The National Environmental Specimen Bank (NESB) has been sampling and cryogenically preserving various wildlife specimens to monitor bio-accumulations of chemical pollutants since 2010. Recently, the NESB set up a plan to develop reference materials at their facility to assure the analytical quality of and validate the analytical methods for their monitoring samples. One of the crucial characteristics of reference materials is intra- and inter-bottle homogeneity. In this study, we used ANOVA for total mercury concentrations in some samples to validate their homogeneities after milling and homogenization. We examined the intra- and inter-bottle homogeneities of two cryogenically-milled samples (Korean mussel (Mytilus coruscus) and black-tailed gull's egg (Larus crassirostris). The variations in the total mercury concentrations were not significantly different intra- and inter-bottle (mussel: F=0.74, p=0.67; gull egg: F=1.96, p=0.10). Additionally, the relative standard deviations of the total mercury concentrations showed low values (mussel: 2.02%, gull egg: 1.78%). Therefore, the cryogenic-milling process statistically proves the homogeneity of the materials of mussels and sea gull eggs for chemical analysis for total mercury.
This study was carried out to evaluate the uncertainty in the analysis of menthol content from the mentholated cigarette. Menthol in the sample cigarette was extracted with methanol containing an anethole as an internal standard, and then analyzed by gas chromatography. As the sources of uncertainty associated with the analysis of menthol, were the following points tested, such as the weighing of sample, the preparation of extracting solution, the pipetting of extracting solution into the sample, the preparation of standard solution, the precision of GC injections for standard & sample solution, the GC response factor of standard solution, the reproducibility of menthol analysis, and the determination of water content in tobacco, etc. For calculating the uncertainties, type A of uncertainty was evaluated by the statistical analysis of a series of observation, and type B by the information based on supplier's catalogue and/or certificated of calibration. Sources of uncertainty were subsequently included and mathematically combined with the uncertainty arising from the assessment of accuracy to provide the overall uncertainty. It was shown that the main source of uncertainty came from the errors in the reproducibility of menthol and water determination, the purity of menthol reference material in the preparation of standard solution, and the precision of GC injections for sample solution. The errors in sample weighing and volume measurement contributed relatively little to the overall uncertainty. The expanded uncertainty in the mentholated cigarettes, Korean and American brand, at 0.95 level of statistical confidence was $\pm$0.06 and $\pm$0.07 mg/g for a menthol content of 1.89 and 2.32 mg/g, respectively.
This paper seeks for the set-up plan in the reasonable target area of environmental assessment. Domestically, the target area of environmental assessment is set up by the adjustment of opinion collection about the assessment scope result of an environmental factor. In a foreign country, the boundary of the target area for a development project is established with the environmental, economical, and social viewpoint based on the standard for significantly environmental impact. Based on the analysis of the present condition at home and abroad, the first set-up plan for the reasonable target area of environmental assessment is the preparation of the detailed term definition related to the target area of environmental assessment. The second is the arrangement of the judgement standard for significantly environmental impact. The quantitative assesment item can apply the present standard or regulation, and the qualitative assessment item can establish the standard based on factual and objective data. The last is the preparation of the reference material or the guideline for deciding the initial target area by the judgement standard and determining the final target area by opinion collection in the stage of scoping.

As a ZnO varistor is subjected to electrical and environmental stresses, it degrades gradually, which may result in power interruption by short circuit. This study investigates changes in the electrical characteristics of ZnO varistors due to deterioration owing to energy absorption, and determines the optimal parameters for on-line diagnosis of the varistor. Two types of varistors were used for an accelerated aging experiment involving the application of the $8/20{\mu}s$ standard lightning impulse current. The electrical characteristics in terms of the reference voltage, total leakage current, resistive leakage current, and third-harmonic component of the total leakage current were measured, and their change rates were analyzed. The results revealed that the total leakage current increased slightly with an increase in the varistor absorbed energy, while the resistive leakage current and the third-harmonic component increased apparently. Therefore, the third-harmonic component of the total leakage current was proposed as the optimal parameter for on-line monitoring of ZnO varistor conditions.
