팔라티노스 및 팔라티노스 시럽의 Rat 에서 급성 독성 (Acute Toxicity of Palatinose and Palatinose Syrup in Rats)

  • Rat에서 팔라티노스 및 팔라티노스 시럽의 급성독성을 평가하였다. 팔라티노스 및 팔라티노스 시럽 각각을 27 g/kg 용량으로 투여하였을때 사망하는 동물은 관찰되지 않았다. 팔라티노스 시럽 웅성 투여군에서 체중증가가 약간 감소하였으나, 독성학적 중요성은 없는 것으로 보인다. 혈액학적 및 혈액생화학적 검사에서도 어떤 유의한 변화가 관찰되지 않았다. 이 결과로 보아, 팔라티노스 및 팔라티노스 시럽은 어떤 현저한 독성증상을 보이지 않았으며, Sprague-Dawley rat에 경구투여시 $LD_{50}$값은 27g/kg 이상인것으로 결론지었다.

Sprague Dawley Rat의 미세병변에서 Moire Artifact를 제거하기 위한 Grid suppression software 사용 후 영상분석 (Image Analysis Using Grid Suppression Software to Remove Moire Artifact from Micro Lesions of Sprague Dawley Rat)

  • Moire artifact는 미세병변과 주파수 대역이 중첩되기 때문에 moire artifact를 제거하는 Image processing software를 사용할 경우 미세 병변의 손실을 가져올 수 있다. 본 연구에서는 SD(Sprague Dawley) Rat에 microcalcification과 microfracture와 같은 미세병변을 임의로 형성하여 영상화하고, reference 영상과 grid suppression software를 사용한 영상, optimizied grid 영상을 비교 분석하였다. 영상은 두 명의 영상의학과 전문의가 컨소시엄을 형성하여 판독하였고, 판독 결과 값은 McNemar's test 이용하여 평가하였다. 73개의 microcalcifications 중 Grid suppression후 13 cases에서, optimized grid를 사용한 영상은 3 cases에서 영상의 손실이 확인되어 Grid suppression후의 영상이 통계적으로 유의하게 영상 손실을 발생하고 있음을 보여주고 있다(p=0.021). 총 53개의 fracture line은 Grid suppression을 시행한 후 영상에서 19 cases가 영상의 손실을 보였고, optimized grid를 사용한 영상에서는 영상손실이 없는 것으로 판독되었다. 따라서 미세병변을 진단하는 영상에 있어 moire artifact를 제거하기 위한 grid suppression software 사용은 신중하게 고려되어야 할 것이다.

호박분말이 Sprague-Dawley 흰쥐에서 인위적으로 유발한 위암 및 유선암에 미치는 영향 (Effects of Pumpkin Powder on Chemically Induced Stomach and Mammary Cancers in Sprague-Dawley Rats)

  • This study was conducted to investigate the effectiveness of pumpkin powder in the diet of experimental animals on chemically induced stomach and mammary canceers. Three weeks old male SpragueDawley rats were randomly allocated to either 1) basal diet+MNNG, 2) basal diet+MNNG+PMC, 3) 2.5% pumpkin powder supplemented diet+MNNG+PMC, or 4) 5.0% pumpkin powder supplemented diet+MNNG+PMC for stomach cancer experiment. And female Sprague-Dawley(5 weeks old) rats were randomly assigned to either 1) basal diet only, 2) basal diet+DMBA, 3) 2.5% pumpkin powder supplemented diet+DMBA, or 4) 5.0% pumpkin powder supplementd diet+DMBA. In both experiments, supplements of pumpkin powder in basal diet decreased body weight of both male and female experimental animals. Pumpkin powder in rat diet decreased significantly(p<.05) chemically induced stomach cancer. With its suppressing effects on stomach cancer, pumpkin powder in diet of experimental rats had decreasing effects on the initiation and development of DMBA-induced mammary cancer. In conclusiion, current study may provide in vivo data to develop health foods using pumpkin. Further studies, however, are essential to clarify the exact role of pumpkin powder in chemically induced stomach and mammary cancers.

Embryonal rhabdomyosarcoma in the abdominal cavity of an aged Sprague-Dawley rat

  • We report a spontaneous embryonal rhabdomyosarcoma in the abdominal cavity of an aged (88-week-old) Sprague-Dawley rat. The animal had a firm lobulated $5{\times}5{\times}4.5cm$ mass in the abdominal cavity that was whitish to tan with necrotic and hemorrhagic plaques. Microscopically, the mass contained nodules with spindle or globoid shaped neoplastic cells with abundant eosinophilic cytoplasm and round or elongated nuclei mixed with other spindle cells with a filamentous appearance and scanty cytoplasm. Multinucleated cells and cross-striations were also observed. The neoplastic cells were positive for vimentin, desmin, and alpha-smooth muscle actin, especially the small spindle cells.

