A Study on the Comparison of Chemical Characterization and Ellagic Acid Content Between Distribution Bokbunja and Korean Native Bokbunja (국내유통 복분자와 토종복분자의 이화학적 특성과 엘라그산 함량 비교연구)

  • The purpose of this study was to compare the chemical properties of the bokbunja distributed in Korea with the content of the bioactive substance ellagic acid. The bokbunja was Rubus coreanus group and Rubus occidentalis group were compared, domestic bokbunja and import bokbunja were compared. In bokbunja, free sugar was 30.89 ± 0.7 mg/g of Rubus coreanus and 29.05 ± 0.87 mg/g of Rubus occidentalis. and 27.28 ± 7.4 mg/g of domestic bokbunja and 21.58 ± 6.73 mg/g of import bokbunja. The free amino acids was 4.50 ± 0.08 mg/g of Rubus coreanus and 5.05 ± 0.08 mg/g of Rubus occidentalis. and 4.13 ± 1.09 mg/g of domestic bokbunja and 3.75 ± 0.31 mg/g of import bokbunja. Validation of the ellagic acid method was confirmed by comparing the retention time and spectrum of the standard and extract using HPLC. The calibration curve (R2) showed linearity of 0.9999. As a result of analyzing the ellagic acid content of each extraction solvent, DMSO and methanol mixture extracts were high, and Rubus coreanus was 2.56 mg/g and Rubus occidentalis was 3.16 mg/g, which was not significantly different (p < 0.05) In addition, the ellagic acid content of domestic bokbunja and import bokbunja was 2.83 mg/g and 2.99 mg/g, which was not significantly different (p < 0.05).

Study of BOD5 Variation Patterns with Flow Regime Alteration in the Tributaries (지류하천의 유황분석을 통한 BOD5 농도변화 유형 분석)

  • We analyzed the variations of water quality with flow regime alterations to determine the characteristics of the stream where the stream management is considerably difficult due to the high variability of the flow rates. In this study, both flow rates and water qualities were monitored at the tributaries, 34 in count, of both Geum River and Sabgyo Lake Basins. The variation of water qualities were divided into 2 types, based on their stream flow rates, known as Type I and Type II. If the water quality of a stream increases during low flow rate periods compared with high flow rate periods, it is classified as Type I; if the water quality of the stream increases during high flow rate periods compared with low flow rate periods, it falls under Type II. The analysis for the variations of water qualities, of all 43 basins, resulted to 24 basins under Type I and Nineteen 19 basins under Type II. The variations of water qualities were analyzed first by using Regression Analysis followed by Statistical Analysis. The average slope of the variations of water qualities and the slope of the standard deviations were 0.00135 and 0.00477, respectively. The Probability Distributions of both Type I and Type II basins were 61.1% and 38.9%, respectively. The basin having a probability distribution of 61.1% and is also known as Type I, increases during periods of low flow rates, due to the presence of point sources. Therefore, the basin should be enforced with stream management. Before the stream management can be implemented in all streams falling under Type II, the sources of contaminants should first be estimated. These contaminants can be classified into two parts, the first is Point source pollution and the second is Non-point source pollution, where the Non-Point source pollution can be sub-divided into two types, with storm runoff and without storm runoff.

Accuracy of one-step automated orthodontic diagnosis model using a convolutional neural network and lateral cephalogram images with different qualities obtained from nationwide multi-hospitals

  • Objective: The purpose of this study was to investigate the accuracy of one-step automated orthodontic diagnosis of skeletodental discrepancies using a convolutional neural network (CNN) and lateral cephalogram images with different qualities from nationwide multi-hospitals. Methods: Among 2,174 lateral cephalograms, 1,993 cephalograms from two hospitals were used for training and internal test sets and 181 cephalograms from eight other hospitals were used for an external test set. They were divided into three classification groups according to anteroposterior skeletal discrepancies (Class I, II, and III), vertical skeletal discrepancies (normodivergent, hypodivergent, and hyperdivergent patterns), and vertical dental discrepancies (normal overbite, deep bite, and open bite) as a gold standard. Pre-trained DenseNet-169 was used as a CNN classifier model. Diagnostic performance was evaluated by receiver operating characteristic (ROC) analysis, t-stochastic neighbor embedding (t-SNE), and gradient-weighted class activation mapping (Grad-CAM). Results: In the ROC analysis, the mean area under the curve and the mean accuracy of all classifications were high with both internal and external test sets (all, > 0.89 and > 0.80). In the t-SNE analysis, our model succeeded in creating good separation between three classification groups. Grad-CAM figures showed differences in the location and size of the focus areas between three classification groups in each diagnosis. Conclusions: Since the accuracy of our model was validated with both internal and external test sets, it shows the possible usefulness of a one-step automated orthodontic diagnosis tool using a CNN model. However, it still needs technical improvement in terms of classifying vertical dental discrepancies.