Because hyperspectral imaging provides detailed spectral information across a broad range of wavelengths, it can be utilized in numerous applications, including environmental monitoring, food quality inspection, medical diagnosis, material identification, art authentication, and crime scene analysis. However, hyperspectral images often contain various types of distortions due to the environmental conditions during image acquisition, which necessitates the proper removal of these distortions through a data preprocessing process. In this study, a preprocessing method was investigated to effectively correct the distortion caused by artificial light sources used in indoor hyperspectral imaging. For this purpose, a halogen-tungsten artificial light source was installed indoors, and hyperspectral images were acquired. The acquired images were then corrected for distortion using a preprocessing that does not require complex auxiliary equipment. After the corrections were made, the results were analyzed. According to the analysis, a statistical transformation technique using mean and standard deviation with reference to a reference signal was found to be the most effective in correcting distortions caused by artificial light sources.
Experimental study on the durability characteristics to examine the feasibility of concrete with high water-tightness and self-healing performance to minimize maintenance of concrete for artificial ground is as follows. 1) When blending agent, swelling agents, and curing accelerator were added on the ternary system cement with blast-furnace slag fine particles and fly ash to give a self-healing property, higher blending strengths by 82% at design standard strength of 24MPa and by 74% at design strength of 30MPa, respectively could be obtained. 2) The permeability test for the specimens having high water-tightness and no shrinkage showed that the permeability was reduced at maximum of 98%. However, the permeability was decreased as the design strength was increased, showing the reduction rate of 87% at the design strength of 50MPa. 3) The depth of carbonation of blast-furnace slag and fly ash was increased in all the specimens compared with those of OPC only. However, as the material age was increased, carbonation penetration depth was decreased compared with the reference blend. 4) Compared with the reference blending using only OPC, the freeze-thaw resistance was higher in the case of blending with 40% of blast-furnace slag and 10% of fly ash at the design standard strength of 50MPa. In addition, the freeze-thaw resistance in general was superior in the design standard strength of 50MPa with the lower water-binder ratio (W/B) as compared with the design standard strength of 24MPa and 30MPa with the high water-binder ratios.
This study was investigated with sample digestion techniques and a reliability of the analytical results for a quantitative analysis of a standard reference material ('urban particulate matter', SRM 1648). The metallic elements were analyzed by inductively coupled plasma mass spectrometry (ICP-MS) with closed vessel microwave acid digestion method and atomic absorption spectroscopy (AAS). Quality control of the overall analytical procedures for metallic element determinations of standard reference material were estimated by analysis of a SRM 1648. Three digestion solutions $(type\;1:\;HNO_3,\;type\;2:\;HNO_3\;and\;H2O_2(4'1)\;mixture,\;type\;3:5.55\%\; HNO_3\;and\;16.75\%\;HCI\;mixture)$ were applied to SRM 1648. As a result, three digestion solutions used in this study are completely unable to digest Cr of SRM 1648. Reliability of Cr and As showed some errors in the digestion with digestion solution type (3) due to the influence of chlorides. Type (3) digestion solutions are sufficient to fulfill the digestion of As as well as Se in SRM 1648. ICP-MS results showed the improvement in accurate and precise determination of some trace elements like Cd, V and Pb in SRM 1648. It is important to use the proper digestion solution for each element to qualify analytical precision.
