Effects of Angelica keiskei Koidzumi and Turmeric Extract Supplementation on Serum Lipid Parameters in Hypercholesterolemic Diet or P-407-Induced Hyperlipidemic Rats (명일엽과 울금 추출물의 투여가 고콜레스테롤식이와 P-407로 유도한 고지혈증쥐의 혈중 지질 함량에 미치는 영향)

  • The effects of administration alone or mixed of Angelica keiskei and turmeric extract on blood lipids were evaluated in hypercholesterolemic diet or P-407-induced hyperlipidemic rats. In the study 1, female Sprague-Dawley rats weighing 160-180 g were divided into each groups and fed high cholesterol diets for 8 weeks. Experimental groups were administered with following diets: basal diet (Normal), high cholesterol diets (1% cholesterol). We did the oral administration for evaluation in experimental groups: C (vehicle), A (angelica extract), T (turmeric extract), AT (angelica extract, turmeric extract/ 1:1 complex). The concentrations of serum total cholesterol, and LDL-cholesterol were decreased by 6.8%, 9.8% in A group, by 22.1%, 28.8% in C group and by 28.2%, 35.6% in AT group, compared to the C group, respectively. HDL-cholesterol levels were not different among the experimental groups. In the study 2, we induced the hypertriglyceridemia in rats by intraperitoneal injection of P-407 (0.5 g/kg) once per three days. From the next day after P-407 injection beginning, we did the oral administration as the study 1. Angelica keiskei extract, turmeric extract and complex extract decreased serum triglyceride by 17.2%, 19.7% and 48.3%, respectively. These results suggested that Angelica keiskei and turmeric extract complex might have synergistic effect in lowering total cholesterol, LDL-cholesterol and triglyceride in hyperlipidemic rats.

Resorbability and histological reaction of bioabsorbable membranes (수종의 흡수성 차단막의 생체 분해도와 조직학적 반응)

  • The major goals of periodontal therapy are the functional regeneration of periodontal supporting structures already destructed by periodontal disease. There have been many efforts to develop materials and therapeutic methods to promote periodontal wound healing. With the development of non-resorbable membrane, GTR has proved to be the representive technique of periodontal regeneration. However, due to various clinical problems of non-resorbable membrane, resorbable membrane was developed and it showed to be clinically effective. The newly developed Para-Dioxanone membrane has a characteristic of non-woven fabric structures which is different from the generally used membranes with structure of mesh form. In addition, Chitosan membrane has been developed to apply its adventage maximally in GTR. Although a number of different types of membranes had been clinically used, researches on absorption rate of membranes were inadequate and limited to subjective opinions. However, since long term period of resorption and space maintenance are required in implant or ridge augmentation, accurate verification of resorption rate is clinically important. In this study, we had implanted Resolut(R), Biomesh(R), Para-Dioxanone membrane and Chitosan membrane (Size : 4mm ${\times}$ 4mm) on dorsal side of Sprague Dawley rat, and sacrified them after 4 weeks, 8 weeks, 12 weeks respectively. Histologic observation was carried out, and the following results were obtained by calculating the objective resorption rate. 1. In case of Resolut(R), external resorption took place initially, followed by internal resorption. Surface area are 5.76${\pm}$2.37$mm^2$, 4.90${\pm}$l.06$mm^2$, 4.90${\pm}$0.98$mm^2$ at 4 weeks, 8 weeks, 12 weeks respectively, and invasion rate of connective tissue to membrane are 31.6${\pm}$4.5%, 52.8${\pm}$9.4%, 56.4${\pm}$5.1% respectively. 2. Biomesh(R) showed a pattern of folding, relatively slow resorption rate with small size of membrane. Surface area are 3.62${\pm}$0.82$mm^2$, 3.63${\pm}$0.76$mm^2$, 4.07${\pm}$1.14$mm^2$ at 4 weeks, 8 weeks, 12 weeks respectively, and invasion rate of connective tissue to membrane are 26.1${\pm}$5.8%, 30.9${\pm}$3.4%, 29.2${\pm}$3.6%, respectively. 3. Para-Dioxanone membrane was surrounded by fibrous conncetive tissue externally, and resorption took place internally and externally. Surface area are 5.96${\pm}$1.05$mm^2$, 4.77${\pm}$10.76$mm^2$, 3.86${\pm}$0.84$mm^2$ at 4 weeks, 8 weeks, 12 weeks respectively, and invasion rate of connective tissue to membrane are 30.7${\pm}$5.1%, 53.3${\pm}$4.4%, 69.5${\pm}$3.1%, respectively. 4. Each fiber of Chitosan membrane was surrounded by connective tissue and showed external resorption pattern. It showed little invasion of inflammatory cells and excellent biocompatability. The resorption rate was relatively slow. Surface area are 6.01${\pm}$2.01$mm^2$, 5.49${\pm}$1.3$mm^2$, 5.06${\pm}$1.38$mm^2$ at 4 weeks, 8 weeks, 12 weeks respectively, and invasion rate of connective tissue to membrane are 31.3${\pm}$3.6%, 38.4${\pm}$3.80%, 39.7${\pm}$5.6%, respectively. Consequently, Para-Dioxanone membrane and Chitosan membrane are found to be clinically effective for their excellent tissue reaction and biocompatibility. Futhermore, the advantage of bone regenerating ability as well as the relatively long resorption period of Chitosan membrane, it might be widely used in implant or ridge augmentation.