Fabrication and application of cell-based microfluidic chip for eye-irritation test of chemicals (화학 물질의 안자극 시험용 세포 기반 미세유체 칩의 제작 및 응용)

  • This study presents the development of cell-based microfluidic chips for the performance of acute eye irritation tests due to chemicals and examined some of their applications. Microfluidic chips were fabricated by photolithography and soft lithography, and they had three compartments with different areas for cell culture. Rabbit corneal epithelial cells were used for the eye irritation test. The death of cells cultured inside the chip was monitored at regular time intervals after treatment with an aqueous solution of chemicals, and the cell death rate constants were calculated based on the viability curve. The performance of the microfluidic chip was verified by examining the effects of cell-cell junctions, cell-substrate adhesion, and initial cell numbers compared to cell death rates. Eye irritation tests were performed at various concentrations of an aqueous solution of sodium dodecyl sulfate (SDS), a standard substance for the eye irritant test. The cells were exposed to the SDS aqueous solution for 300 s, and the resulting eye irritation was assessed by cell viability. Finally, the equation for calculating the toxicity score (TS) was derived based on the weighting factor for each compartment in the chip. The cell-based microfluidic chip developed in this study may be used for eye irritation tests from chemicals used in cosmetics and pharmaceuticals.

Development of LC-MS/MS analytical methods for metalaxyl in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai (LC-MS/MS를 이용한 생약 백출 및 우슬 중 Metalaxyl 잔류분석법 개발)

  • A new rapid and simple method for metalaxyl in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai has been developed and validated. This study was conducted to develop a method for analyzing metalaxyl by a method based on QuEChERS using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Samples were extracted with acetonitrile and purified using amino-propyl (NH2) Solid Phase Extraction cartridge. The method limit of quantitation (MLOQ) was 0.01 mg/kg. The linearity of matrix-matched calibration curve (r2) was ≥0.99 at the calibration range of 0.001-0.05 mg/kg. For recovery test, Atractylodes macrocephala Koidzumi or Achyranthes japonica Nakai was treated with standard solutions at MLOQ and 10MLOQ levels. Recovery rates were in the range of 88.1-109.1% with <5.5% coefficient of variation. This established analytical method was fully validated. Based on these results, it can contribute to improving the safety of residual pesticides in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai.

Hypoalbuminemia and Albumin Replacement during Extracorporeal Membrane Oxygenation in Patients with Cardiogenic Shock

  • Background: Extracorporeal membrane oxygenation (ECMO) has been widely used in patients with cardiorespiratory failure. The serum albumin level is an important prognostic marker in critically ill patients. We evaluated the efficacy of using pre-ECMO serum albumin levels to predict 30-day mortality in patients with cardiogenic shock (CS) who underwent venoarterial (VA) ECMO. Methods: We reviewed the medical records of 114 adult patients who underwent VA-ECMO between March 2021 and September 2022. The patients were divided into survivors and non-survivors. Clinical data before and during ECMO were compared. Results: Patients' mean age was 67.8±13.6 years, and 36 (31.6%) were female. The proportion of survival to discharge was 48.6% (n=56). Cox regression analysis showed that the pre-ECMO albumin level independently predicted 30-day mortality (hazard ratio, 0.25; 95% confidence interval [CI], 0.11-0.59; p=0.002). The area under the receiver operating characteristic curve of albumin levels (pre-ECMO) was 0.73 (standard error [SE], 0.05; 95% CI, 0.63-0.81; p<0.001; cut-off value=3.4 g/dL). Kaplan-Meier survival analysis showed that the cumulative 30-day mortality was significantly higher in patients with a pre-ECMO albumin level ≤3.4 g/dL than in those with a level >3.4 g/dL (68.9% vs. 23.8%, p<0.001). As the adjusted amount of albumin infused increased, the possibility of 30-day mortality also increased (coefficient=0.140; SE, 0.037; p<0.001). Conclusion: Hypoalbuminemia during ECMO was associated with higher mortality, even with higher amounts of albumin replacement, in patients with CS who underwent VA-ECMO. Further studies are needed to predict the timing of albumin replacement during ECMO.

A study on the separation and determination of the rare earth Elements by the AG® 50W-X8 cation exchange resin (AG® 50W-X8 양이온교환수지를 이용한 희토류원소의 분리와 분석에 관한 연구)

  • Methods to separate 14 rare earth elements (REEs) and yttrium by the $AG^{(R)}$ 50W-X8 cation exchange resin, and to determine REEs by inductively coupled plasma atomic emission spectrophotometry (ICP-AES) were described. Ion exchange capacities of REEs on the resin were so high that the REEs were quantitatively ion exchanged under the flow rate of 0.3~1.0 mL/min at pH 1~6. The breakthrough capacity curve of the REEs showed that ion exchange capacities of light REEs (Cerium group) were greater than that of the heavy REEs (Yttrium group). When $200{\mu}g$ of each REEs was ion exchanged on 100 mg of resin, most of the heavy REEs were quantitatively desorbed with 10 mL of 2.0 M of $HNO_3$, while most of the light REEs with 30 mL. The method was applied to the monazite sample. The REEs could be separated from matrix, since ion exchange capacities of matrix ions of Ca, Ti, Mg, Mn were much lower than that of the REEs. However the relative standard deviations of the analytical results by the present method were not improved, as high as 1~5%.

