Getah virus (GETV) infection causes sporadic outbreaks of mild febrile illness in horses and reproductive failure in pigs. In this study, we established a reverse transcription polymerase chain reaction (RT-PCR) method to detect GETV from suspected virus-infected samples. The reaction conditions were optimized and validated by using RNA extracted from GETV propagated in cell culture. A GETV-specific GED4 primer set was designed and used to amplify a 177 bp DNA fragment from a highly conserved region of the E1 glycoprotein gene in the GETV genome. RT-PCR performed with this primer set revealed high sensitivity and specificity. In the sensitivity test, the GED4 primer set detected GETV RNA at the level of $10^{2.0}\;TCID_{50}/mL$. In the specificity test, the GED4 primer set amplified only a single band of PCR product on the GETV RNA template, without non-specific amplification, and exhibited no cross-reactivity with other viral RNAs. These results suggest that this newly established RT-PCR method is useful for accurate identification of GETV infection in animals.
Glial fibrillary acidic proteins (GFAP) are a group of intermediate filaments that are distributed in the cytoplasm of glial cells. GFAP immunoreactivity (GFAP-IR) increase after central and peripheral nerve injuries. The purpose of this study was to determine change of GFAP-IR in rat trigeminal ganglion satellite cells following the axotomy of inferior alveolar nerve(IAN). The immunohistochemistry was carried out using the avidin-biotin-peroxidase complex(ABC) method. 1. Control group : Astrocytes in central root of trigeminal ganglion had strong GFAP-IR, but satellite cells of trigeminal ganglion occasionally had GFAP-IR. The patterns of reactivity in satellite cells of trigeminal ganglion were not concenturated in any specific region of trigeminal ganglion. 2. Three day group after IAN axotomy : There were highly GFAP-IR in satellite cells of trigeminal ganglion in mandibular region. GFAP-IR in maxillary and ophthalmic regions were less intense compared to mandibular region. 3. Seven day group after IAN axotomy : GFAP-IR that were increased compared to control group were seen in the mandibular region. But GFAP-IR were less intense compared to three day group. These results suggest that GFAP-IR increase in specific region of trigeminal ganglion following peripheral axotomy. therefore we suppose that GFAP study offer research tool in trigeminal neuralgia.
It is well known that a long-term exposure to a loud noise environment affects performance, since it distracts attention, and also is able to evoke stress accompanied by negative emotional states. The purpose of this study was to analyze dynamics of subjective and physiological variables during long-lasting (30 min) exposure to intensive white noise (85 dB[A]). Physiological signals on 23 college students were recorded by BIOPAC, Grass Neurodata systems and AcqKnowledge 3.5 software. Autonomic variables, namely skin conductance level (SCL), non-specific SCR number (N-SCR), inter-beat intervals in ECG (RR intervals), heart rate variability index (HF/LF ratio of HRV), respiration rate (RESP), and skin temperature (SKT) were analyzed on 5 min epoch basis. Psychological assessment (subjective rating of stress level) was also repeated on every 5 min basis. Regression and correlation analyses were employed to trace the time course of the dynamics of the subjective and autonomic physiological variables and their relationship. Results showed that intense noise evokes subjective stress with associated autonomic nervous system responses. However, it was shown that physiological variables endure specific changes in the process of exposure to loud white noise. Discussed are probable psychophysiological mechanisms mediating reactivity to long-term auditory stimulation of high intensity.