Potent Antimutagenic Activity of Caryophyllene Oxide for Aflatoxin $B_1\;(AFB_1)$ and 2-Amino-3-Methyl-imidazo[4,5-f]Quinoline (IQ) (Aflatoxin $B_1\;(AFB_1)$과 2-amino-3-methylimidazo[4,5-f]quinoline (IQ)에 대한 caryophyllene oxide의 항돌연변이성)

  • Substantial amount of caryophyllene oxide (CPO) is present in the essential oils of traditionally-used folk medicinal plants and herbal spices. The CPO, produced via chemical and/or enzymatic reaction of caryophyllene (CP), has largely being used as a flavoring component and exhibited a variety of biological activities. Now, we report the antimutagenic activity of CPO determined by Ames's preincubation test. S-9 fraction was prepared from the liver of rats treated with Arochor 1254. Anatoxin $B_1\;(AFB_1)$ and 2-amino-3-methylimidazo[4,5-f]quinoline (IQ) were used as mutagens. Reduction of mutagenicity of $AFB_1$ or IQ for S. typhimurium TA98 and TA100 by CPO was found to be a dose-dependant manner. CPO (500 ${\mu}g/plate$) reduced mutagenicity of AEB1 for S. typhimurium TA98 and TA100 to 89% and 71%, respectively. For IQ, similar results were observed against S. typhimurium TA98 and TA100, resulting in the inhibition percentage of 77% and 51%, respectively. CP also reduced mutagenicity of AEB1 and IQ for S. typhimurium TA98 and TA100, but the reduction rate was somewhat lowered relative to that of CPO. These results indicate that CPO could be developed as a potent antimutagenic flavoring agent.

Effects of Kale Juice Powder on Serum Lipids, Folate and Plasma Homocysteine Levels in Growing Rats (케일녹즙 분말식이가 흰쥐의 혈중 지질, 엽산 및 호모시스테인 수준에 미치는 효과)