The Comparative Analysis of Reservoir Capacity of Chungju Dam based on Multi Dimensional Spatial Information (다차원 공간정보 기반의 충주댐 저수용량 비교분석)

  • Dam is very important facility in water supply and flood control. Therefore study needs to analyze reservoir capacity accurately to manage Dam efficiently. This study compared time series reservoir capacity using multi-dimensional spatial information to Chungju Dam reservoir and major conclusions are as follows. First, LiDAR and multi beam echo sounder survey were carried out in land zone and water zone of Dam reservoir area. And calibration process was performed to enhance the accuracy of survey data and it could be constructed that multi dimensional spatial information which was clearly satisfied with the standard of tolerance error by validation with ground control points. Reservoir capacity by water level was calculated using triangle irregular network from detailed topographic data that was constructed by linked with airborne LiDAR and multi beam echo sounder data, and curve equation of reservoir capacity was developed through regression analysis in 2008. In the comparison of the reservoir capacity of 2008 with those of 1986 and 1996, the higher water level goes, total reservoir capacity of 2008 showed decrease because of the increase of sediment in reservoir. Also, erosion and sediment area could be analyzed through calculating the reservoir capacity by the range of water level. Especially the range of water level as 130.0~135.0 which is the upper part of average water level, showed the highest erosion characteristics during 1986~2008 and 1996~2008 and it is considered that the erosion of reservoir slant by heavy rainfall is major reason.

Comparison of Milling and Flour Quality Characteristics of Foreign Wheat and Korean Wheat

  • This study was investigated to compare the milling and physicochemical characteristics of six Korean wheat cultivars (Keumkang, KK; Jokyung, JK; Goso, GS; Joongmo2008, JM; Baekkang, BK; Saekeumkang, SKK) and five foreign wheat classes (Australian standard white wheat, ASW; Australian hard, AH; US northern spring, NS; US hard red winter, HRW; Soft wheat, SW). Korea and foreign wheat grains were milled using a Buhler MLU-202. Flour moisture, ash, protein, gluten, sedimentation, particle size, solvent retention capacity (SRC) and dough properties of flour were analyzed. Results showed that the hard wheats had a greater total flour yield and reduction fraction yield than the soft wheats regardless of the country. However, there were in the milling characteristics between the US and Korean soft wheats. GS, a soft wheat in Korea, had the lowest flour yield (59.6%) and the highest bran fraction yield (21.4%). The particle sizes of flour by milling fraction were B1>B2>B3 for the largest, and the R1〈R2〈R3 for the smallest. Particle size, ash, protein contents and the values of lactic acid SRC showed highly correlated with flour yield. The gluten-performance-index (GPI) is the ratio of the lactic acid SRC value to the sum of sodium carbonate and sucrose SRC values, and it has been used as a quality indicator for overall performance potential of flour. GPI values differed depending on the wheat variety or class, JM (0.82) was the highest value, and SKK (0.56) and SW (0.59) were low. The curve pattern of the Mixolab result also gives a quality indication of the flour sample. JM and NS flour had similar pattern at water absorption and gluten strength parameters and BK and HRW had similar viscosity patterns. These results will enable further study for blending Korean wheat cultivar to improve the flour quality.

Genetic Characteristics and Anthocyanin Content of Basil(Ocimum basilicum L.)

  • Basil(Ocimum basilicum L.) is an annual herb that grows wild in hot and humid regions such as tropical Asia and Africa. This study was carried out for resource discovery, propagation, and DB construction of aromatic plants. In order to evaluate the genetic characteristics, 28 kinds of basil seeds were supplied from the Center for Genetic Resources. For basil characteristics, planting date, plant height, growth shape, leaf length, leaf shape, petiole color, petiole, stem color, flower color, after harvest, Ml length including roots, flower length, stem thickness and biological weight were measured. For anthocyanin analysis, only basil with purple color was selected, anthocyanins were extracted with 60% ethanol containing 1% citric acid, a standard quantitative curve was prepared with cyanidin-3-glucoside, and absorbance was measured at 525 nm. Basil planting started around June 16, and the flowering period lasted for a total of 18 days from July 19 to August 5. On August 6, when flowering was completed, morphological characteristics including flower color were measured at the site by an object. The measured basil plant length was 26.6-59.6 cm, leaf length 2.0-7.9 cm, leaf width 0.7-5.2 cm, and petiole 1.3-3.9 cm. The growth pattern of basil was generally straight, medium in shape. The color of the leaves was observed evenly by mixed green, purple, purple and green, and the recesses in the shape of the leaf were also conspicuously observed. A total of three basil flower colors were observed: white, light pink(149P 10P/84), and purple(37V 2.5RP 4/12). During the second growth survey after harvest, the total length including the basil roots is 26.7-52.0 cm, the flower length is 8.2-29.3cm, the stem thickness is 7.1-15.9mm, the number of stem nodes is 3-12 nodes, and the total weight of the basil was measured to be about 218-1540 g. There are a total of 16 types of basil expressed in purple, and the anthocyanin content was measured to be 73.74 ~ 380.72 mg/100g.