The high theoretical specific capacity of lithium-sulfur (Li-S) batteries makes them a more promising energy storage system than conventional lithium-ion batteries (LIBs). However, the slow kinetics of the electrochemical conversion reaction seriously hinders the utilization of Li-S as an active battery material and has prevented the successful application of Li-S cells. Therefore, exploration of alternatives that can overcome the sluggish electrochemical reaction is necessary to increase the performance of Li-S batteries. In this work, an ionic liquid (IL) is proposed as a functional additive to promote the electrochemical reactivity of the Li-S cell. The sluggish electrochemical reaction is mainly caused by precipitation of low-order polysulfide (l-PS) onto the positive electrode, so the IL is adopted as a solubilizer to remove the precipitated l-PS from the positive electrode to promote additional electron transfer reactions. The ILs effectively dissolve l-PS and greatly improve the electrochemical performance by allowing greater utilization of l-PS, which results in a higher initial specific capacity, together with a moderate retention rate. The results presented here confirmed that the use of an IL as an additive is quite effective at enhancing the overall performance of the Li-S cell and this understanding will enable the construction of highly efficient Li-S batteries.

Barley yellow dwarf virus (BYDV) has been a major viral pathogen causing significant losses of cereal crops including oats worldwide. It spreads naturally through aphids, and a rapid, specific, and reliable diagnostic method is imperative for disease monitoring and management. Here, we established a rapid and reliable method for isothermal reverse transcription recombinase polymerase amplification (RT-RPA) combined with a lateral flow strips (LFS) assay for the detection of BYDV-infected oat samples based on the conserved sequences of the BYDV coat protein gene. Specific primers and a probe for RT-RPA reacted and optimally incubated at 42℃ for 10 min, and the end-labeled amplification products were visualized on LFS within 10 min. The RT-RPA-LFS assay showed no cross-reactivity with other major cereal viruses, including barley mild mosaic virus, barley yellow mosaic virus, and rice black streaked dwarf virus, indicating high specificity of the assay. The sensitivity of the RT-RPA-LFS assay was similar to that of reverse transcription polymerase chain reaction, and it was successfully validated to detect BYDV in oat samples from six different regions and in individual aphids. These results confirm the outstanding potential of the RT-RPA-LFS assay for rapid detection of BYDV.

Psychological assessment (subjective rating of stress level) was also repeated every 5 min. Statistical analysis was employed to trace the time course of the dynamics of subjective and autonomic physiological variables and their relationships. Results showed that the intense noise evoked subjective stress as well as associated autonomic nervous system responses. However it was shown that physiological variables endured specific changes in the process of exposure to the loud white noise. Discussed were probable psychophysiological mechanisms mediating reactivity to long-term auditory stimulation of high intensity, namely short-term activation, followed by transient adaptation (with relatively stable autonomic balance) and then a subsequent wave of arousal due to tonic sympathetic dominance.
The emergence of rice black-streaked dwarf virus (RBSDV) poses a significant threat to global cereal crop cultivation, necessitating the urgent development of reliable detection and quantification techniques. This study introduces a reliable approach for the precise and sensitive quantification of the RBSDV in cereal crop samples, employing a reverse transcription digital polymerase chain reaction (RT-dPCR) assay. We assessed the specificity and sensitivity of the RT-dPCR assay proposed for precise RBSDV detection and quantification. Our findings demonstrate that RT-dPCR was specific for detection of RBSDV, with no cross-reactivity observed with other viruses infecting cereal crops. The RT-dPCR sensitivity was over 10 times that of RT-quantitative PCR (RT-qPCR). The detection limit of RT-dPCR was 0.096 copies/㎕. In addition, evaluation of RT-dPCR assay with field samples was conducted on 60 different cereal crop samples revealed that RT-dPCR (45/60) exhibited superior accuracy compared with RT-qPCR (23/60). In this study, we present a specific and accurate RT-dPCR assay for the detection and quantification of RBSDV.
