Bacillus polymyxa No.26, was purified and characterized. The enzyme production was increased after a logarithmic phase of bacterial growth and paralleled with the onset of bacterial sporulation. By applying anion exchange chromatography and gel filtration the enzyme was purified 16.7-fold and had a specific activity of 285.7 units/mg. Two enzyme activities were eluted on a column of DEAE-Sephadex chromatography, and they were designated as E-I for a major enzyme peak and E-II for a minor peak. Of them, E-I enzyme peak was further purified by using gel chromatography. The molecular mass of this enzyme was determined to be 64, 000 daltons and consisted of a single subunit, showing an isoelectric point of 8.9. The enzyme was able to attack specifically the $\alpha$-l, 4-glycosidic linkages in soluble starch and caused its complete hydrolysis to maltose and $\beta$-limited dextrin. This amylolytic enzyme displayed a temperature optimum at $45^\circ{C}$ and a pH optimum at 7.0. The amino acid composition of the purified enzyme was quite similar to the other bacterial $\beta$-amylases reported. Surprisingly, the purified enzyme from this aerobe only exhibited hydrolytic activity on soluble starch, not on starch granules. The degradation of from starch by $\beta$-amylase was greatly stimulated by pullulanase addition. These results differentiated from other $\beta$-amylases reported. Based on a previous result that showed the enzyme system involves in effective degradation of raw starch granules, this result strongly suggested that the purified enzyme (E-I) can be a synergistic part of starch granule-digestion and E-II plays a crucial role in digestion of starch granules.

Influence of carbon type and carbon to nitrogen ratio on the biochemical methane potential, pH, and ammonia nitrogen in anaerobic digestion

  • Organic waste used as a feedstock in the anaerobic digestion (AD), it includes carbon and nitrogen. Carbon and nitrogen have an effect on the various digestive characteristics during AD, however, the study is rare about those of the interaction. This study investigates the influence of carbon type and carbon to nitrogens (C/N ratios) on the AD characteristics of organic waste. Experimental treatments involved a combination of three carbon types with three C/N ratios. The AD tests were carried out using a 125-mL serum bottle at a constant temperature of 37℃ and moisture 95% for 18 days. Degradation pattern shows the difference among three-carbon treatments, the starch group was faster than other groups. Maximum methane production date was similar between starch (9.96 ± 0.05 day) and xylan group (10.0 ± 0.52 day), those of the cellulose group (14.6 ± 1.80 day) was slower than other groups (p < 0.05). The lag phase was only affected by the carbon type (p < 0.05). Ammonia nitrogen was mainly affected by nitrogen concentration regardless of carbon type (p < 0.05). This study showed that xylan is useful as feedstock in order to decrease the lag phase, and it showed that ammonia was independently affected by the nitrogen concentration.

쌀 입자크기가 흰쌀죽의 이화학적 특성에 미치는 영향 (Effect of Grain Size on the Physicochemical Properties of Rice Porridge)

  • The objective of this study was to investigate the effect of grain size on the physicochemical properties of rice porridge. Here, the grain size of the rice was classified as whole grain, half grain, and flour by traditional Korean cooking methods. The viscosity of the rice flour porridge was highest for the among the three different grain size porridges. In the amylographs, the increase in viscosity for the whole grain porridge was higher than that of the rice flour porridge during cooling. The soluble solid and reducing sugar contents of the rice porridges increased according to the rice grain size, while the blue value decreased. The SDI (starch digestion index) increased according to the rice grain size. The RDS (rapidly digestible starch) was highest while the SDS (slowly digestible starch) the lowest in the rice flour porridge. The morphologies of the rice porridges were examined by SEM and showed a smoother surface and more exudated gelatinized granules in the rice flour than in the whole grain rice porridge. In conclusion, rice porridges made from the smallest possible grain size such as flour may be helpful for people with weaker digestive systems such as infants, the elderly, and hospital patients.