  • The purpose of this study was to investigate the effects of green vegetable (kale) juice powder supple-mentation on lipid profiles, plasma homocysteine, folate and vitamin $B_{12}$ in rats fed cholesterol (Chol. ) or Chol. free diet. 7-week old male Sprague Dawley rats (n=40) were divided into 4 groups, and experimental diets containing control diet group (CO), control diet plus 0.5 wt$ \% $ Chol. (CC), control diet added with 5 wt$ \% $ kale (KO), control diet added with 5 wt$ \% $ kale plus 0.5 wt$ \% $ Chol. (KC) were fed for 8 wks. Plasma homocysteine level was examined by amino acid analyzer and serum folate and vitamin $B_{12}$ level were measured by com-petitive radioimmunoassay methods. In various serum lipid profiles, TG level was lower in kale juice powder groups (KO, KC) compared to the corresponding groups (CO, CC) (p<0.001). In Chol. supplemented groups (CC, KC), HDL-Chol. level was lower (p<0.001) and LDL-Chol. level was higher (p<0.05) than Chol. free diet. HDL-Chol. level was higer (p<0.05) in kale juice powder groups. HDL/LDL ratio was lower in Chol. supplemented groups (CC, KC) and tended to be higher in kale juice powder groups (KO, KC) Serum folate and vitamin $B_{12}$ levels were not affected by dietary Chol. and kale juice powder supplementation. Plasma homocysteine level was not affected by dietary Chol. and kale juice powder supplementation, too. Serum folate level was positively correlated with serum vitamin $B_{12}$ level (r=0.5632, p<0.001), but plasma homocysteine level was not significantly correlated with any serum folate, vitamin $B_{12}$ and Chol. levels, respectively. In summary, kale juice powder supplementation have improved serum lipid profiles by increasing the HDL level and decreasing the TG level and have not altered homocysteine level under the sufficient supply of folate and vitamin B complex relating with the homocysteine metabolism.

Effects of Dietary Oligosaccharide on the Blood Glucose and Serum Lipid Composition in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐의 혈당 및 혈중 지질조성에 미치는 Oligosaccharide의 영향)

  • This study was conducted to examine the effects of dietary oligosaccharide on the blood glucose and serum lipid composition in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 150$\pm$10g were randomly assigned to one normal and four STZ-induced diabetic groups. Diabetic groups were classified to basal diet(DM group) 10% xylooligosaccharide diet(DM-XO group) 10% isomaltooligosaccharide(DM-IMO group) and 10% fructooligosaccharide (DM-FO). Diabetes was experimentally induced by intravenous injection of 50 mg/kg of body weight of STZ in citrate buffer (pH 4.3) after feeding of experimental diets for 4 weeks. The rats were fed with experimental diet for further 4 weeks in diabetic state. The oligosaccharide diets were not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the weights of liver kidney and small intestine but the weight of cecum was significantly increased on the groups of xylooligosaccharide and isomaltooligosaccharide diet. The levels of oral glucose tolerance test was more effectively improved by DM-XO group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. Activities of two intestinal enzymes such as lactase and sucrase in DM-XO and DM-FO groups were lower than that of DM group while activity of maltase was lower only in DM-XO in DM-FO groups than that of DM-group respectively. The levels of serum triglyceride in DM-XO group were lower than that of DM-group respectively. The levels of serum triglyceride in DM-XO groups were lower than that of DM group however was no significant differences among the oligosaccharide groups. These results suggest that dietary oligosaccharide may act as functional food to be capable of improving carbohydrate and lipid metabolism in diabetic rats.

Hepatic Oxygen Free Radical and Alcolhol Metabolizing Enzyme Activities in Rats Fed Diets Supplemented with Lycium chinense Ethanol Extract (흰쥐에 있어서 구기자 추출물 첨가식이가 간조직의 유해산소 및 알콜대사 효소활성에 미치는 영향)

  • To investigate the oxygen free radical and alcohol metabolizing system in liver of rats fed diets with 30% ethanol extract of Lycium chinense (LCEE), Sprague-Dawley male rats weighing 225~235g have been fed a diet supplemented with 2% or 4% LCEE for a month. The rats fed LCEE supplemented diets gained less body weight compared with the control, and had no remarkable changes of liver function. In rats fed 2% LCEE supplemented diet, hepatic cytochrome P450 contents appeared to be increased, but catalase (204.88$\pm$20.06 $H_2O$$_2$nmoles/mg protein/min), and superoxide dismutase (13.18$\pm$0.74 Unit/mg protein) activities were significantly increased compared with control 120.28$\pm$26.99 $H_2O$$_2$nmoles/mg protein/min and 10.49$\pm$0.80 Units/mg protein). There was no difference in hepatic glutathione content, glutathione peroxidase, and glutathione-S-transferase ctivities between the rats fed LCEE suplemented diets and the control diet. On the other hand, hepatic alcohol dehydrogenase activity were not changed by LCEE feeding, but hepatic aldehyde dehydrogenase activities were significantly increased in rats fed both 2 and 4% LCEE diets(5.01$\pm$0.21 and 4.47$\pm$0.06 $\mu$moles NADPH/mg protein/min) compared with control (3.28$\pm$0.21 $\mu$moles NADPH/mg protein/min) and its Vmax value was 1.9 fold increased in rats fed 2% LCEE and 1.5 fold in those fed 4% LCEE compared with control. In conclusion, it is likely that rats receiving a diet supplemented with LCEE may have the oxygen free radicals and alcohol detoxication potential.