Spontaneous Peripheral Ameloblastic Odontoma in a Male Sprague-Dawley Rat

  • Peripheral ameloblastic odontoma is a rare variant of odontogenic tumor occurring in the extraosseous region. The present report describes a spontaneous tumor in male Sprague-Dawley (SD) rats. The clinically confirmed nodule in the right mandibular region was first observed when the rat was 42 weeks and remained until the terminal sacrifice date when the animal was 48 weeks of age. At necropsy, a well demarcated nodule, approximately $2.5{\times}2.0{\times}2.0cm$, protruded from the ventral area of the right mandible. The nodule was not attached to mandibular bone and was not continuous with the normal teeth. Histopathologically, the tumor was characterized by the simultaneous occurrence of an ameloblastomatous component and composite odontoma-like elements within the same tumor. The epithelial portion formed islands or cords resembling the follicle or plexiform pattern typical of ameloblastoma and was surrounded by mesenchymal tissue. Formation of eosinophilic and basophilic hard tissue matrix (dentin and enamel) resembling odontoma was observed in the center of the tumor. Mitotic figures were rare, and areas of cystic degeneration were present. Immunohistochemically, the epithelial component was positive for cytokeratin AE1/AE3 (CK AE1/AE3), and the mesenchymal component and odontoblast-like cells were positive for vimentin, in the same manner as in normal teeth. On the basis of these findings, the tumor was diagnosed as a peripheral ameloblastic odontoma in an extraosseous mandibular region in a SD rat. In the present study, we report the uncommon spontaneous peripheral ameloblastic odontoma in the SD rat. We also discuss here the morphological characteristics, origin, histochemical, and immunohistochemical features for the diagnosis of this tumor.

2-Bromopropane에 의한 유발된 Sprague-Dawley 랫트의 고환위축의 병리학적 관찰 및 Flow Cytometry를 이용한 검사 (Histopathological Observation and Flow Cytometry Analysis of Testicular Atrophy Induced by 2-Bromopropane On the Sprague-Dawley Rat)

  • This study was carried out to evaluate the testicular toxicity of 2-bromopropane (2-BP), which recently caused occupational intoxication on the reproductive and hematopoietic system in Koreans, using light microscopy, immunohistochemistry and flow cytometry. 10 weeks old male Sprague-Dawley (SD) rats were treated with 0.5 g/㎏/day of 2-BP orally for 8 consecutive weeks. The testes of the rats were vascularly perfused with Karnovsky's solution or immersed in Bouin's solution, embedded in plastic and evaluated with light microscopy. And relative proportions of haploid, diploid, and tetra-ploid states of DNA ploidy in the testicular cell suspensions of the SD rats were examined by flow cytometry. 2-BP induced severe testicular atrophy, depletion and degeneration of spermatogonia, spermatocytes, and spermatids and mild hyperplasia of Leydig cells without significant morphological changes. The Leydig cell hyperplasia was confirmed by immunohistochemistry using proliferating cell nuclear antigen (PCNA). The immunopositive cells against PCNA were observed in the nuclei oj some interstitial cells. Relative proportions of haploid states of DNA ploidy decreased in the atrophic testicular cell suspensions comparing with those of the control. In conclusion, 2-BP induced testicular atrophy with Leydig cell hyperplasia as examined by histopathology, immunohistochemistry and DNA flow cytometry.

Dietary Conjugated Linoleic Acid did not Affect on Body Fatness, Fat Cell Sizes and Leptin Levels in Male Sprague Dawley Rats

  • This study was designed to observe the effect of conjugated linoleic acid (CLA) supplementation on body fatness, fat cell sizes and leptin levels in male Sprague Dawley rats. Following weaning, forty rats were divided into 4 groups beef tallow (BT), fish oil (FO). beef tallow with CLA supplementation (BTC), and fish oil with CLA supplementation (FOC) group. For four weeks, all rats were fed experimental diets containing 12% of total dietary fat (w/w) with or without 1% CLA. After 4 weeks, the animals were sacrificed; the total carcass fat, plasma leptin levels, epididymal fat pad weights and fat cell sizes in adipose tissue were measured. CLA supplementation did not significantly affect the rat's body weights, total body fat, epididymal fat pad weights, and fat cell sizes. CLA also did not have a significant effect on plasma leptin levels. These results suggest that CLA supplement was not an effective way to reduce the body weights of male Sprague Dawley rats.

수영과 저출력 레이저 치료가 Freund's Complete Adjuvant 유발 흰쥐의 관절염 치유에 미치는 영향 (The Effect of Swimming and Low Power Laser on the Healing of the Freund's Complete Adjuvant Induced Arthritis in Rat)

  • Purpose: This study was to find that what mechanism take effects that was Adjuvant Induced Arthritis in Sprague-Dawley rat and then treated the swimming and low power laser. Methods: Adjuvant Induced Arthritis was induced 24 Sprague-Dawley rat by the subcutaneous injection of a 0.2ml Freund's Complete Adjuvant into the right hind paw and right knee joint. Second injection used of 0.05ml Freund's Complete Adjuvant by same method. Arthritic rat were divided 3 groups; arthritic swimming group, arthritic laser group and case control group. The author performed several experimental tests which were the hind paw thickness, step length, knee joint space, activity of enzyme. Results: Hind paw thickness decreased in swimming and laser group. Left step length and knee joint space increased in swimming and laser. Conclusion: Swimming and low power laser therapy on the Adjuvant Induced Arthritis in rats does effective for the rheumatic arthritis therapy by decrease of hind paw thickness, increase of opposite side step length, increase of activity of albumin and IgG and increase of knee joint space.