오염 지하수 처리를 위한 획기적인 방법으로 높은 반응성을 가진 나노영가철이 최근 주목을 받고 있다. 그러나 미세한 분말형태로 인하여 현장공법에 적용이 제한되어 나노영가철을 입자형태의 지지체에 부착시켜서 투수성을 유지하여 실용성을 높인 방법이 제시되었다. 본 연구에서는 지지체의 존재 하에 생성된 나노영가철이 제조방법에 따라 어떤 특성의 변화를 보이는지 분석하고 이에 따른 반응성의 차이를 평가하였다. 지지체로는 이온교환수지를 선택하였으며 분산제를 포함한 에탄올/물의 이중용매를 사용하여 나노영가철을 제조한 후 그것의 물리적, 화학적 특성 및 반응성을 기존의 물만을 사용하여 제조한 경우와 비교하였다. 이중용매 상에서 제조된 반응물질은 비표면적이 $38.10m^2/g$, 철함량이 22.4 mg Fe/g 으로 수용액상에서 제조된 반응물질 보다 비표면적과 철 함량이 각각 20%와 23% 증가하였으며 질산성질소를 이용한 반응성 평가에서도 $0.462h^{-1}$의 반응상수값을 보이며 61% 증가한 반응성을 나타냈다. 단위표면의 반응상수 또한 증가하여 단위 면적에서의 반응성도 향상된 나노영가철을 얻을 수 있었다. 제조된 반응물질은 높은 반응성과 지지체의 사용으로 실제 오염 지하수 등의 처리에서 유용하게 사용될 수 있을 것으로 기대되어진다.
혈중 항원 특이적 IgE 검사와 피내시험으로 집먼지 진드기 (house dust mites, HDM)에 양성 반응을 보인 20두의 아토피성 피부염으로 진단된 개를 대상으로, 말초혈단핵구 (peripheral blood mononuclear cells, PBMC)를 채취하여 HDM항원에 대한 반응을 검토하였다. PBMC를 분리하여 HDM항원으로 자극한 결과 20두 중 13두 (65%)에서 항원 특이적이 림프구의 증식반응을 확인할 수 있었다. HDM 항원에 증식반응은 아토피성 피부염군에서 대조군에 비해 유의적으로 높은 반응이 확인되었다 (P=0.007). 또한, HDM에 대한 반응은 혈중의 IgE 농도와 유의적으로 상관관계를 나타내었으며 (P=0.035), 이는 체내에서 항원 특이적인 IgE의 생산을 촉진하는 작용을 반영하는 지표가 될 수 있다고 생각되었다. 이러한 결과로, 말초혈액중에 존재하는 HDM 항원 특이적인 림프구는 개의 아토피성 피부염의 병태생리에 관여하고 있는 것으로 시사되었다.
Serum immunoglobulins (Igs) from Israeli carp were purified using affinity chromatography. Fish were immunized with purified mouse IgG, and the specific fish antibodies were purified from the immune serum on a mouse IgG-immobilized agarose gel. Rabbit anti-Israeli carp Igs (R $\alpha$ I. carp Igs) antibodies were produced following hyperimmunization with mouse IgG specific carp antibodies. SDS-PAGE analysis under reducing condition showed that Israeli carp Igs were composed of two $\mu$-like heavy chains with about 82 and 50 kd, respectively, and one light chain with about 25 kd. On immunoblotting analysis, however, R $\alpha$ I. carp Igs failed to react with the light chain. When both protein A and protein G-purified normal carp Ig were compared with mouse IgG-specific Israeli carp Ig, no significant structural differences among them were observed. To investigate if there is any homology between other fish Ig molecules, cross-reactivity of R $\alpha$ I. carp Igs against Ig molecules from 6 different fish sera and mouse control serum was checked on immunoblotting analysis. As a result, R $\alpha$ I. carp Igs responded to Israeli carp, common carp, and tilapia Ig molecules. In flow cytometry study, however, R $\alpha$ I. carp Igs appeared to recognize 42.0%, 35.8% and <5% of Israeli carp, common carp and tilapia $Ig^+$ head kidney cells, respectively. The result suggests the heterogeneity between receptor Igs on B-like lymphocytes and soluble Igs in serum. It is crucial to obtain pure fish Igs to produce reagent antibodies as tools for the study on their specific immune responses.