Effect of Paecilomyes japonica on Lipid Metabolism In Rats Fed High Fat Diet (눈꽃동충하초가 고지방 식이를 섭취한 흰쥐의 지질대사에 미치는 영향)

  • The effects of Paecilomyes japonica on weight gains, food intakes, food efficiency ratios, serum and hepatic lipid concentrations, serum protein levels and serum enzyme activities, were studied in adult male rats. Sprague-Dawley rats,35 weeks old, were given four different types of diets for a succeeding period of five weeks: either a normal diet (5% corn oil), a control diet (high fat; 5% corn oil + 15% lard), a PF diet (control diet + 3% fruiting body of Paecilomyes japonica), or a PM diet (control diet+.3% mycelium of Paecilomyes japonica). The body weight gains, hepatic weights and food efficiency ratios of rats fed the PF or PM diets were significantly lower than those fed the control diet, but were similar to those fed the normal diet. The concentrations of hepatic total lipids, cholesterol and triglyceric, and serum triglyceride, of rats given the PF or PM diets were significantly lower than those given the control diet. Hut the concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol and phospholipid in the serum of rats fed the control, PF or PM diets were significantly higher than those fed the normal diet. In the serum of rats fed the PF diet, the HDL-cholesterol/total cholesterol ratio was significantly higher and the atherogenic index was significantly lower than those fed the control diet, while such effect was not observed in rats fed the PM diet. The alkaline phosphatase activity in the serum of rats fed the control and PM diets was more significantly decreased compared to rats fed the PF and normal diet. No differences were noted in the weights of the pancreas, kidney and heart, the serum concentrations of glucose, hemoglobin and albumin, and the activities of GOT, GPT and ${\gamma}$-GTP, among the rats on all the experimental diets. In conclusion, the rats fed the PF or PM diets maintained normal body and hepatic weights. Despite of the high intake of fats in the PF and PM diets, the concentrations of hepatic total lipids, cholesterol and triglyceride, and serum triglyceride were decreased.

Effects of Vegetable Sprout Power Mixture on Lipid Metabolism in Rats Fed High Fat Diet (새싹채소 혼합분말이 고지방 식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of vegetable sprout powder on serum and adipose tissue lipid metabolism in rats fed high-fat diet for 4 weeks for induction hyperlipidemic model rat. Weight-matched male Sprague-Dawley rats were assigned to five groups according to dietary fat level (10% or 20% of diet wt.) and mixture of vegetable sprout powder levels (5% or 10% 10% or 20% of diet wt.). Vegetable sprout powder was the mixture of same amounts of dried barley, broccoli, rapeseed, alfalfa, radish, mustard, buckwheat and brussels sprouts. Experimental groups were normal fat diet with 5% cellulose (NF-C), high fat diet without fiber (HF-N), high fat diet with 5% cellulose (HF-C), HF-C diet with 5% vegetable sprout powder (HF-CSL), and HF-C diet with 10% vegetable sprout powder (HF-CSH). The body weight of HF-N group increased 16% compared with the NF-C group, while it was decreased by 15% and 22% for HF-CSL group and HF-CSH group, respectively. Fat mass and fat cell size of adipose tissue were lower in HF-CSL group and HF-CSH group compared with HF-C group, and lower in HF-CSH group compared with HF-CSL group. Serum triglyceride, total cholesterol and LDL-cholesterol contents were markedly decreased by vegetable sprout powder containing diet, while the serum HDL-cholesterol and phospholipid contents were higher in vegetable sprout powder containing diet in a dose-dependent manner. Leptin and insulin levels in serum showed a decrease in HF-CSH group. Significantly increased contents of triglyceride, total cholesterol, LDL-cholesterol, leptin and insulin in the serum of HF-N group were returned to normal or even below normal levels by feeding 10% vegetable sprout powder diet. The increased activities of NADP-malate dehydrogenase (ME), glucose-6-phosphate dehydrogenase (G6PDH) and 6-phosphogluconate dehydrogenase (6PGDH) and lipoprotein lipase (LPL) in adiposetissue by HF-N group were decreased to the activity of normal fat group by feeding vegetable sprout powder in a dose-dependant manner. These results indicate that lipid metabolism in rats fed high-fat diet was suppressed by feeding vegetable sprout powder.