In vitro Digestibility of Cooked Noodle Products

  • The in vitro digestive properties of 6 domestic noodle products (spaghetti, somyeon, ramyeon, dangmyeon, naengmyeon, and jjolmyeon) were compared after cooking under the manufacture's recommended cooking conditions. The kinetic constant (k), representing the rate of hydrolysis at the initial digestion stage, was highest in the somyeon noodles (0.1151), followed by naengmyeon (0.0954), and was lowest in the spaghetti (0.0421). However, the concentration of starch ($C_{\infty}$) hydrolyzed over 2 hr was not different between the spaghetti (96.22) and the somyeon (96.40), indicating that different digestion behaviors occurred in each type of noodle, even though the amounts of digested starch were similar. The ramyeon, dangmyeon, and naengmyeon noodles showed relatively lower $C_{\infty}$ values than the spaghetti and the somyeon noodles. The spaghetti had the highest amount of slowly digestible starch (SDS, 43%) and the lowest glycemic index (GI, 87.8), whereas the somyeon had the lowest SDS value (9.6%) and the highest or (93.0). The digestibility differences among the noodles were attributed to differences in their flour compositions and manufacturing processes.

Soy Oligosaccharides and Soluble Non-starch Polysaccharides: A Review of Digestion, Nutritive and Anti-nutritive Effects in Pigs and Poultry

  • Soybean contains a high concentration of carbohydrates that consist mainly of non-starch polysaccharides (NSP) and oligosaccharides. The NSP can be divided into insoluble NSP (mainly cellulose) and soluble NSP (composed mainly of pectic polymers, which are partially soluble in water). Monogastric animals do not have the enzymes to hydrolyze these carbohydrates, and thus their digestion occurs by means of bacterial fermentation. The fermentation of soybean carbohydrates produces short chain fatty acids that can be used as an energy source by animals. The utilization efficiency of the carbohydrates is related to the chemical structure, the level of inclusion in the diet, species and age of the animal. In poultry, soluble NSP can increase digesta viscosity, reduce the digestibility of nutrients and depress growth performance. In growing pigs, these effects, in particular the effect on gut viscosity, are often not so obvious. However, in weaning piglets, it is reported that soy oligosaccharides and soluble NSP can cause detrimental effects on intestinal health. In monogastrics, consideration must be given to the anti-nutritive effect of the NSP on nutrient digestion and absorption on one hand, as well as the potential benefits or detriments of intestinal fermentation products to the host. This mirrors the needs for i) increasing efficiency of utilization of fibrous materials in monogastrics, and ii) the maintenance and improvement of animal health in antibiotic-free production systems, on the other hand. For example, ethanol/water extraction removes the low molecular weight carbohydrate fractions, such as the oligosaccharides and part of the soluble pectins, leaving behind the insoluble fraction of the NSP, which is devoid of anti-nutritive activities. The resultant product is a high quality soy protein concentrate. This paper presents the composition and chemical structures of carbohydrates present in soybeans and discusses their nutritive and anti-nutritive effects on digestion and absorption of nutrients in pigs and poultry.

효소변형 전분기반 하이드로젤 모델 프리믹스 내 탑재된 커큐민의 소화과정 중 안정성 (Stability of the enzyme-modified starch-based hydrogel model premix with curcumin during in vitro digestion)

  • 본 연구에서는 커큐민 함유 에멀션의 안정성을 위해 필드하이드로젤로 쌀전분(RS), 1시간 효소처리한 전분(1GS), 24시간 효소처리한 전분(24GS), 96시간 효소처리한 전분(96GS)을 사용하였으며 이를 이용한 모델 프리믹스로 커큐민 탑재 필드하이드로젤 분말(FHP)을 제조하여 탑재된 커큐민의 안정성 및 in vitro 소화 후 커큐민 보유율의 변화를 관찰하였다. FHP는 커큐민 함유 에멀션분말(EMP)과 비교하여 재분산시 크리밍 현상없이 분산안전성을 보여주었다. 재분산 하이드로젤은 고온에서 녹으면서 오히려 액적(droplet)의 유착(coalescence)과 응집(flocculation)이 가속화되어 에멀션에 비해 열안정성이 떨어졌으나 UV 안정성은 RS-FHP와 1GS-FHP에서 유의적인 커큐민 보호 효과를 나타냈다. GS-FHP는 in vitro 소화 중 기름방울의 응집 및 유착이 발견되지 않았으며 소화 후 에멀션의 유상에 용해된 커큐민을 성공적으로 보호함을 확인하였다. 따라서, GS-FHP는 분말형태 프리믹스 개발에 새로운 소재로 사용 가능할 것이며 커큐민 외에 다른 다양한 소수성 기능성물질에 대해서도 적용을 확장할 수 있을것으로 생각된다.