Aspartate aminotransferase (AST; E.C. 2.6.1.1), a vitamin B6-dependent enzyme, preferentially promotes the mutual transformation of aspartate and ${\alpha}$-ketoglutarate to oxaloacetate and glutamate. It plays a key role in amino acid metabolism and has been widely recommended as a biomarker of liver and heart damage. Our study aimed to evaluate the extensive preparation of AST and its application in quality control in clinical laboratories. We describe a scheme to express and purify the 6His-AST fusion protein. An optimized sequence coding AST was synthesized and transformed into Escherichia coli BL21 (DE3) strain for protein expression. Ideally, the fusion protein has a volumetric productivity achieving 900 mg/l cultures. After affinity chromatography, the enzyme activity of purified AST reached 150,000 U/L. Commutability assessment between the engineered AST and standard AST from Roche suggested that the engineered AST was the better candidate for the reference material. Moreover, the AST showed high stability during long-term storage at $-20^{\circ}C$. In conclusion, the highly soluble 6His-tagged AST can become a convenient tool for supplying a much better and cheaper standard or reference material for the clinical laboratory.
For reliable determination of mechanical characteristics of adhesively bonded joints used e.g. as input data for computer-aided design of complex components, the thick-adherend tensile-shear test according to ISO 11003-2 is the most important material testing method. Although the total displacement of the joint is measured across the polymer layer directly in the overlap zone in order to minimize the influence of the stepped adherends, the substrate deformation must be taken into account within the framework of the evaluation of the shear modulus and the maximum shear strain, at least when high-strength adhesives are applied. In the standard ISO 11003-2 version of 1993, it was prescribed to perform the substrate deformation correction by means of testing a one-piece reference specimen. The authors, however, pointed to the excessive demands on the measuring accuracy of the extensometers connected with this technique in industrial practice and alternatively proposed a numerical deformation analysis of a dummy specimen. This idea of a mathematical correction was included in the revised ISO 11003-2 version of 2001 but in the simplified form of an analytical method based on Hooke's law of elasticity for small strains. In the present work, it is shown that both calculation techniques yield considerably discordant results. As experimental assessment would require high-precision distance determination (e.g. laser extensometer), finite element analyses of the deformation behavior of the bonded joint are performed in order to estimate the accuracy of the obtained substrate deformation corrections. These simulations reveal that the numerical correction technique based on the finite element deformation modeling of the reference specimen leads to considerably more realistic results.

The authentication of the physico-chemical properties of ginsenosides reference materials as well as qualitative and quantitative batch analytical data based on validated analytical procedures is a prerequisite for certifying good manufacturing practice (GMP). Ginsenoside Rb1 and Rg1, representing protopanaxadiol and protopanaxatriol ginsenosides, respectively, are accepted as marker substances in quality control standards worldwide. However, the current analytical methods for these two compounds recommended by Korean, Chinese, European, and Japanese pharmacopoeia do not apply to red ginseng preparations, particularly the extract, because of the relatively low content of the two agents in red ginseng compared to white ginseng. In manufacturing fresh ginseng into red ginseng products, ginseng roots are exposed to a high temperature for many hours, and the naturally occurring ginsenoside Rb1 and Rg1 are converted to artifact ginsenosides such as Rg3, Rg5, Rh1, and Rh2 during the heating process. The analysis of ginsenosides in commercially available ginseng products in Korea led us to propose the inclusion of the (20S)- and (20R)-ginsenoside Rg3, including ginsenoside Rb1 and Rg1, as additional reference materials for ginseng preparations. (20S)- and (20R)-ginsenoside Rg3 were isolated by Diaion HP-20 adsorption chromatography, silica gel flash chromatography, recrystallization, and preparative HPLC. HPLC fractions corresponding to those two ginsenosides were recrystallized in appropriate solvents for the analysis of physico-chemical properties. Documentation of those isolated ginsenosides was achieved according to the method proposed by Gaedcke and Steinhoff. The ginsenosides were subjected to analyses of their general characteristics, identification, purity, content quantification, and mass balance tests. The isolated ginsenosides showed 100% purity when determined by the three HPLC systems. Also, the water content was found to be 0.534% for (20S)-Rg3 and 0.920% for (20R)-Rg3, meaning that the net mass balances for (20S)-Rg3 and (20R)-Rg3 were 99.466% and 99.080%, respectively. From these results, we could assess and propose a full spectrum of physico-chemical properties of (20S)- and (20R)-ginsenoside Rg3 as standard reference materials for GMP-based quality control.