Effects of Persimmon (Diospyros kaki Thunb) Syrup Extracted from Persimmon and Persimmon By-products on Blood Glucose Level (감(Diospyros kaki Thunb) 또는 감부산물에서 추출한 감시럽의 혈당수치에 미치는 영향)

  • The purpose of this study was to measure the blood glucose level and glycemic index (GI) in response to persimmon (Diospyros kaki Thunb) syrup extracted from persimmon and extract of persimmon by-products. Major component analyses of persimmon syrup I (PSI, 95:5 mixture of purified persimmon syrup and non-purified persimmon syrup) and persimmon syrup II (PSII, 50:50 mixture ratio of purified persimmon syrup and non-purified persimmon syrup) were $0.3{\pm}0.1$ and $0.6{\pm}0.2$ mg/g for total polyphenolic compounds and $70.6{\pm}0.6$ and $66.6{\pm}1.6%$ for total carbohydrates, respectively. Blood glucose responses of PSI and PSII were determined using both normal ICR mice and streptozotocin (STZ)-induced diabetic male Sprague-Dawley (SD) rats. Further, oral glucose tolerance test (OGTT) was performed on diabetic rats to assess the effects of the experimental diets. Blood glucose response and OGTT showed that blood glucose levels were significantly lower in mice and diabetic rats fed PSI and PSII compared to those fed diets of sugar, maple syrup, or honey. The GIs of healthy volunteers in response to PSI and PSII were calculated to be 51.9 and 35.7, respectively. On the contrary, the GIs of healthy volunteers fed diets including sugar, maple syrup, or honey were 52.6, 20.0, and 93.0, respectively. These results suggest that persimmon syrup can be used for both the treatment of diabetics and healthy people due to its beneficial effects on blood glucose level.

Effect of Supplementation of Korean Native Chicken Egg Fed with Citrus Peel on Profiles in Sprague-Dawley Rats (감귤 껍질을 급여한 토종닭 계란이 흰쥐의 혈액 성상에 미치는 영향)

  • In this experiment, three different diets were produced to investigate the effects of Jeju native chicken eggs fed with citrus peel on the profiles of rats. The first diet did not contain any eggs (TS), the second diet contained 10% eggs and no citrus peels (T0), and the third diet contained 10% chicken egg with citrus peels (T1). These diets were provided to 11-week-old male rats for four weeks. The weight gain, feed intake and feed efficiency were not significantly different between the three treatment groups, TS, T0, and T1. The weight of the liver was significantly higher in T0 and T1 than TS (p<0.05), but the weights of the kidney and epididymal fat pad were not significantly different between the TS, T0, and T1 groups. The total lipid, phospholipid, triglycerides, total cholesterol and LDL cholesterol did not significantly difference among the TS, T0, and T1 groups. The HDL cholesterol and HDL cholesterol/ total cholesterol of T1 were the highest among the groups, and the atherogenic index of T1 was the lowest among the groups (p<0.05). The total protein of TS was significantly lower in T0 and T1, and the albumin of T1 was the highest among the groups (p<0.05). The albumin/globulin ratio of TS was significantly higher in T0 and T1, but the creatinine of T0 was significantly higher in T0 and T1 (p<0.05). The blood sugar and blood pigment were not significantly different between the TS, T0, and T1 groups. The $\gamma$-GTP of T1 was the lowest among the groups (p<0.05), but the ALT, AST and ALP did not show significant difference among the TS, T0, and T1 groups.