Treatment of Starch Wastewater by Anaerobic Digestion Combined with Hollow Fiber UF

  • Anaerobic digester coupled with hollow fiber membrane unit. Treatment of starch waste with anaerobic digester-membrane system was studied. $0.17\;m^2$ area of hollow fiber membrane unit of known pore size was immersed into laboratory-scale anaerobic digestion system. The gas production was about $0.74\;m^3/kg$ COD treated. The COD removal efficient was about 80-95% depending on the hydraulic retention time. Crossflow ultrafiltration as Post treatment to anaerobic filter. The study conducted with different membrane pore size indicated that membrane with 1,000,000 molecular weight cut-off size gave a higher COD removal efficiency in the range of 83-87% while giving a study flux of $120-130\;L/m^2\;{\cdot}\;h$. A study was conducted to see the long term clogging effect of membrane also.

재료에 따른 인삼닭죽의 in vitro 단백질 및 전분 분해율과 물리적 특성 (Effect of Ingredients on In vitro Digestibility and Physical Properties of Ginseng-Chicken Meat Porridge)

  • To determine the nutritional quality and physical properties of ginseng-chicken meat porridge, 10 kinds of ginsengchicken meat porridge samples containing waxy and/or non-waxy rice were analyzed for in vitro protein digestibility and their degree of starch hydrolysis. Viscosity and spreadness were determined for the gelatinized pastes of the porridge samples. Microphotographs of the starch granules and pastes were studied to confirm structural changes in the rice starch during cooking. The starch paste from non-waxy rice porridge had higher viscosity than the starch paste from the waxy rice porridge; however, in the case of the ginseng-chicken meat porridge, the difference in viscosity was negligible. Microphotograph comparisions between the waxy rice porridge and non-waxy rice porridge indicated apparent differences in the shapes of their starch granules and gels. The granule surface of the non-waxy rice was very rough while that of the waxy rice was very smooth; this difference would lead to organoleptical discrepancy. The added ginseng increased the protein digestibility of the chicken meat; however, the protein digestibility of the ginseng-chicken meat porridge was lower than that of the chicken meat or rice porridge due to inhibited protein digestion by the gelatinized starch. Finally, the rice porridge had increased starch hydrolysis with additions of chicken meat and vegetables.

Effects of Synchronization of Energy and Nitrogen Release in the Rumen on the Starch Disappearance in the Gastrointestinal Tract and Growth Performance of Hanwoo Steers

  • The objectives of experiments were to compare directly the effects of synchrony of slowly fermented (corn and corn gluten meal; C-CGM) and of rapidly fermented ingredients (barley and soybean meal; B-SBM) in the rumen on starch disappearances in gastrointestinal tracts (Experiment 1) and growth performance (Experiment 2) of Hanwoo steers in the feedlot barn. In experiment 1, four Hanwoo steers ($288{\pm}21$ kg) fitted with ruminal and "T" shaped duodenal cannula were placed in one pen with Calan gate and assigned randomly to a duplicate $2{\times}2$ Latin square design. In experiment 2, eight intact Hanwoo steers ($311{\pm}8$ kg) were assigned randomly to one of two pens with Calan gate to evaluate the effect of the same diets as like in experiment 1 on growth performance. There were no differences in ruminal pH, ammonia and total VFA concentrations between treatments. Percentage of apparent ruminal starch disappearance was 33.3% unit lower (p<0.05) for steers fed C-CGM than for steers fed B-SBM diets and this difference resulted in 268% higher (p<0.05) in duodenal starch flows for steers fed C-CGM diet than for steers fed B-SMB diet. There was significant increase (p<0.05) in quantity (927 vs. 400 g/d) of corn starch digested post-ruminally compared to barley starch. However, percentage of starch apparently digested post-ruminally was 8% higher (p=0.1) in steers fed fast synchrony diet with B-SBM than in steers fed slow synchrony diets with C-CGM. The differences of percentage and amount of starch apparently digested post-ruminally between C-CGM and B-SBM diets did not affect rice straw DM intake, average daily gain (ADG) and feed efficiency. In conclusion, there is some uncertainty in regards to the relationship between site of starch digestion and DM intake, ADG, and feed efficiency in this study.
